High-voltage electrical stimulation improves nerve regeneration after sciatic crush injury.

BACKGROUND: Peripheral nerve injury causes prolonged functional limitation being a clinical challenge to identify resources that accelerates its recovery. OBJECTIVES: To investigate the effect of high-voltage electrical stimulation (HVES) on the morphometric and functional characteristics of the regenerated nerve after crush injury in rats. METHODS: Twenty Wistar rats were randomly allocated into 4 groups: Control (CON) - without injury and without HVES; Denervated (D) - sciatic nerve crush only; Denervated + HVES - sciatic nerve crush and HVES; SHAM - without injury but HVES. The HVES and S

Electrophysiological and theoretical analysis of melatonin in peripheral nerve crush injury.

Electrophysiological and theoretical studies have been performed to investigate peripheral nerve injuries and nerve regeneration. The aim of this work is to evaluate the effect of melatonin functionally and electrophysiologically on peripheral nerve crush injury. Adult male Wistar rats (n=32, 200+/-50 g) were randomly allocated into four groups. Sciatic crush was constituted on left sciatic nerves. Treatment groups received intraperitoneal melatonin at doses of 5 and 20mg/kg for 21 days. Functional nerve recovery was evaluated using sciatic functional index (SFI) every week during the experime

Effects of intrathecal administration of FK506 after sciatic nerve crush injury.

Although various administration routes of FK506 have been published, intrathecal administration of FK506 has not previously been reported in the literature. A daily dose of 0.05 mg/kg of FK506 was given (a small dose compared with those reported in the available literature). The authors used this small dose to obtain lower immunosuppression and neurotoxicity, and a higher axonal regeneration rate. A total number of 40 female Wistar rats were used and randomly divided into four groups: control, sham, FK506-treated, and vehicle-treated. Sciatic nerve regeneration was evaluated by walking track a

Nimodipine promotes regeneration and functional recovery after intracranial facial nerve crush.

The calcium flow inhibitor, nimodipine, has been shown to promote motor neuron survival in the facial nucleus after intracranial facial nerve transection. However, it has not been known whether the neuroprotective effects primarily involve survival of nerve cell bodies or outgrowth and/or myelination of nerve fibers. Here, we studied the effects of nimodipine in a different injury model in which the facial nerve was unilaterally crushed intracranially. This lesion caused complete anterograde degeneration and partial retrograde degeneration that were studied with a combination of several stereo

Flunarizine enhances functional recovery following sciatic nerve crush lesion in rats.

We have used the rat sciatic nerve crush (SNC) injury model to assess the neuroprotective effects of flunarizine (FNZ), a calcium channel antagonist and a vasodilator. The animals were treated with FNZ for various durations following SNC (0.33 mg/kg per day, i.p). Employing the physical disector method, we quantitated the rates of neuron loss in the dorsal root ganglion and spinal cord and protective effects of FNZ. FNZ treatment following SNC reduced neuron loss up to 86.6 and 82.5% in DRG sensory and spinal cord motor neurons, respectively. Functional recovery following SNC with or without F

Tubulization increases axonal outgrowth of rat sciatic nerve after crush injury.

It is important to develop methods which increase nerve regeneration since restoration of function following injury to peripheral nerves often requires outgrowth of the injured axons over long distances. In this study, axonal outgrowth after bilateral crush injury to the sciatic nerve of the rat was measured. One group with large-diameter nonpermeable silicone tubes and one group with large diameter permeable silicone tubes applied around the crush site on one side had regeneration following nerve injury compared to controls on the other side. The length of regeneration of the regenerating axo

Mechanoreceptors in rat glabrous skin: redevelopment of function after nerve crush.

In the glabrous skin of the rat's hindfoot the same triple set of low-threshold mechanoreceptors is present as has been found in other mammals: slowly adapting (SA), rapidly adapting (RA), and very rapidly adapting Pacinian corpuscle-like (PC) receptors. Their functional characteristics were examined in normal rats and compared with those of sensitive mechanoreceptors found in the glabrous skin of the foot 2-24 wk after crush of the plantar nerves, resulting in regeneration of the transected nerve fibers. After 2 wk of nerve regeneration, low-threshold RA and SA cutaneous mechanoreceptors reap

Surgical Methods in Postmetamorphic Xenopus laevis: Optic Nerve Crush Injury Model.

Many human optic neuropathies lead to crippling conditions resulting in partial or complete loss of vision. While the retina is made up of several different cell types, retinal ganglion cells (RGCs) are the only cell type connecting the eye to the brain. Optic nerve crush injuries, wherein RGC axons are damaged without severing the optic nerve sheath, can serve as a model for traumatic optical neuropathies as well as some progressive neuropathies such as glaucoma. In this chapter, we describe two different surgical methods for establishing an optic nerve crush (ONC) injury in the postmetamorph

Analysis of Visual Recovery After Optic Nerve Crush in Adult Zebrafish.

Zebrafish can successfully regenerate axons after optic nerve crush (ONC). Here, we describe two different behavioral tests to map visual recovery: the dorsal light reflex (DLR) test and the optokinetic response (OKR) test. The DLR is based on the tendency of fish to orient their back to a light source, and it can be tested by rotating a flashlight around the dorsolateral axis of the animal or by measuring the angle between the left/right body axis and the horizon. The OKR, in contrast, consists of reflexive eye movements triggered by motion in the visual field of the subject and is measured b

Metabolomics dataset of mouse optogenetic axon regeneration after optic nerve crush.

This metabolite dataset was collected from transgenic murine retinal ganglion cells (RGC) expressing bacterial channelrhodopsin labeled with fluorescent protein. Mice were subjected to optic nerve crush (ONC) with subsequent RGC stimulation of channelrhodopsin with blue light (promoting regeneration) or non-stimulation (control). ONC induces retinal ganglion cell degeneration over time with progressive loss of axons. In transgenic bacterial channelrhodopsin expressing RGC cells, light stimulation promotes regeneration of ONC axons. Genetically matched wild-type uninjured optic nerves were anal

Hematogenous Macrophages Contribute to Fibrotic Scar Formation After Optic Nerve Crush.

Although glial scar formation has been extensively studied after optic nerve injury, the existence and characteristics of traumatic optic nerve fibrotic scar formation have not been previously characterized. Recent evidence suggests infiltrating macrophages are involved in pathological processes after optic nerve crush (ONC), but their role in fibrotic scar formation is unknown. Using wild-type and transgenic mouse models with optic nerve crush injury, we show that macrophages infiltrate and associate with fibroblasts in the traumatic optic nerve lesion fibrotic scar. We dissected the role of

Cinnamaldehyde Enhanced Functional Recovery after Sciatic Nerve Crush Injury in Rats.

Peripheral nerve injury is a common clinical issue induced by trauma, tumor, and damage caused by treatment. Such factors create chemical and inflammatory alterations at the injury site, which increase nerve deterioration. Thus, minimizing these modifications can lead to nerve protection after injury. The present study sought to evaluate the possible improvement in nerve regeneration and enhancement of functional outcomes by cinnamaldehyde (Cin) administration following sciatic nerve crush in a rat model. Rats (n = 48) were distributed into 6 groups, including sham, injury, DMSO (vehicle group

VGLUT1 synapses and P-boutons on regenerating motoneurons after nerve crush.

Stretch-sensitive Ia afferent monosynaptic connections with motoneurons form the stretch reflex circuit. After nerve transection, Ia afferent synapses and stretch reflexes are permanently lost, even after regeneration and reinnervation of muscle by motor and sensory afferents is completed in the periphery. This loss greatly affects full recovery of motor function. However, after nerve crush, reflex muscle forces during stretch do recover after muscle reinnervation and reportedly exceed 140% baseline values. This difference might be explained by structural preservation after crush of Ia afferen

Reduced Renshaw recurrent inhibition after neonatal sciatic nerve crush in rats.

Renshaw recurrent inhibition (RI) plays an important gated role in spinal motion circuit. Peripheral nerve injury is a common disease in clinic. Our current research was designed to investigate the change of the recurrent inhibitory function in the spinal cord after the peripheral nerve crush injury in neonatal rat. Sciatic nerve crush was performed on 5-day-old rat puppies and the recurrent inhibition between lateral gastrocnemius-soleus (LG-S) and medial gastrocnemius (MG) motor pools was assessed by conditioning monosynaptic reflexes (MSR) elicited from the sectioned dorsal roots and record

Dynamic change of Prohibitin2 expression in rat sciatic nerve after crush.

As a novel cell cycle inhibitor, PHB2 controls the G1/S transition in cycling cells in a complex manner. Its aberrant expression is closely related to cell carcinogenesis. While its expression and role in peripheral nervous system lesion and repair were still unknown. Here, we performed an acute sciatic nerve crush (SNC) model in adult rats to examine the dynamic changes of PHB2. Temporally, PHB2 expression was sharply decreased after sciatic nerve crush and reached a valley at day 5. Spatially, PHB2 was widely expressed in the normal sciatic nerve including axons and Schwann cells. While afte

Innate immune network in the retina activated by optic nerve crush.

PURPOSE: Innate immunity plays a role in many diseases, including glaucoma and AMD. We have used transcriptome profiling in the mouse to identify a network of genes involved in innate immunity that is present in the normal retina and that is activated by optic nerve crush (ONC). METHODS: Using a recombinant inbred (RI) mouse strain set (BXD, C57BL/6 crossed with DBA/2J mice), we generate expression datasets (Illumina WG 6.2 arrays) in the normal mouse retina and 2 days after ONC. The normal dataset is constructed from retinas from 80 mouse strains and the ONC dataset is constructed from 62 st

Temporal-spatial expressions of Spy1 in rat sciatic nerve after crush.

As a novel cell cycle protein, Spy1 enhances cell proliferation, promotes the G1/S transition as well as inhibits apoptosis in response to UV irradiation. Spy1 levels are tightly regulated during mammary development, and overexpression of Spy1 accelerates tumorigenesis in vivo. But little is known about the role of Spy1 in the pathological process of damage and regeneration of the peripheral nervous system. Here we established a rat sciatic nerve crush (SNC) model to examine the spatiotemporal expression of Spy1. Spy1 expression was elevated gradually after sciatic nerve crush and peaked at da

Spatial arrangements of dorsal root ganglion cells after sciatic nerve crush.

UNLABELLED: The present study explores 3D spatial arrangements of the A- and B-cells within the fifth lumbar dorsal root ganglion (DRG) of rat in order to test for differences after sciatic nerve crush (SNC). The pair correlation and cross-correlation functions were estimated through counting dipole probes superimposed on DRG sections. The data showed that the A- and B-cells were clustered at distances below 40 and 35 μm in sham-operated animals, respectively. These values were 35 and 25 μm in SNC animals. Beyond these distances, the cells of the control group were arranged neither clustered n

Y-P30 confers neuroprotection after optic nerve crush in adult rats.

The survival-promoting peptide, Y-P30, has been shown to be neuroprotective and stimulates neurite outgrowth in vitro. In this study, we examined whether the peptide increases survival and induces axon outgrowth of retinal ganglion cells after an incomplete optic nerve crush. A single intraocular injection of the peptide directly after optic nerve crush increased the number of retinal ganglion cells that preserved an axonal connection with the superior colliculus in the adult rat by more than 50%. However, administration of Y-P30 into the vitreous or optic nerve had no effect on the number of

Pioglitazone promotes peripheral nerve remyelination after crush injury through CD36 upregulation.

In our previous study, we found that CD36-deficient mice showed significant delays in peripheral nerve remyelination after sciatic nerve crush injury and suggested that CD36 played an important role in the restoration of injured peripheral nerves. The aim of this study was to investigate whether CD36 upregulation can promote peripheral nerve remyelination. We made crush injury that caused demyelination and mild axonal degeneration to sciatic nerves and investigated the effect of pioglitazone (PIO) on the remyelination post-injury in C57Bl/6 wild-type and CD36-deficient mice.
