Exploring the systemic delivery of a poorly water-soluble model drug to the retina using PLGA nanoparticles.

During the drug development process, many pharmacologically active compounds are discarded because of poor water solubility, but nanoparticle-based formulations are increasingly proposed as a solution for this problem. We therefore studied the distribution of nanoparticulate carriers and the delivery of their poorly water-soluble cargo to a structure of the central nervous system, the retina, under naive and pathological conditions. The lipophilic fluorescent dye coumarin 6 (Cou6) was encapsulated into poly(lactic-co-glycolic acid) PLGA nanoparticles (NPs). After intravenous administration in

Design of 3-aminophenol-grafted polymer-modified zinc sulphide nanoparticles as drug delivery system.

Zinc sulphide (ZnS) nanoparticles were synthesized by the coprecipitation method. The ZnS nanoparticle surface was polymerized with allyl glycidyl ether (AGE), and 3-aminophenol was then deposited as a ligand on nanosorbent. The modified nanosorbent was investigated with Fourier transform infrared spectroscopy and thermogravimetric analysis. The particle size of the modified nanosorbent was studied with scanning electron microscopy. Some characteristic factors of the adsorption process such as pH and time were investigated for famotidine using the modified nanosorbent. The equilibrium adsorpti

Recent advances and challenges of repurposing nanoparticle-based drug delivery systems to enhance cancer immunotherapy.

Cancer immunotherapy is an attractive treatment option under clinical settings. However, the major challenges of immunotherapy include limited patient response, limited tumor specificity, immune-related adverse events, and immunosuppressive tumor microenvironment. Therefore, nanoparticle (NP)-based drug delivery has been used to not only increase the efficacy of immunotherapeutic agents, but it also significantly reduces the toxicity. In particular, NP-based drug delivery systems alter the pharmacokinetic (PK) profile of encapsulated or conjugated immunotherapeutic agents to targeted cancer ce

How Nanoparticle Physicochemical Parameters Affect Drug Delivery to Cells in the Retina via Systemic Interactions.

Minor changes in the composition of poloxamer 188-modified, DEAE-dextran-stabilized (PDD) polybutylcyanoacrylate (PBCA) nanoparticles (NPs), by altering the physicochemical parameters (such as size or surface charge), can substantially influence their delivery kinetics across the blood-retina barrier (BRB) in vivo. We now investigated the physicochemical mechanisms underlying these different behaviors of NP variations at biological barriers and their influence on the cellular and body distribution. Retinal whole mounts from rats injected in vivo with fluorescent PBCA NPs were processed for ret

The mucoadhesive and gastroretentive properties of hydrophobin-coated porous silicon nanoparticle oral drug delivery systems.

Impediments to intestinal absorption, such as poor solubility and instability in the variable conditions of the gastrointestinal (GI) tract plague many of the current drugs restricting their oral bioavailability. Particulate drug delivery systems hold great promise in solving these problems, but their effectiveness might be limited by their often rapid transit through the GI tract. Here we describe a bioadhesive oral drug delivery system based on thermally-hydrocarbonized porous silicon (THCPSi) functionalized with a self-assembled amphiphilic protein coating consisting of a class II hydrophob

Potential Applications of Natural Components of Traditional Chinese Medicine Delivery via Nanoparticle Drug Delivery Systems in the Treatment of Alzheimer's Disease.

Alzheimer's disease (AD), a primary neurodegenerative disorder, is characterized by amyloid-β plaques and tau hyperphosphorylation-induced neurofibrillary tangles. Current treatments only alleviate symptoms, and Aβ monoclonal antibodies raise safety concerns in clinical use. Natural components (NCs) of Traditional Chinese Medicine (TCM) (eg, curcumin, quercetin, berberine, resveratrol) exhibit multi-target neuroprotective effects in AD, but poor blood-brain barrier (BBB) penetration and low bioavailability limit clinical use. Recent strategies to enhance TCM delivery include NP-based nanoparti

Solid lipid nanoparticle-based dissolving microneedles: A promising intradermal lymph targeting drug delivery system with potential for enhanced treatment of lymphatic filariasis.

Источник пометил публикацию как отозванную

Conventional oral therapy of lymphatic filariasis drugs is only effective to kill microfilariae in the bloodstream, but is often ineffective to kill adult filarial (macrofilariae) in the complex anatomy of the lymphatic system. The encapsulation of drugs into lipid-based nanoparticles with sizes of <100nm, and administration intradermally, could be used to enhance lymphatic uptake. Therefore, we developed an innovative approach, using solid lipid nanoparticles (SLNs) and dissolving microneedles (MNs) to deliver antifilariasis drugs, namely doxycycline, diethylcarbamazine and albendazole, intra

Nanoparticles as Drug Delivery Systems in Intravesical Therapy for Non-muscle Invasive Bladder Cancer: A Narrative Review.

Non-muscle invasive bladder cancer (NMIBC) accounts for the majority of newly diagnosed bladder cancer (BC) cases and is known for its high recurrence rate after initial treatment. The current standard of care includes the transurethral resection of bladder tumor (TURBT), followed by adjuvant intravesical therapy. However, therapeutic outcomes of intravesical therapies are often limited by the physiological characteristics of the urinary bladder, including continuous urine production and voiding, as well as the presence of the bladder permeability barrier (BPB), which reduces drug retention ti

Controlled release and antibioticeffect of vancomycin-loaded poly(N-isopropylacrylamide) nanoparticles as a smart drug delivery system.

Numerous studies have addressed the use of vancomycin (VA) to effectively treat bacterial infections. However, VA is known to cause side effects when administered intravenously. Herein, monodisperse poly(N-isopropylacrylamide) (PNIPAAm) hollow nanocapsules were synthesized at the interface of a water-in-oil (W/O) single emulsion via Shirasu porous glass (SPG) membrane emulsification and UV-initiated polymerization. In water solutions, the PNIPAAm nanocapsules were able to encapsulate VA and form a new nanoscale water-soluble drug delivery system, namely, PNIPAAm-VA. In vitro experiments showed

Polyglycerol-Based Lipids: A Next-Generation Alternative to PEG in Lipid Nanoparticles for Advanced Drug Delivery Systems.

Polyethylene glycol (PEG) is widely used to provide a stealth effect in various therapeutic applications. However, an increased occurrence of anti-PEG antibodies in patients can lead to immunological side effects and accelerated blood clearance, reducing the efficiency of PEG-based delivery vectors. One of these PEG-containing vectors is lipid nanoparticles (LNPs), which effectively deliver nucleic acids. Their high potential for treating a great number of diseases in the future was already showcased in the mRNA vaccines developed during the COVID-19 pandemic. Thus, the aim of our study was to
