Countering the Faltering Ability of Warming Oceans to Absorb CO2 by Adding Silica and Other Nanoparticles.

Diatoms play a central role in marine CO2-fixation and the marine food web, yet their survival is increasingly threatened by intensifying marine heatwaves. Here, we demonstrate that engineered nanoparticles (ENPs) can significantly enhance diatom thermotolerance. The diatom Skeletonema costatum pre-exposed to 0.1 mg/L SiO2 NPs exhibited significantly higher survival rates (from 43 to 64-71%), increased biomass, enhanced photosynthetic pigment content, and a bolstered antioxidant system compared to untreated, heat-stressed controls. Multiomics analyses revealed that SiO2 NPs precondition the ce

Factors regulating methyl mercury uptake in a cyanobacterium.

The simultaneous addition of dithiothreitol (DTT), mercaptoethanol, and glutathione (30 microM each) and CH3Hg+ to Nostoc calcicola cells reduced CH3Hg+ uptake in the order GSH > DTT > mercaptoethanol. However, the preexposure of cyanobacterial cells to similar thiols resulted in different pattern of CH3Hg+ uptake in the sequence: GSH > mercaptoethanol > DTT. Light-grown cyanobacterial cells demonstrated a faster initial uptake of CH3Hg+ (rate 0.619 mumol CH3Hg+ mg-1 protein min-1, 10 min) with a biphasic pattern saturating at 30 min (bioconcentration factor = 2.7 x 10(3)). 3-(3,4-Dichlorophen

Lesser-Known Cyanotoxins: A Comprehensive Review of Their Health and Environmental Impacts.

Cyanobacteria, also known as blue-green algae, are a diverse phylum of photosynthetic, Gram-negative bacteria and one of the largest microbial taxa. These organisms produce cyanotoxins, which are secondary metabolites that can have significant impacts on both human health and the environment. While toxins like Microcystins and Cylindrospermopsins are well-documented and have been extensively studied, other cyanotoxins, including those produced by Lyngbya and Nostoc, remain underexplored. These lesser-known toxins can cause various health issues in humans, including neurotoxicity, hepatotoxicit

Toxicological effects of cyanobacterial metabolites on zebrafish larval development.

Freshwater cyanobacteria are known worldwide for their potential to produce toxins. However, these organisms are also found in marine, terrestrial and extreme environments and produce unique compounds, other than toxins. Nevertheless, their effects on biological systems are still barely known. This work tested extracts of different cyanobacterial strains against zebrafish (Danio rerio) larvae and analyzed their metabolomic profiles using liquid chromatography combined with mass spectrometry. Strains Desertifilum tharense, Anagnostidinema amphibium, and Nostoc sp. promoted morphological abnorma

Abundance, Diversity and Metabolomic Profiles of Soil Cyanobacteria-Dominated Microbial Communities Under Long-Term Differential Management in an Olive Orchard.

This study assessed the effects of 22 years of sustainable (Smng) versus conventional (Cmng) management on cyanobacteria-enriched microbial communities in a Mediterranean olive orchard. Surface soils were selectively cultivated in nitrogen-free Bristol medium to obtain cultivable cyanobacteria-dominated, nitrogen-fixing assemblages for molecular and metabolomic analyses. Cyanobacteria-enriched assemblages derived from Smng soils showed greater phylogenetic diversity and distinct community clustering under N-free cultivation conditions. At the cyanobacterial genus level, Cmng-derived communitie

Structure, biosynthesis, and bioactivity of nostolysamides.

A recent genome mining study identified class II lanthipeptides encoded in Nostoc punctiforme PCC 73102 that contain acyl groups conjugated to Lys side chains. The structure and bioactivity of these peptides, named nostolysamides, were not determined. In this study, we produced the nostolysamides by co-expression of the NpuA precursor peptide with an N-terminal SUMO tag with the class II lanthipeptide synthetase NpuM in Escherichia coli. All four lanthionine and methyllanthionine residues were shown to have the DL configuration by Marfey's analysis. Tandem mass spectrometry and mutagenesis stu

Natural Pigments and Biogas Recovery from Microalgae Grown in Wastewater.

This study assessed the recovery of natural pigments (phycobiliproteins) and bioenergy (biogas) from microalgae grown in wastewater. A consortium of microalgae, mainly composed by Nostoc, Phormidium, and Geitlerinema, known to have high phycobiliproteins content, was grown in photobioreactors. The growth medium was composed by secondary effluent from a high rate algal pond (HRAP) along with the anaerobic digestion centrate, which aimed to enhance the N/P ratio, given the lack of nutrients in the secondary effluent. Additionally, the centrate is still a challenging anaerobic digestion residue s

Assessing activity of Hepatitis A virus 3C protease using a cyclized luciferase-based biosensor.

Hepatitis A is an acute infection caused by Hepatitis A virus (HAV), which is widely distributed throughout the world. The HAV 3C cysteine protease (3Cpro), an important nonstructural protein, is responsible for most cleavage within the viral polyprotein and is critical for the processes of viral replication. Our group has previously demonstrated that HAV 3Cpro cleaves human NF-κB essential modulator (NEMO), a kinase required in interferon signaling. Based on this finding, we generated four luciferase-based biosensors containing the NEMO sequence (PVLKAQ↓ADIYKA) that is cleaved by HAV 3Cpro an

Detection and Quantification of Label-Free Infectious Adenovirus Using a Switch-On Cell-Based Fluorescent Biosensor.

Reliable and fast viral detection and quantification protocols are a requirement for the advance of basic research and clinical approaches with wild type or recombinant viruses. However, available cell-based assays are either time-consuming or require labeled viral particles, which may alter virus biology or pose safety issues in clinical applications. Since adenoviruses constitute a major healthcare burden but also, when engineered, widely used vectors in vaccination and gene and oncolytic therapies, herein we developed a genetically encoded switch-on fluorescent biosensor consisting of a cyc

Metagenomic insights into microbial biodeterioration of terracotta in the 18th-century-old temple Jor Mandir.

Microbial colonisation represents a significant yet underexplored factor contributing to the biodeterioration of terracotta heritage structures in tropical environments. Here, we present a comprehensive multi-analytical investigation of the Jor Mandir complex, an eighteenth-century terracotta monument in eastern India. Culture-based analysis revealed morphologically distinct cyanobacteria that were not detected by metagenomics, underscoring the complementarity of the two approaches. Amplicon sequencing of a composite biofilm sample revealed a microbial community dominated by Cyanobacteria (44.

A Förster Resonance Energy Transfer (FRET)-Based Immune Assay for the Detection of Microcystin-LR in Drinking Water.

Cyanobacteria bloom is the term used to describe an abnormal and rapid growth of cyanobacteria in aquatic ecosystems such as lakes, rivers, and oceans as a consequence of anthropic factors, ecosystem degradation, or climate change. Cyanobacteria belonging to the genera Microcystis, Anabaena, Planktothrix, and Nostoc produce and release toxins called microcystins (MCs) into the water. MCs can have severe effects on human and animal health following their ingestion and inhalation. The MC structure is composed of a constant region (composed of five amino acid residues) and a variable region (comp

Current states and challenges of salt-affected soil remediation by cyanobacteria.

Natural and human activities lead to soil degradation and soil salinization. The decrease of farmlands threatens food security. There are approximately 1 billion ha salt-affected soils all over of world, which can be made available resources after chemical, physical and biological remediation. Nostoc, Anabaena and other cyanobacterial species have outstanding capabilities, such as the ability to fix nitrogen from the air, produce an extracellular matrix and produce compatible solutes. The remediation of salt-affected soil is a complex and difficult task. During the past years, much new researc

New Approach for Controlling Snail Host of Schistosoma mansoni, Biomphalaria alexandrina with Cyanobacterial Strains-Derived C-Phycocyanin.

Schistosomiasis is one of the major communicable diseases of public health and socioeconomic importance in the developing world. It is a waterborne disease in which Biomphalaria alexandrina snails are known to be the intermediate molluscan host for Schistosoma mansoni: the causative agent of human intestinal schistosomiasis. Therefore, snail control is one of the cornerstones of schistosomiasis control programs. Several methods have been used to eliminate snail hosts. One of these methods is chemical molluscicides, which have undesirable effect to nontarget organisms. Consequently, the search
