Contribution of 1425G/A polymorphism in protein kinase C-Eta (PRKCH) gene and brain white matter lesions to the risk of sudden sensorineural hearing loss in a Japanese nested case-control study.

Protein kinase C-eta (PRKCH) gene has been recently identified as a susceptible risk locus for cerebral infarction and hemorrhage in the Asian populations. The inner ear artery, a usual branch of anterior inferior cerebellar artery, is an end artery with minimal collaterals, therefore, the inner ear is particularly vulnerable to ischemia. The potential association between the development of stroke and sudden sensorineural hearing loss (SSNHL) has been implied. The authors hypothesized that the PRKCH polymorphism predisposing to stroke is associated with SSNHL risk, in view of brain magnetic re

Detection of Feline Coronavirus Membrane Gene Based on Conventional Revere Transcription-Polymerase Chain Reaction, Nested Reverse Transcription-Polymerase Chain Reaction, and Reverse Transcription-Quantitative Polymerase Chain Reaction: A Comparative Study.

Feline coronavirus (FCoV) is a major pathogen causing feline infectious peritonitis (FIP), a lethal disease in cats, necessitating accurate diagnostic methods. This study developed and compared novel primers targeting the FCoV membrane (M) gene for enhanced detection. Specific primers were designed for the M gene and their performance evaluated using reverse transcription-PCR (RT-PCR), nested RT-PCR, and reverse transcription-quantitative PCR (RT-qPCR) on 80 clinical effusion samples from cats suspected of FIP. Specificity of assays was tested against other feline viruses, with sensitivity bei

Transcriptional interference drives intronic polyadenylation at the endogenous H13/Mcts2 locus.

Over a tenth of mammalian genes are nested within other host genes, raising the question of how both can be co-expressed when concurrent transcription occurs in overlapping genetic space. DNA methylation at nested intragenic CpG island promoters can prevent such conflict by silencing the nested gene. Conversely, when these promoters lack DNA methylation and are active, transcriptional interference is widely assumed to occur, but this has not been mechanistically demonstrated at endogenous mammalian loci. Here, we demonstrate transcriptional interference at an endogenous mammalian host/nested l

Atypical nested 22q11.2 duplications between LCR22B and LCR22D are associated with neurodevelopmental phenotypes including autism spectrum disorder with incomplete penetrance.

BACKGROUND: Chromosome 22q11.2 is susceptible to genomic rearrangements and the most frequently reported involve deletions and duplications between low copy repeats LCR22A to LCR22D. Atypical nested deletions and duplications are rarer and can provide a valuable opportunity to investigate the dosage effects of a smaller subset of genes within the 22q11.2 genomic disorder region. METHODS: We describe thirteen individuals from six families, each with atypical nested duplications within the central 22q11.2 region between LCR22B and LCR22D. We then compared the molecular and clinical data for pat

Development and Evaluation of a Dual-Target One-Step Nested PCR for the Detection of Spotted Fever Group Rickettsia spp. in Ticks.

Spotted fever group (SFG) rickettsioses are tick-borne infectious diseases caused by more than 30 Rickettsia species. As ticks may harbor and transmit multiple pathogens during a single blood meal, sensitive and specific molecular detection methods are essential for early diagnosis. Conventional nested PCR is commonly used but is time-consuming and prone to cross-contamination due to multiple amplification steps. This study evaluated a dual-target one-step nested PCR assay developed as a rapid alternative to conventional nested PCR for SFG Rickettsia detection. Gene-specific primers targeting

Incomplete premating and postmating reproductive barriers between two parapatric populations of a social spider mite.

Closely related species with overlapping distributions often show premating reproductive barriers to avoid hybridization. Stigmaeopsis miscanthi (Saito) is a social spider mite infesting Chinese silver grass, and the species consists of two parapatric groups with frequent contacts within the contact zone. They differ in male-male aggressiveness, male morphology, female diapause traits, and life history parameters. There is incomplete but strong post-mating reproductive isolation between the two groups, and their DNA sequences are slightly different, suggesting that they diverged recently. In t

Legionella SBT applied directly to respiratory samples as a rapid molecular epidemiological tool.

Legionnaires' disease (LD) is an atypical pneumonia caused by the inhalation of Legionella. The methods used for the diagnosis of LD are direct culture of respiratory samples and urinary antigen detection. However, the sensitivity of culture is low, and the urinary antigen test is specific only for L. pneumophila sg1. Moreover, as no isolates are obtained, epidemiological studies cannot be performed. The implementation of Nested-sequence-based typing (Nested-SBT) makes it possible to carry out epidemiological studies while also confirming LD, especially in cases caused by non-sg 1. Sixty-two r

Designing and developing of high-resolution melting technique for separating different types of Toxoplasma gondii by analysis of B1 and ROP8 gene regions.

BACKGROUND: Determination of Toxoplasma gondii genotypes plays an important role in the health management and epidemiology of toxoplasmosis. We developed HRM analysis to differentiate genotypes of T. gondii using the B1 and ROP8 genes, through comparing the sensitivity and specificity of both genes and methods used for the detection of T. gondii. METHODS: A total of 96 DNA samples of muscle tissue of livestock and poultry brain tissue with three standard strains RH (type I), PRU (type II) and VEG (type III) were prepared and analyzed. Comparison of real-time PCR and nested PCR for toxoplasmosis diagnosis in toxoplasmic retinochoroiditis patients.

BACKGROUNDS: PCR is a proper technique that significantly improves toxoplasmosis diagnosis. However, a more sensitive technique is required. This study compared real-time PCR with nested PCR using B1, SAG-4, and MAG-1 bradyzoite genes to diagnose toxoplasmosis in toxoplasmic retinochoroiditis patients. METHODS: Blood samples were collected from 10 patients with active toxoplasmic chorioretinal lesions and 10 healthy individuals. Blood samples including peripheral blood mononuclear cells (PBMCs), serum and whole blood samples were used for DNA extraction. Serum was also used to detect anti-tox

Comparative sensitivity of PCR primer sets for detection of Cryptosporidium parvum.

Improved methods for detection of Cryptosporidium oocysts in environmental and clinical samples are urgently needed to improve detection of cryptosporidiosis. We compared the sensitivity of 7 PCR primer sets for detection of Cryptosporidium parvum. Each target gene was amplified by PCR or nested PCR with serially diluted DNA extracted from purified C. parvum oocysts. The target genes included Cryptosporidium oocyst wall protein (COWP), small subunit ribosomal RNA (SSU rRNA), and random amplified polymorphic DNA. The detection limit of the PCR method ranged from 10(3) to 10(4) oocysts, and the

The hematopoietic function, histological characteristics, and transcriptome profiling of Wanxi white geese ovary during nesting and late-laying stages.

Despite several factors influencing reproduction in geese, but the precise molecular mechanisms of egg cessation are not fully understood. In the present study, the hematopoietic parameters and serum hormone levels in Wanxi white geese were analyzed. RNA-Seq was utilized to identify the differentially expressed mRNAs (DEGs) and lncRNAs (DE lncRNAs) in the ovarian tissues associated with nesting in geese during the late-laying and nesting periods. Triglyceride (TG) and alkaline phosphatase (ALP) levels were higher in late-laying geese, while white blood cell (WBC), neutrophil (NEU), hemoglobin

Viruses and virus satellites of haloarchaea and their nanosized DPANN symbionts reveal intricate nested interactions.

Nested symbioses, including hyperparasitism in which parasites exploit other parasites within a host, are common in nature. However, such nested interactions remain poorly studied in archaea. Here we characterize this phenomenon in ultra-small archaea of the candidate phylum Nanohaloarchaeota, members of the DPANN superphylum (named after its first representative phyla: Diapherotrites, Parvarchaeota, Aenigmarchaeota, Nanoarchaeota and Nanohaloarchaeota) that form obligate interactions with halophilic archaea of the class Halobacteria. We reconstructed the viromes from geothermally influenced s

Genome-wide comparative analysis reveals distinct evolutionary patterns of overlapping genes in prokaryotes and eukaryotes.

Overlapping genes are widely distributed across the genomes of nearly all living organisms. Although thousands of overlapping genes have been identified and studied in several species, a comprehensive annotation of these genes is lacking, and the forces shaping their evolution remain poorly understood. Here, we present a comparative analysis of overlapping genes in 253 prokaryotic genomes, 115 plant genomes, and 111 animal genomes. We find that co-oriented overlapping genes predominate in prokaryotes, whereas nested overlapping genes are the most common type in eukaryotes. In prokaryotes, over

Identification and phylogenetic analysis of novel Piroplasmida detected in the two-toed sloth (Choloepus didactylus).

This study investigated the pathological and parasitological causes of death in two two-toed sloths (Choloepus didactylus) housed at a zoo in Hokkaido, Japan. Genomic DNA was extracted from the clotted blood of these animals and subsequently examined by nested PCR assays targeting the 18S rRNA and β-tubulin genes. The nucleotide sequences of these genes were determined and a phylogenetic analysis of the 18S rRNA gene was performed. One sample was detected positive in nested PCR. Sequencing analysis revealed that the detected parasite was closely related to the order Piroplasmida and was subseq

Serendipitous isolation of non-Vibrio bacterial strains carrying the cholera toxin gene from environmental waters in indonesia.

We initially attempted to isolate a Vibrio cholerae O1 El Tor biotype that carries a novel variant of the cholera toxin gene (ctxAB) from environmental waters of Indonesia, where the seventh cholera pandemic by V. cholerae O1 El Tor biotype began. Nested PCR targeting the gene revealed that a total of eight strains were found to carry ctxAB. However, sequencing of the 16S rRNA genes of these isolates showed they were not V. cholerae but were either Klebsiella, Enterobacter, Pantoea, or Aeromonas. Subsequent nested PCR assays targeting all genes known to be encoded on the CTX phage (i.e., zot,

Detection and characterization of Cryptosporidium species and genotypes in three chicken production systems in Brazil using different molecular diagnosis protocols.

The objective of this study was to determine the occurrence of Cryptosporidium spp. in domestic chickens raised in different chicken production systems in Brazil using three nested PCR protocols. The purification and concentration of oocysts present in 190 fecal samples from chickens raised in extensive, semi-intensive and intensive production systems were accomplished by centrifugal flotation in Sheather's solution and were followed by the extraction of genomic DNA. The detection and molecular characterization of Cryptosporidium species and genotypes were performed using three nested polymera
