STX1B variant-specific synaptic dysfunction is associated with network hyperexcitability in human iPSC-derived neurons.

BACKGROUND: Variants in STX1B/syntaxin-1B are linked to a spectrum of fever-associated epilepsy syndromes. While studies in murine models have provided mechanistic insights, their relevance to human disease in a heterozygous context may be limited. METHODS: We investigated two pathogenic STX1B variants using isolated single neurons and neuronal network cultures derived from patient-specific induced pluripotent stem cells. These carried either a de novo p.G226R variant, associated with severe developmental epilepsy, or an InDel variant (p.K45delinsRCMIE/p.L46M) linked to a transient familial s

Business model, pricing, and transparency in neurosurgery and neurology journals.

The business model of scientific journals - gold open access (OA), hybrid, or subscription - creates different financial incentives for publishers, since in the author-pays OA model publication fees (Article Processing Charges, APCs) are collected upon acceptance. This descriptive audit surveys 109 neurosurgery and neurology journals from six major publishing groups (Elsevier, Springer Nature/Wolters Kluwer, Wiley, SAGE, Taylor & Francis, Thieme) and verifies, journal by journal, business model, notoriety indicator (Journal Impact Factor or Scimago SJR), publication fee, and, where publicly av

Access techniques for EVAR: percutaneous techniques and working with small arteries.

Access is a key component of endovascular aneurysm repair (EVAR.) The femoral artery is the most common site of access and is traditionally exposed via open cutdown. Percutaneous access is possible by placing suture-mediated closure devices before up-sizing to large-bore sheaths. Comparison of open cutdown (n = 2,802) with percutaneous femoral access (n = 1,781) in elective EVAR cases using the American College of Surgeons National Surgical Quality Improvement Program database shows that the main advantage of percutaneous access is decreased operative time (159 ± 63 min v 150 ± 68 min; P < .05

Achieving universal health coverage for people with stroke in South Africa: protocol for a scoping review.

INTRODUCTION: Stroke is the second most common cause of death after HIV/AIDS and a significant health burden in South Africa. The extent to which universal health coverage (UHC) is achieved for people with stroke in South Africa is unknown. Therefore, a scoping review to explore the opportunities and challenges within the South African health system to facilitate the achievement of UHC for people with stroke is warranted. METHODS AND ANALYSIS: The scoping review will follow the approach recommended by Levac, Colquhoun and O'Brien, which includes five steps: (1) identifying the research questi

The Mapping of Meditation-Based Interventions with Emphasis on Brain-Derived Neurotrophic Factor in Hypertension: A Scoping Review.

OBJECTIVE: Hypertension is a global target for noncommunicable diseases, and meditation-based interventions (MBIs) benefit patients with hypertension (HTN). The primary objective of this scoping review is to map the globally published MBI studies on patients with HTN. The secondary goal is to identify the role of brain-derived neurotrophic factor (BDNF) in HTN. METHODS: Based on the Arksey and O'Malley protocol of the Joanna Briggs Institute framework for scoping review, 5 electronic databases were searched with search terms related to HTN and meditation. The open-access articles in the Engli

Journal of Global Health's Guidelines for Reporting Analyses of Big Data Repositories Open to the Public (GRABDROP): preventing 'paper mills', duplicate publications, misuse of statistical inference, and inappropriate use of artificial intelligence.

In recent years, global accessibility to large 'big data' repositories that enable 'open research' - such as the UK Biobank, National Health and Nutrition Examination Survey (NHANES), and Global Burden of Disease (GBD) datasets - has created unprecedented opportunities for researchers worldwide to conduct secondary data analyses. This development is particularly beneficial for early-career researchers in low- and middle-income countries (LMICs), as it lets them access large and otherwise costly datasets without the need for local infrastructure, potentially curbing brain drain. However, throug

Greetings from your predatory journal! What they are, why they are a problem, how to spot and avoid them.

Predatory publishers, also known as counterfeit, deceptive, or fraudulent, are organisations that exploit the open-access scholarly model by charging hefty article processing charges (APCs), often without the scientific rigour and ethical processes offered by legitimate journals. Their rising prevalence is of concern to the scientific community, as the consequences of falling victim to them can negatively impact academic integrity and reputation, and render an author's work worthless and untrustworthy. Common characteristics include inappropriate marketing and misrepresentation of services by

Developing treatment guidelines for myasthenia gravis.

A task force of the Myasthenia Gravis Foundation of America recently published a formal consensus statement intended to be a treatment guide for clinicians caring for myasthenia gravis (MG) patients worldwide. Its development was stimulated by the fact that there is generally no accepted standard of care for MG, and no one treatment is best for all MG patients. Also, there are few randomized trials of treatments in current use, and the generalizability of the few trials that have been successful may be difficult. Fifteen international experts in MG participated in the consensus process, which

Gastrointestinal tract access for enteral nutrition in critically ill and trauma patients: indications, techniques, and complications.

BACKGROUND: Enteral nutrition (EN) is a widely used, standard-of-care technique for nutrition support in critically ill and trauma patients. OBJECTIVE: To review the current techniques of gastrointestinal tract access for EN. METHODS: For this traditional narrative review, we accessed English-language articles and abstracts published from January 1988 through October 2012, using three research engines (MEDLINE, Scopus, and EMBASE) and the following key terms: "enteral nutrition," "critically ill," and "gut access." We excluded outdated abstracts. RESULTS: For our nearly 25-year search perio

Automated Processes and Artificial Intelligence in Generating Candidates for Oncology Drug Repurposing: Three-Year Scoping Review of Data.

Oncology conditions are increasingly defined by their molecular profiles, and drug repurposing exploits this new evidence to identify new therapeutic uses of authorized/investigational medicinal products outside their original indication(s). This scoping review mapped original research published between January 2022 and December 2024 to determine the impact of automated processes and artificial intelligence in generating oncology candidates for drug repositioning, and 42 individual projects met the eligibility criteria and were analyzed. The included studies demonstrate extensive use of comput

Free Fall Acrobatics to Reduce Neck Loads During Parachute Opening Shock: Evaluation of an Intervention (ACROPOSE).

INTRODUCTION: Neck pain is a widespread health problem in the skydiver athlete population, epidemiologically linked to repeated exposure to parachute opening shock (POS). During POS, a parachutist is subjected to considerable deceleration forces. This study aims to evaluate the use of preventive free fall acrobatics to reduce the biomechanical load on the neck of parachutists during parachute opening. METHODS AND ANALYSIS: Interventional study with a cross-over, within-subject, repeated measures design. Two consecutive skydives are made on the same day with random ordering of either an 'inter

[The initial experience in supervised injecting facilities in Denmark].

Intravenous drug abuse is a major health concern. The National Board of Health estimates the number of injecting drug users (IDUs) in Denmark to be 13,000. Supervised injecting facilities (SIF) reduce the risk behaviour and bacterial infections and also increase the rate of detoxification and access to health care. The first SIF in Denmark is driven by volunteers and it opened in September 2011. In the first six months there were 1,139 visits. As in earlier studies the IDUs were mainly males with a long history of drug use. Unlike in previously published studies cocaine was the most commonly i

The International Committee on Classification of Corneal Dystrophies (IC3D) was founded in 2005 to address difficulties arising from the outdated nomenclature for corneal dystrophies (CD) and to correct misconceptions in the literature. For each of the 22 CDs, a separate template was created to represent the current clinical, pathological and genetic knowledge of the disease. In addition, each template contains representative clinical photographs as well as light and electron microscopic images and, if available, confocal microscopic and coherence tomographic images of the respective CD. Identifying randomized clinical trials in Spanish-language dermatology journals.

INTRODUCTION: The necessary foundation for good clinical practice lies in knowledge derived from clinical research. Evidence from randomized clinical trials (RCTs) is the pillar on which decisions about therapy are based. OBJECTIVE: To search exhaustively and rigorously to identify RCTs in dermatology journals published in Spanish. METHODS: We located dermatology journals through the following search engines and indexes: PubMed, LILACS, SciELO, Periódica, Latindex, Índice Médico Español, C-17, IBECS, EMBASE, and IMBIOMED. The publication of impaired doctors' identity by Australian and New Zealand tribunals: law, practice, and reform.

For doctors with mental health or substance use disorders, publication of their name and sensitive medical history in disciplinary decisions may adversely impact their health and may reinforce barriers to accessing early support and treatment. This article challenges the view that naming impaired doctors or disclosing the intimate details of their medical condition in disciplinary decisions always serves the public interest in open justice. We analysed and compared the approach of Australian and New Zealand health tribunals to granting orders that suppress the name and/or medical history of im
