Metabolomic changes in the mouse retina after optic nerve injury.

In glaucoma, although axonal injury drives retinal ganglion cell (RGC) death, little is known about the underlying pathomechanisms. To provide new mechanistic insights and identify new biomarkers, we combined latest non-targeting metabolomics analyses to profile altered metabolites in the mouse whole retina 2, 4, and 7 days after optic nerve crush (NC). Ultra-high-performance liquid chromatography quadrupole time-of-flight mass spectrometry and liquid chromatography Fourier transform mass spectrometry covering wide spectrum of metabolites in combination highlighted 30 metabolites that changed

Optic nerve as a source of activated retinal microglia post-injury.

Using mice expressing green fluorescent protein (GFP) from a transgenic CD11c promoter we found that a controlled optic nerve crush (ONC) injury attracted GFPhi retinal myeloid cells to the dying retinal ganglion cells and their axons. However, the origin of these retinal myeloid cells was uncertain. In this study we use transgenic mice in conjunction with ONC, partial and full optic nerve transection (ONT), and parabiosis to determine the origin of injury induced retinal myeloid cells. Analysis of parabiotic mice and fate mapping showed that responding retinal myeloid cells were not derived f

Optic nerve trauma with unilateral edema as a result of head injury.

We encountered a patient who suffered a head injury that translated to the optic nerves, leading to dramatic unilateral right optic nerve edema. The decedent was a 20-year-old unsecured passenger in a convertible. The car collided with a pickup truck. The patient survived for 8 h. At autopsy, a comminuted skull fracture involving the right frontal bone including the right orbital plate was found. The right optic nerve measured 1.2 cm in diameter, compared to only 0.4 cm for the left optic nerve. Microscopically, the right optic nerve was markedly edematous, but the nerve fibers and nuclei were

[Optic nerve injury from the medical expertise point of view].

The optic nerve can be injured directly or indirectly. Direct injury occurs in association with orbital or craniocerebral injury with interruption of anatomical and functional optic nerve integrity. Indirect injuries occur when the force of impact is transmitted to the optic nerve. In the indirect injury group the mostly false diagnosed is traumatic opticoneuropathy especially after whiplash injury. According to Quebec classification, after whiplash injury Ist to IIIrd degree, ophthalmologic examination or visual field testing is not necessary, because there is no possibility of optic nerve in

[A pathological study on indirect optic nerve injury in rabbit eyes].

OBJECTIVE: To study the pathological changes after indirect optic nerve injury. METHODS: Animal models (72 rabbits, 144 eyes) for intracanalicular optic nerve injury was developed by keeping the optic canal intact. The pathological changes of optic nerve were observed under light microscope and transmission electron microscope and the number of myelinated axons was enumerated by computer-aided image analyzer. The data were analyzed by using variance of SAS software. RESULTS: The model contributed much valuable information regarding nerve edema, loss of myelin, myelin regeneration and changes

Gangliosides attenuate axonal loss after optic nerve injury.

Gangliosides have been shown to be capable of protecting nerve tissue from mechanical and biochemical insults and promoting their repair. The present study provides morphologic evidence that monosialogangliosides attenuate the degenerative process at the distal stump of the rat optic nerve after crush injury. Injured rat optic nerves were treated for 7 days after injury with daily intraperitoneal injections of monosialogangliosides (30 mg/kg/day), and compared with untreated injured controls with respect to the number of viable axons 2 and 4 weeks after injury, as indicated by transmission ele

Functional recovery and morphological changes after injury to the optic nerve.

Physiological and morphological parameters of optic nerve lesions followed by functional recovery are discussed in detail. To study functional and/or morphological recovery processes, a recently developed model of the 'graded optic nerve crush' is compared with other models of lesioning. It is concluded that the optic nerve crush model is a valuable tool when studying brain repair mechanisms. First results show that functional recovery depends on the initial preservation of a sufficiently large population of ganglion cells in the retina. It takes place despite the progressive loss of retinal g

[Direct optic nerve and retinal injury caused by retrobulbar injections].

The authors report on 2 patients with direct damage to the optic nerve and retina: In a 25-year-old deaf-mute patient a peripheral iridectomy was performed after lid and retrobulbar anesthesia. The amaurosis noticed immediately after surgery was caused by direct injury to the optic nerve. A 69-year-old patient with high myopia was treated with retrobulbar steroid injections. The globe was perforated and a series of 4 retinal defects from the periphery to close to the optic nerve head were created. An unhealed retinal detachment developed. The importance of informing the patient preoperatively

[Advances in research on the role of immune cells in optic nerve injury].

Optic nerve injury can result in the loss of retinal ganglion cells (RGCs) and their axons, representing a significant cause of irreversible vision impairment. Immune response is a common step following injury, and it often exhibits contrasting effects in optic nerve pathologies. Immune cells play a crucial role in this process, and understanding the differentiation of various immune cells post-injury, mitigating their neurotoxicity, and directing them towards a beneficial outcome for the protection of RGCs and axons are vital for optic nerve preservation. This paper provides a comprehensive r

Metabolomics dataset of zebrafish optic nerve regeneration after injury.

Zebrafish (Danio rerio) have the capacity for successful adult optic nerve regeneration. In contrast, mammals lack this intrinsic ability and undergo irreversible neurodegeneration seen in glaucoma and other optic neuropathies. Optic nerve regeneration is often studied using optic nerve crush, a mechanical neurodegenerative model. Untargeted metabolomic studies within successful regenerative models are deficient. Evaluation of tissue metabolomic changes in active zebrafish optic nerve regeneration can elucidate prioritized metabolite pathways that can be targeted in mammalian systems for thera

Retinal transcriptome of neonatal mice after optic nerve injury.

BACKGROUND: The axonal growth capacity of retinal ganglion cells decreases dramatically within the first day of birth, and the axonal regeneration after injury in mature mammals is very limited. Here, this study aimed to delineate the transcriptomic changes associated with altered axonal growth capacity and to identify the key genes associated with axonal regeneration by the RNA sequencing (RNA-Seq) analysis. METHODS: The whole retinas from the mice of embryonic day (E) 20, postnatal day (P) 1 and P3 were collected at 6 hours after optic nerve crush (ONC). Differentially expressed genes (DEGs

Isolated optic nerve oedema as unusual presentation of electric injury.

A 45-year-old man with no significant medical history presented following an electric current injury (380 V). He developed multiple systemic injuries including third degree burns and after 1 week of hospitalisation he reported unilateral visual changes. Examination suggested the presence of optic nerve oedema without evidence of haemorrhage, exudate or vessel abnormality. This was considered to be related to the electric shock. A trial of corticosteroids was considered. He was followed up to 5 months in clinic and was noted to have developed unilateral optic atrophy and no other systemic manif

Optic nerve avulsion following accidental penetrating orbital injury.

Traumatic enucleation with optic nerve avulsion following accidental penetrating orbital injury is a rare phenomenon. A 35 years old young active man was suddenly traumatized in his left orbit after falling down on a boat following collision of two running boats having no search light in a dark night. The patient was examined few hours after the event. The patient presented with severe pain around left periorbital region. He never lost consciousness. On examination right eye revealed no abnormality. There was left periorbital swelling with blood clots. Left eyeball was hanging from the orbit w

Elevated intraocular pressure and optic nerve injury models in the rat.

Models of experimentally elevated intraocular pressure in rats provide valuable opportunities to discover and study mechanisms of pressure-induced optic nerve damage. The structure and vasculature of the rat optic nerve head have several anatomic similarities and differences from the primate that allow useful comparisons and insights into human glaucoma. Specifically, the ultrastructural relationship between astrocytes, retinal ganglion cell axons and the connective tissues of the optic nerve head appear quite similar to the primate, and have a high potential for revealing cellular mechanisms

Axonal and vascular changes following injury to the rat's optic nerve.

The optic nerve of the rat has been examined by light and electron microscopy, and also for vascular permeability to fluorescently labelled albumin, 2 days to 34 weeks after crushing in the orbit. The operation was usually followed by loss of 20-70% of the retinal ganglion cells. Axons could be followed from the retina into the optic nerve at all postoperative times, but they always ended anterior to the lesion. Evidence of feeble regenerative growth of optic axons was seen in the first few postoperative weeks: bifurcating fibres and fibres that turned back towards the eye were present within
