[Investigation of Cryptosporidium oocysts in elementary school students in Mersin].

Cryptosporidium parvum, a protozoon, is an obligate intracellular parasite which can cause fatal diarrheal disease in immunocompromised individuals. Cryptosporidium parvum and Cryptosporidium hominis are usually known to be transmitted from fecally contaminated drinking and tap waters. Because oocysts can be detected in asymptomatic healthy individuals and no safe and effective therapy for cryptosporidial enteritis is available, the importance of cryptosporidiosis is increased. In this study, stool specimens (n=72) were collected from children, 8-12 years in age, in four elementary school in M

A rapid method for producing highly purified Cryptosporidium parvum oocysts.

Most procedures that have been described for purifying Cryptosporidium parvum oocysts are designed to either identify the parasites in clinical specimens or isolate oocysts from a small volume of feces from infected animals. The present study describes a rapid method for purifying high numbers of C. parvum oocysts from feces of infected calves that contains minimal contaminating fecal material and bacteria. The isolation method is based on differential flotation of C. parvum oocysts in NaCl, followed by ether extraction to solubilize lipids in calf feces. This procedure regularly yields > 10(9

Adhesion kinetics of viable Cryptosporidium parvum oocysts to quartz surfaces.

The transport and deposition (adhesion) kinetics of viable Cryptosporidium parvum oocysts onto ultrapure quartz surfaces in a radial stagnation point flow system were investigated. Utilizing an optical microscope and an image-capturing device enabled real time observation of oocyst deposition behavior onto the quartz surface in solutions containing either monovalent (KCl) or divalent (CaCl2) salts. Results showed a significantly lower oocyst deposition rate in the presence of a monovalent salt compared to a divalent salt. With a monovalent salt, oocyst deposition rates and corresponding attach

Inactivation of Cryptosporidium parvum oocysts in cider by flash pasteurization.

Cryptosporidium parvum is a well-recognized pathogen of significant medical importance, and cider (apple juice) has been associated with foodborne cryptosporidiosis. This study investigated the effect of flash pasteurization on the viability of contaminant C. parvum oocysts. Cider inoculated with oocysts was heated at 70 or 71.7 degrees C for 5, 10, or 20 s, and oocyst viability was measured by a semiquantitative in vitro infectivity assay. By infecting multiple wells of confluent Madin-Darby bovine kidney cells with serial dilutions of heat-treated oocysts and examining infected cells by indi

[Improvement of amplification method for Cryptosporidium parvum oocysts from mice].

OBJECTIVE: To establish a simplified amplification method for obtaining a large number of purified Cryptosporidium parvum oocysts from infected C57BL/6N mice. METHODS: All mice in the experimental groups were immunosuppressed by given different concentrations of dexamethasone phosphate added in drinking water throughout the experiment. The recovery and purity of the oocysts obtained using different purification methods was compared. The infectivity of the oocysts obtained from the same origin but different animals and different purification methods in a bovine fallopian tube epithelial cell c

Morphological and genetic characterisation of Cryptosporidium oocysts from domestic cats.

Faecal samples were collected from domestic cats in the metropolitan area of the city of Perth, Western Australia, and screened for the presence of Cryptosporidium by both microscopy and PCR. Of 162 samples screened, two were positive for Cryptosporidium (a prevalence of 1.2%). Sample Ct33 was from an 18-month-old female and sample Ct131 from a 12-month-old female. Morphological studies revealed oocysts with an average size of 4.6 x 4.0 microm, smaller in size than isolates typically seen in humans (5.0 x 4.5 microm). Sequence analysis of PCR products showed sequences from cat isolates to be d

Fuchsin fluorescence and autofluorescence in Cryptosporidium, Isospora and Cyclospora oocysts.

Cryptosporidium parvum and Isospora belli oocysts stained with carbol-fuchsin, as in a modified Ziehl Neelsen technique, fluoresce bright red under green light (546nm). Cryptosporidium oocysts tend to fluoresce more brightly the less intensely stained they appear under transmitted light; this is not the case with Isospora. Fuchsin-stained Cyclospora cayetanensis oocysts fluoresce rather dimly, but those not taking the dye retain their typical autofluorescence. Cryptosporidium and Isospora oocysts are also autofluorescent, appearing violet under u.v. light (365 nm), and green under violet (405

Flow cytometric detection of Cryptosporidium oocysts in human stool samples.

Cryptosporidium parvum is an important pathogen that causes diarrhea in virtually all human populations. Improved diagnostic methods are needed to understand the risk factors, modes of transmission, and impact of cryptosporidiosis. In the present study, we fluorescently labeled and counted C. parvum oocysts by flow cytometry (FC) and developed a simple and efficient method of processing human stool samples for FC analysis. Formed stool (suspended in phosphate-buffered saline) from an asymptomatic, healthy individual was seeded with known concentrations of oocysts, and oocysts were labeled with

Infectivity of preserved Cryptosporidium parvum oocysts for immunosuppressed adult mice.

The present study was undertaken to determine the infectivity of Cryptosporidium parvum oocysts for immunosuppressed adult C57BL/6N mice after the oocysts had been stored from 1-48 months at 4 degrees C in 2.5% potassium dichromate. All mice inoculated with oocysts 1-18 months old developed patent infections, while mice inoculated with older oocysts remained uninfected. The prepatent period was extended from 2 to 6 or 7 days as the storage time for oocysts increased. The finding that C. parvum oocysts remain infective for mice for at least 18 months offers important economic and time-saving ad

Occurrence of oocysts of Cryptosporidium sp. in Larus spp. gulls.

Between November 1990 and February 1991 101 gull faecal samples, collected in central Scotland, and 50 cloacal lavages, from gulls captured at two refuse tips near Durham, England were examined for the presence of Cryptosporidium sp. oocysts. Five of 101 (c 5%) of faecal samples and 11 of 50 (22%) of cloacal lavages contained oocysts, of which 64% and 83%, respectively were considered viable when examined with propidium iodide and 4'-6-diamidino-2-phenylindole. Since there is insufficient evidence to ascribe these oocysts to a recognized species they are therefore referred to as Cryptosporidiu

Detection of Cryptosporidium oocysts in sputum during screening for mycobacteria.

We report a case in which Cryptosporidium oocysts were detected during routine auramine screening of a sputum specimen for acid-fast bacilli. The patient was a 28-year-old male with acquired immune deficiency syndrome and intestinal cryptosporidiosis. Microbiology laboratory workers should be alert to the possibility that these oocysts may be present in sputum specimens from patients with acquired immune deficiency syndrome.

Cryptosporidium Oocyst Purification Using Discontinuous Gradient Centrifugation.

Many laboratory studies in cryptosporidial research require a source of purified oocysts. Sources can include experimentally infected laboratory animals or from samples collected from naturally infected animals and from clinical cases of human cryptosporidiosis. Purification of oocysts can be accomplished with readily available laboratory equipment including table-top centrifuges and microcentrifuges. Following purification, oocysts can be stored in antibiotic-supplemented buffers or in 2.5% aqueous potassium dichromate for over 6 months. Ultimately, oocyst viability and infectivity declines t

[Study of prevalence of Toxoplasma gondii oocysts in the cats of Nouakchott].

The aim of the work was to evaluate the prevalence of Toxoplasma gondii oocyst shedding on a sample of 100 cats in five districts of Nouakchott, Mauritania. The faecal flotation method revealed that 23% ± 0.08 of cats which underwent the test excreted oocysts and the prevalence was influenced by age and sex. Excretion rates were significantly higher in neighborhoods of Basra (27.8 ± 0.2), Elmina (25 ± 0.13) and Netegue (20 ± 0.35). The average parasite number was less than 5 oocysts/5 g faeces. Hence, the results suggest that cats have an important role in the transmission of the zoonosis in N

Survival of Cryptosporidium parvum oocysts in the presence of hydrated lime.

To investigate the effects of hydrated lime on the survival of Cryptosporidium oocysts, the percentage viability of oocysts was assessed using fluorescent in situ hybridisation. In the absence of lime and with lime at a concentration of 1 per cent, there was a gradual decline in oocyst viability during the 10-day trial. Although the addition of 5 or 10 per cent lime caused the total number of oocysts to decrease, there appeared to be an increase in the proportion of potentially viable oocysts.

Outbreak of waterborne cryptosporidiosis associated with low oocyst concentrations.

Water treatment plants in the United Kingdom at significant risk of cryptosporidiosis and using conventional filtration methods have been required to install 24-h monitoring systems since April 2000. No major waterborne outbreaks have been described since 2001. Small outbreaks have been associated with water. This paper describes such an outbreak. Data from a local multi-agency surveillance system was used to describe the outbreak, including mapping cases against water supply zones. A case-control study investigated hypotheses raised. All cases were genotype 1. Early cases were in the supply z

Occurrence of Cryptosporidium oocysts and Giardia cysts in sewage in Norway.

Samples of sewage influent from 40 sewage treatment works (STW) throughout Norway were examined for Cryptosporidium oocysts and Giardia duodenalis cysts. Both parasites were detected frequently (80% of STW were Cryptosporidium positive; 93% of STW were Giardia positive) and at maximum concentrations of > 20,000 parasites/liter. The data suggest giardiasis is more widespread, and/or occurs with greater infection intensity, than cryptosporidiosis in Norway. STW serving higher person equivalents were more likely to be positive and had higher parasite concentrations. Parasite concentrations were u

Detection and differentiation of Cryptosporidium oocysts in water by PCR-RFLP.

Consumption of contaminated water has been implicated as a major source of Cryptosporidium infection in various outbreak investigations and case control studies. Surveys conducted in various regions of the United States demonstrated the presence of Cryptosporidium oocysts in 67-100% of wastewaters, 24-100% of surface waters, and 17-26.8% of drinking waters. The identity and human infective potential of these waterborne oocysts are not known, although it is likely that not all oocysts are from human-infecting Cryptosporidium species. Likewise, the source of the oocyst contamination is also not

Diarrhoea associated with cryptosporidial oocyst shedding in a quarterhorse stallion.

Cryptosporidial oocyst shedding was detected in a 2.5-year-old Quarterhorse stallion with diarrhoea. Based on the detection of cryptosporidial oocysts, and the absence of other aetiological agents or enteropathogens, cryptosporidiosis was presumptively diagnosed. This case was unusual because cryptosporidial oocyst shedding is primarily found in some immunocompromised or immature horses, and has not been reported in healthy mature horses.
