Phenotypic characterization of Mycoplasma synoviae induced changes in the metabolic and sensitivity profile of in vitro infected chicken chondrocytes.

In infectious synovitis caused by Mycoplasma synoviae chicken chondrocytes (CCH) may come into direct contact with these bacteria that are also capable of invading CCH in vitro. In this study, phenotype microarrays were used to evaluate the influence of Mycoplasma synoviae on the global metabolic activity of CCH. Therefore, CCH were cultured in the presence of 504 individual compounds, spotted in wells of 11 phenotype microarrays for eukaryotic cells, and exposed to Mycoplasma synoviae membranes or viable Mycoplasma synoviae. Metabolic activity and sensitivity of normal cells versus infected c

Influence of Different Tetracycline Antimicrobial Therapy of Mycoplasma (Mycoplasma synoviae) in Laying Hens Compared to Tea Tree Essential Oil on Table Egg Quality and Antibiotic Residues.

The food of animal origin that is the most consumed is the table egg, but laying hens treated with antibiotics can produce eggs contaminated with antibiotic residues. Residues of antibiotics may present a risk for consumer health. Keeping in mind that laying hens almost always suffer from Mycoplasma (Mycoplasma synoviae), for which they are treated with antibiotics, high-quality egg production is even harder. Our research aimed to investigate the influence of three different antibiotics compared to the tea tree (Melaleuca alternifolia) essential oil administered to naturally infected laying he

Development and evaluation of a recombinase-aided amplification-lateral flow dipstick assay for rapid and sensitive detection of Mycoplasma synoviae.

BACKGROUND: Mycoplasma synoviae (MS) is a critical avian pathogen responsible for enormous economic losses in global poultry production. Timely prevention and control of MS epidemics rely heavily on rapid, sensitive and accurate diagnostic approaches, while conventional detection methods remain time-consuming and complicated. Therefore, we developed a visual on-site detection system based on recombinase-aided amplification combined with lateral flow dipstick for MS (RAA-LFD-MS assay). METHODS: According to the design rules for RAA primers and exo-probes, specific primers and a probe were desi

Development of molecular assays for the rapid and cost-effective determination of fluoroquinolone, macrolide and lincosamide susceptibility of Mycoplasma synoviae isolates.

Mycoplasma synoviae infection occurs worldwide, leading to considerable economic losses in the chicken and turkey industry due to infectious synovitis, respiratory diseases and eggshell apex abnormalities. Control programs against M. synoviae infection are based on eradication, vaccination and medication with antimicrobial agents. Prudent use of antibiotics can be improved greatly by the determination of antibiotic susceptibility prior to the treatment. However, the conventional broth or agar microdilution is very labor-intensive and time-consuming method. Thus, there is an increasing need for

European ring test on the differentiation of wild-type Mycoplasma synoviae and the MS-H vaccine strains in clinical samples.

The control and prevention of Mycoplasma (M.) synoviae infections have been significantly improved by the use of the MS-H live vaccine strain. This vaccine has been used globally for decades; therefore, many technical approaches have been published or commercialized for the differentiation of MS-H from field strains. However, key problems have been identified related to DIVA (differentiating infected from vaccinated animals) testing and monitoring of vaccination in many European laboratories and poultry farms. To address these concerns, the reliability of protocols for the differentiation of t

Comparative genomic analyses of Mycoplasma synoviae vaccine strain MS-H and its wild-type parent strain 86079/7NS: implications for the identification of virulence factors and applications in diagnosis of M. synoviae.

Mycoplasma synoviae is an economically important avian pathogen worldwide, causing respiratory disease, infectious synovitis, airsacculitis and eggshell apex abnormalities in commercial chickens. Despite the widespread use of MS-H as a live attenuated vaccine over the past two decades, the precise molecular basis for loss of virulence in this vaccine is not yet fully understood. To address this, the whole genome sequence of the vaccine parent strain, 86079/7NS, was obtained and compared to that of the MS-H vaccine. Except for the vlhA expressed region, both genomes were nearly identical. Thirt

Residues Ser83, Arg85, Tyr88, Asn124, and Lys192 of C-Terminal Lipid-Associated Membrane Hemagglutinin Affect Mycoplasma synoviae Agglutination of Erythrocytes.

Mycoplasma synoviae (M. synoviae) is an avian pathogen responsible for respiratory disease and synovitis. The VlhA (variable lipoprotein hemagglutinin) family of surface adhesins plays a critical role in host cell attachment, yet the specific residues and structural determinants governing this interaction remain incompletely understood. In this study, we selected the C-terminal lipid-associated membrane hemagglutinin (LAM HA) domain within the VlhA family as the bait protein, based on its conserved C-terminal region. Yeast two-hybrid screening identified 18 LAM HA-interacting host proteins, an

Recombinant adenovirus expressing pdhβ-pdhD fusion protein produces robust immune responses and partial protection against Mycoplasma Synoviae challenge in chickens.

Mycoplasma synoviae (MS) stands as a pivotal pathogen, responsible for triggering arthritis and airsacculitis in both chickens and turkeys. Given the pressing need for safe and efficacious vaccine candidates, we engineered recombinant adenoviruses expressing a fusion antigen. This antigen consisted of the pyruvate dehydrogenase E1 subunit beta (pdhβ) and dihydrolipoyl dehydrogenase (pdhD) of MS. We then systematically evaluated the immune effect and protective efficacy of these recombinant adenoviruses against MS challenge in a chicken model. Our results demonstrated the successful constructio

Enhancing Mycoplasma synoviae control: A novel quantitative real-time PCR approach to differentiate MS-H vaccine strain and field strains.

Mycoplasma synoviae (MS) is a prevalent pathogen in poultry, causing substantial economic losses. Vaccination is a crucial strategy for controlling MS infections. In China, a temperature-sensitive live vaccine, Vaxsafe®MS (MS-H strain), was introduced to address the rising prevalence of MS since 2017. Accurate differentiation between the MS-H vaccine strain and field strains is essential for the effective application of the vaccine and the eradication of MS in poultry. In this study, a duplex TaqMan real-time quantitative PCR (qPCR) method was first developed based on a single nucleotide polym

Mycoplasma synoviae surface-located elongation factor G functions as a cytoadhesin to promote adhesion to synovial sheath cells through binding to vimentin.

Mycoplasma synoviae infection has caused serious economic losses to the poultry industry worldwide. The molecular mechanism by which M. synoviae colonizes the synovium and induces synovitis is unclear. In this study, desthiobiotin pull-down and liquid chromatography-tandem mass spectrometry analyses were used to screen M. synoviae membrane proteins that bind the membrane proteins of synovial sheath cells (SSCs). Among the 128 screened proteins, elongation factor G (EF-G) of M. synoviae was identified as a surface-located protein using colony blotting and dual fluorescence analyses. The immunog

Molecular detection of Mycoplasma synoviae and avian reovirus infection in arthritis and tenosynovitis lesions of broiler and breeder chickens in Santa Catarina State, Brazil.

Infectious arthritis or tenosynovitis in broiler and breeder chickens results in major loss of productivity because of reduced growth and downgrading at processing plants. The most common causative agents of avian infectious arthritis are the bacterium Mycoplasma synoviae and avian reoviruses (ARVs) (family Reoviridae, genus Orthoreovirus). In this study, we evaluated the occurrence of these two pathogens in arthritis or tenosynovitis lesions of broilers and breeder flocks in southern Brazil using molecular detection. Tissue sections from tibiotarsal joints with visible lesions from 719 broile
