Mesiodens with an unusual morphology and multiple impacted supernumerary teeth in a non-syndromic patient.

Supernumerary teeth are a relatively frequent disorder of odontogenesis characterized by an excess number of teeth. Mesiodens is the most common type of supernumerary tooth found in the premaxilla between the two central incisors. They can be supplemental (resembling natural teeth), conical, tuberculate or molariform. We present the case of a 19 year-old girl who presented with a mesiodens of an unusual morphology and multiple impacted supernumerary teeth not associated with any syndrome.

Additional criteria for EPR dosimetry using tooth enamel.

Currently, EPR measurements are based on the assumption that odontogenesis (the series of events between the bud formation stage until the complete maturation of the tooth) is finished as soon as the tooth erupts. Consequently, it is also assumed that the hydroxyapatite concentration of the enamel (source of free radicals) does not depend on tooth age. However, the present work provides evidence that odontogenesis does not end after tooth eruption but continues for several years after eruption. Fifty-nine molars and pre-molars were analyzed by EPR spectroscopy. Tooth enamel samples were irradi

Differential Expression of Long Noncoding RNAs in Normal and Inflamed Human Dental Pulp.

INTRODUCTION: Dental pulp inflammation is an excellent model for the interaction between tissue inflammation and regenerative processes. It is worthwhile to better understand molecular signaling of repair and regeneration in inflammatory processes. Emerging evidence suggests that long noncoding RNA (lncRNA) participates in immune system inflammatory processes. Here we investigate the expression of lncRNAs in pulpitis, the inflammation of dental pulp tissue, and identify lncRNAs that possibly participate in inflammation responses and odontogenesis. METHODS: Integrated comparative lncRNA microa

Exploring the genetics, mechanisms, and therapeutic innovations in non-syndromic tooth agenesis.

Tooth agenesis is the congenital absence of one or more teeth in the normal series due to failures during dental development in the odontogenesis process. Although tooth development mechanisms are more precise in the literature, the etiology of non-syndromic tooth agenesis remains partially unknown. Mutations in genes that regulate the transcription factors involved in tooth development are associated with this condition. Despite advances in genetic research, questions remain about whose understanding might enable more precise and customized treatments. This study aimed to explain the molecula

Bidirectional Differentiation of Human-Derived Stem Cells Induced by Biomimetic Calcium Silicate-Reinforced Gelatin Methacrylate Bioink for Odontogenic Regeneration.

Tooth loss or damage is a common problem affecting millions of people worldwide, and it results in significant impacts on one's quality of life. Dental regeneration with the support of stem cell-containing scaffolds has emerged as an alternative treatment strategy for such cases. With this concept in mind, we developed various concentrations of calcium silicate (CS) in a gelatin methacryloyl (GelMa) matrix and fabricated human dental pulp stem cells (hDPSCs)-laden scaffolds via the use of a bioprinting technology in order to determine their feasibility in promoting odontogenesis. The X-ray dif

Retinoic Acid Signaling and the Zebrafish Dentition During Development and Evolution.

Explaining how the extensive diversity in form of vertebrate teeth arose in evolution and the mechanisms by which teeth are made during embryogenesis are intertwined questions that can merit from a better understanding of the roles of retinoic acid (RA) in tooth development. Pioneering studies in rodents showed that dietary vitamin A (VA), and eventually RA (one of the major active metabolites of VA), are required for proper tooth formation and that dentin-forming odontoblast cells seem to be especially sensitive to changes in RA levels. Later, rodent studies further indicated that RA signalin

Expression, gene regulation, and roles of Fisp12/CTGF in developing tooth germs.

Odontogenesis involves multiple events, including tissue-tissue interactions, cell proliferation, and cell differentiation, but the underlying mechanisms of regulation are far from clear. Because Fisp12/CTGF is a signaling protein involved in similar events in other systems, we asked whether it is expressed in developing tooth germs and what roles it may have. Indeed, Fisp12/CTGF transcripts were first expressed by dental laminas, invaginating epithelium, and condensing mesenchyme at the bud stage, and then became abundant in enamel knot and preameloblasts. Fisp12/CTGF was present not only in

Generation of a MSX1 knockout human embryonic stem cell line using CRISPR/Cas9 technology.

The MSX1 gene encodes a transcriptional repressor and plays important roles in limb-pattern formation, craniofacial development, and odontogenesis during vertebrate embryogenesis. Previous studies demonstrated that human MSX1 mutations are associated with tooth agenesis, orofacial clefting, and nail dysplasia. Here, we generated a MSX1 knockout cell line from human embryonic stem cell (hESC) line (H9) by CRISPR/cas9-mediated gene targeting. This cell line may serve as a valuable in vitro cell model for MSX1 mutation-related diseases and help to gain more insight into the biological function of

Interrupted development of dentition in children receiving bone marrow transplantation for acute lymphocytic Leukemia: a case series.

This case series depicts dental anomalies that may develop in children who have undergone bone marrow transplantation (BMT) for acute lymphoblastic leukemia (ALL). The most common finding in these patients was root stunting; other abnormalities included microdontia, hypodontia, taurodontia, caries, enamel pearls, and pulpal calcification. Recognition of these adverse effects of BMT on odontogenesis, as demonstrated on panoramic radiograph images, will allow healthcare providers to explain to parents and patients the possible dental outcomes associated with BMT and to optimize their dental heal

Region-specific gene expression profiling of early mouse mandible uncovered SATB2 as a key molecule for teeth patterning.

Mammalian dentition exhibits distinct heterodonty, with more simple teeth located in the anterior area of the jaw and more complex teeth situated posteriorly. While some region-specific differences in signalling have been described previously, here we performed a comprehensive analysis of gene expression at the early stages of odontogenesis to obtain complete knowledge of the signalling pathways involved in early jaw patterning. Gene expression was analysed separately on anterior and posterior areas of the lower jaw at two early stages (E11.5 and E12.5) of odontogenesis. Gene expression profil

A rare case of dens evaginatus and dens invaginatus in the same tooth, with a review of treatment options.

Dens evaginatus (DE) and dens invaginatus (DI) are rare developmental anomalies of tooth formation. These variations in odontogenesis have been individually observed and reported in the literature. This report describes the radiographic appearance and management of a 16-year-old boy who came to the clinic with a rare occurrence of both DE and DI in the same tooth and examines the criteria used in identifying and classifying such anomalies. This report also discusses the importance of early identification and efficient management of dental anomalies, which can prevent irreversible pulpal pathol

The role of epithelium in the development, structure and function of the tissues of tooth support.

The roles of epithelium in the development, structure and function of the tissues of tooth support are reviewed. Epithelium is involved in initiating odontogenesis which includes the tissues of tooth support and this role is discussed. Particular attention is paid to Hertwig's epithelial root sheath and its participation in the formation of the hyaline layer on the root surface as well as its possible role in initiating the differentiation of cementoblasts. The possible functions of the epithelial cell rests are reviewed and it is concluded that as yet no function can be ascribed to them. Evid

Trps1 transcription factor regulates mineralization of dental tissues and proliferation of tooth organ cells.

Mutations of the TRPS1 gene cause trichorhinophalangeal syndrome (TRPS), a skeletal dysplasia with dental abnormalities. TRPS dental phenotypes suggest that TRPS1 regulates multiple aspects of odontogenesis, including the tooth number and size. Previous studies delineating Trps1 expression throughout embryonic tooth development in mice detected strong Trps1 expression in dental mesenchyme, preodontoblasts, and dental follicles, suggesting that TRPS dental phenotypes result from abnormalities in early developmental processes. In this study, Trps1+/- and Trps1-/- mice were analyzed to determine

Rethinking Ameloblastic Fibroma and Fibro-odontoma: A Serie of 6 Cases and Reclassification Proposal.

PURPOSE: The classification of mixed odontogenic tumors-specifically ameloblastic fibroma (AF), ameloblastic fibro-odontoma (AFO), and odontoma-remains controversial. The current WHO classification emphasizes the presence of dental hard tissues but overlooks the distinction between aberrant inductive activity and maturation resembling normal odontogenesis. This has led to diagnostic ambiguity and inconsistent management strategies. This study aims to propose a biologically grounded reclassification based on histological, developmental, and molecular criteria. METHODS: We conducted a conceptua
