A lobster cysteine protease with immunoreactivity and activities of calcitonin and CGRP.

The high concentrations of molecules immunologically related to salmon calcitonin (CT) and/or to human calcitonin gene-related peptide (CGRP) in the oesophagus of the norway lobster Nephrops norvegicus have been examined. In the present study. We report the purification of these molecules by means of a specific radioimmunoassay for calcitonin and calcitonin gene related peptide. The immunoreactive molecules were tested for their functional similarities with CT and CGRP. This was investigated by measuring their ability to interact with CGRP and CT radioreceptor assays and to stimulate the adeny

Differential modulation of chemical and electrical components of mixed synapses in the lobster stomatogastric ganglion.

1. Two pairs of neurons in the pyloric network of the spiny lobster, Panulirus interruptus, communicate through mixed graded chemical and rectifying electrical synapses. The anterior burster (AB) chemically inhibits and is electrically coupled to the ventricular dilator (VD); the lateral pyloric (LP) and pyloric (PY) neurons show reciprocal chemical inhibition and electrical coupling. We examined the effects of dopamine (DA), serotonin (5HT) and octopamine (Oct) on these mixed synapses to determine the plasticity possible with opposing modes of synaptic interaction. 2. Dopamine increased net i

Zinc ion enhances the blocking potency of synthetic analogs of spider toxin (JSTX) on the glutamate receptor.

The effect of synthetic spider toxin analogs containing aza-crown in combination with zinc and copper was studied on the lobster neuromuscular synapse. The suppressive action of N4-type spider toxin analog (N4) on excitatory postsynaptic potentials (EPSPs) was markedly enhanced in the presence of 10(-5)-10(-4) M Zn2+. Cu2+ (10(-4) M) also had a potentiating effect on the suppression of EPSPs by N4 but to a lesser degree than Zn2+. While N6-type spider toxin analog (N6) suppressed EPSPs more effectively than N4, additional suppression was not pronounced in the presence of Zn2+ or Cu2+. The resu

Effect of a spider toxin (JSTX) on excitatory postsynaptic current at neuromuscular synapse of spiny lobster.

1. We studied the blocking properties of a spider (Nephila clavata) toxin (JSTX) purified from venom on the spiny lobster neuromuscular junction. 2. When a small amount of JSTX was applied to the neuromuscular junction, the excitatory postsynaptic potential (EPSP) was partially suppressed. The amplitude of EPSPs remained at a steady level for several hours during the washing of the preparation, showing that the action of JSTX is irreversible. 3. We recorded the excitatory postsynaptic current (EPSC) from synaptic site using a macro-patch electrode. The amplitude of EPSC increased linearly with

All-or-none control of the bursting properties of the pacemaker neurons of the lobster pyloric pattern generator.

In Crustacea the central pattern generator for the pyloric motor rhythm (filtration to the midgut) is known to be located within the stomatogastric ganglion (STG); its cycling activity is known to be organized by three endogenous burster neurons acting as pacemakers and driving 11 follower neurons. In Homarus, recordings from the isolated stomatogastric nervous system (Fig. 1) indicate that (1) the pyloric output can be generated only when the STG is afferented (i.e., connected to the more rostral oesophageal and commissural ganglia) (Fig. 2) and (2) the deafferentation of the STG results in a

Localization of horseradish peroxidase-alpha-bungarotoxin binding in crustacean axonal membrane vesicles and intact axons.

A conjugate of alpha-bungarotoxin with horseradish peroxidase was used to visualize alpha-bungarotoxin binding sites at the fine structural level in isolated axonal membrane vesicles from lobster walking leg nerve. These plasma membrane vesicles have previously been shown to exhibit saturable binding of [3H]nicotine and [3H]acetylcholine. Binding of the toxin was identified in the axon plasma membrane and could be blocked by pretreatment with excess free alpha-bungaratoxin or d-tubocurarine. Binding sites for alpha-bungarotoxin were identified by the same technique in sections of intact nerve

Acute and long-term manganese exposure and subsequent accumulation in relation to idiopathic blindness in the American lobster, Homarus americanus.

Manganese (Mn) is a hypoxic reactive metal commonly found in marine sediments. Under hypoxic conditions the metal becomes fully reduced to Mn2+ and is biologically available to the benthic community for uptake. Mn is also a potent neurotoxin and it may play a role in the etiology of idiopathic blindness that has been observed in American lobsters. An acute exposure study was designed to expose American lobster, Homarus americanus, to 0, 20, 80, 150, and 300 mg L-1 (ppm) for 96 hs to explore disparities in Mn accumulation among several tissues: optic nerve, brain, hepatopancreas, muscle, hemoly

Ultrasonic extraction followed by sonolysis-ozonolysis as a sample pretreatment method for determination of reactive arsenic toward sodium tetrahydroborate by flow injection-hydride generation AAS.

A new sample pretreatment method based on ultrasonic extraction in HCI medium and subsequent oxidation of the extracts by sonozone (i.e., sonolysis-ozonolysis) has been developed for determination of reactive arsenic toward sodium tetrahydroborate [mainly As(III) + As(V)] by flow injection-hydride generation atomic absorption spectrometry. This method avoids the use of intensive treatments with concentrated and corrosive acids, high pressures, and temperatures that are inherent with traditional wet or dry ashing procedures and entails reduced waste production and reagent consumption. Presynaptic afferent inhibition of lobster olfactory receptor cells: reduced action-potential propagation into axon terminals.

Action-potential propagation into the axon terminals of olfactory receptor cells was measured with the use of voltage-sensitive dye imaging in the isolated spiny lobster brain. Conditioning shocks to the olfactory nerve, known to cause long-lasting suppression of olfactory lobe neurons, allowed the selective imaging of activity in receptor cell axon terminals. In normal saline the optical signal from axon terminals evoked by a test stimulus was brief (40 ms) and small in amplitude. In the presence of low-Ca2+/high-Mg2+ saline designed to reduce synaptic transmission, the test response was unch

Suppressive control of the crustacean pyloric network by a pair of identified interneurons. II. Modulation of neuronal properties.

In the lobster Homarus, the 2 identified PS neurons have a strong suppressive modulatory effect on the activity of the pyloric network in the STG (Cazalets et al., 1990). In the present paper, we consider the effects of PS on individual pyloric neurons isolated from their partners in the network by cell photoinactivation and synaptic blockade. Three types of PS action are described: (1) a transient, EPSP-mediated depolarization of the PD, VD, and AB neurons; (2) a long-lasting hyperpolarization concomitant with a loss of oscillatory properties in the PD and LP neurons; (3) a long-lasting depol

Targets of octopamine action in the lobster: cyclic nucleotide changes and physiological effects in hemolymph, heart and exoskeletal muscle.

We have examined three potential targets of octopamine action of the lobster--hemolymph, heart and exoskeletal muscle--and found that each tissue responds to the amine with increases in cyclic adenosine 3': 5'-monophosphate (cAMP) and changes in measurable physiological phenomena. A highly specific octopamine-stimulated increase in cAMP is observed in hemolymph, and this effect is probably due to the presence of a unique octopamine receptor on hematocytes. Pharmacological studies show that synephrine is a potent agonist of the hemolymph receptor for octopamine and that the alpha adrenergic rec

Direct determination of methylmercury and inorganic mercury in biological materials by solid sampling-electrothermal vaporization-inductively coupled plasma-isotope dilution-mass spectrometry.

This paper reports on the use of solid sampling-electrothermal vaporization-inductively coupled plasma mass spectrometry (SS-EIV-ICPMS) for the direct and simultaneous determination of methylmercury and inorganic mercury in biological materials. The main advantage of this fast and sensitive method is that no sample preparation is required. In this way, the sample throughput can be considerably increased, problems of contamination and analyte losses are kept to a minimum and, even more important, the original chemical form of the different analyte species in the solid samples is preserved. Organomercury determination in biological reference materials: application to a study on mercury speciation in marine mammals off the Faröe Islands.

The potential use of graphite furnace atomic absorption spectrometry (GF-AAS) for the organic mercury determination in marine biological tissues was evaluated. Following its isolation by acid extraction in toluene, organic mercury was recovered in aqueous thiosulfate and measured by GF-AAS. The detection limit was 0.01 microgram Hg/g (as methyl mercury). Analyses were conducted on three reference standard materials certified for their methyl mercury content, DOLT-1, DORM-1, and TORT-1, provided by the National Research Council of Canada. The method resulted in very good recovery and reproducib

Mechanisms of gastric rhythm generation in the isolated stomatogastric ganglion of spiny lobsters: bursting pacemaker potentials, synaptic interactions, and muscarinic modulation.

1. The gastric central pattern generator (CPG), located in the stomatogastric ganglion (STG) of the spiny lobster (Panulirus interruptus), is nonrhythmic when deprived of neuromodulatory inputs from anterior ganglia. Leaving these inputs intact in vitro can sustain a gastric rhythm but also introduces numerous, uncontrolled and largely unknown modulatory and synaptic influences that greatly complicate analysis of this CPG. 2. Here we induced gastric rhythms in the isolated STG, by superfusing a specific modulator, the muscarinic agonist, pilocarpine. Muscarinic agents sustain vigorous gastric

[Red soft coral-induced allergic symptoms observed in spiny lobster fishermen].

Among spiny lobster fishermen on the Pacific coast of Miyazaki Prefecture, the authors found two cases with bronchial asthma due to occupational sensitization with red soft corals (Dendronephthytia nipponica). In this report are presented these two cases, and the results of further investigation from allergic viewpoint on 72 subjects of the same occupation. Among these fishermen, such symptoms as sneezing, nasal discharge, cough, asthmatic attacks, conjunctival hyperamia, and dermatitis on removal of catches (red soft corals) from a dragnet were seen during spiny lobster fishing season, from S

Electrically coupled pacemaker neurons respond differently to same physiological inputs and neurotransmitters.

The two pyloric dilator (PD) motor neurons and the single anterior burster (AB) interneuron are electrically coupled and together comprise the pacemaker for the pyloric central pattern generator of the stomatogastric ganglion of the lobster, Panulirus interruptus. Previous work (31) has shown that the AB neuron is an endogenously bursting neuron, while the PD neuron is a conditional burster. In this paper the effects of physiological inputs and neurotransmitters on isolated PD neurons and AB neurons were studied using the lucifer yellow photoinactivation technique (33). Pattern generation in the lobster (Panulirus) stomatogastric ganglion. I. Pyloric neuron kinetics and synaptic interactions.

There are a number of perspectives gained from a quantitative analysis of the pyloric system which may be applicable to other simple pattern generators: 1. The system is organized around a dominant, endogenously-bursting neuron group, and its properties are tailored to that dominance. In particular, synaptic strengths and firing frequencies of that group appear just sufficient to suppress postsynaptic "follower" cells if the latter are not too highly excited. 2. Repetitive firing properties of follower neurons are such as to facilitate their switch-like mode of activity. This includes pacemake

Selective allergy to lobster in a case of primary sensitization to house dust mites.

Allergy to only 1 kind of seafood is uncommon. We report a case of selective allergy to lobster. We studied a 30-year-old man who suffered generalized urticaria, facial erythema, and pharyngeal pruritus after eating lobster. He had a more than 10-year history of mild persistent asthma and sensitization to house dust mites. The study was performed by skin prick test, and prick-prick test, oral food challenge, specific immunoglobulin (Ig) E determinations by CAP (Phadia, Uppsala, Sweden) and ADVIA-Centaur (ALK-Abelló, Madrid, Spain), and IgE-immunoblotting. The patient's serum recognized 2 aller

Monoamine control of the pacemaker kernel and cycle frequency in the lobster pyloric network.

The monoamines dopamine (DA), serotonin (5HT), and octopamine (Oct) can each sculpt a unique motor pattern from the pyloric network in the stomatogastric ganglion (STG) of the spiny lobster Panulirus interruptus. In this paper we investigate the contribution of individual network components in determining the specific amine-induced cycle frequency. We used photoinactivation of identified neurons and pharmacological blockade of synapses to isolate the anterior burster (AB) and pyloric dilator (PD) neurons. Bath application of DA, 5HT, or Oct enhanced cycle frequency in an isolated AB neuron, wi

Alpha-pompilidotoxin (alpha-PMTX), a novel neurotoxin from the venom of a solitary wasp, facilitates transmission in the crustacean neuromuscular synapse.

A new neurotoxin, named alpha-pompilidotoxin (alpha-PMTX) has been found in the venom of the solitary wasp Anoplius safnariensis. In the neuromuscular synapse of the lobster walking leg preparation, alpha-PMTX (10-100 micro/M) caused great enhancement of both the excitatory and inhibitory postsynaptic potentials. Recordings of the excitatory post synaptic currents (EPSCs) at the synaptic sites showed that alpha-PMTX reversibly and dose-dependently potentiates EPSCs. Alpha-PMTX may act primarily on the presynaptic membrane but the mode of action of the toxin is clearly different from other know
