Factors involved in inflammation-induced developmental white matter damage.

Developmental white matter damage is a brain pathology associated with several long-term neurological disorders. An inflammatory insult has been suggested as the major instigating event. This study investigated the relative influence of inflammation, blood-brain barrier permeability and glial ontogeny in white matter damage. Systemic inflammation was induced in Monodelphis domestica (opossum) by serial intraperitoneal injections of lipopolysaccharide at different stages of brain development. Volume of white matter was estimated for the external capsule. Blood-brain barrier permeability was ass

Ontogenesis of the scapula in marsupial mammals, with special emphasis on perinatal stages of didelphids and remarks on the origin of the therian scapula.

The development of the scapula was studied in embryonic and postnatal specimens of Monodelphis domestica and perinatal specimens of Philander opossum, Caluromys philander, and Sminthopsis virginiae using histological sections and 3D reconstructions. Additionally, macerated skeletons of postnatal M. domestica were examined. This study focused on the detachment of the scapulocoracoid from the sternum and on the acquisition of a supraspinous fossa, a supraspinatus muscle, and a scapular spine, all these events associated with the origin of the therian shoulder girdle. In none of the specimens is

UV-induced melanoma cell lines and their potential for proteome analysis: a review.

We have established cell lines from benign cutaneous melanocytic lesions and from melanoma-affected lymph nodes of monodelphis domestica, the laboratory opossum (a South American marsupial now widely maintained in captive colonies for experimental purposes). Unlike melanoma cell lines currently available from humans and other mammals, the opossum lines are derived from cells transformed in vivo by experimentally controlled exposure to ultraviolet B (UVB) radiation of known spectral composition. Differences in the patterns of protein expression among cell lines at different stages of the UVB-in

Low-dose ultraviolet exposure early in development can lead to widespread melanoma in the opossum model.

Suckling young of opossums (Monodelphis domestica) were exposed to ultraviolet radiation (UVR, predominantly UVB: 290-320 nm) in part to determine an optimal protocol for induction and progression of melanoma in this species. In all, 620 litters were introduced to one of seven protocols. The lowest dose (175 J/m2) administered three times a week for almost three weeks led to the highest incidence of melanotic lesions with melanoma potential (8.1%) among young (5-month-old) adults. Among 101 much older animals (> 17 months at necropsy), 43% showed metastatic melanoma to the lymph nodes and almo

Divergent DNA methylation dynamics in marsupial and eutherian embryos.

Based on seminal work in placental species (eutherians)1-10, a paradigm of mammalian development has emerged wherein the genome-wide erasure of parental DNA methylation is required for embryogenesis. Whether such DNA methylation reprogramming is, in fact, conserved in other mammals is unknown. Here, to resolve this point, we generated base-resolution DNA methylation maps in gametes, embryos and adult tissues of a marsupial, the opossum Monodelphis domestica, revealing variations from the eutherian-derived model. The difference in DNA methylation level between oocytes and sperm is less pronounc

The relationships of marsupial-dwelling Viannaiidae and description of Travassostrongylus scheibelorum sp. n. (Trichostrongylina: Heligmosomoidea) from mouse opossums (Didelphidae) from French Guiana.

The trichostrongylid nematode Travassostrongylus scheibelorum sp. n. from the Linnaeus' mouse opossum, Marmosa murina (Linnaeus) (type host), and the woolly mouse opossum, Marmosa demerarae (Thomas), from French Guiana is described. The nematodes have a synlophe with ridges frontally oriented from right to left, six dorsal and six ventral, at midbody; seven dorsal and seven ventral posterior to the vulva, and two cuticular thickenings within the lateral spaces; a long dorsal ray and a pointed cuticular flap covering the vulva. This is the 12th species of Travassostrongylus Orloff, 1933, which

Genome-wide Characterization of Non-shared Sequences among Amphora-shaped Giant Viruses.

Giant viruses are distinguished not only by their large particle size, but also by their extensive genomes, often reaching megabase levels. Many sequences within these genomes are considered to have been introduced by hosts, surrounding organisms, or other viruses. Since the natural hosts of many giant viruses remain unidentified, analyzing sequences potentially derived from other organisms may aid in clarifying their hosts. In the present study, we identified eukaryote-homologous sequences by isolating those not shared among viruses, an aspect previously overlooked. Our primary focus was on p

The Potential Connection between Molecular Changes and Biomarkers Related to ALS and the Development and Regeneration of CNS.

Neurodegenerative diseases are one of the greatest medical burdens of the modern age, being mostly incurable and with limited prognostic and diagnostic tools. Amyotrophic lateral sclerosis (ALS) is a fatal, progressive neurodegenerative disease characterized by the loss of motoneurons, with a complex etiology, combining genetic, epigenetic, and environmental causes. The neuroprotective therapeutic approaches are very limited, while the diagnostics rely on clinical examination and the exclusion of other diseases. The recent advancement in the discovery of molecular pathways and gene mutations i

A comparative examination of odontogenic gene expression in both toothed and toothless amniotes.

A well-known tenet of murine tooth development is that BMP4 and FGF8 antagonistically initiate odontogenesis, but whether this tenet is conserved across amniotes is largely unexplored. Moreover, changes in BMP4-signaling have previously been implicated in evolutionary tooth loss in Aves. Here we demonstrate that Bmp4, Msx1, and Msx2 expression is limited proximally in the red-eared slider turtle (Trachemys scripta) mandible at stages equivalent to those at which odontogenesis is initiated in mice, a similar finding to previously reported results in chicks. To address whether the limited domain

GnRH neurons and other cell populations migrating from the olfactory neuroepithelium.

Cell migration from the olfactory neuroepithelium to the brain has been widely studied during vertebrate development. Immunocytochemical analysis has revealed that many of the migrating cells contain GnRH (Gonadotropin-Releasing Hormone). The GnRH positive cells migrate from the medial olfactory placode, steam along the nasal septum, cross the basal forebrain and reach the hypothalamic and septal areas from where they control the release of hypophyseal gonadotropic peptides. A peculiar feature of these cells is that they start expressing GnRH during migration. We have analysed the presence of

Development and migration of olfactory neurones in the nervous system of the neonatal opossum.

The neonatal opossum (Monodelphis domestica) was used to assess how different populations of cells are generated in the olfactory region, and how they migrate along pathways to the central nervous system. Developing nerve cells were immunocytochemically labelled using antisera directed against two specific markers of olfactory receptor neurones: olfactory marker protein (OMP) and the dipeptide carnosine. In new-born opossums both carnosine and OMP are already co-expressed in primary olfactory neurones and in those axons that extend towards the olfactory bulb. Expression of these markers in olf

The UT family of MHC class I loci unique to non-eutherian mammals has limited polymorphism and tissue specific patterns of expression in the opossum.

BACKGROUND: The Major Histocompatibility Complex (MHC) class I family of genes encode for molecules that have well-conserved structures, but have evolved to perform diverse functions. The availability of the gray, short-tailed opossum, Monodelphis domestica whole genome sequence has allowed for analysis of MHC class I gene content in this marsupial. Utilization of a novel method to search for MHC related domain structures revealed a previously unknown family of MHC class I-related genes. These genes, named UT1-17, are clustered on chromosome 1 in the opossum, unlinked to the MHC region. UT gen

Symmetry breakage in the development of one-armed gonads in nematodes.

Whereas the hermaphrodite gonad of Caenorhabditis elegans has two symmetric arms (didelphy), the female/hermaphrodite gonad of many nematode species features a single anterior arm (monodelphy). We examined how gonadal cell lineages and intercellular signalling evolve to generate these diverse structures. In C. elegans, the two arms develop symmetrically from two somatic precursor cells, Z1 (anterior) and Z4 (posterior). Each first gives rise to one distal tip cell (which promotes arm growth and germ line proliferation), two ovary precursors and three uterine precursors in the center of the dev

Variation in social feeding behavior and interactions among Caenorhabditis nematodes.

The ability to respond to complex stimuli and environmental cues is essential for organisms to survive and reproduce. Responding to a wide range of stimuli requires a neuronal network that can integrate cues and execute behavioral responses. Evolution of behaviors occurs ubiquitously in most established ecological niches, even among closely related species. To uncover the genetic and neuronal drivers of evolving behaviors, we have taken advantage of the large and relatively ancient divergence in the Caenorhabditis genus to ask how different Caenorhabditis nematodes respond to environmental sti

Evidence from plasma progesterone concentrations for male-induced ovulation in the brush-tailed bettong, Bettongia penicillata.

Female brush-tailed bettongs, Bettongia penicillata, were housed with either an intact or vasectomized male or isolated from males in the peripartum period. Development of the quiescent corpus luteum formed at the post partum oestrus was initiated by removing the pouch young. Blood samples for analysis of plasma progesterone were collected from the females 2 days before removal of pouch young, daily for 5 or 6 days and then 2-3 times each week until 19 days after removal of pouch young. Plasma progesterone profiles were similar in pregnant and nonpregnant cycles. There was an early progesteron
