The pathobiology of Dupuytren's contracture: effects of prostaglandins on myofibroblasts.

The in vitro response of myofibroblasts to prostaglandins F2alpha (a vasoconstrictor) and E2 (a vasodilator) were evaluated in specimens obtained from the Dupuytren's nodules of 12 patients. Fibroblasts from four control samples of palmar fascia were similarly tested. This study demonstrated the ability of prostaglandin F2alpha to induce significant contraction of myofibroblasts. Prostaglandin E2 was noted to cause significant relaxation of myofibroblasts. The contractile/relaxation responses of control fibroblasts to these prostaglandins were minimal.

The pathogenesis of Dupuytren's contracture: contractile mechanisms of the myofibroblasts.

The role of myofibroblasts in the pathogenesis of Dupuytren's contracture was investigated by light and electron microscopic histochemical methods. Dupuytren's myofibroblasts contain an intracellular contractile mechanism that is driven by the dephosphorylation of adenosine triphosphate. Our study of calcium adenosinetriphosphatase (ATPase) activities verifies that the site of this energy system is on the myofilaments of the myofibroblasts. The degree of ATPase activity, as determined by cell counts, appeared to correlate with the residual contracture as predicted by the Legge and McFarlane Ou

Possible role of myofibroblasts in the pathogenesis of Dupuytren's contracture.

In the contracting palmar aponeurosis of patients suffering from Dupuytren's disease myofibroblasts are revealed by electron microscopical examination. These cells display abnormal foldings of the nucleus, microfilaments resembling the myofilaments in smooth muscle cells, rough endoplasmic reticulum occupying circumscribed portions of the cytoplasm, free ribosomes, well-developed Golgi apparatus, numerous mitochondria, occasionally lipid droplets, cilia-like formations, hemidesmosomes and maculae adherens (desmosomes) between the cells and formation of basement membrane. According to literary

[The role of myofibroblasts in the pathogenesis of Dupuytren's contracture].

The involved palmar aponeurosis of 30 patients in different stages of Dupuytren's disease was removed and examined by light and electron microscopy. The specimens were obtained by Z-plasty and limited fasciectomy. The light microscopic slides of the early stages of the disease showed areas rich in cells and surrounded by a network of collagen fibres. In the late phase of the lesion mature scar tissue was observed. By electron microscopy fibroblast-like cells--so-called myofibroblasts were seen in the early stages of the lesion. These cells possess, in addition to nuclear deformations, the foll

Inflammatory myofibroblastic tumor of the prostate.

INTRODUCTION: Inflammatory myofibroblastic tumors are borderline malignant soft tissue tumors primarily affecting the lungs and pelvic organs. This report presents a rare case of an inflammatory myofibroblastic tumor originating from the prostate gland in a young male. CASE PRESENTATION: A 20-year-old man developed gross hematuria and dysuria, revealing a prostatic mass. Pathological examination of a biopsy displayed spindle-shaped myofibroblast proliferation and an infiltrate of inflammatory cells, leading to a diagnosis of inflammatory myofibroblastic tumor. Following fertility preservation

Uterine inflammatory myofibroblastic tumor.

Inflammatory myofibroblastic tumor (IMT) is recognized as a true neoplasm of unknown etiology, but its pathogenesis is related to abnormalities in the ALK gene. This is an uncommon tumor with a wide anatomic distribution and often constitutes a challenging diagnosis owing to its histological similarities with other tumors. Uterine IMTs are rare and their detailed characteristics should be described based on case reports and small case series. Thus, we performed a comprehensive review of the literature showing that uterine IMTs show a wide range of age at diagnosis (median, 39 years), and a sym

Unraveling SSc Pathophysiology; The Myofibroblast.

Systemic sclerosis (SSc) is a severe auto-immune disease, characterized by vasculopathy and fibrosis of connective tissues. SSc has a high morbidity and mortality and unfortunately no disease modifying therapy is currently available. A key cell in the pathophysiology of SSc is the myofibroblast. Myofibroblasts are fibroblasts with contractile properties that produce a large amount of pro-fibrotic extracellular matrix molecules such as collagen type I. In this narrative review we will discuss the presence, formation, and role of myofibroblasts in SSc, and how these processes are stimulated and

Mesenteric inflammatory myofibroblastic tumors.

Inflammatory myofibroblastic tumors (IMTs), also known as inflammatory pseudotumors and inflammatory fibrosarcomas, are uncommon mesenchymal tumors composed of myofibroblastic spindle cells admixed with lymphocytes, plasma cells and eosinophils. Once thought to be reactive, these lesions are now considered to be neoplastic. These tumors can occur throughout the body, most commonly in the lung, mesentery and omentum. Patients commonly present with painless abdominal mass or with intestinal obstruction. IMTs may be multicentric, have a high local recurrence rate and may metastasize in rare cases

Laryngeal inflammatory myofibroblastic tumor.

Inflammatory myofibroblastic tumor seldom involves the larynx, as only about 50 to 60 cases have been described in the literature. Even though these tumors are often not aggressive, they have the potential for invasion and local recurrence. We describe the case of a 27-year-old man who was admitted to an emergency department with signs of upper airway obstruction secondary to an obstructive mass. Histology identified the mass as an inflammatory myofibroblastic tumor of the subglottis. The patient underwent an emergency tracheotomy followed by a partial laryngectomy. During 14 months of follow-

Inflammatory myofibroblastic tumor of the bladder.

We illustrate a case of an inflammatory myofibroblastic tumor (IMT) involving the bladder in a woman with dysuria and review the literature and differential diagnosis. Inflammatory myofibroblastic tumor, also referred to as pseudosarcomatous myofibroblastic proliferation, is a rare lesion that can arise in the genitourinary system and is characterized by a fascicular arrangement of myofibroblasts with admixed inflammatory cells and slitlike vessels. Urinary bladder IMT can be a diagnostic pitfall because its histologic features (brisk mitoses, invasion into muscularis propria, and prominent nu

Inflammatory myofibroblastic tumor in colon.

Inflammatory myofibroblastic tumor (IMT) is an uncommon mesenchymal solid tumor commonly documented in children and young adults. Here, we report a case of IMT in colon confirmed pathologically after laparoscopic anterior resection. A 35-year-old man presented with anal bleeding after defecation for 2 weeks. Colonoscopy demonstrated a mass with shallow ulceration in the central area and irregular margin accompanied by intact mucosa in the descending colon. Computer tomography showed a well-demarcated and homogenous solitary mass in the descending colon. We performed laparoscopic anterior resec

Inflammatory myofibroblastic tumor of the trachea.

This report described a 2-year-old boy who presented with severe respiratory distress and stridor. Bronchoscopy and CT revealed a mass in the left anterolateral tracheal wall and histopathology showed a tracheal inflammatory myofibroblastic tumor. Initial removal by rigid bronchoscopy resulted in prompt recurrence of the tumor. Therefore he underwent tracheal surgical resection. A bronchoscopy at 12 months after surgery did not show any recurrence sign.

The role of the myofibroblast in Dupuytren's contracture.

A case report of Dupuytren's contracture illustrates the functional and light and electron microscopic characteristics of the myofibroblast. The myofibroblast is now associated with an increasing number of diseases, and is believed to be a morphologic and functional variant of the fibroblast, which shows changes typical of smooth-muscle cells. Its presence may be suspected by light microscopy, but its ultrastructure is characteristic. Its distinctive electron microscopic features are a fibrillar system in the cytoplasm similar to that of a smooth muscle, nuclear deformations indicative of cont

Subepithelial myofibroblasts enhance stem cell regeneration after intestinal epithelial damage.

When intestinal stem cells (ISCs) are damaged, other epithelial cells de-differentiate to restore the ISC compartment. Immune, nerve and lymphatic endothelial cells contribute signals that promote this response, but it is unclear whether or how the native mesenchymal niche senses and responds to ISC loss. In homeostasis, trophocytes and smooth muscle beneath intestinal crypts secrete BMP inhibitors (BMPi) and R-spondins to foster ISC self-renewal, whereas sub-epithelial myofibroblasts (SEMFs) concentrated at crypt tops promote differentiation through bone morphogenetic proteins (BMPs). We asse

MYOFIBROBLASTS HINDER RECOVERY OF HASHIMOTO THYROIDITIS IN THE ULTRASTRUCTURAL LEVEL.

BACKGROUND: Hashimoto thyroiditis (HT) is an autoimmune disorder associated with hypothyroidism. Lymphocyte infiltration leading to thyroid follicular cell destruction is counteracted by increased collagen production, deposition and scarring. However, only recently a specific subpopulation of modified fibroblasts with contractile properties, namely "myofibroblasts" (MFBs) have been linked to HT. AIM: Our ultrastructural study aims to delineate the presence and contribution of MFBs to the fibrotic milieu of HT. MATERIAL AND METHODS: Tissue biopsies were obtained from 5 HT-diagnosed patients a

Albendazole ameliorates aerobic glycolysis in myofibroblasts to reverse pulmonary fibrosis.

BACKGROUND: Idiopathic pulmonary fibrosis (IPF) is a chronic and lethal lung disorder for which effective treatments remain limited. Recent investigations revealed a potential link between altered glucose metabolism and the activation of fibroblasts, the key cells responsible for generating and depositing extracellular matrix proteins within the lung interstitium during IPF development. METHOD: In this study, we aimed to investigate the potential therapeutic impact of albendazole on fibroblast to myofibroblast transition in IPF. We assess albendazole's effectiveness in attenuating the activat
