Antidepressant-like effects of fish, krill oils and Vit B12 against exposure to stress environment in mice models: current status and pilot study.

Oxidative stress has significant role in pathophysiology of any kind of depression through actions of free radicals, non-radical molecules, and unbalancing antioxidant systems in body. In the current study, antidepressant responses of fish oil (FO), Neptune krill oil (NKO), vitamin B12 (Vit B12), and also imipramine (IMP) as the reference were studied. Natural light was employed to induce stress in the animals followed by oral administration of the drugs for 14 days. The antidepressant effect was assessed by tail suspension test (TST) and forced swimming test (FST), antioxidant enzymes and oxi

Coreless exoplanets induced by metal-silicate miscibility.

The rocky interiors of exoplanets are generally expected to differentiate into a silicate mantle and a metallic core due to the immiscibility of metal and silicate under typical planetary formation conditions. Here, we use large-scale molecular dynamics simulations driven by machine-learning potentials of ab initio quality to explore the phase behavior of metallic liquids and silicate melts under the extreme pressure-temperature conditions of the exoplanetary interiors. The results map out the solvus closure curve, above which a single homogeneous liquid phase of the iron and silicate mixture

Identification of dicarbonyl and L-xylulose reductase as a therapeutic target in human chronic kidney disease.

An imbalance of nephroprotective factors and renal damaging molecules contributes to development and progression of chronic kidney disease (CKD). We investigated associations of renoprotective factor gene expression patterns with CKD severity and outcome. Gene expression profiles of 197 previously reported renoprotective factors were analyzed in a discovery cohort in renal biopsies of 63 CKD patients. Downregulation of dicarbonyl and L-xylulose reductase (DCXR) showed the strongest association with disease progression. This significant association was validated in an independent set of 225 pat

A reassessment of soluble urokinase-type plasminogen activator receptor in glomerular disease.

It has been suggested that soluble urokinase receptor (suPAR) is a causative circulating factor for and a biomarker of focal and segmental glomerulosclerosis (FSGS). Here we undertook validation of these assumptions in both mouse and human models. Injection of recombinant suPAR in wild-type mice did not induce proteinuria within 24 h. Moreover, a disease phenotype was not seen in an inducible transgenic mouse model that maintained elevated suPAR concentrations for 6 weeks. Plasma and urine suPAR concentrations were evaluated as clinical biomarkers in 241 patients with glomerular disease from t

Single cell transcriptomics identifies focal segmental glomerulosclerosis remission endothelial biomarker.

To define cellular mechanisms underlying kidney function and failure, the KPMP analyzes biopsy tissue in a multicenter research network to build cell-level process maps of the kidney. This study aimed to establish a single cell RNA sequencing strategy to use cell-level transcriptional profiles from kidney biopsies in KPMP to define molecular subtypes in glomerular diseases. Using multiple sources of adult human kidney reference tissue samples, 22,268 single cell profiles passed KPMP quality control parameters. Unbiased clustering resulted in 31 distinct cell clusters that were linked to kidney

Global glomerulosclerosis with nephrotic syndrome; the clinical importance of age adjustment.

Globally sclerotic glomeruli (GSG) occur with both normal aging and kidney disease. However, it is unknown whether any GSG or only GSG exceeding that expected for age is clinically important. To evaluate this, we identified patients with a glomerulopathy that often presents with nephrotic syndrome (focal segmental glomerulosclerosis, membranous nephropathy, or minimal change disease) in the setting of the Nephrotic Syndrome Study Network (NEPTUNE), China-Digital Kidney Pathology (DiKiP), and the Southeast Minnesota cohorts. Age-based thresholds (95th percentile) for GSG based on normotensive l

Angiotensin II up-regulates sodium-glucose co-transporter 2 expression and SGLT2 inhibitor attenuates Ang II-induced hypertensive renal injury in mice.

Clinical trials indicate that sodium/glucose co-transporter 2 (SGLT2) inhibitors (SGLT2i) improve kidney function, yet, the molecular regulation of SGLT2 expression is incompletely understood. Here, we investigated the role of the intrarenal renin-angiotensin system (RAS) on SGLT2 expression. In adult non-diabetic participants in the Nephrotic Syndrome Study Network (NEPTUNE, n=163), multivariable linear regression analysis showed SGLT2 mRNA was significantly associated with angiotensinogen (AGT), renin, and angiotensin-converting enzyme (ACE) mRNA levels (P<0.001). In vitro, angiotensin II (A

Using PROMIS® to create clinically meaningful profiles of nephrotic syndrome patients.

OBJECTIVE: Nephrotic syndrome (NS) is a kidney disease known to adversely impact health-related quality of life (HRQOL). Patient-reported outcome (PRO) measures are commonly used to characterize HRQOL and the patient disease experience. This study aims to improve the interpretability and clinical utility of the Patient-Reported Outcomes Measurement Information System® (PROMIS®) by identifying distinct meaningful HRQOL profiles in children and adults with NS. METHOD: Patients were from 2 prospective NS cohort studies (PROMIS-II®: 121 children; NEPTUNE: 40 children and 219 adults) with data fro

The APOL1 p.N264K variant is co-inherited with the G2 kidney disease risk variant through a proximity recombination event.

Black Americans are 3-4 times more likely to develop nondiabetic kidney disease than other populations. Exclusively found in people of recent African (AFR) ancestry, risk variants in Apolipoprotein L1 (APOL1) termed G1 and G2 contribute significantly to this increased susceptibility. Our group and others showed that a missense variant in APOL1, rs73885316 (p.N264K, "M1"), is remarkably protective against APOL1 kidney disease when co-inherited with the G2 risk allele. Since the distance between the M1 and G2 variants is only 367 base pairs, we initially suspected that 2 independent mutation eve

A giant planet transiting a 3-Myr protostar with a misaligned disk.

Astronomers have found more than a dozen planets transiting stars that are 10-40 million years old1, but younger transiting planets have remained elusive. The lack of such discoveries may be because planets have not fully formed at this age or because our view is blocked by the protoplanetary disk. However, we now know that many outer disks are warped or broken2; provided the inner disk is depleted, transiting planets may thus be visible. Here we report observations of the transiting planet IRAS 04125+2902 b orbiting a 3-million-year-old, 0.7-solar-mass, pre-main-sequence star in the Taurus Mo

Follistatin-Like-1 (FSTL1) Is a Fibroblast-Derived Growth Factor That Contributes to Progression of Chronic Kidney Disease.

Our understanding of the mechanisms responsible for the progression of chronic kidney disease (CKD) is incomplete. Microarray analysis of kidneys at 4 and 7 weeks of age in Col4a3-/- mice, a model of progressive nephropathy characterized by proteinuria, interstitial fibrosis, and inflammation, revealed that Follistatin-like-1 (Fstl1) was one of only four genes significantly overexpressed at 4 weeks of age. mRNA levels for the Fstl1 receptors, Tlr4 and Dip2a, increased in both Col4a-/- mice and mice subjected to unilateral ureteral obstruction (UUO). Hydroxypropyl-β-cyclodextrin protects from kidney disease in experimental Alport syndrome and focal segmental glomerulosclerosis.

Studies suggest that altered renal lipid metabolism plays a role in the pathogenesis of diabetic kidney disease and that genetic or pharmacological induction of cholesterol efflux protects from the development of diabetic kidney disease and focal segmental glomerulosclerosis (FSGS). Here we tested whether altered lipid metabolism contributes to renal failure in the Col4a3 knockout mouse model for Alport Syndrome. There was an eight-fold increase in the cholesterol content in renal cortexes of mice with Alport Syndrome. This was associated with increased glomerular lipid droplets and cholestero

Prediction of thermally driven quasi-1D superionic states in carbon hydride under giant planetary conditions.

Planetary interiors experience high-pressure-high temperature conditions that give rise to unconventional states of matter, reshaping our understanding of planetary dynamics and the generation of magnetic fields. Here, using first-principles computational simulations in combination with machine-learning interatomic potentials, we predict a distinct atomic state, termed a quasi-1D superionic phase, that emerges in a stable carbohydride (CH) compound under giant planetary interior conditions. This phase originates from temperature-induced transformations and features a chiral carbon framework in

Enterococcus exceedances related to environmental variability at New Jersey ocean beaches.

Microbial pollution at ocean beaches is a global public health problem that can be exacerbated by excessive rainfall, particularly at beaches adjacent to urban areas. Rain is acknowledged as a predictive factor of Enterococcus levels at NJ beaches, but to date no study has explicitly examined the link. Here, five beaches (156 observations) in Monmouth County, NJ, with storm drain outflows present were sampled for Enterococcus and water quality during dry and wet periods. Hypotheses included (1) beaches differ in Enterococcus levels, (2) Enterococcus is present year-round, and (3) Enterococcus

Evaluation of performance criteria for simulation of submaximal steady-state cycling using a forward dynamic model.

The objectives of this study were twofold. The first was to develop a forward dynamic model of cycling and an optimization framework to simulate pedaling during submaximal steady-state cycling conditions. The second was to use the model and framework to identify the kinetic, kinematic, and muscle timing quantities that should be included in a performance criterion to reproduce natural pedaling mechanics best during these pedaling conditions. To make this identification, kinetic and kinematic data were collected from 6 subjects who pedaled at 90 rpm and 225 W. Intersegmental joint moments were

Novel Scalar-on-matrix Regression for Unbalanced Feature Matrices.

Image features that characterize tubules from digitized kidney biopsies may offer insight into disease prognosis as novel biomarkers. For each subject, we can construct a matrix whose entries are a common set of image features (e.g., area, orientation, eccentricity) that are measured for each tubule from that subject's biopsy. Previous scalar-on-matrix regression approaches which can predict scalar outcomes using image feature matrices cannot handle varying numbers of tubules across subjects. We propose the CLUstering Structured laSSO (CLUSSO), a novel scalar-on-matrix regression technique tha

Longitudinal analysis of blood pressure and lipids in childhood nephrotic syndrome.

BACKGROUND: In the current study, longitudinal BP and lipid measurements were examined in a NEPTUNE cohort of children with newly diagnosed nephrotic syndrome (cNEPTUNE). We hypothesized that hypertensive BP and dyslipidemia would persist in children with nephrotic syndrome, regardless of steroid treatment response. METHODS: A multi-center longitudinal observational analysis of data obtained from children < 19 years of age with new onset nephrotic syndrome enrolled in the Nephrotic Syndrome Study Network (cNEPTUNE) was conducted. BP and lipid data were examined over time stratified by disease

External Validation of a Urinary Biomarker Risk Score for the Prediction of Steroid Responsiveness in Adults With Nephrotic Syndrome.

INTRODUCTION: In idiopathic nephrotic syndrome, response to corticosteroids remains the best indicator of prognosis. Noninvasive markers to predict a patient's response to steroids would allow improved prognostication and a more personalized approach to management. We have previously derived a urinary biomarker risk score which can differentiate steroid sensitive nephrotic syndrome (SSNS) from steroid resistant nephrotic syndrome (SRNS) in children. The goal of this study was to validate this previously derived biomarker risk score in a cohort of steroid-naïve adult patients, to determine whet
