Label-free photothermal optical coherence microscopy to locate desired regions of interest in multiphoton imaging of volumetric specimens.

Biochip-based research is currently evolving into a three-dimensional and large-scale basis similar to the in vivo microenvironment. For the long-term live and high-resolution imaging in these specimens, nonlinear microscopy capable of label-free and multiscale imaging is becoming increasingly important. Combination with non-destructive contrast imaging will be useful for effectively locating regions of interest (ROI) in large specimens and consequently minimizing photodamage. In this study, a label-free photothermal optical coherence microscopy (OCM) serves as a new approach to locate the des

Accelerating Cancer Histopathology Workflows with Chemical Imaging and Machine Learning.

UNLABELLED: Histopathology has remained a cornerstone for biomedical tissue assessment for over a century, with a resource-intensive workflow involving biopsy or excision, gross examination, sampling, tissue processing to snap frozen or formalin-fixed paraffin-embedded blocks, sectioning, staining, optical imaging, and microscopic assessment. Emerging chemical imaging approaches, including stimulated Raman scattering (SRS) microscopy, can directly measure inherent molecular composition in tissue (thereby dispensing with the need for tissue processing, sectioning, and using dyes) and can use ar

Label-free imaging of amyloid plaques in Alzheimer's disease with stimulated Raman scattering microscopy.

One of the key pathological features of Alzheimer's disease (AD) is the existence of extracellular deposition of amyloid plaques formed with misfolded amyloid-β (Aβ). The conformational change of proteins leads to enriched contents of β sheets, resulting in remarkable changes of vibrational spectra, especially the spectral shifts of the amide I mode. Here, we applied stimulated Raman scattering (SRS) microscopy to image amyloid plaques in the brain tissue of an AD mouse model. We have demonstrated the capability of SRS microscopy as a rapid, label-free imaging modality to differentiate misfold

Live-Cell Bioorthogonal Chemical Imaging: Stimulated Raman Scattering Microscopy of Vibrational Probes.

Innovations in light microscopy have tremendously revolutionized the way researchers study biological systems with subcellular resolution. In particular, fluorescence microscopy with the expanding choices of fluorescent probes has provided a comprehensive toolkit to tag and visualize various molecules of interest with exquisite specificity and high sensitivity. Although fluorescence microscopy is currently the method of choice for cellular imaging, it faces fundamental limitations for studying the vast number of small biomolecules. This is because common fluorescent labels, which are relativel

Mirror‑Induced Field Compression Drives Tunable Strong Exciton-Plasmon Polaritons in WS2/h-BN/Au Heterostructures.

Deep‑subwavelength confinement and strong coupling at visible frequencies are central to scalable nanophotonic and quantum technologies. While transition metal dichalcogenides are promising candidates for such confinement, achieving tunable strong coupling often relies on patterned plasmonic nanostructures or chemical modification of the excitonic material. Furthermore, in metal-coupled systems, the distinct role of metallic mirror-induced electromagnetic confinement is often experimentally obscured by exciton-plasmon energy hybridization. Here we introduce a vdW-integrated, chemically non-inv

Characterization of pathological thyroid tissue using polarization-sensitive second harmonic generation microscopy.

Polarization-sensitive second harmonic generation (SHG) microscopy is an established imaging technique able to provide information related to specific molecular structures including collagen. In this investigation, polarization-sensitive SHG microscopy was used to investigate changes in the collagen ultrastructure between histopathology slides of normal and diseased human thyroid tissues including follicular nodular disease, Grave's disease, follicular variant of papillary thyroid carcinoma, classical papillary thyroid carcinoma, insular or poorly differentiated carcinoma, and anaplastic or un

Two-photon microscopy of fungal keratitis-affected rabbit cornea ex vivo using moxifloxacin as a labeling agent.

Two-photon microscopy (TPM) is a three dimensional (3D) microscopic technique based on nonlinear two-photon fluorescence, which has been tested as an alternative to reflectance confocal microscopy (RCM) for detecting fungal keratitis via optical imaging. Although TPM provided images with better contrast than RCM for fungal keratitis, its imaging speed was relatively low because of weak intrinsic signal. Moxifloxacin, a Food and Drug Administration (FDA)-approved antibiotic, was recently used as a cell-labeling agent for TPM. In this study, moxifloxacin was used to label fungal cells for TPM im

High accuracy astigmatic-focusing system for laser targets.

An accurate location of the focal position with respect to a solid target is a key task for different applications, for instance, in laser driven plasma acceleration for x-ray generation where minimum required intensities are above 1014W/cm2. For such practical applications, new approaches for focus location and target delivery techniques are needed to achieve the required intensity, repeatability, and stability. There are different techniques to accomplish the focusing and target positioning task such as interferometry-, microscopy-, astigmatism-, and nonlinear-optics-based techniques, with t

Tunable polaritonic topologies generated by non-local photonic modes.

Photonic skyrmions are topological textures that exhibit remarkable resilience to environmental perturbations and support deeply subwavelength features, making them promising candidates for high-resolution microscopy, optical computing devices and ultrahigh-density information encoding. However, in contrast to free-space optical skyrmions, all existing approaches to generate polaritonic field skyrmions are limited by a lack of dynamic tunability. In general, without engineering the phase of the incident light, both their lattice site diameter and total topological charges remain fixed after fa

A strategic guide of techniques for biomedical and tissue engineering applications to measure mechanical properties of soft matter, eye and skin.

A comprehensive understanding of tissue mechanics at the microscale is critical for advancing personalised therapies, controlled drug release, and tissue engineering. Characterising the mechanical properties of complex, soft biological materials-particularly multilayered and anisotropic organs such as the skin and eye-remains a significant challenge due to their variable water content and scale-dependent behaviour. Traditional continuum models and linear material responses often fail to capture the dynamic and nonlinear nature of these tissues under physiologically relevant conditions. This re

Multiphoton Luminescence Imaging: A Label-Free Tool for Visualizing the Long-Term Fate of Gold Nanoparticles in Tissue.

Gold nanoparticles (AuNPs) are increasingly used in applications across the biomedical domain, yet their long-term biodistribution and biocompatibility remain poorly understood. Conventional brightfield microscopy imaging techniques often fail to detect AuNPs due to optical diffraction limits and lack of chromogenic contrast. Understanding the biodistribution and ultimate fate of these nonbiodegradable NPs is crucial for further development of AuNP-based therapeutics and diagnostics. Here, we present a label-free multiphoton luminescence (MPL) imaging workflow that enables sensitive detection

Above-twofold quantum super-resolution microscopy enabled by multiple idler passes with entangled biphotons.

Spatial resolution defines the fundamental limit of how precisely optical imaging systems can distinguish details within an object. Recent advances in quantum imaging have surpassed classical resolution limits by exploiting entanglement. However, experimental demonstrations of quantum enhancement above two times using entangled biphotons have thus far been limited to nonimaging experiments that use replication of the object or nonlinearity-both are impractical and inefficient. Here, without resorting to either, we enhance the classical resolution using entangled photon pairs by either two- or

Sensing Volatile Pollutants with Spin-Coated Films Made of Pillar[5]arene Derivatives and Data Validation via Artificial Neural Networks.

Different types of solvents, aromatic and aliphatic, are used in many industrial sectors, and long-term exposure to these solvents can lead to many occupational diseases. Therefore, it is of great importance to detect volatile organic compounds (VOCs) using economic and ergonomic techniques. In this study, two macromolecules based on pillar[5]arene, named P[5]-1 and P[5]-2, were synthesized and applied to the detection of six different environmentally volatile pollutants in industry and laboratories. The thin films of the synthesized macrocycles were coated by using the spin coating technique

A Three-Dimensional Organic-Inorganic Hybrid Perovskite-Type Molecular Ferroelectric Material [3.2.2-H2dabcn]Rb(NO3)3.

Replacing halide ions with nitrate ions to construct hybrid molecular ferroelectrics provides a new strategy for optimizing the properties of ferroelectric materials. In this work, an organic-inorganic hybrid ferroelectric crystal [3.2.2-H2dabcn]Rb(NO3)3 (1) is successfully synthesized by reacting the spherical molecule 1,5-diazabicyclic[3.2.2]nonane with RbNO3 in a 1:1 molar ratio in dilute nitric acid solution. The compound exhibits a phase transition above room temperature (317 K), accompanied by a pronounced step-like dielectric anomaly and reversible nonlinear optical switching behavior.

Ex vivo nonlinear microscopy imaging of Ehlers-Danlos syndrome-affected skin.

Ehlers-Danlos syndrome (EDS) is the name for a heterogenous group of rare genetic connective tissue disorders with an overall incidence of 1 in 5000. The histological characteristics of EDS have been previously described in detail in the late 1970s and early 1980s. Since that time, the classification of EDS has undergone significant changes, yet the description of the histological features of collagen morphology in different EDS subtypes has endured the test of time. Nonlinear microscopy techniques can be utilized for non-invasive in vivo label-free imaging of the skin. Among these techniques,
