The molecular weights of plasma and intestinal kallikreins in rats.

1. The molecular weights of kallikreins of rat intestine and rat plasma have been estimated using gel filtration. 2. Extracts of pooled tissue from rat jejunum, ileum, caecum and colon activated by autolytic processes gave a single peak of kallikrein activity with a molecular weights of 33000. 3. Acid-activated rat plasma gave two peaks of kallikrein activity with molecular weight of 125000 and 61500. 4. Rat intestinal tissue contains a kinin forming enzyme having a molecular weight similar to those of glandular kallikreins and different from those of the rat plasma kallikreins.

Advances in functional photoisomerisable low-molecular-weight π-gelators.

Functional photoresponsive π-gelators represent a new generation of low-molecular-weight gelators (LMWGs) that self-assemble through noncovalent interactions, including hydrogen bonding, π-π stacking, and van der Waals forces. Photoresponsive gelators incorporate photochromic groups that reversibly restructure upon light irradiation, offering tunability of the gel-sol phase transitions. This responsiveness provides an external control over the gel behavior with non-invasive stimuli such as light (UV or visible light), making them interesting candidates for sensors, optomechanical actuators, en

Exploring high molecular weight components in Tityus serrulatus venom.

Our study identified high-molecular-weight compounds from Tityus serrulatus venom (TsV), and most of them have not yet been well explored. TsV was fractionated using FPLC system with different columns, analyzed by SDS-PAGE, and characterized by MALDI-TOF/TOF. Our study showed that TsV contains several high-molecular-weight compounds, including CRISPs, metalloproteinase and hyaluronidase. We show how these molecules can be obtained from TsV, enabling future studies about their molecular structures and biological actions, expanding knowledge about this venom.

Molecular Study on Twin Cohort with Discordant Birth Weight.

The increased rate of twinning has pointed out newer challenges in clinical practices related to gestational complications, intrauterine growth restriction, perinatal mortality, and comorbidities. As a twin pregnancy progresses, the increased demand for oxygen supply can easily disrupt the redox homeostasis balance and further impose a greater challenge for the developing fetuses. A substantial birth-weight difference acts as an indicator of a deficit in oxygenation or blood flow to one of the fetuses, which might be related to a low bioavailable nitric oxide level. Therefore, in this study, w

Isolation of High-Molecular-Weight DNA Using Organic Solvents.

Perhaps the most basic of all procedures in molecular cloning is the purification of nucleic acids. The key step, the removal of proteins, can often be performed simply by extracting aqueous solutions of nucleic acids with phenol:chloroform and chloroform.

Low-Molecular-Weight Heparins as Immunomodulators in Dermatology Practice.

Low-molecular-weight heparins (LMWHs) have some effects on cell proliferation and inflammation beyond mere anticoagulation. They have been tried on treatment of a few dermatological disorders such as lichen planus, skin wound healing, recurrent aphtous stomatitis, chronic urticaria, and contact hypersensitivity. LMWHs are generally well-tolerated drugs, rarely can lead to severe reactions. In this article, we will review the novel indications of LMWHs in dermatology practice and common skin reactions associated with their use.

Low-molecular-weight heparin in patients with advanced cirrhosis.

BACKGROUND & AIMS: The use of low-molecular-weight heparins (LMWH) in patients with advanced liver diseases is frequently avoided because of the enhanced risk of bleeding complications. However, many patients with impaired liver function are at a high risk of thrombosis or have an indication for therapeutic anticoagulation. Therefore, the aim of this study was to evaluate the pharmacokinetics of LMWH in patients with cirrhosis. METHODS: Eighty-four consecutive patients with cirrhosis and a clinical indication for prophylactic or therapeutic anticoagulation were included. The LMWH doses were c

Spontaneous hemothorax following anticoagulation with low-molecular-weight heparin.

OBJECTIVE: To report a case of spontaneous hemothorax following anticoagulation with low-molecular-weight heparin (LMWH) for the management of suspected pulmonary embolism. CASE SUMMARY: A 66-year-old man with a background history of breast carcinoma was admitted with pleuritic chest pain. He was initially managed as a suspected case of pulmonary embolism. Dalteparin, an LMWH, was started at a maximum dose of 18,000 units subcutaneously once daily, according to British national prescribing guidelines. On day 4, following 3 doses of dalteparin, the patient developed acute respiratory distress

Hospital guidelines for use of low-molecular-weight heparins.

OBJECTIVE: To describe the development of guidelines for initial use of low-molecular-weight heparins (LMWHs) and other anticoagulants in acute-care hospitals that are part of a national group purchasing organization (GPO). DATA SOURCES: A systematic literature search (1970-December 2001) was conducted to identify evidence on the efficacy of various anticoagulants for initial therapy in deep-vein thrombosis and pulmonary embolism, and in treatment of acute coronary syndrome. A group consensus method was then used to develop guidelines. Guidelines were reviewed and revised by an internal exper

Heparin and low molecular weight heparin: background and pharmacology.

The various LMWHs available for therapeutic use have multiple mechanisms of action, most of which are similar to the mechanisms of UH. The relative potencies of expression of the mechanisms differ between LMWH and UH and among specific LMWHs. The pharmacokinetics of LMWHs and UH are often measured according to the results of anti-Xa assays, although the correlation between anti-Xa levels and the antithrombotic activities of the drugs is questionable. Animal models of thrombosis give some information regarding the antithrombotic efficacy of different LMWHs when compared with UH and with other L

Chromatographic determination of high-molecular weight impurities in amoxicillin.

The separation of high molecular weight impurities in amoxicillin has been examined by three methods of gel filtration chromatography (GFC) and reversed phase liquid chromatography (RPLC). Specificity of GFC was checked by applying the method for the determination of amoxicillin and its related impurities and linking of GFC and RPLC. The result shows that for the determination of polymers the gel filtration chromatography is a rather simple separation mode as compared to RPLC. Regarding the three gel filtration chromatographic methods, the separation on the Superdex peptide column is advantage

[Comments about dosing of low molecular weight heparins (LMWH)].

Low molecular weight heparins (LMWHs) have different schedules for dosage according to the official descriptions and recommendations prepared by the particular manufacturer. Therapeutic regimens are mainly depending on bodyweight, even if data concerning the possible effects body composure (i.e. in obesity, lean body mass) are apparently more or less missing. Prophylactic doses are either based on bodyweight (or just an approximation, weight-frames) or mainly perioperative cases on the assessment of the surgical risk. Manufacturers are busy to supply easy and convenient way to approach dosage

Low molecular weight heparin in unstable coronary artery disease.

Since the recognition that any thrombosis overlying a ruptured atherosclerotic plaque is a central component of the pathogenesis of unstable coronary artery disease (CAD), a number of antithrombotic treatment strategies have been investigated in randomised clinical trials. Aspirin reduces the occurrence of symptomatic and silent ischaemia, myocardial infarction (MI) and death in patients with unstable CAD, both in the acute phase and during continued long-term treatment and is now considered routine therapy. The addition of unfractionated heparin infusion further reduces cardiac events during

Thromboprophylaxis with low molecular weight heparin (dalteparin) in pregnancy.

Venous thromboembolism remains an important cause of maternal mortality. For women at risk during pregnancy, the recommended venous thromboembolismprophylaxis is unfractionated heparin. Low molecular weight heparins, such as dalteparin, also may be suitable, but randomised trials have not been performed. Pregnant women (105) with confirmed previous or current thromboembolism were randomised to receive either unfractionated heparin twice daily (mean 20569 IU/day) or dalteparin once daily (mean 4631 IU anti-factor Xa units/day) subcutaneously for thromboprophylaxis during pregnancy and postpartu

Low molecular weight components from black widow spider venom.

Highly purified alpha-latrotoxin from the black widow spider venom (alpha-LTX) consists of two polypeptides with mol. wts of 130,000 and 8000 (LMWP). We have isolated two low mol. wt proteins LMWP and LMWP2 from the low mol. wt fraction of this venom. The chemical properties of these proteins and partial amino acid sequence of novel protein LMWP2 were studied. By means of i.v. or intracerebroventricular injections into mice it was shown that low mol. wt components of the venom at concentrations of 2.3 mg/kg and 0.8 mg/kg, respectively, did not possess any direct toxic effect on vertebrates. Low molecular weight proteins in human malignant hyperthermic muscle.

Malignant hyperthermia (MH) is a pharmacogenetic disorder provoked by volatile anesthetics and depolarizing muscle relaxants. The preoperative diagnosis of MH is difficult because it requires a large muscle biopsy and a laboratory dedicated to such diagnostic studies. The authors performed electrophoresis of six muscles taken from MH patients or their relatives to determine whether the protein composition is different from normal muscle. MH muscle was found to contain large amounts of two low molecular weight proteins (15,000 daltons and 13,500 daltons) that are not present in normal muscle. A

Stable low molecular weight DNA in xeroderma pigmentosum cells.

Xeroderma pigmentosum (XP) cells from several complementation groups contained more low molecular weight DNA upon alkaline sucrose gradient centrifugation than did other human cells examined. Under conditions in which only 5% of the DNA in normal cells sedimented at 16 S or less, 20% of the DNA in XP cells from complementation group A sedimented at 16 S or less. Because cells were layered directly onto the gradients for lysing of cells and denaturing of the DNA, it appears that this low molecular weight material is due to naturally existing gaps or alkali-sensitive sites, or both, in the cellu
