Administration of the nitrosamine carcinogen N-nitroso-bis (2-oxopropyl) amine (BOP) by subcutaneous injection (5 mg/kg/week) led to the development of invasive pancreatic ductular adenocarcinoma in 100% of normal Syrian hamsters by 24 weeks. Pretreatment of a second group of hamsters with the beta-cell toxin streptozotocin in a diabetogenic dose (50 mg/kg i.p. A pancreatic ductal carcinoma, designated CBP, was established as a transplantable tumor line in the CB/SsLak inbred strain of Syrian golden hamsters. We review the results of the 'Enhanced' Ames Test (EAT) for assessment of the mutagenicity of N-nitrosamines, integrating the data from six key studies. High (>98 %) purities of the nitrosamines were confirmed by LC-MS and glutathione co-incubation to avoid false-positive results. The most sensitive strains were Salmonella typhimurium TA1535 and Escherichia coli WP2 uvrA (pKM101), which detect primarily GC→AT transitions, typically induced by O6-alkylguanine DNA adducts. Cholangiocarcinoma is a common hepatobiliary carcinoma in Thailand. It is believed that both chronic exposure to liver fluke infestation and nitrosamine exposure are the two main underlying factors leading to the carcinogenesis. Here, the author further extrapolates and proposes a new hypothesis based on the environmental ecological data that the stimulation of fresh water fish by contaminated pesticide in water reservoirs might be a possible background of the high prevalence of cholangiocarcinoma in Thailand.
