Seeing in Tandem: Cis and Trans-Regulation of Tandemly-Duplicated Cone Opsin Genes in Zebrafish.

The molecular basis for vertebrate color vision is expression of spectrally-selective opsin-based photopigments within distinct cone photoreceptor populations. In several primates including humans, and in numerous fish species, genes encoding duplicated, spectrally-divergent cone opsin proteins may be positioned in tandem arrays. This article reviews studies of the regulation of differential expression of these duplicated cone opsin genes in humans and fish, with an emphasis on knowledge derived from the zebrafish model. In humans, the long wavelength-sensitive/medium wavelength-sensitive (LWS

From pineal photoreception to optogenetics: functional colour opponency based on a pineal bistable opsin.

Most animals sense light through opsins, which are photosensitive G protein-coupled receptors that utilize light information to regulate various physiological functions. In vertebrates, a representative example of such functions is vision, which is based on photoreception by visual opsins expressed in rod and cone photoreceptor cells of the retina. Accumulating photobiological evidence has gradually revealed the significance of the molecular properties of these opsins, as well as those of signal transduction-related molecules such as transducin, rhodopsin kinases, and arrestins, and their coll

Consequences of losing metamorphosis on the evolution of opsin and phototransduction gene expression in the eyes of Brook Salamanders.

Metamorphosis can provide an opportunity for animals with complex life cycles to transition between differing environments and still maintain optimal functionality across ontogeny. The structure and function of the amphibian visual system generally remodels to overcome differences in media (water, air) and fluctuations of available light. This in turn causes a shift in the selective pressures acting on the visual system at each life stage. Few comparative studies have evaluated how metamorphosis drives the evolution and expression of visual system genes across closely related species that occu

Therapeutic Efficacy of Multi-Characteristic Opsin Gene Therapy in a Mouse Model of Stargardt Disease.

Optogenetic gene therapy-based treatment offers a unique approach to bypass dysfunctional or degenerated photoreceptors in retinal degenerative disorders. Ambient light-activatable multi-characteristic opsin (MCO) targeted to bipolar cells of the retina has demonstrated partial vision restoration in animal models of retinitis pigmentosa (RP). Here, we describe the potential therapeutic efficacy of intravitreally delivered AAV-carried MCO-010 in a mouse model of Stargardt disease. MCO-010 treatment led to significantly improved behavioral outcomes in the visually guided radial arm water maze. F

A key spectral tuning site of UV-sensitive vertebrate non-visual opsin Opn5.

Opsins are photoreceptive proteins responsible for visual and non-visual photoreceptions in animals. In general, vertebrates have multiple visual and non-visual opsins whose spectral sensitivities range from the UV to the red region. Among these opsins, Opn5 has been widely identified in vertebrates from fishes to primates and functions as a non-visual opsin in various tissues, including the retina and brain. Vertebrate Opn5 has been characterized as a UV-sensitive bistable opsin. Thus, Opn5 provides one of the molecular mechanisms determining the short wavelength limit that vertebrates can de

Hypothalamic opsin 3 suppresses MC4R signaling and potentiates Kir7.1 to promote food consumption.

Mammalian opsin 3 (OPN3) is a member of the opsin family of G-protein-coupled receptors with ambiguous light sensitivity. OPN3 was first identified in the brain (and named encephalopsin) and subsequently found to be expressed in other tissues. In adipocytes, OPN3 is necessary for light responses that modulate lipolysis and glucose uptake, while OPN3 in human skin melanocytes regulates pigmentation in a light-independent manner. Despite its initial discovery in the brain, OPN3 functional mechanisms in the brain remain elusive. Here, we investigated the molecular mechanism of OPN3 function in th

Eye-Mimicked Neural Network Composed of Photosensitive Neural Spheroids with Human Opsin Proteins.

An in vitro model, composed of the short-wavelength human opsins and rhodopsins, is created. Two types of photosensitive neural spheroids are transfected for selective reaction under bluish-purple and green lights. These are employed to two devices with intact neuron and neural-spheroid to study the interaction. By photostimulation, the photosensitive spheroid initiated photoactivation, and the signal generated from its body is transmitted to adjacent neural networks. Specifically, the signal traveled through the axon bundle in narrow gap from photosensitive spheroid to intact spheroid as an e

Distinct Opsin 3 (Opn3) Expression in the Developing Nervous System during Mammalian Embryogenesis.

Opsin 3 (Opn3) is highly expressed in the adult brain, however, information for spatial and temporal expression patterns during embryogenesis is significantly lacking. Here, an Opn3-eGFP reporter mouse line was used to monitor cell body expression and axonal projections during embryonic and early postnatal to adult stages. By applying 2D and 3D fluorescence imaging techniques, we have identified the onset of Opn3 expression, which predominantly occurred during embryonic stages, in various structures during brain/head development. In addition, this study defines over twenty Opn3-eGFP-positive n

A novel small molecule chaperone of rod opsin and its potential therapy for retinal degeneration.

Rhodopsin homeostasis is tightly coupled to rod photoreceptor cell survival and vision. Mutations resulting in the misfolding of rhodopsin can lead to autosomal dominant retinitis pigmentosa (adRP), a progressive retinal degeneration that currently is untreatable. Using a cell-based high-throughput screen (HTS) to identify small molecules that can stabilize the P23H-opsin mutant, which causes most cases of adRP, we identified a novel pharmacological chaperone of rod photoreceptor opsin, YC-001. As a non-retinoid molecule, YC-001 demonstrates micromolar potency and efficacy greater than 9-cis-r

Opsin-G11-mediated signaling pathway for photic entrainment of the chicken pineal circadian clock.

Light is a major environmental signal for entrainment of the circadian clock, but little is known about the intracellular phototransduction pathway triggered by light activation of the photoreceptive molecule(s) responsible for the phase shift of the clock in vertebrates. The chicken pineal gland and retina contain the autonomous circadian oscillators together with the photic entrainment pathway, and hence they represent useful experimental models for the clock system. Here we show the expression of G11alpha, an alpha subunit of heterotrimeric G-protein, in both tissues by cDNA cloning, Northe

A novel rod-like opsin isolated from the extra-retinal photoreceptors of teleost fish.

We have isolated a novel opsin from the pineal complex of Atlantic salmon (Salmo salar) and from the brain of the puffer fish (Fugu rubripes). These extra-retinal opsins share approximately 74% identity at the nucleotide and amino acid level with rod-opsins from the retina of these species. By PCR, we have determined that the novel rod-like opsin is not expressed in the salmon retina, and the retinal rod-opsin is not expressed in the salmon pineal. Phylogenetic analysis suggests that the rod-like opsins arose from a gene duplication event approximately 205 million years ago, a time of consider

Parapinopsin, a novel catfish opsin localized to the parapineal organ, defines a new gene family.

Multiple sites of extraretinal photoreception are present in vertebrates, but the molecular basis of extraretinal phototransduction is poorly understood. This study reports the cloning of the first opsin specifically expressed in the directly photosensitive pineal and parapineal of cold-blooded vertebrates. This opsin, identified in channel catfish and termed parapinopsin, defines a new gene family of vertebrate photopigments and is expressed in a majority of parapinealocytes and a subset of pineal photoreceptor cells. Parapinopsin shows a caudal-rostral gradient of expression within the pinea

Cloning and characterization of the pineal gland-specific opsin gene of marine lamprey (Petromyzon marinus).

We have cloned and sequenced the pineal gland-specific opsin (P opsin) gene, p(Pm), of marine lamprey, P. marinus. The deduced P opsin, P(Pm), consisting of 444 aa, is about 90 aa longer than the orthologous opsins of chicken, pigeon, and American chameleon. The evolution of P(Pm) is characterized not only by a significantly higher rate of aa replacement than the other P opsins but also by a larger number of unique aa replacements at highly conserved sites in the nonTM region. These changes in P(Pm) might have been important in achieving marine lamprey-specific interaction between P(Pm) and ot

Expression of opsin mRNA in normal and vitamin A deficient retinas of the sphingid moth Manduca sexta.

Two distinct opsin-encoding cDNAs, designated MANOP1 and MANOP2, were isolated as 3' fragments from the sphingid moth Manduca sexta. They were obtained by reverse transcription of retinal RNA and amplification with the polymerase chain reaction (PCR) using a degenerate primer designed to an amino-acid sequence conserved in arthropod opsins. The cDNA fragments labelled bands at approximately 1.8 kb on Northern blots of retinal RNA extracts. Levels of opsin message were compared in retinas from normal moths, whose diets were fortified with carotenoid precursors of the Manduca rhodopsin chromopho

Loss of the systemic vitamin A transporter RBPR2 affects the quantitative balance between chromophore and opsins in visual pigment synthesis.

The distribution of dietary vitamin A/all-trans retinol (ROL) throughout the body is critical for maintaining retinoid function in peripheral tissues and for generating visual pigments for photoreceptor cell function. ROL circulates in the blood bound to the retinol binding protein 4 (RBP4) as RBP4-ROL. Two membrane receptors, RBPR2 in the liver and STRA6 in the eye are proposed to bind circulatory RBP4 and this mechanism is critical for internalizing ROL into cells. Here, we present a longitudinal investigation towards the importance of RBPR2 and influence of the diet on systemic retinoid hom

Axin1 stabilizes S-opsin and maintains cone photoreceptor survival by inhibiting GSK3β activity.

To ensure high phototransduction efficiency in the retina, the precise subcellular localization of signaling molecules must be tightly orchestrated by scaffold proteins. Aberrant localization of these scaffold proteins not only disrupts the transition of photoelectrical signals but also triggers endoplasmic reticulum (ER) stress, which leads to photoreceptor apoptosis. However, it is unknown how these proteins are localized to specific subcellular compartments of photoreceptors or how protein mislocalization is coupled with apoptotic signaling. Herein, we observed a specific spatiotemporal exp

The role and signal pathways of opsin 3 in the skin: from light perception to pathophysiological mechanisms.

Opsins are a class of classic G-protein-coupled receptor superfamily members, traditionally known to play a central role in phototransduction in the retina. Recent studies have revealed that various opsins are widely expressed in the skin, making the skin an important peripheral light-sensing organ. Among these, opsin 3 (OPN3), as one of the most abundantly expressed and functionally diverse photosensitive proteins in mammalian skin, has attracted increasing attention. This narrative review systematically outlines the expression profile of OPN3 among photoreceptors in the skin. Building on thi

Exceptional Visual-Opsin Coexpression and Phenotypic Diversity in Outer-Retinal Photoreceptors of Caenophidian Snakes.

Snakes are a valuable yet understudied taxon for investigating evolutionary adaptations in the vertebrate retina. They possess up to three visual pigments: a short-wavelength-sensitive opsin (SWS1), a medium/long-wavelength-sensitive opsin (LWS), and rhodopsin (RH1). Nocturnal snakes have duplex retinas containing both rod and cone photoreceptors, whereas diurnal caenophidian ("advanced") snakes exhibit simplex "all-cone" retinas, lacking morphologically typical rods. In this study, we analyzed photoreceptor morphology in the retinas of caenophidian snakes using high-resolution scanning electr

Blue cone monochromatism: clinical findings in patients with mutations in the red/green opsin gene cluster.

BACKGROUND: X-linked blue cone monochromatism (BCM) has to be differentiated from x-linked cone dystrophy and autosomal recessive rod monochromatism. METHODS: In nine male patients with congenital cone dysfunction (one family, six single cases; age range: 9-55 years), mutations in the red/green opsin gene cluster were confirmed. Clinical findings were analyzed retrospectively. RESULTS: In one family and three single cases, a single red-green hybrid gene was found carrying a Cys203Arg mutation. Two patients had multiple opsin genes, a red/green hybrid gene and at least one green pigment gene,
