Organization and neurochemistry of vagal preganglionic neurons innervating the lower esophageal sphincter in ferrets.

The motor control of the lower esophageal sphincter (LES) is critical for normal swallowing and emesis, as well as for the prevention of gastroesophageal reflux. However, there are surprisingly few data on the central organization and neurochemistry of LES-projecting preganglionic neurons. There are no such data in ferrets, which are increasingly being used to study LES relaxation. Therefore, we determined the location of preganglionic neurons innervating the ferret LES, with special attention to their relationship with gastric fundus-projecting neurons. The neurochemistry of LES-projecting ne

Ayahuasca treatment reduces 50-kHz ultrasonic vocalizations and modifies mesocorticolimbic monoamine neurochemistry in rats.

Rats emit 50-kHz ultrasonic vocalizations (USVs), a widely used measure of drug-induced motivational and affective effects and dopaminergic activity. Although psychedelic drugs are promising for addiction treatment, the effects of ayahuasca (AYA) on 50-kHz ultrasonic vocalizations (USVs) remain unclear. We investigated whether acute AYA treatment attenuates lisdexamfetamine (LDX)-induced USVs, hyperlocomotion and monoaminergic content in adult male Wistar rats. Rats were pretreated with vehicle (VEH) or LDX (10 mg/kg). Forty minutes later, they received VEH or AYA (240 mg/kg) treatment. Behavi

Unique infrared thermographic profiles and altered hypothalamic neurochemistry associated with mortality in endotoxic shock.

Neonatal sepsis results in significant morbidity and mortality, but early detection is clinically challenging. In a neonatal rat model of endotoxic shock, we characterised unique infrared thermographic (IRT) profiles in skin temperature that could identify risk of later mortality. Ten-day old rats were placed in a thermally stable isolette and IRT images of cranial (TCR), scapula (TSC) and rump (TRU) skin temperature were obtained continuously for 8 h following an intraperitoneal injection of lipopolysaccharide (LPS) or saline. LPS resulted in ∼74 % mortality (designated as non-survivors, LPSN

Cytotoxicity of ventricular cerebrospinal fluid from Parkinson patients: correlation with clinical profiles and neurochemistry.

Other investigators have reported that the cerebrospinal fluid (CSF) from patients with Parkinson's disease (PD) might contain endogenous dystrophic factors. Using CSF samples drawn from individual PD patients during surgery, we investigated the toxic effect of ventricular CSF (vCSF) on the growth of PC12 cells and the correlation between the clinical profiles of the patients and CSF neurochemistry. Ventricular CSF samples from 28 patients with PD or essential tremor (ET) were collected during ventriculography for stereotactic pallidotomy or thalamotomy. PC12 cells were incubated with 20% vCSF

The dual incretin agonist tirzepatide blunts nicotine-induced behaviours and neurochemistry in male mice.

BACKGROUND AND PURPOSE: Nicotine dependence remains one of the leading causes of morbidity and mortality worldwide, with high relapse rates and post-cessation weight gain representing persistent barriers to sustained abstinence. The incretin hormones glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) have emerged as novel treatment targets, as they appear to influence both reward-related behaviours associated with addictive substances and body weight regulation. Whether recently developed multi-receptor incretin agonists such as tirzepatide, a long-acting du

Fundamental neurochemistry review: Glutamatergic dysfunction as a central mechanism underlying flavivirus-induced neurological damage.

The Flaviviridae family comprises positive-sense single-strand RNA viruses mainly transmitted by arthropods. Many of these pathogens are especially deleterious to the nervous system, and a myriad of neurological symptoms have been associated with infections by Zika virus (ZIKV), West Nile virus (WNV), and Japanese encephalitis virus (JEV) in humans. Studies suggest that viral replication in neural cells and the massive release of pro-inflammatory mediators lead to morphological alterations of synaptic spine structure and changes in the balance of excitatory/inhibitory neurotransmitters and rec

Abnormal hippocampal neurochemistry in smokers: evidence from proton magnetic resonance spectroscopy at 3 T.

OBJECTIVE: In animals, nicotine, the primary psychoactive constituent of tobacco smoke, reduces neurogenesis and increases cell loss in both hippocampus and cortex. Accordingly, tobacco smoking has been linked to reduced performance on cognitive paradigms requiring attention and working memory in humans. However, few prior studies have tested for evidence of structural brain alterations in human tobacco smokers. In this study, proton magnetic resonance spectroscopy was used to assess the effects of chronic smoking on neuronal integrity of the hippocampus and anterior cingulate cortex (ACC). M

Cocaethylene exposure during the brain growth spurt period: brain growth restrictions and neurochemistry studies.

The concurrent use of alcohol and cocaine has recently attracted attention in the medical research field due to the prevalence of this drug abuse pattern and the exclusive formation of a pharmacologically active substance, cocaethylene (CE). This is the first study to examine the neuroteratogenic effects of cocaethylene exposure during the brain growth spurt (part of the third trimester equivalent) on brain growth restrictions and neurochemical profiles. For the brain growth restrictions study, three groups of artificially reared rat pups were given daily injections of 0, 10 or 20 mg/kg cocaet

Long-lasting alterations in behavior and brain neurochemistry following continuous low-level LSD administration.

Groups of rats were administered either 80 micrograms LSD-25 continuously over seven days using subcutaneous minipumps, or were given the same total amount of drug in seven daily injections, or were administered vehicle. When tested long after cessation of drug administration, persisting alterations in behavior and brain were found in the continuous LSD groups. In social open-field tests, this consisted of decreased social distance between animals; this effect increased upon repeated testing. In uptake of labeled ligands, this was reflected predominantly by decreased 3H-LSD binding in several

Evidence in support of using a neurochemistry approach to identify therapy for both epilepsy and associated depression.

The present study aimed to develop a neurochemistry-based single or adjuvant therapy approach for comprehensive management of epilepsy and associated depression employing pentylenetetrazole-kindled animals. Kindling was induced in two-month-old male Swiss albino mice by administering a subconvulsant pentylenetetrazole dose (35mg/kg, i.p.) at an interval of 48±2h. These kindled animals were treated with saline and sodium valproate (300mg/kg/day, i.p.) for 15days. Except for the naïve group, all other groups were challenged with pentylenetetrazole (35mg/kg, i.p.) on days 5, 10, and 15 to evaluat

The neurochemistry of decreased sound tolerance: A magnetic resonance spectroscopy (MRS) study of misophonia and hyperacusis.

This study examined the concentrations of excitatory and inhibitory neurotransmitters (glutamate and GABA respectively) in two regions of interest (auditory and insular cortices) and a control region (visual cortex) to test the hypothesis that decreased sound tolerance would be linked to increased cortical excitability (more glutamate and/or less GABA). Misophonia (negative emotional responses to specific sounds) and hyperacusis (finding sounds atypically loud) were separately considered and key comorbidities excluded (tinnitus, migraine, autism). A MEGA-PRESS sequence was used for auditory co

Signatures of exposure to childhood trauma in young adults in the structure and neurochemistry of the superior temporal gyrus.

BACKGROUND: Childhood trauma (CT) has been linked to increased risk for mental illness in adulthood. Although work in experimental animals has shown that early life stressors can affect inhibitory and excitatory neurotransmission in adult rodents, with possible excitotoxic effects on local grey matter volumes (GMV), the neurobiological mechanisms that mediate this relationship in humans remain poorly understood. AIM: To examine glutamate and gamma-aminobutyric acid (GABA) metabolite concentrations and potential excitotoxic effects on GMV, in adults who experienced CT. METHODS: Fifty-six youn

Region-specific effects of repeated ketamine administration on the presynaptic GABAergic neurochemistry in rat brain.

A growing body of evidence indicates that clinical use of ketamine as a promising antidepressant can be accompanied by psychotic-like side effects. Although, the generation of such effects is thought to be attributed to dysfunction of prefrontal GABAergic interneurons, the mechanism underlying ketamine's propsychotic-like action is not fully understood. Due to wide spectrum of behavioral abnormalities, it is hypothesized that ketamine action is not limited to only cortical GABA metabolism but may also involve alterations in other functional brain areas. To test it, we treated rats with ketamin

Empirical evidence for gut microbial influence on human brain neurochemistry via the gut-brain axis.

The gut microbiome produces metabolites with potential neuroactive properties, many of which act locally within the gut. While preclinical studies suggest these microbial pathways can influence cognitive and emotional processes, human evidence remains limited. This study investigates associations between gut microbiome-derived neuroactive functional potential and in vivo brain neurotransmitter concentrations in healthy young females. Using proton magnetic resonance spectroscopy (¹H-MRS), we quantified GABA and glutamate levels in the dorsolateral prefrontal cortex (dlPFC), anterior cingulate c

Transcranial focused ultrasound stimulation enhances semantic memory by modulating brain morphology, neurochemistry and neural dynamics.

The ventromedial anterior temporal lobe (ATL) is a core transmodal hub for semantic memory, yet non-invasive modulation of this region has remained challenging. Transcranial ultrasound stimulation (TUS) offers high spatial precision suitable for deep brain targets. In this study, we investigated whether theta-burst TUS (tbTUS) to the ventromedial ATL enhances semantic memory, using a multimodal neuroimaging approach-magnetic resonance spectroscopy (MRS), functional MRI (fMRI), and voxel-based morphometry (VBM). Compared to control stimulation, tbTUS improved semantic task performance. MRS show

State-dependent reactivity of anterior cingulate cortex neurochemistry and downstream autonomic arousal in intrusive thinking.

Unwanted intrusive thoughts play a key role in the onset, maintenance, and relapse of stress-related psychopathological conditions and are usually accompanied by a physiological fight-or-flight response. However, the mechanisms underlying the persistence of this maladaptive process remain unclear. This study employed a multimodal approach, integrating proton magnetic resonance spectroscopy, resting-state functional connectivity, and heart rate monitoring, to investigate gamma-aminobutyric acid (GABA) and glutamate-mediated neurometabolism during intrusive thinking, alongside central and periph

Protective effects of prucalopride in MPTP-induced Parkinson's disease mice: Neurochemistry, motor function and gut barrier.

Evidence suggests constipation precedes motor dysfunction and is the most common gastrointestinal symptom in Parkinson's disease (PD). 5-HT4 receptor (5-HT4R) agonist prucalopride has been approved to treat chronic constipation. Here, we reported intraperitoneal injection of prucalopride for 7 days increased dopamine and decreased dopamine turnover. Prucalopride administration improved motor deficits in 1-methyl-4-phenyl-1,2,3,6-tetrathydropyridine (MPTP)-induced PD mouse models. Prucalopride treatment also ameliorated intestinal barrier impairment and increased IL-6 release in PD model mice.

Altered neurochemistry in the anterior white matter of bipolar children and adolescents: a multivoxel 1H MRS study.

Abnormalities within frontal lobe gray and white matter of bipolar disorder (BD) patients have been consistently reported in adult and pediatric studies, yet little is known about the neurochemistry of the anterior white matter (AWM) in pediatric BD and how medication status may affect it. The present cross-sectional 3T 1H MRS study is the first to use a multivoxel approach to study the AWM of BD youth. Absolute metabolite levels from four bilateral AWM voxels were collected from 49 subjects between the ages of 8 and 18 (25 healthy controls (HC); 24 BD) and quantified. Our study found BD subje

Mechanisms of dopaminergic and serotonergic neurotransmission in Tourette syndrome: clues from an in vivo neurochemistry study with PET.

Tourette syndrome (TS) is a neuropsychiatric disorder with childhood onset characterized by motor and phonic tics. Obsessive-compulsive disorder (OCD) is often concomitant with TS. Dysfunctional tonic and phasic dopamine (DA) and serotonin (5-HT) metabolism may play a role in the pathophysiology of TS. We simultaneously measured the density, affinity, and brain distribution of dopamine D2 receptors (D2-R's), dopamine transporter binding potential (BP), and amphetamine-induced dopamine release (DA(rel)) in 14 adults with TS and 10 normal adult controls. We also measured the brain distribution a
