Process-driven flavour modulation in soybean-based alternative proteins via Neurospora crassa coculture fermentation.

Soybean-derived ingredients are widely utilised in plant-based foods and protein formulations but are often limited by lipid-derived off-flavours, particularly undesirable "beany" and "green" notes. Microbial fermentation offers a clean-label strategy to improve flavour while maintaining processing sustainability. In this study, a factorial experimental design was applied to optimise solid-state fermentation of soybeans using two distinct coculture systems: a fungal-fungal system (Neurospora crassa-Rhizopus oryzae), and a fungal-bacterial system (Neurospora crassa-Lactiplantibacillus plantarum

Nucleotide sequence of pho-4+, encoding a phosphate-repressible phosphate permease of Neurospora crassa.

The nucleotide (nt) sequence of the Neurospora crassa pho-4+ gene, which encodes a phosphate-repressible phosphate permease, has been determined. The gene specifies a protein of 590 amino acids (aa) and contains two introns. Two RNA transcripts of 3.3 and 2.4 kb have been identified, and transcription start points (tsp) and termination sites and/or processing sites have been located. The 3.3-kb message is initiated about 890 nt upstream from the tsp for the 2.4-kb transcript. A hydropathy profile of the aa sequence suggests ten to twelve membrane-spanning helices with a large hydrophilic domai

Gamma-ray inactivation of conidia from heterokaryons of Neurospora crassa containing UV-sensitive mutations.

Two-component heterokaryons were formed with the fungus Neurospora crassa. The UV-sensitive mutations upr-I, uvs-4, and uvs-6 were utilized. Conidia produced by these heterokaryons were exposed to gamma-rays and survival curves were established for the three conidial fractions produced by each heterokaryon. Results showed that upr-I, when included in only one nuclear component, did not affect the sensitivity of any conidial fraction; however, when included in both components, all three conidial fractions exhibited two- to four-fold decreases in survival at the 30 krad exposure. The uvs-4 mutat

Co-Production of Mycoprotein, Carotenoids, and B Vitamins From Cheese Whey by Neurospora intermedia.

Cheese whey, a nutrient-rich dairy byproduct that requires management due to its high organic load, was valorized using an integrated biorefinery platform with the edible fungus Neurospora intermedia to simultaneously produce mycoprotein, carotenoids, and B vitamins (B1, B2, B3, B6). Initial optimization employed a one-factor-at-a-time (OFAT) approach to evaluate the effects of whey concentration, initial pH, KNO3, salt supplementation, trace elements, precursor, and pellet-inducing agents (CaCl2, glycerol, and Tween 80). Subsequent optimization via response surface methodology (RSM) revealed

Regulation of Neurospora Catalase-3 by global heterochromatin formation and its proximal heterochromatin region.

Catalase-3 (CAT-3) constitutes the main catalase activity in growing hyphae of Neurospora crassa, and its activity increases during exponential growth or is induced under different stress conditions. Although extensive progress has been made to identify catalase regulators, the regulation mechanism of CAT-3 at the chromatin level still remains unclear. Here, we aim at investigating the molecular regulation mechanisms of cat-3 at the chromatin level. We found that CAT-3 protein levels increased in mutants defective in proper global heterochromatin formation. Bioinformatics analysis identified a

Isolation and characterization of a mitochondrially synthesized polypeptide from Neurospora crassa cni-1 mutant.

A polypeptide with a molecular weight of 8 500 (HP 8 500) was isolated from the mitochondrial membrane of the nuclear mutant cni-1 of Neurospora crassa. This mutant is characterized by a cyanide-insensitive respiration and by a deficiency in the cytochromes aa3 and b. The polypeptide is synthesized on mitochondrial ribosomes. It has an extremely hydrophobic character; it is insoluble in aqueous media in the absence of sodium dodecylsulfate and is soluble in acid chloroform/methanol. It lacks histidine. The polar amino acids lysine, arginine, aspartic acid, glutamic acid, serine and threonine m

Compressible but not redundant: knockout recovery and assay-panel architecture of the Neurospora crassa deletion phenome.

Classical phenotype assays remain central to functional genetics, but a deletion phenome reflects both assay design and the set of knockouts that can be recovered for phenotyping. We integrated current Neurospora crassa knockout-availability records with a published complete-data phenome of 1,168 deletion strains scored across 10 growth and developmental assays. Among 8,892 genes with interpretable current knockout status, 7,239 had an available homokaryon and 1,653 were represented only as heterokaryons. Within iJDZ836 model genes, Dreyfuss-predicted essential genes were strongly enriched amo

Characterization of the Polysaccharide and Protein Composition of Neurospora crassa Conidium, Perithecium, and Ascospore Cell Walls.

We carried out carbohydrate composition and carbohydrate linkage analyses on Neurospora crassa cell walls from vegetative hyphae, conidia (asexual spores), perithecia (female mating structures), and ascospores (sexual spores). We show that the composition and structure of the cell wall undergo dramatic changes during the N. crassa life cycle. All the cell walls contain β-1,3-glucan and mixed β-1,3-/β-1,4-glucan (lichenin). The conidia cell walls and perithecia cell walls also contain α-1,3-glucan. β-1,3-glucan was the most abundant polysaccharide in the vegetative hyphae cell walls and mixed β

Regulation by blue light of the fluffy gene encoding a major regulator of conidiation in Neurospora crassa.

The development of asexual spores, that is, the process of conidiation, in the fungus Neurospora crassa is increased by light. The fluffy (fl) gene, encoding a major regulator of conidiation, is activated by light. We describe here a detailed characterization of the regulation by blue light of fl in vegetative hyphae. This induction requires the white collar complex (WCC) while the FLD protein acts as a dark repressor of fl transcription. We show that the WCC directly regulates fl transcription in response to blue light after transiently binding the promoter. We propose that fl is repressed by

The Goodwin model: simulating the effect of light pulses on the circadian sporulation rhythm of Neurospora crassa.

The Goodwin oscillator is a minimal model that describes the oscillatory negative feedback regulation of a translated protein which inhibits its own transcription. Now, over 30 years later this scheme provides a basic description of the central components in the circadian oscillators of Neurospora, Drosophila, and mammals. We showed previously that Neurospora's resetting behavior by pulses of temperature, cycloheximide or heat shock can be simulated by this model, in which degradation processes play an important role for determining the clock's period and its temperature-compensation. Increased viability of a strain of Neurospora crassa after treatment with ultraviolet radiation or other agents.

A strain of Neurospora crassa with low viability but near normal germination shows a two- to sixfold increase in the number of colonies formed after treatment with ultraviolet radiation, X radiation, nitrous acid, or heat (50 C). Effective dosages of these agents produce moderate killing of the wild type.

The Neurospora crassa Pangenome: A Robust Framework for Population-Scale Analysis and Structural Variant Discovery.

Neurospora crassa is a widely distributed ascomycete with high genetic diversity, yet reliance on limited reference genomes has hindered a comprehensive understanding of its genetic landscape. To address this limitation, we integrated the functional annotation of the FGSC2225 genome with a comprehensive comparative genomic analysis of N. crassa strains. FGSC2225 gene and transposable element (TE) proportions mirrored those of FGSC2489, though TE levels were significantly higher than those in sister species Sordaria macrospora. Phylogenetic analysis resolved the N. crassa population into two pr

Light-dependent regulation of the gene cut-1 of Neurospora, involved in the osmotic stress response.

The gene cut-1 from Neurospora encodes a protein of the haloacid dehalogenase (HAD) family, a group of enzymes usually associated to phosphatase or phosphotransferase activities. Loss of cut-1 function results in an osmosensitive phenotype and its transcription is induced by high osmotic pressure or heat-shock, but not by other stressing conditions tested. Unexpectedly, cut-1 transcript levels markedly decrease following illumination in the wild type and in some mutants tested, such as bd and vvd. The reduction was less pronounced in wc-1 and wc-2 mutants, indicating that this photoresponse is

GMP-stimulation of the cyanide-insensitive mitochondrial respiration in heat-shocked conidia of Neurospora crassa.

In mitochondria of heat-shocked conidia of Neurospora exogenous NADH and succinate were oxidized mainly via the alternative, hydroxamate-sensitive pathway (70%) and only 30% via the cytochromic, cyanide-sensitive pathway which was predominant in untreated conidia; the alternative oxidase pathway was markedly stimulated by guanosine 5'-monophosphate (GMP).

A high molecular weight dihydro-orotate dehydrogenase of Neurospora crassa. Purification and properties of the enzyme.

Some of the unusual molecular and catalytic properties of a high molecular weight dihydro-orotate dehydrogenase (DHOD) from Neurospora crassa have been determined. Comparison of the properties of this enzyme with the properties of the soluble biosynthetic enzyme of prokaryotes has revealed several important differences. The fungal enzyme is located in a mitochondrial membrane in a position consistent with linkage with the respiratory chain through ubiquinone (Miller, R. W.: Arch. Biochem, Biophys. 146, 256-270 (1971)). Release of the enzyme from the membrane results in a solubilized protein co

Regulation of mitochondrial membrane assembly in Neurospora crassa. Transient expression of a respiratory mutant phenotype.

Cultures of mutant cni-1, a chromosomal mutant of Neurospora crassa, undergo a marked change in respiratory properties as the age of the culture increases. Early log phase cultures have a high level of respiration that is insensitive to inhibition by cyanide or antimycin A. Late log and stationary phase cultures have reduced rates of respiration. A high percentage of this respiration is inhibited by cyanide. Mitochondria from early log phase cni-1 have an excess of cytochrome c and little or no detectable cytochrome aa3. Mitochondria from late log and stationary phase cultures have levels of c

Carbon and nitrogen limitation increase chitosan antifungal activity in Neurospora crassa and fungal human pathogens.

Chitosan permeabilizes plasma membrane and kills sensitive filamentous fungi and yeast. Membrane fluidity and cell energy determine chitosan sensitivity in fungi. A five-fold reduction of both glucose (main carbon (C) source) and nitrogen (N) increased 2-fold Neurospora crassa sensitivity to chitosan. We linked this increase with production of intracellular reactive oxygen species (ROS) and plasma membrane permeabilization. Releasing N. crassa from nutrient limitation reduced chitosan antifungal activity in spite of high ROS intracellular levels. With lactate instead of glucose, C and N limita
