Astrocytic Regulation of aberrant perineuronal net formation in Mecp2 -null Neocortex.

Rett syndrome (RTT), caused by mutations in MECP2 , is a complex neurological disorder characterized by myriad physiological disruptions, including early closure of the critical period of developmental plasticity and precocious formation of perineuronal nets (PNNs). PNNs are lattice-like substructures of extracellular matrix (ECM) that enwrap specific subpopulations of neurons. PNNs are essential in the modulation of neuronal plasticity and brain maturation, and their enzymatic disruption can partially restore plasticity in adults and improve memory. Although precocious PNN formation is well-e

Behavioral decomposition reveals rich encoding structure employed across neocortex in rats.

The cortical population code is pervaded by activity patterns evoked by movement, but it remains largely unknown how such signals relate to natural behavior or how they might support processing in sensory cortices where they have been observed. To address this we compared high-density neural recordings across four cortical regions (visual, auditory, somatosensory, motor) in relation to sensory modulation, posture, movement, and ethograms of freely foraging male rats. Momentary actions, such as rearing or turning, were represented ubiquitously and could be decoded from all sampled structures. H

Resonance properties of GABAergic interneurons in immature GAD67-GFP mouse neocortex.

Subthreshold resonance is a characteristic membrane property of different neuronal classes, is critically involved in the generation of network oscillations, and tunes the integration of synaptic inputs to particular frequency ranges. In order to investigate whether neocortical GABAergic interneurons show resonant behavior already during early postnatal development, we performed whole-cell patch-clamp recordings from visually identified interneurons in supragranular layers of parietal regions in coronal neocortical slices from postnatal day (P) P6-P13 GAD67-GFP knock-in mice. Subthreshold reso

Modeling depression: social dominance-submission gene expression patterns in rat neocortex.

Gene expression profiles in the cortex of adult Long-Evans rats as a function of a stressful social loss and victory in inter-male fighting encounters were examined. This social dominance and subordination model has been postulated to simulate early changes in the onset of depression in the losers. Microarrays were fabricated containing 45mer oligonucleotides spotted in quadruplicate and representing 1178 brain-associated genes. Dynamic range, discrimination power, accuracy and reproducibility were determined with standard mRNA "spiking" studies. Gene expression profiles in dominant and subord

Abnormalities in neuronal maturation in Rett syndrome neocortex: preliminary molecular correlates.

In correspondence with the severe cognitive impairment and autistic features of Rett syndrome (RS), multiple anomalies of the cerebral cortex that include generalized reductions in dendritic arborizations and in cholinergic markers have been found. Considering the potential role of neurotransmitters in cortical differentiation, we have studied the relationship between cholinergic deficit and dendritic protein expression in RS and in a relevant animal model. Dendritic development is characterized by the sequential expression of cytoskeletal proteins whose levels remain relatively stable in adul

Pharmacological reduction of electrophysiological diaschisis after photothrombotic ischemia in rat neocortex.

Focal cerebral lesions in the rat brain induced by photothrombosis cause hyperexcitability of the surrounding brain. This can be demonstrated in brain slices taken from animals several days after lesioning, by analysis of field potential responses to paired-pulse stimulation. We now investigated whether and how these remote effects of a cortical lesion can be modified pharmacologically. Application of the NMDA receptor antagonist, MK-801 ((+)-5-methyl-10, 11-dihydro-5H-dibnzo[a,d]cyclohepten-5,10-imine), was shown to block induction of immediate early genes and activation of astrocytes as evid

Ultrastructural changes in the mouse fetal neocortex following chronic maternal alcoholization.

Female mice (RAP strain) were alcoholized for 30-50 days before mating and during pregnancy until killing, with a 20% solution of ethanol administered instead of drinking water. From foetuses of 16, 18, 20 days and from newborn puppies on day 1 parietal neocortex fragments were excised and examined electronmicroscopically. Chronic maternal alcoholization induces in the neocortex of mouse foetuses and newborn puppies various ultrastructural changes: swelling of mitochondria with the disappearance of cristae and vacuolation, both in the capillary endothelium and in the cells of the neural tissue

Cortical hem-like progenitors contribute pyramidal neurons to the neocortex and hippocampus.

The cortical hem is a signaling center at the midline of the embryonic telencephalon that provides instructive cues to the adjacent cortical primordium and serves as the hippocampal organizer, inducing multiple distinct hippocampal field identities. The cortical hem consists of neuroepithelial progenitors, and gives rise to diverse neuronal and non-neuronal cell types, including Cajal-Retzius cells, hippocampal pyramidal neurons, and choroid plexus epithelial cells. Here, we use two independent Cre drivers, Wnt3aCre and Lmx1aCre, to examine the lineages from the cortical hem. We report that th

A Key Regulator of Dendritic Morphology in Supragranular Neocortex Impacts Mismatch Negativity.

BACKGROUND: Cytoskeletal structure and neuronal function are heavily intertwined, and altered pyramidal neuron morphology (e.g., reduced dendritic arbors) has been consistently identified in the neocortex of individuals with psychiatric disorders. A missense mutation in Kalrn (Kalrn-PT) enhances activation of RhoA, a cytoskeletal regulator, and leads to adolescent-onset dendritic regression in supragranular auditory cortex. To investigate the relationship between this spatiotemporally altered cytoskeletal structure and psychiatric-relevant dysfunction, we examine the functional impacts of this

Imbalanced neurogenesis and gliogenesis in the developing neocortex of mice lacking the proteoglycan Tsukushi.

Development of the mammalian neocortex relies on a tightly controlled schedule, wherein cortical progenitors initially expand by symmetric divisions, then sequentially generate neurons of the deep and superficial cortical layers by asymmetric divisions, and finally give rise to glial cell populations. This sequence of events is orchestrated by a complex network of transcriptional regulators and extracellular signals ensuring a proper balance between cortical expansion and the production of neurons and glia. Although the transcription factors governing progenitor self-renewal, neurogenesis and

Neocortex Formation in a Tapered Wedge Stem is Not Indicative of Complications or Worse Outcomes.

BACKGROUND: The formation of sclerotic bone, a neocortex, distally surrounding total hip arthroplasty (THA) stems may commonly be seen on radiographs around proximally porous coated stems but can be confused with loosening. The goal of this study was to determine the prevalence of the neocortex finding and whether it associated with worse outcomes after THA. METHODS: A retrospective review of 825 patients with a single tapered wedge stem was performed. Radiographs at 1-year, as well as final follow-up were reviewed for evidence of sclerotic bone (neocortex) surrounding the stem in all 14 Grue

A network of genetic repression and derepression specifies projection fates in the developing neocortex.

Neurons within each layer in the mammalian cortex have stereotypic projections. Four genes-Fezf2, Ctip2, Tbr1, and Satb2-regulate these projection identities. These genes also interact with each other, and it is unclear how these interactions shape the final projection identity. Here we show, by generating double mutants of Fezf2, Ctip2, and Satb2, that cortical neurons deploy a complex genetic switch that uses mutual repression to produce subcortical or callosal projections. We discovered that Tbr1, EphA4, and Unc5H3 are critical downstream targets of Satb2 in callosal fate specification. Thi

Area-specific resonance of excitatory networks in neocortex: control by outward currents.

During disinhibition or low [Mg++](o) buffer, 7-14 Hz ( approximately 10 Hz) oscillations are generated by excitatory networks of interconnected pyramidal cells in motor (agranular) cortex but are absent in barrel (granular) cortex. Here we studied if the inability of barrel cortex to produce approximately 10 Hz oscillations during these conditions is because barrel cortex networks lack the necessary cellular mechanisms or, alternatively, because those mechanisms are inhibited by outward currents. The results show that blockers of slowly inactivating voltage-dependent K+ currents unmask approx

Attraction exerted in vivo by grafts of embryonic neocortex on developing thalamic axons.

In a previous study we provided evidence that embryonic (E) day 16 frontal cortical cells grafted into the occipital cortex of newborn rats receive inputs from the ventrolateral (VL) and ventromedial (VM) thalamic nuclei which, normally, project to the frontal cortex (25). The present study was designed to examine further the conditions of development of the thalamic innervation of heterotopic neocortical grafts. We demonstrate that VL/VM axons do not provide transitory aberrant input to the occipital cortex either in intact newborn animals or in rats having received neonatal occipital lesion

Manipulation of neural precursors in situ: induction of neurogenesis in the neocortex of adult mice.

Over the past three decades, research exploring potential neuronal replacement therapies have focused on replacing lost neurons by transplanting cells or grafting tissue into diseased regions of the brain. Over most of the past century of modern neuroscience, it was thought that the adult brain was completely incapable of generating new neurons. However, in the last decade, the development of new techniques has resulted in an explosion of new research showing that neurogenesis, the birth of new neurons, normally occurs in two limited and specific regions of the adult mammalian brain, and that

Functional consequences of embryonic neocortex transplanted to rats with prefrontal cortex lesions.

In four experiments we reexamined the recent report by Labbe, Firl, Mufson, and Stein (1983) that fetal cortical tissue transplanted to an aspirative prefrontal cortical cavity in rats can ameliorate the learning impairments induced by the aspirative lesions. Healthy surviving grafts from young (E16) embryonic donors had no immediate effects on the rats' impairments in T-maze alternation, spatial navigation in the Morris swimming pool task, or locomotor activity, and they produced even greater impairments than the lesions alone when all three tests were conducted after longer (3-5 month) survi

GABAergic circuits reflect different requirements for sensory integration in postnatal mouse neocortex.

Information transfer in mammalian cerebral cortex is dependent on GABAergic interneuron circuits that are widely assumed to be uniform across the neocortex. Here, we demonstrate that this does not hold true during postnatal life prior to the onset of active sensory exploration in mice. During this time, somatostatin interneurons differentially contribute to sensory-evoked activity in primary somatosensory (S1BF) and visual (V1) cortices. In S1BF, somatostatin interneurons provide a mechanism for feedforward control of sensory responses as opposed to the feedback role observed in V1. This funct

Celf4 controls mRNA translation underlying synaptic development in the prenatal mammalian neocortex.

Abnormalities in neocortical and synaptic development are linked to neurodevelopmental disorders. However, the molecular and cellular mechanisms governing initial synapse formation in the prenatal neocortex remain poorly understood. Using polysome profiling coupled with snRNAseq on human cortical samples at various fetal phases, we identify human mRNAs, including those encoding synaptic proteins, with finely controlled translation in distinct cell populations of developing frontal neocortices. Examination of murine and human neocortex reveals that the RNA binding protein and translational regu
