Nerve growth factor is expressed in rat femoral vein.

PURPOSE: Entubulization is a well known method of nerve repair for defects too large to be reconstructed by direct suturing without tension. Vein grafts and alloplastic tubes have been used for entubulization in peripheral and cranial nerves, but the mechanism by which they promote healing is poorly understood. The overall hypothesis of this laboratory is that nerve growth factor (NGF) plays an important role in nerve regeneration after entubulization with a vein graft. The purpose of this pilot study was to localize NGF protein expression in the rat femoral vein. MATERIALS AND METHODS: Sciat

Melatonin increases nerve growth factor in mouse submandibular gland.

The effect of melatonin administration on nerve growth factor (NGF) was studied in the submandibular glands of adult Swiss male mice. Melatonin injection, at 1 microg daily for 30 days, resulted in an increase in the NGF content as detected by immunohistochemistry. The submandibular gland weight and the area of the granular convoluted tubules, which contained NGF, were also increased significantly. These effects were not observed when the dose of melatonin was increased to 10 and 50 microg daily. None of the melatonin treatments used influenced the weights or histology of the testes or seminal

Highly sensitive enzyme immunoassays for beta-nerve growth factor.

A comparison was made between three different strategies for measuring beta-nerve growth factor (NGF) by fluorometric enzyme immunoassay. The substrate used was 4-methylumbelliferyl-beta-galactoside and the enzyme reaction was followed in a Microfluor plate reader (Dynatech). After optimizing incubation times, concentrations, buffers, pH, and washings, a primary anti-NGF antibody directly conjugated to beta-galactosidase gave the best detection limit (2 X 10(-17) M) of purified mouse NGF (Mr 26,000) in a two-site sandwich assay. Biotinylated secondary antibodies followed by streptavidin conjug

Binding of nerve growth factor receptor in sympathetic ganglia.

(125)I-Labeled nerve growth factor binds to membrane preparations from superior cervical ganglia of rabbit in a specific fashion indicative of an interaction with the physiological receptor. Of various tissues examined, binding was observed only in the superior cervical ganglia, a presumed target organ of nerve growth factor. Binding of labeled factor is displaced by nonradioactive factor but not by several other peptide hormones. Binding to ganglia of preparations of nerve growth factor treated with N-bromosuccinimide closely parallels their biological activity. Binding of nerve growth factor

Immunological properties of nerve growth factors.

Antisera were raised against nerve growth factors isolated from mouse salivary gland and five venoms representative of the three main families of poisonous snakes. Immunochemical cross-reactivity was assessed from the ability of the antisera to inhibit the biological activities of the heterologous antigens. The high and low molecular weight forms of the salivary gland factors were found to be immunologically identical but distinct from the snake venom factors. The snake venom factors, on the other hand, were immunologically closely related to each other but not identical.

Sorafenib protects human optic nerve head astrocytes from light-induced overexpression of vascular endothelial growth factor, platelet-derived growth factor, and placenta growth factor.

OBJECTIVES: Growth factors, such as vascular endothelial growth factor (VEGF), platelet-derived growth factor (PDGF), and placenta growth factor (PlGF) are key players in the development of diabetic retinopathy, age-related macular degeneration, and other retinal neovascular diseases. Glial cells provide a significant source of retinal growth factor production under physiologic and pathologic conditions. Cumulative light exposure has been linked to increased retinal growth factor expression. Previous reports indicate that sorafenib, an oral multikinase inhibitor, might have a beneficial effect

Effects of vascular endothelial growth factor on nerve regeneration in acellular nerve grafts.

The purpose of this study was to evaluate the effects of human recombinant vascular endothelial growth factor (VEGF-165) on peripheral nerve axonal sprouting and elongation following peripheral nerve injury and repair. Two-centimeter nerve gaps were created in rat peroneal nerves and repaired with either peripheral nerve autografts, acellular peripheral nerve isografts, or VEGF-165-treated acellular peripheral nerve isografts. Four months postoperatively, the peroneal nerves were harvested and histomorphometric analysis was performed. The reinnervated extensor digitorum longus (EDL) muscles we

Monoclonal antibodies against Vipera lebetina venom nerve growth factor cross-react with other snake venom nerve growth factors.

Nerve growth factor (NGF) was isolated from the venom of Vipera lebetina and was purified to homogeneity as judged by SDS gel electrophoresis. The biologically active NGF was used to immunize BALB/c mouse, and the spleen cells from immunized mouse were fused with mouse PAI myeloma cells. Forty-seven hybrid cell lines, secreting monoclonal antibodies to V. lebetina NGF, were isolated and nine of them purified from ascitic fluids. The isolated antibodies define two partially overlapping epitopes of the V. lebetina NGF which are not involved in the biological activity of the molecule. Both epitop

Unraveling the molecular basis of snake venom nerve growth factor: human TrkA recognition through molecular dynamics simulation and comparison with human nerve growth factor.

INTRODUCTION: Neurodegenerative diseases pose significant challenges owing to the limited number of effective therapies. Nerve growth factor (NGF) plays a crucial role in neuronal survival and differentiation through tropomyosin receptor kinase A (TrkA). Although snake venom NGF (sNGF) has been studied for its ability to activate TrkA, the binding modes and associated dynamics remain unclear compared to those of human NGF (hNGF). Herein, we explored the possibilities of NGFs from Daboia russelii and Naja naja as potential therapeutic alternatives to hNGF by comparing the structural similaritie

Nerve growth factor facilitates regeneration across nerve gaps: morphological and behavioral studies in rat sciatic nerve.

The rat sciatic nerve does not possess a high potential for regeneration through silastic tubes when the interstump nerve gap is greater than 10 mm. In this study, the effect of NGF treatment on regeneration of the rat sciatic nerve in 10- and 15-mm silastic chambers was compared. In addition, regeneration in 15-mm silastic chambers was compared to regeneration in 15-mm semipermeable chambers. Sections of tubing were implanted and filled with NGF or a control solution of cytochrome C (Cyt. C). Tube implants were removed at various postoperative times and regeneration was assessed histologicall

Targeting nerve growth factor for pain relief: pros and cons.

Nerve growth factor (NGF) is a neurotrophic protein that has crucial roles in survival, growth and differentiation. It is expressed in neuronal and non-neuronal tissues. NGF exerts its effects via two types of receptors including the high affinity receptor, tropomyosin receptor kinase A and the low affinity receptor p75 neurotrophin receptor highlighting the complex signaling pathways that underlie the roles of NGF. In pain perception and transmission, multiple studies shed light on the effects of NGF on different types of pain including inflammatory, neuropathic, cancer and visceral pain. Als

Nerve growth factor protects salivary glands from irradiation-induced damage.

AIMS: Radiotherapy has become a basic treatment modality for head and neck cancer. However, radiotherapy results in inevitable side effects, particularly radiation sialadenitis, that significantly impairs quality of life. A previous study indicated that nerve growth factor (NGF) has a radio-protective effect, but the mechanism was not determined in salivary glands. In this study, we explored the functional role and mechanism regarding how NGF protects salivary glands against IR-induced damage. MAIN METHODS: Human salivary gland (HSG) cells and C57BL/6 mice were selected to establish an IR-ind

Nerve growth factor: a neuroimmune crosstalk mediator for all seasons.

Neurotrophic factors comprise a broad family of biomolecules - most of which are peptides or small proteins - that support the growth, survival and differentiation of both developing and mature neurons. The prototypical example and best-characterized neurotrophic factor is nerve growth factor (NGF), which is widely recognized as a target-derived factor responsible for the survival and maintenance of the phenotype of specific subsets of peripheral neurons and basal forebrain cholinergic nuclei during development and maturation. In addition to being active in a wide array of non-nervous system c
