Pharmacotherapy of Pythium insidiosum keratitis - a contemporary update.

INTRODUCTION: Pythium insidiosum keratitis is an emerging, vision-threatening corneal infection that is increasingly recognized worldwide, particularly in tropical and agrarian regions. Owing to its close clinical resemblance to fungal keratitis, delayed diagnosis and inappropriate antifungal therapy frequently result in rapid stromal destruction, therapeutic keratoplasty, and poor visual outcomes. Unlike true fungi, Pythium insidiosum is an oomycete lacking ergosterol in its cell membrane, rendering conventional antifungal agents largely ineffective and necessitating a paradigm shift toward t

Toxic effects in Aphanomyces brasiliensis and zebrafish embryos caused by oomyceticides.

In fish farming, high losses occur during egg incubation and larviculture due to diseases caused by oomycetes. This study aimed to identify the oomycete species that occurs in zebrafish Danio rerio eggs and to evaluate the oomyceticidal effect of copper sulfate, bronopol and methylene blue on the mycelial growth of this organism, as well as to determine the lethal and sublethal toxicity of these compounds in embryos of D. rerio. The isolates were cultivated in yeast-starch medium to determine the concentration necessary to inhibit mycelial growth by 50% (IC50) and 100% (minimum oomyceticidal c

Large-scale tubular and tubular-lamellar intracellular membrane systems of fungi.

Filamentous fungi and oomycetes have four large intracellular membrane systems consisting of tubes, lamellae, or a combination thereof. Such elongated membrane structures can extend for tens of micrometers in hyphal cells, branching and connecting into bundles and networks. These systems are represented by tubular vacuoles, elongated mitochondria, endoplasmic reticulum, and macroinvaginations of the plasma membrane. They are most developed in xylotrophic and mycorrhiza-forming basidiomycetes. This review examines three membrane systems, excluding the macroinvaginations. Various morphotypes of

Immune-pathological response induced by Saprolegnia parasitica in Pangasianodon hypophthalmus and assessment of clinical efficacy of clotrimazole.

Saprolegnia parasitica causes significant morbidity, mortality and economic loss in fish hatcheries and growing culture systems, especially during winter. The present study examined clinical and histopathological alterations in Pangasianodon hypophthalmus experimentally infected with the oomycete and evaluated the clinical efficacy of antifungal drug clotrimazole as bath treatment. Besides development of off-white cotton wool-like fungal growth on fins and skin, invasion of hyphae into underlying musculature and muscle fibre necrosis, experimental infection also induced gill lamellar hyperplas

Lytic Polysaccharide Monooxygenases as Chitin-Specific Virulence Factors in Crayfish Plague.

The oomycete pathogen Aphanomyces astaci, also known as "crayfish plague", is an obligate fungal-like parasite of freshwater crustaceans and is considered responsible for the ongoing decline of native European crayfish populations. A. astaci is thought to secrete a wide array of effectors and enzymes that facilitate infection, however their molecular mechanisms have been poorly characterized. Here, we report the identification of AA15 lytic polysaccharide monooxygenases (LPMOs) as a new group of secreted virulence factors in A. astaci. We show that this enzyme family has greatly expanded in A.

Immunofluorescence of the epizootic ulcerative syndrome pathogen, Aphanomyces invadans, using a monoclonal antibody.

A monoclonal antibody (MAb), designated 3gJC9, was raised against a protein antigen of Aphanomyces invadans, the oomycete pathogen that causes epizootic ulcerative syndrome (EUS). The antigen was expressed on the surface of hyphae and secreted extracellularly. MAb 3gJC9 did not cross-react with other oomycete or fungal pathogens of fish, although it did react to the crayfish plague pathogen A. astaci. The MAb was used for immunofluorescent staining on histological sections of fish infected with EUS, and was found to be more sensitive than conventional staining methods for detecting A. invadans

Development of a Quantitative PCR Assay for Early Detection and Quantification of Phytopythium vexans in Kiwifruit Plant and Soil Affected by Vine Decline Syndrome.

Kiwifruit vine decline syndrome is a soilborne disease affecting Actinidia fruit trees in perennial cropping systems. Since its emergence in 2012, studies have increasingly identified the oomycete Phytopythium vexans as a major causative agent of the disease. Phytopythium vexans is also implicated in complex soilborne disease systems of woody perennial crops, including replant disease in apple and pear. To date, most molecular assays for the detection of P. vexans target the nuclear ribosomal internal transcribed spacer, a region that is ill-suited for distinguishing between closely related oo

Light suppresses sporulation and epidemics of Peronospora belbahrii.

Peronospora belbahrii is a biotrophic oomycete attacking sweet basil. It propagates asexually by producing spores on dichotomously branched sporophores emerging from leaf stomata. Sporulation occurs when infected plants are incubated for at least 7.5h in the dark in moisture-saturated atmosphere at 10-27°C. Exposure to light suppresses spore formation but allows sporophores to emerge from stomata. Incandescent or CW fluorescent light of 3.5 or 6 µmoles.m(2).s(-1) respectively, caused 100% inhibition of spore formation on lower leaf surface even when only the upper leaf surface was exposed to l

Thiamine cross-feeding drives microbial coexistence in the leaf microbiome.

Microbiome homeostasis is crucial for host health and ecosystem function, yet the molecular and ecological mechanisms underlying community assembly and stability remain elusive. Here we uncover a conserved yeast-oomycete association that promotes their coexistence in the leaf microbiome. Using a continental-scale microbiome survey, we identified an asymmetric mutualistic interaction between two eukaryotic hub microbes: the yeast Dioszegia hungarica and the obligate oomycete Albugo laibachii. We show that Dioszegia facilitates Albugo colonization by supplying thiamine via a dedicated membrane p

Modern Taxonomy as the Foundation for Identifying and Managing Fungal and Fungus-Like Plant Pathogens.

Reliable, up-to-date fungal and oomycete taxonomy underpins every aspect of plant pathology by enabling accurate pathogen identification, robust host-range analyses, clear assessment of geographic spread, and the development of effective disease management strategies. Yet, despite major advances in systematics and molecular phylogenetics, the integration and use of current names in applied plant pathology remains inconsistent. We discuss best practices for accurate identification, combining morphology, molecular data, and phylogenetic frameworks, and examine challenges arising from the use of

A Modular Fusion Protein Strategy Confers Broad-Spectrum Resistance Against Phytophthora infestans, Alternaria solani, and Ralstonia solanacearum.

Potato production is severely threatened by the oomycete Phytophthora infestans, the fungus Alternaria solani, and the bacterium Ralstonia solanacearum; however, strategies capable of simultaneously managing these three pathogen classes remain relatively scarce. In this study, we designed a series of signal peptide (SP)-fused recombinant constructs containing four functional modules with distinct antimicrobial and immune-inducing properties: GAFP1-2×FYVE, an oomycete-inhibiting antimicrobial protein; BbAFP1-ErBD, a fungal-suppressive protein fragment; the pathogen-associated molecular pattern

Clinically Important Emerging and Uncommon Fungal Pathogens in Asia: Epidemiology, Antifungal Resistance and Challenges in Diagnosis and Management.

Fungal infections in Asia are influenced by warm, humid climates, high population density and expanding healthcare systems, which together facilitate the emergence, transmission and detection of both common and rare pathogens. While Candida and Aspergillus species remain the leading causes of invasive fungal disease, increasing attention has focused on uncommon yeasts and rare moulds, many of which show intrinsic or acquired antifungal resistance. Advances in molecular diagnostics and susceptibility testing have improved identification of emerging pathogens, including uncommon Candida species

Producing cell-free culture broth of rhamnolipids as a cost-effective fungicide against plant pathogens.

Biosurfactants of rhamnolipids have been enthusiastically investigated for substitutes of synthetic agrochemicals against plant pathogens. However, all such studies have been conducted on rhamnolipids with high purity which have limitations due to high costs. This paper focused on the applicability of rhamnolipid-containing cell-free culture broth. It was found that rhamnolipids in cell-free culture broth of Pseudomonas aeruginosa ZJU211 were largely composed of di-rhamnolipid and mono-rhamnolipid with the ratio varying with culture time. After 96 h of fermentation, the mass ratio of di-rhamno

Structure guided development of phenoxyethanol based N-aryloxyethyl heterocycles for the control of Phytophthora infestans.

BACKGROUND: Late blight caused by the oomycete Phytophthora infestans remains one of the most devastating diseases threatening global potato and tomato production. Overuse of a narrow range of fungicides has sped up resistance, making it urgent to develop new agents that work differently and are safer. RESULTS: In this study, a series of N-aryloxyethyl heterocycles were rationally designed by integrating the aryloxyethyl scaffold of phenoxyethanol with bioactive heterocyclic frameworks. Among the 40 synthesized derivatives, compound 8c exhibited the highest anti-oomycete activity against P. i

Genome Re-Sequencing and Functional Analysis Reveal an α-1,3-Glucosyltransferase Conferring Metalaxyl Resistance in Phytophthora sojae.

Phytophthora and allied oomycete pathogens pose a perennial challenge to global food security through their devastating impact on crop systems. While metalaxyl has demonstrated remarkable efficacy in controlling Phytophthora diseases since its introduction decades ago, the persistent emergence of metalaxyl-resistant strains has severely compromised its field efficacy. Elucidating the genetic determinants underlying resistance mechanisms is critical to developing surveillance strategies and sustainable countermeasures against evolving oomycete resistance. Through experimental evolution, we gene

High Levels of Insensitivity to Phosphonate Fungicides in Pseudoperonospora humuli.

Phosphonate (phosphite; HPO3-2) is fungicidal against oomycetes and certain other organisms. The Fungicide Resistance Action Committee has deemed phosphonate to be at low risk of resistance development, and reduced sensitivity to phosphonate has been reported only occasionally in plant pathogens. Reduced sensitivity to the fungicide fosetyl-Al was documented in the hop downy mildew pathogen, Pseudoperonospora humuli, in the early 2000s, but disease caused by insensitive isolates could still be managed commercially if the fungicide rate was doubled from 2.24 to 4.48 kg/ha. In this research, we

eDNA monitoring as a tool for evaluating the reintroduction of Austropotamobius pallipes after a crayfish plague outbreak.

The crayfish plague, a severe disease caused by the oomycete Aphanomyces astaci, is responsible for most population declines of susceptible crayfish in Europe. This pathogen has been devastating native populations of Austropotamobius pallipes since the 1970s in the Iberian Peninsula. In this study, we report a massive mortality event in one of the most important Spanish populations of A. pallipes. We aimed to: (i) identify the cause of the mortality, and (ii) evaluate the reintroduction viability of the species. Over the course of six months, we used environmental DNA (eDNA) and traditional tr

Ingadosides A-C, acacic acid-type saponins from Inga sapindoides with potent inhibitory activity against downy mildew.

As part of a project aiming at the discovery of environmentally friendly alternatives to copper in organic agriculture, a 96% ethanolic extract from the leaves of Inga sapindoides showed potent inhibitory activity against grapevine downy mildew (Plasmopara viticola) in vitro (MIC100 25 μg/mL). Separation of the n-BuOH soluble fraction by silica gel column chromatography followed by a combination of RP18 and HILIC HPLC resulted in the isolation of a series of bidesmosidic saponins characterized by the presence of a monoterpenoid unit attached to a triterpenoid aglycone, a p-methoxycinnamoyl res

Innovative Delivery of Cu(II) Ions by a Nanostructured Hydroxyapatite: Potential Application in Planta to Enhance the Sustainable Control of Plasmopara viticola.

Downy mildew caused by Plasmopara viticola is probably the most serious disease affecting grapevine (Vitis vinifera), and it is capable of causing consistent yield losses. In organic viticulture, the only acceptable and effective means to control the disease is by applications of copper-based fungicides. However, the use of copper in agriculture is expected to be further restricted by European countries because of its critical ecotoxicological and phytotoxicological profile. Research on ways to reduce the effective amounts of copper by developing innovative formulations as well as optimization

Self-limiting outbreak of crayfish plague in an Austropotamobius pallipes population of a river basin in the Abruzzi region (central Italy).

Crayfish plague, caused by the oomycete Aphanomyces astaci, is a serious disease of European freshwater crayfish and has eliminated entire populations in several European countries. In September 2011, mortality was observed among the Austropotamobius pallipes population of a river basin in the Abruzzi region (central Italy), and A. astaci DNA was detected by PCR in dead crayfish. A systematic survey was carried out to evaluate the spread and the effects of the plague in the river basin. The source of the outbreak remained unknown since North American crayfish species, which frequently act as s
