Creutzfeldt-Jakob disease with plaques and paired helical filaments.

A 32-year-old man with rapidly progressive dementia, pyramidal signs, myoclonic jerks and dystonic movements died following brain biopsy. neuropathological examination revealed minimal neuronal loss accompanied by mild spongiform change and astrocytic reaction. Numerous plaques and neurofibrillary tangles composed of paired helical filaments dominated the ultrastructural picture. This patient had features of both Creutzfeldt-Jakob disease and Alzheimer's disease, providing additional support for the existence of an overlap between these disorders.

The dissolution of language in Pick's disease with neurofibrillary tangles: a case study.

This paper is a detailed retrospective history of a patient who began to have difficulty with speaking and comprehension in 1967, and whose neuropathological examination at time of death 12 1/2 years later was consistent with a diagnosis of Pick's disease, complicated by neurofibrillary tangles. It follows the deteriorating course of his language abilities in the context of relatively less-impaired general cognitive abilities, using two sources of information. The first is his own written record of his deteriorating abilities, shown in letters and notes he wrote over this period. The second is

[Classical and ultrastructural neuropathology of aging processes in the human: a critical review (author's transl)].

Although the primary interest in the course of studies on aging is focused on problems of function, morphology has an important role to play as a marker and as a control. Processes concomitant with brain aging are multiple and diverse. They ought to be correlated individually with functional changes. Statistical problems in population sampling for reference morphological studies are difficult. Bias is often a consequence of unstated underlying theories (Brain aging: a physiological process v. one of many pathological processes). Morphological criteria do not allow close application of animal m

Topography of Pick inclusion bodies in hippocampi of demented patients. A quantitative study.

Topographic analysis was performed on the distribution of argyrophilic inclusions in hippocampal neurones of patients with Pick's dementia. A semiautomated scanning stage microscope linked potentiometrically to an XY pen recorder permitted the plotting of cytoarchitectonic "scattergrams" from the sequentially screened hippocampal formations. The density of Pick body-bearing nerve cells per cubic mm. of (pyramidal) cortex was quantified by measuring the area of each of six "zones" with a digitizer and programmable calculator. The ranking orders of relative severity showed that neurones in Rose'

CS2 binding to rat spinal neurofilaments.

The binding of CS2 sulphur to rat spinal cord neurofilaments was studied 3 and 6 h after the intraperitoneal injection of 1.3 mmol of CS2 in control and phenobarbitone pretreated rats. The binding of CS2 carbon was studied similarly 3 h after the injection. The binding of CS2 sulphur was 31% higher in the control neurofilaments than that in the phenobarbitone pretreated group. Half of the neurofilament associated CS2 sulphur was bound to protein in both groups as studied by electrophoresis. Approximately 30% of the neurofilament associated CS2 sulphur was detected 6 h after the injection of th

Phytochemical Analysis, Antioxidant Activity and Bioassay-Guided Isolation of Acetylcholinesterase and Butyrylcholinesterase Inhibitors from Horsfieldia polyspherula Bark (Myristicaceae).

Alzheimer's disease (AD) is a neurodegenerative condition brought on by aging and characterised by progressive decline in cognitive function and abnormalities in the central cholnergic system. β-amyloid deposits, neurofibril tangle aggregation, oxidative stress or reduced level of acetylcholine are a few causes that have been linked to AD. In this study, the bioassay-guided isolation from ethyl acetate (EtOAc) extract of Horsfieldia polyspherula bark led to the isolation of nine compounds namely, 16-phenylhexadecanoic acid (1), undecylbenzene (2), 3,4-dihydroxybenzoic acid (3), dodecanoic acid

[Neuropathological aspects of normal and abnormal aging].

Several senile changes in the central nervous system in cadavers were examined. The pattern of extension of Alzheimer's neurofibrillary tangles (NFT) and senile plaques (SP) in the olfactory bulbs of 100 specimens was examined during routine autopsy by immunohistochemical staining. NFT were first observed in the anterior olfactory nucleus after the age of 60, and incidence rose with increasing age. Senile plaques were found in the nucleus when there were many SP in the cerebral cortex. Of 25 non-demented amyotrophic lateral sclerosis patients, SP were found in the cerebral cortices of 10, and

Responses in the posterior lobe of the rat cerebellum to electrical stimulation of cutaneous afferents to the snout.

1. Responses in the cortex of the posterior lobe of the cerebellum to electrical stimulation of afferent fibres from the skin of the snout have been analysed in decerebrate and pentobarbitone-anaesthetized rats by means of surface and microelectrode records. Single shocks were applied either to the exposed follicles of the mystachial vibrissae or to the infraorbital branch of the trigeminal nerve. 2. In decerebrate rats responses were mediated only via mossy fibre afferents. Stimulation of one side of the snout yielded responses with mean latency 2.4 ms throughout the uvula (largest ipsilatera

Neurofibrillary changes after acute experimental lead poisoning.

Following acute tetraethyllead intoxication in rabbits classic neurofibrillary tangles develop. As studied by electron microscopy, such tangles develop in hydropic degenerating neurons and consist primarily of bundles of 200 A diameter smooth tubules of enormous length. In two of 16 animals a few twisted tubules (periodicity of 800 A) also could be found. The significance of these lead-induced twisted tubules, so characteristic of Alzheimer's pre-senile dementia in man, is discussed.

A histopathological study on senile changes in the human olfactory bulb.

Senile changes in the human olfactory bulb were studied histopathologically in 133 individuals ranging in age from 40 to 91 years with a mean age of 64.3 years. Neurofibrillary tangles (NFTs) in the olfactory bulb were observed in 47 subjects aged over 50 years. The frequency of NFTs was 35.3% (47/133) in total and 40.5% (47/116) among subjects 50 years of age or older. The frequency of individuals with NFTs increased linearly with age. Senile plaques (SPs) were observed in 5 individuals over 60 years old at a frequency of 3.8% (5/133) in total and 5.7% (5/88) among individuals 60 years old or

Early progressive supranuclear palsy: pathology and clinical presentation.

Progressive supranuclear palsy (PSP) is a syndrome of supranuclear ophthalmoplegic palsy, pseudobulbar palsy, rigidity of the limbs, nuchal dystonia, and dementia in which the gaze palsy is the hallmark of the disease. Most neurologists are reluctant to consider the diagnosis unless visual problems exist. Since the earliest complaints of PSP are said to be variable and subtle, accurate diagnosis is often delayed and initial pathologic changes of the disease not well studied. Two patients came to autopsy with dementia, gait disturbances, and/or dysarthria but no eye findings by history or physi

Cytoskeletal immunohistochemistry of Alzheimer's disease.

The first half of this paper is devoted to a review on the cytoskeletal immunocytochemistry of the various morphological changes in Alzheimer's disease (AD), i.e. neurofibrillary tangles (NFT), senile plaques (SP), granulovacuolar degeneration (GV) and Hirano bodies (HB). In the second half it is demonstrated that sera raised against the paired helical filaments (PHF) in NFT, and monoclonal antibodies to phosphorylated epitopes on neurofilament proteins, also stain structures within neuronal perikarya without PHFs in some diseases. These include Pick bodies and swollen cells in Pick's disease,

[Cerebral degeneration in Down's syndrome].

Clinical and neuropathological findings of a 63-year-old male and a 25-year-old female with Down's syndrome are presented. Neuropathological examination of the older patient revealed intense features of Alzheimer's disease or senile dementia, including congophilic angiopathy and extensive mineral deposits in the globus pallidus and in the white matter of the cerebellum. In the hippocampus of the younger patient, plaque-like bodies and a few neurofibrillary tangles were found. From a survey of the cases hitherto reported in the literature it appears that among patients over 50 years of age it i

Altered tau and neurofilament proteins in neuro-degenerative diseases: diagnostic implications for Alzheimer's disease and Lewy body dementias.

The neuronal cytoskeleton is one of the most profoundly altered organelles in late life neuro-degenerative disorders that are characterized by progressive impairments in cognitive abilities. The elucidation of the protein building blocks of these organelles as well as advances in understanding how these proteins become altered in Alzheimer's disease (AD) and other less common dementing illnesses, i.e., diffuse Lewy body disease (DLBD) or the Lewy body variant of AD (LBVAD), will provide insights into the molecular basis of these disorders. Within, we review evidence that normal adult human tau
