Mitochondrial degeneration in dystrophic neurites of senile plaques may lead to extracellular deposition of fine filaments.

Recent data show that amyloid precursor protein accumulates inside axons after disruption of fast axonal transport, but how this leads to mature plaques with extracellular amyloid remains unclear. To investigate this issue, primitive plaques in prefrontal cortex of aged rhesus monkeys were reconstructed using serial section electron microscopy. The swollen profiles of dystrophic neurites were found to be diverticula from the main axis of otherwise normal neurites. Microtubules extended from the main neurite axis into the diverticulum to form circular loops or coils, providing a transport pathw

Perfluorodecalin modifies the pattern of cell arrangement and induces loss of neurites in rat retinal cultures.

Perfluorodecalin (PFD), a high specific weight, water-immiscible perfluorocarbon, previously studied as a potential blood substitute, now is used widely in the field of ophthalmic surgery as a tool for maneuvering intraocular tissues and as a short- or medium-term vitreous substitute. In in vivo experiments, several types of lesions in retinal tissue have been described in conjunction with long-term PFD treatment. To better evaluate the biological effects of PFD on retinal cells, we tested it on primary cultures of rat retina seeded on special cyclopore wells that allow the culture to be fed f

Qualitative and quantitative differences in senile plaque dystrophic neurites of Alzheimer's disease and normal aged brain.

We have investigated the relationship of plaque dystrophic neurites expressing beta-amyloid precursor protein (beta APP) to those bearing markers of neurofibrillary degeneration (tau), or accumulating the synaptic protein chromogranin A (CgA). In cortical and hippocampal plaques in Alzheimer's disease (AD) beta APP colocalized with CgA in a neuritic population largely distinct from the subset of neurites labeled by tau. Putaminal plaques generally incorporated only CgA/beta APP, but not tau neurites, and with a rare exception cerebellar plaques were not associated with neurites. Neocortical an

Growth cones and the formation of central and peripheral neurites by sensory neurones in amphibian embryos.

We have examined the primary growth of neurites by Rohon-Beard sensory neurones in embryos of Xenopus, Rana, and Ambystoma. Using transmission electron microscopy we have confirmed that some central longitudinal neurites first grow directly under the basal lamina which surrounds the spinal cord in Xenopus embryos. Using scanning electron microscopy of dissected embryos we have followed the growth of neurites from the spinal cord to their target tissue, the skin. Comparisons between the three genera are made and detailed observations on growth cones during fasciculation, on the somites and on t

Hypoxia preconditioned serum (HPS) and platelet-rich plasma (PRP) promote proliferation, neurite network formation, and neurite elaboration in N2a cells.

BACKGROUND: The use of autologous growth factors, such as those contained in Hypoxia-Preconditioned Serum (HPS) and Platelet-Rich Plasma (PRP), represents a promising strategy to enhance regenerative processes in injured nerves. As an initial in vitro study, we investigated the effects of these secretomes on the neuroblastoma cell line N2a. METHODS: HPS and PRP were first compared with normal serum (NS) using a protein microarray to detect neurologically related growth factors. Subsequently, N2a cells were cultured with three different secretome concentrations (0.1%, 1%, and 10%) for up to 96

Inhibition of dynein but not kinesin induces aberrant focal accumulation of neurofilaments within axonal neurites.

Studies from several laboratories indicate that the microtubule motors kinesin and dynein respectively participate in anterograde and retrograde axonal transport of neurofilaments. Inhibition of dynein function by transfection with a construct expressing dynamitin or intracellular delivery of anti-dynein antibodies accelerates anterograde transport, which has been interpreted to indicate that the opposing action of both motors mediates the normal distribution of neurofilaments along axons. Herein, we demonstrate that, while expression of relatively low levels of exogenous dynamitin indeed acce

Cholinergic differentiation of rat sympathetic neurons in culture: effects of factors applied to distal neurites.

Cholinergic properties are induced in sympathetic neurons by several factors applied to entire neurons in culture. Evidence from work with the rat sweat gland model indicates that factors located in target tissues can induce cholinergic differentiation in vivo. We now report that when leukemia inhibitory factor (LIF), heart cell-conditioned medium (HCCM), or dermal fibroblast-conditioned medium (DFCM) is applied to only distal neurites in compartmented cultures of rat sympathetic neurons, the neurons exhibit an increase in specific choline acetyltransferase activity and a concomitant decrease

Sympathetic neurons extend neurites in a culture medium containing cyanide and dinitrophenol but not iodoacetate.

A culture medium circulated through the rat heart and supplemented with insulin, transferrin and nerve growth factor leads to a massive proliferation of neurite outgrowth from neurons of peripheral sympathetic ganglia of the chick embryo. Addition of 1 mM cyanide or 50 microM dinitrophenol to such medium for 2 days had no adverse effect on the neurite outgrowth and ATP content of these neurons. However, 0.5 microM iodoacetate lowered ATP content 65% without affecting the number of surviving neurons up to 2 days. Only when ATP content was reduced to 80% by 2.5 microM iodoacetate was the number

Calcium influx is necessary for optimal regrowth of transected neurites of rat sympathetic ganglion neurons in vitro.

The calcium ion serves many major roles in cellular processes, and therefore, its specific role in the regrowth of neurites after transection has been difficult to define. In the present study the effect of calcium on regrowth of transected rat superior cervical ganglion neurons in culture independent of its effects on neuronal survival was examined. Superior cervical ganglion neurons in culture for 10-14 days were preloaded with the calcium chelator 1,2 bis-(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid acetoxymethyl ester (BAPTA-AM), and then the neurites were transected. The chelation of

Filamented hydrogels as tunable conduits for guiding neurite outgrowth.

Anisotropic scaffolds with unidirectionally aligned fibers present an optimal solution for nerve tissue engineering and graft repair. This study investigates the application of filamented light (FLight) biofabrication to create hydrogel matrices featuring highly aligned microfilaments, facilitating neurite guidance and outgrowth from encapsulated chicken dorsal root ganglion (DRG) cells. FLight employs optical modulation instability (OMI) to rapidly and safely (<5 s) fabricate hydrogel constructs with precise microfilament alignment. The tunability of FLight matrices was demonstrated by adjust

Analyses of Neurite and Spine Formation by In Utero Electroporation in Mice.

Dendrite morphology and dendritic spines are key features of the neuronal networks in the brain. Abnormalities in these features have been observed in patients with psychiatric disorders and mouse models of these diseases. In utero electroporation is an easy and efficient gene transfer system for developing mouse embryos in the uterus. By combining with the Cre-loxP system, the morphology of individual neurons can be clearly and sparsely visualized. Here, we describe how this labeling system can be applied to visualize and evaluate the dendrites and dendritic spines of cortical neurons.

Research advances on neurite outgrowth inhibitor B receptor.

Neurite outgrowth inhibitor-B (Nogo-B) is a membrane protein which is extensively expressed in multiple organs, especially in endothelial cells and vascular smooth muscle cells of blood vessels and belongs to the reticulon protein family. Notably, its specific receptor, Nogo-B receptor (NgBR), encoded by NUS1, has been implicated in many crucial cellular processes, such as cholesterol trafficking, lipid metabolism, dolichol synthesis, protein N-glycosylation, vascular remodelling, angiogenesis, tumorigenesis and neurodevelopment. In recent years, accumulating studies have demonstrated the stat

Intracellular amyloid and the neuronal origin of Alzheimer neuritic plaques.

Genetic analysis of familial forms of Alzheimer's disease (AD) causally links the proteolytic processing of the amyloid precursor protein (APP) and AD. However, the specific type of amyloid and mechanisms of amyloid pathogenesis remain unclear. We conducted a detailed analysis of intracellular amyloid with an aggregation specific conformation dependent monoclonal antibody, M78, raised against fibrillar Aß42. M78 immunoreactivity colocalizes with Aß and the carboxyl terminus of APP (APP-CTF) immunoreactivities in perinuclear compartments at intermediate times in 10month 3XTg-AD mice, indicating

Analysis of gene expression during neurite outgrowth and regeneration.

BACKGROUND: The ability of a neuron to regenerate functional connections after injury is influenced by both its intrinsic state and also by extrinsic cues in its surroundings. Investigations of the transcriptional changes undergone by neurons during in vivo models of injury and regeneration have revealed many transcripts associated with these processes. Because of the complex milieu of interactions in vivo, these results include not only expression changes directly related to regenerative outgrowth and but also unrelated responses to surrounding cells and signals. In vitro models of neurite ou

Isolation and characterization of infiltrates in the nerves of patients with neuritic leprosy.

A study was done on the characteristics of infiltrating cells in the nerves of 9 patients with pure neuritic leprosy, by preparing a single cell suspension. The patients had no skin lesion. Histopathological examination revealed that 2 of the 9 nerves showed granulomas characteristics of tuberculoid leprosy, while the remaining 7 had features of lepromatous granulomas. In the nerves showing tuberculoid granulomas, a high proportion of lymphocytes were T cells as they formed rosettes with sheep erythrocytes and only a few percent were EAC rosette forming cells. On the other hand, the nerves sho

[Viral serology in ear diseases of presumably neuritic origin].

A viral etiology is often advanced to explain the origin of Bell's palsy, sudden deafness and certain vertigos. Virology findings were compared in 72 patients with either chicken pox, herpes, zona or other "viral" infections, where the viral origin was not absolutely clear, and a population of healthy volunteers. Results confirmed the indefinite viral nature of these affections. However, recent studies have shown that definite viral disorders are not always associated with an increase in antibody levels, and the intervention of a virus should not be totally discounted in some of these affectio
