Enzymatic synthesis of isopropyl myristate using immobilized lipase from Bacillus cereus MTCC 8372.

A purified alkaline thermo-tolerant bacterial lipase from Bacillus cereus MTCC 8372 was immobilized on a Poly (MAc-co-DMA-cl-MBAm) hydrogel. The hydrogel showed approximately 94% binding capacity for lipase. The immobilized lipase (2.36 IU) was used to achieve esterification ofmyristic acid and isopropanol in n-heptane at 65 degrees C under continuous shaking. The myristic acid and isopropanol when used at a concentration of 100 mM each in n-heptane resulted in formation of isopropyl myristate (66.0 +/- 0.3 mM) in 15 h. The reaction temperature below or higher than 65 degrees C markedly reduce

Sub- or Supercritical CO2‑Assisted Melting Point Depression of Free Fatty Acids and Triglycerides for Microencapsulation Processes in Sustainable Veterinary Applications.

The process based on the particles from gas-saturated solutions (PGSS) using supercritical fluids offers a green alternative to conventional lipid-based encapsulation methods, which typically require higher thermal energy. This study presents experimental data including onset (first melting), completion (last melting), and freezing temperatures for the binary system formed by fatty acids and carbon dioxide at pressures range from 5 to 35 MPa. The results revealed notable melting point depressions across all tested compounds, with the greatest depression observed for myristic acid (ΔT = 16.36 K

Palmitic acid, but not other long-chain saturated fatty acids, increases S100B protein and TNF-α secretion by astrocytes.

Obesity is a health problem that involves fat accumulation in adipose and other tissues and causes cell dysfunction. Long-chain saturated fatty acids can induce and propagate inflammation, which may also contribute to the brain alterations found in individuals with obesity. Fatty acids accumulate in astrocytes in situations of blood‒brain barrier disruption, such as inflammatory conditions. Furthermore, the increase in tumor necrosis factor-alpha (TNF-α) and S100 calcium-binding protein B (S100B) secretion is considered an essential component of the inflammatory response. We hypothesize that t

Effect of human serum albumin on drug metabolism: structural evidence of esterase activity of human serum albumin.

Human serum albumin (HSA) is the most abundant plasma protein in the human body with a plasma concentration of 0.6mM. HSA plays an important role in drug transport and metabolism. Enzymatic activity of HSA on different substrates or drugs has been studied and documented. The structural mechanism of this activity, however, is unknown. In this study, we have determined the crystal structures of HSA-myristate in a complex of aspirin and of salicylic acid, respectively. The crystal structure of HSA-myristate-aspirin illustrates that aspirin transfers acetyl group to Lys199 and is hydrolyzed into s

Qualitative and Quantitative Analysis of Regional Cerebral Free Fatty Acids in Rats Using the Stable Isotope Labeling Liquid Chromatography-Mass Spectrometry Method.

Free fatty acids serve as important bioactive molecules in the brain. They are involved in message transfer in the brain. There are many reports available in the literature regarding the role of cerebral fatty acids in message transfer; however, most of the studies are mainly focused on limited fatty acid species or only a few specific brain regions. To understand the relationship between cerebral functions and free fatty acids, it is necessary to investigate the distribution of the free fatty acids among different regions in the whole brain. In this study, free fatty acids were extracted from

Comparison of the Thermal Behavior and Chemical Composition of Milk Powders of Animal and Plant Origin.

The present study aims to perform a comparative analysis of the chemical composition and thermal behavior of two distinct milk types, namely animal and plant-based. The thermal analysis revealed the presence of the following classes of compounds: hydrocarbons, heterocycles, aldehydes, ketones, amines and alcohols. All types of milk contain saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs) and polyunsaturated fatty acids (PUFAs), though the relative proportions of these vary depending on the specific milk type. Animal milk powders contain SFAs, including palmitic, stearic, and m

Uveal vs. cutaneous melanoma. Origins and causes of the differences.

BACKGROUND: Fatty acid composition of serum lipids is associated with cardiovascular disease. As attenuated endothelium-dependent vasodilation (EDV) is an early event in atherosclerosis, we investigated the relationships between endothelial vasodilatory function and the proportion of serum fatty acids, reflecting dietary fat quality, in 74 healthy men and women, aged 20-30 years. DESIGN: Endothelium-dependent vasodilation and endothelium-independent vasodilation (EIDV) was studied in the forearm during local administration of methacholine (2 and 4 micro g min-1) and nitroprusside (5 and 10 mi

Long-Chain Fatty Acids Inhibit the Human Bitter Taste Receptor TAS2R16 in a Structure-Dependent Manner.

Bitterness limits the development of palatable foods containing bioactive ingredients. This study investigated whether long-chain fatty acids (LCFAs) modulate the human bitter taste receptor TAS2R16 using a HEK293T cell-based calcium assay with salicin as an agonist. None of the tested LCFAs directly activated TAS2R16. Under co-treatment, only oleic acid inhibited salicin-induced calcium responses, shifting the salicin EC50 from 1.82 ± 0.11 mM to 3.55 ± 1.22 mM at 30 μM (p < 0.01). Pretreatment with all LCFAs inhibited TAS2R16 concentration-dependently. Reliable IC50 values were obtained for p

Effect of different fatty acids on Neisseria gonorrhoeae viability.

Neisseria gonorrhoeae (GC) is the agent of one of the most common bacterial sexually transmitted infections worldwide. The possible development of 'untreatable' infections points out the need for antibiotic-sparing methods to reduce the number of gonococcal infections. In this context, fatty acids are interesting candidates as next-generation antibacterial agents. The aim of this study was to investigate the bactericidal effects of selected fatty acids on GC viability, as well as to observe their biological effects by means of transmission electron microscopy. The cytotoxicity of these compoun

Labeling of adipocyte membranes by sulfo-N-succinimidyl derivatives of long-chain fatty acids: inhibition of fatty acid transport.

Sulfo-N-succinimidyl derivatives of the long-chain fatty acids, oleic and myristic, were synthesized and covalently reacted with isolated rat adipocytes. The plasma membrane proteins labeled by these compounds and the effect of labeling on the transport of long-chain fatty acids were investigated. Sulfo-N-succinimidyl oleate (SSO) and myristate (SSM) inhibited the transport of fatty acids (by about 70%). Inhibition of fatty acid transport was not a result of alterations in cell integrity, as intracellular water volume was not changed. It did not reflect effects on fatty acid metabolism, since

Simultaneous Determination of Essential Oil Components and Fatty Acids in Fennel using Gas Chromatography with a Polar Capillary Column.

Cultivated and wild growing samples of fennel (Foeniculum vulgare Mill., Apiaceae) from R. Macedonia were studied for their volatiles and fatty acid composition. The main essential oil components isolated via hydrodistillation were: trans-anethole (>80%), estragole (< 6%), limonene (< 6%), anisaldehyde (< 1%) and 0.5 % fenchone. An alternative method for characterization of both the non-polar volatile and non volatile fractions was developed using n-hexane and dichloromethane (3:1, v/v) in a Soxhlet extraction followed by transesterification. The obtained extracts were then characterized and t

Maternal separation-induced changes in breast milk fatty acid composition are associated with altered gut microbiota and serotonergic gene expression in neonatal mice.

Maternal separation (MS) is a widely used model of early-life stress, yet the biological pathways transmitting maternal stress signals to offspring remain incompletely understood. Breast milk represents a primary biological interface between mother and infant, and stress-induced changes in milk composition may influence offspring development through the gut microbiota-brain axis. Here, we investigated whether MS alters the breast milk metabolome and whether such changes are associated with offspring gut microbiota composition, serum metabolite profiles, and serotonin-related gene expression in

Non-destructive detection of multiple fatty acids in fuzzy cottonseeds based on near-infrared spectroscopy and chemometrics.

As an important oil resource, the fatty acids composition of cottonseed significantly influences its nutritional value and industrial application. This study develops a rapid and non-destructive detection method for nine kinds of fatty acids of fuzzy cottonseeds based on near-infrared spectroscopy (NIRS) coupled with chemometrics. To enhance the performance of the model, proposed the optimization strategy by integrating three preprocessing methods: derivative, Savitzky - Golay smoothing, and standard normal variate, and confirming parameters by grid searching. Moreover, employs SHapley Additiv
