Mechanical Considerations for Electrospun Nanofibers in Tendon and Ligament Repair.

Electrospun nanofibers possess unique qualities such as nanodiameter, high surface area to volume ratio, biomimetic architecture, and tunable chemical and electrical properties. Numerous studies have demonstrated the potential of nanofibrous architecture to direct cell morphology, migration, and more complex biological processes such as differentiation and extracellular matrix (ECM) deposition through topographical guidance cues. These advantages have created great interest in electrospun fibers for biomedical applications, including tendon and ligament repair. Electrospun nanofibers, despite

PLLA-PEG-TCH-labeled bioactive molecule nanofibers for tissue engineering.

By mimicking the native extracellular matrix, electrospun nanofibrous scaffolds (ENSs) can provide both chemical and physical cues to modulate cell adherence and differentiation and to promote tissue regeneration while retaining bioresorbable and biocompatible properties. In this study, ENSs were developed to deliver multiple biomolecules by loading them into the core-sheath structure and/or by conjugating them to the nanofiber surfaces. In this work, poly(L-lactide)-poly(ethylene glycol)-NH(2) and poly(L-lactide) were emulsion electrospun into nanofibers with a core-sheath structure. A model

Electrospinning thermoplastic polyurethane-contained collagen nanofibers for tissue-engineering applications.

Electrospinning is a new method used in tissue engineering. It can spin fibers in nanoscale by electrostatic force. A series of thermoplastic polyurethane (TPU)/collagen blend nanofibrous membranes was prepared with different weight ratios and concentrations via electrospinning. The two biopolymers used 1,1,1,3,3,3,-hexafluoro-2-propanol (HFP) as solvent. The electrospun TPU-contained collagen nanofibers were characterized using scanning electron microscopy (SEM), XPS spectroscopy, atomic force microscopy, apparent density and porosity measurement, contact-angle measurement, mechanical tensile

Electrospun chitosan-P(LLA-CL) nanofibers for biomimetic extracellular matrix.

Chitosan-poly(L-lactic acid-co-epsilon-caprolactone)(50:50) (P(LLA-CL)) (CS/P(LLA-CL)) blends were electrospun into nanofibers using 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and trifluoroacetic acid (TFA) as solvents. Chitosan, which is difficult to electrospin into nanofibers, could be easily electrospun into nanofibers with addition of a small portion of P(LLA-CL). The fiber diameter depended on both the polymer concentration and the blend ratio of chitosan to P(LLA-CL). The average fiber diameter increased with increasing polymer concentration and decreasing the blend ratio of chitosan to P

Cavity and waveguide quantum electrodynamics with atoms and optical nanofibers.

Optical nanofibers provide a versatile platform for exploring strong light-matter interactions at the single-quantum level, a central goal of quantum optics. Their subwavelength diameter supports tightly confined guided modes with strong evanescent fields, enabling efficient coupling between photons and nearby atoms. By incorporating fiber Bragg gratings, nanofibers can form fully fiber-integrated high-finesse cavities, combining strong atom-photon coupling with efficient and low-loss optical input-output interfacing in the setting of cavity quantum electrodynamics (QED). In parallel, nanofibe

Advancing Antimicrobial Peptide Delivery Through Nanofibers: Principles, Challenges, and Perspective.

Antimicrobial peptides (AMPs) are emerging as a promising class of drugs for treating multidrug-resistant illnesses. However, their clinical applicability is hampered by issues including instability, fast disintegration, and low bioavailability. Nanofibers, due to their distinct structural and functional features, provide a potent platform for improving the delivery and therapeutic effectiveness of AMPs. This study investigates the concepts that underpin the integration of AMPs with nanofibers, such as electrospinning processes, encapsulating strategies, and controlled release mechanisms. The

Electrospun nanofibers: Focus on local therapeutic delivery targeting infectious disease.

Whether it be due to genetic variances, lack of patient adherence, or sub-optimal drug metabolism, the risk of antibiotic resistance from medications administered systemically continues to pose significant challenges to fighting infectious diseases. Ideally, infections would be treated locally for maximal efficacy while minimizing off-target effects. The electrospinning of biomaterials has recently facilitated the creation of electrospun nanofibers as an alternative delivery vehicle for local treatment. This review describes electrospun nanofiber applications to locally target various infectio

Preparation of PLGA-Coated Porous Silica Nanofibers for Drug Release.

Fibrous materials have unique applications in drug release and biomedical fields. This study reports on the preparation of porous silica nanofibers, using organic nanofibers as templates, and their use for drug release. Different from the commonly used electrospinning method, the organic nanofibers are produced via a self-assembly approach between melamine and benzene-1,3,5-tricarboxylic acid. Silica is then coated on the organic nanofibers via homogenization in a silica sol, a freeze-drying process, and then a sol-gel process. In order to regulate the surface area and mesopore volume of silic

Preparation of poloxamer-based nanofibers for enhanced dissolution of carvedilol.

Polymer nanofibers have become increasingly important for improvement of dissolution and bioavailability of poorly soluble drugs, representing a great challenge in pharmaceutical development. Here, we introduced a new concept of using amphiphilic polymers as fundamental excipients in electrospun nanofibers, which would improve drug solubilization and accelerate its release. Hydrophilic poloxamer-based nanofibers were developed as a novel drug delivery system for carvedilol. These nanofibers were electrospun from different liquid carvedilol dispersions, as carvedilol (nano)suspensions or ethano

Angiogenic peptide nanofibers repair cardiac tissue defect after myocardial infarction.

UNLABELLED: Myocardial infarction remains one of the top leading causes of death in the world and the damage sustained in the heart eventually develops into heart failure. Limited conventional treatment options due to the inability of the myocardium to regenerate after injury and shortage of organ donors require the development of alternative therapies to repair the damaged myocardium. Current efforts in repairing damage after myocardial infarction concentrates on using biologically derived molecules such as growth factors or stem cells, which carry risks of serious side effects including the

In vitro cardiomyocyte-driven biogenerator based on aligned piezoelectric nanofibers.

Capturing the body's mechanical energy from the heart, lungs, and diaphragm can probably meet the requirements for in vivo applications of implantable biomedical devices. In this work, we present a novel contractile cardiomyocyte (CM)-driven biogenerator based on piezoelectric nanofibers (NFs) uniaxially aligned on a PDMS thin film. Flexible nanostructures interact with the CMs, as a physical cue to guide the CMs to align in a specific way, and create mechanical interfaces of contractile CMs and piezoelectric NFs. As such, the cellular construct features specific alignment and synchronous cont

A bioengineered peripheral nerve construct using aligned peptide amphiphile nanofibers.

Peripheral nerve injuries can result in lifelong disability. Primary coaptation is the treatment of choice when the gap between transected nerve ends is short. Long nerve gaps seen in more complex injuries often require autologous nerve grafts or nerve conduits implemented into the repair. Nerve grafts, however, cause morbidity and functional loss at donor sites, which are limited in number. Nerve conduits, in turn, lack an internal scaffold to support and guide axonal regeneration, resulting in decreased efficacy over longer nerve gap lengths. By comparison, peptide amphiphiles (PAs) are mole
