Nipah Virus C and W Proteins Contribute to Respiratory Disease in Ferrets.

UNLABELLED: Nipah virus (NiV) is a highly lethal paramyxovirus that recently emerged as a causative agent of febrile encephalitis and severe respiratory disease in humans. The ferret model has emerged as the preferred small-animal model with which to study NiV disease, but much is still unknown about the viral determinants of NiV pathogenesis, including the contribution of the C protein in ferrets. Additionally, studies have yet to examine the synergistic effects of the various P gene products on pathogenesis in animal models. Using recombinant NiVs (rNiVs), we examine the sole contribution of…

Vertical transmission and fetal replication of Nipah virus in an experimentally infected cat.

A female adult cat developed clinical disease 13 days after subcutaneous inoculation with Nipah virus (NiV) and was discovered to be pregnant at necropsy. Viral genome was detected in a variety of specimens, including blood, serum, tonsil swabs, and urine, up to 3 days before the onset of disease. Samples collected postmortem, including placenta, uterine fluid, and fetal tissues, were also positive for NiV genome, and the placenta and uterine fluid contained high levels of recoverable virus. The high levels of viral shedding in the adult combined with fetal viral replication suggests that both…

A dual-glycoprotein-targeting bispecific nanoantibody confers potent neutralization against Nipah virus.

Nipah virus (NiV) is a biosafety level 4 pathogen that has consistently posed a significant threat to public health. The two envelope proteins on the viral surface, the receptor-binding protein (RBP) and the fusion (F) protein, are the primary targets of neutralizing antibodies. However, the use of antibodies targeting a single viral antigen often leads to the emergence of resistant strains. Here, by panning phage display library, we obtained a C-type nanoantibody (C-Nab), G-3-1, targeting the head domain of RBP. The half-life-extended G-3-1-HLE effectively protected hamsters against authentic…

Persistent T Cell Immunity Following Nipah Virus Infection: Evidence From Malaysian Survivors.

Nipah virus (NiV) induces strong humoral immunity, but the durability of NiV-specific memory T-cell responses remains unclear. We characterized cellular immunity approximately 25 years after infection in 4 survivors of the 1998 Malaysian outbreak. PBMCs were stimulated with overlapping peptide mini-pools spanning the NiV fusion (NiV-F) and attachment (NiV-G) glycoproteins, and responses were assessed using enzyme-linked immune absorbent spot and intracellular cytokine staining. Durable cytokine-producing T-cell responses were detected, with immunodominant epitopes in the NiV-G ectodomain. Surv…

Cell-Free Expression of Nipah Virus Transmembrane Proteins for Proteoliposome Vaccine Design.

Membrane proteins expressed on the surface of enveloped viruses are potent antigens in a vaccine, yet are difficult to produce and present d … of membrane proteins into nanoscale liposomes. Here, we develop a vaccine candidate for the deadly Nipah virus (NiV), a highly lethal virus listed by the World Health Organization as a priority pathogen, by cell-free expressing two full-length Nipah virus membrane proteins. We demonstrate that both NiV fusion protein (NiV F) and NiV glycoprotein (NiV G) can …

Favipiravir (T-705) protects against Nipah virus infection in the hamster model.

Nipah and Hendra viruses are recently emerged bat-borne paramyxoviruses (genus Henipavirus) causing severe encephalitis and respiratory dise … administration of favipiravir for 14 days fully protected animals challenged with a lethal dose of Nipah virus. This first successful treatment of henipavirus infection in vivo with a small molecule drug suggests that favipiravir should be further evaluated as an antiviral treatment option for henipavirus infections.

Late clinical and magnetic resonance imaging follow up of Nipah virus infection.

The Nipah virus is a newly identified paramyxovirus responsible for an outbreak of fatal encephalitis in Malaysia and Singapore. This paper reports the follow up clinical and magnetic resonance imaging findings in 22 affected subjects. Of 13 patients with encephalitis, one died, one was lost to follow up, and seven recovered. Among the four remaining patients, one had residual sixth nerve palsy, another suffered from severe clinical depression, and a third patient had evidence of retinal artery occlusion. One patient with delayed onset Horner syndrome had a single lesion in the cervical spinal…

Inactivated Recombinant Rabies Virus Displaying the Nipah Virus Envelope Glycoproteins Induces Systemic Immune Responses in Mice.

Nipah virus (NiV) causes severe, lethal encephalitis in humans and pigs. However, there is no licensed vaccine available to prevent NiV infection. In this study, we used the reverse genetic system based on the attenuated rabies virus strain SRV9 to construct two recombinant viruses, rSRV9-NiV-F and rSRV9-NiV-G, which displayed the NiV envelope glycoproteins F and G, respectively. Following three immunizations in BALB/c mice, the inactivated rSRV9-NiV-F and rSRV9-NiV-G alone or in combination, mixed with the adjuvants ISA 201 VG and poly (I:C), were able to induce the antigen-specific cellular …

Development and Immunogenic Evaluation of a Recombinant Vesicular Stomatitis Virus Expressing Nipah Virus F and G Glycoproteins.

Nipah virus (NiV) is a highly pathogenic bat-borne zoonotic pathogen that poses a significant threat to human and animal health, with fatality rates exceeding 70% in some outbreaks. Despite its significant public health impact, there are currently no licensed vaccines or specific therapeutics available. Various virological tools-such as reverse genetics systems, replicon particles, VSV-based pseudoviruses, and recombinant Cedar virus chimeras-have been widely used to study the molecular mechanisms of NiV and to support vaccine development. Building upon these platforms, we developed a replicat…

Nipah virus matrix protein promotes NF-κB activation by targeting multiple signaling modulators.

The Nipah virus (NiV) matrix (M) protein, essential for viral assembly, has been increasingly recognized for its potential immunomodulatory functions. However, its role in modulating the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) signaling pathway-a central regulator of inflammation and innate immunity-remains poorly defined. Here, we demonstrate that NiV-M significantly promotes activation of the classical NF-κB pathway. Our data showed that NiV-M interacts with multiple cellular signaling molecules of the NF-κB pathway, including tripartite motif containing 25 (TR…

Measures to prevent and treat Nipah virus disease: research priorities for 2024-29.

Nipah virus causes highly lethal disease, with case-fatality rates ranging from 40% to 100% in recognised outbreaks. No treatments or licensed vaccines are currently available for the prevention and control of Nipah virus infection. In 2019, WHO published an advanced draft of a research and development roadmap for accelerating development of medical countermeasures, including diagnostics, therapeutics, and vaccines, to enable effective and timely emergency response to Nipah virus outbreaks. This Personal View provides an update to the WHO roadmap by defining current research priorities for dev…

Rapid detection of Nipah virus using the one-pot RPA-CRISPR/Cas13a assay.

Nipah virus (NiV) is a zoonotic pathogen with airborne transmission and high case fatality rates in humans. There is currently no treatment or vaccine against NiV infection approved for humans or animals, therefore early diagnosis is the key to control any potential outbreaks. In this study, we developed an optimized one-pot assay using recombinase polymerase amplification (RPA) coupled to CRISPR/Cas13a for the molecular detection of NiV. The one-pot RPA-CRISPR/Cas13a assay for NiV detection was specific and did not cross-react against other selected (re)-emerging pathogens. The sensitivity of…

Cytokine Induction in Nipah Virus-Infected Primary Human and Porcine Bronchial Epithelial Cells.

During the Nipah virus (NiV) outbreak in Malaysia, pigs and humans were infected. While pigs generally developed severe respiratory disease due to effective virus replication and associated inflammation processes in porcine airways, respiratory symptoms in humans were rare and less severe. To elucidate the reasons for the species-specific differences in NiV airway infections, we compared the cytokine responses as a first reaction to NiV in primary porcine and human bronchial epithelial cells (PBEpC and HBEpC, respectively). In both cell types, NiV infection resulted in the expression of type I…

Development and analytical validation of a dual-gene isothermal assay for Nipah virus detection.

Nipah virus (NiV) is a highly pathogenic zoonotic virus characterized by fatal encephalitis and severe respiratory illness, with case-fatality rates reaching 75%. Given the lack of licensed vaccines or therapeutics, rapid and accurate detection is paramount for outbreak containment. While real-time reverse transcription polymerase chain reaction (qRT-PCR) remains the gold standard, its requirement for sophisticated laboratory infrastructure limits its utility in resource-constrained field settings. In this study, we developed and validated a rapid, colorimetric dual-target reverse transcriptio…

Nipah virus and the lessons of COVID-19: are we prepared for the next pandemic threat?

The coronavirus disease 2019 (COVID-19) pandemic has exposed profound and persistent weaknesses in global preparedness, including fragmented surveillance systems, governance failures, inequitable access to countermeasures, and erosion of public trust. Although SARS-CoV-2 has dominated global attention, other high-consequence zoonotic pathogens with pandemic potential have remained comparatively neglected. Among these, the Nipah virus (NiV) represents a particularly serious threat, characterized by high case-fatality rates, zoonotic spillover from wildlife reservoirs, documented human-to-human …

In silico screening of herbal phytochemicals to develop a Rasayana for immunity against Nipah virus.

BACKGROUND: The first emergence of the Nipah virus (NiV) in 1998 from Malaysia became a major concern when it came to light and resurfaced on different occasions thereafter. NiV is a bat-borne zoonotic and pleomorphic virus that causes severe infection in human and animal hosts. Studies revealed fruit bats are the major reservoirs as natural hosts and pigs as intermediate hosts for the spread of this infection. This became a major concern as the disease was characterized by high pathogenicity varying from 40% to 80% depending on its acuteness. Moreover, the solemnity lies in the fact that the …

Characterization of the antiviral and inflammatory responses against Nipah virus in endothelial cells and neurons.

Nipah virus (NiV) is a highly pathogenic paramyxovirus which causes fatal encephalitis in up to 75% of infected humans. Endothelial cells and neurons are important cellular targets in the pathogenesis of this disease. In this study, viral replication and the innate immune responses to NiV in these cell types were measured. NiV infected endothelial cells generated a functionally robust IFN-beta response, which correlated with localization of the NiV W protein to the cytoplasm. There was no antiviral response detected in infected neuronal cells. NiV infection of endothelial cells induced a signi…
