Mineralocorticoid receptor stimulation effects on spatial memory in healthy young adults: A study using the virtual Morris Water Maze task.

OBJECTIVES: Stress hormones such as cortisol are known to influence a wide range of cognitive functions, including hippocampal based spatial memory. In the brain, cortisol acts via two different receptors: the glucocorticoid (GR) and the mineralocorticoid receptor (MR). As the MR has a high density in the hippocampus, we examined the effects of pharmacological MR stimulation on spatial memory. METHODS: Eighty healthy participants (40 women, 40 men, mean age=23.9years±SD=3.3) completed the virtual Morris Water Maze (vMWM) task to test spatial encoding and spatial memory retrieval after receivi

Alamandine enhanced spatial memory in rats by reducing neuroinflammation and altering BDNF levels in the hippocampus and prefrontal cortex.

Our study aims to determine the effects of alamandine, the newest component of the renin-angiotensin system, on cognitive functions, neuroinflammation, and oxidative stress in the pathophysiology of depression. 35 male Sprague dawley rats, three months old, weighing between 300 and 350 g, were used. The chronic, unpredictable mild stress model of depression was performed. Experimental animals were divided into five groups: control (C), depression (D), alamandine (50 µg/kg, ip) (D + ALA), A779 (300 µg/kg, ip) (D + A779), and both alamandine and A779 treatment groups (D + ALA + A779). Sex-related differences in cognition after severe murine traumatic brain injury: A Morris water maze study evaluating learning and memory.

BACKGROUND: A number of sex-related outcomes following severe traumatic brain injury (TBI) appear to principally favor females. However, sex-related differences in post-TBI learning and memory remain underexplored. We hypothesized that females realize greater cognitive recovery than males following severe TBI. METHODS: CD1 male (n = 12) and female (n = 12) mice were randomized to controlled cortical impact (severe TBI: impactor tip diameter, 3 mm; impact velocity, 6 m/s; depth, 1 mm; dwell time, 100 milliseconds) or sham craniotomy and followed for 14 days. Body weight loss recovery was measu

Adult neurological function following neonatal hypoxia-ischemia in a mouse model of the term neonate: water maze performance is dependent on separable cognitive and motor components.

BACKGROUND AND PURPOSE: Hypoxic-ischemic injury in term neonates remains a significant cause of long-term neurological morbidity. The post-natal day 10 (P10) mouse is accepted as a model for the term human. This study was designed to assess the relationships between the duration of hypoxia-ischemia (HI) on P10 and the structural and functional neurological deficits that appear in the adult mouse as a consequence. METHODS: Post-natal day 10 129T2xC57Bl/6 F1 hybrid mice were subjected to 0, 45, 60 or 75 min of hypoxia-ischemia using the Rice-Vannucci model. Beginning on P50 these mice were test

Administration of ethanolic extract of Erythrophleum ivorense (A Chev.) stem bark to male Wistar rats alters brain areas involved in motor coordination, behavior, and memory.

ETHNOPHARMACOLOGICAL RELEVANCE: Erythrophleum ivorense (A Chev.) is a common plant in the tropics. Its use as ordeal poison in folklore medicine is controversial. The incoordination and behavioral changes following consumption are often associated with guilt. This study is aimed at dispelling or upholding this belief by investigating the actions of E. ivorense on the brain and behavior using rat model. MATERIALS AND METHODS: Sixty male Wistar rats were equally divided into five groups; control group received distilled water, test groups were administered 10, 20, 30 and 40 mg/kg ethanolic extr

Acute alcohol exposure during neurulation: Behavioral and brain structural consequences in adolescent C57BL/6J mice.

Prenatal alcohol exposure (PAE) can induce physical malformations and behavioral abnormalities that depend in part on thedevelopmental timing of alcohol exposure. The current studies employed a mouse FASD model to characterize the long-term behavioral and brain structural consequences of a binge-like alcohol exposure during neurulation; a first-trimester stage when women are typically unaware that they are pregnant. Time-mated C57BL/6J female mice were administered two alcohol doses (2.8g/kg, four hours apart) or vehicle starting at gestational day 8.0. Male and female adolescent offspring (po

Prostaglandin endoperoxide synthase 2 regulates neuroinflammation to mediate postoperative cognitive dysfunction in mice.

Prostaglandin endoperoxide synthase 2 (PTGS2) is a rate-limiting enzyme of prostaglandin (PGs) production, mediating perioperative inflammatory response. This study aimed to explore the mechanisms underlying the involvement of PTGS2 in postoperative cognitive dysfunction (POCD). Transient bilateral common carotid artery occlusion (tBCCAO) was performed to induce POCD. The Morris water maze test was used to assess the cognitive function. PTGS2 expression in the hippocampus and plasma was measured. Hippocampal RNA sequencing was performed to determine the pathological basis of POCD. In vivo, the

Electroacupuncture Ameliorates Cognitive Impairment Through the Inhibition of NLRP3 Inflammasome Activation by Regulating Melatonin-Mediated Mitophagy in Stroke Rats.

Previous studies found that electroacupuncture (EA) at the Shenting (DU24) and Baihui (DU20) acupoints alleviates cognitive impairment in cerebral ischemia-reperfusion (I/R) injury rats. Nonetheless, the mechanisms of the anti-inflammatory effects of EA are unclear. Cerebral I/R injury was induced in rats by middle cerebral artery occlusion (MCAO). Following I/R injury, the rats underwent EA therapy at the Shenting (DU24) and Baihui (DU20) acupoints for seven successive days. The Morris water maze test, magnetic resonance imaging (MRI) and molecular biology assays were utilized to assess the e

Effects of perinatal diet and prenatal stress on the behavioural profile of aged male and female rats.

The present work studies whether chronic prenatal stress (PS) influences the long-term sex-dependent neuropsychological status of offspring and the effects of an early dietary intervention in the dam. In addition, dams were fed with either a high-fat sugar diet (HFSD) or methyl donor supplemented diet (MDSD). PS procedure did not affect body weight of the offspring. MDSD induced decreases in body weight both in male and female offspring (1 month) that were still present in aged rats. HFSD induced an increase in body weight both in male and female offspring that did not persist in aged rats. In

Antidepressant-like and memory-enhancing effects of the N-type calcium channel blocker ziconotide in rats.

The lack of oral or injectable formulations of ziconotide (ω-conotoxin peptide), a novel analgesic agent, limits research on potential neurobehavioral protective properties of this substance, including antidepressant-like effects. Here we expose rats to a stress paradigm that induces depression and memory impairment to assess the effects of ziconotide treatment. Ziconotide was administered intracerebroventricular (i.c.v.) to rats undergoing stereotaxic surgery at a single dose (1 μg/rat) or in repeated long-term applications (dosage groups: 0.1, 0.3, and 1 μg/rat).
