Quantitative analysis of the optogenetic excitability of CA1 neurons.

INTRODUCTION: Optogenetics has emerged as a promising technique for modulating neuronal activity and holds potential for the treatment of neurological disorders such as temporal lobe epilepsy (TLE). However, clinical translation still faces many challenges. This in-silico study aims to enhance the understanding of optogenetic excitability in CA1 cells and to identify strategies for improving stimulation protocols. METHODS: Employing state-of-the-art computational models coupled with Monte Carlo simulated light propagation, the optogenetic excitability of four CA1 cells, two pyramidal and two

Optogenetic approaches to therapy for inherited retinal degenerations.

Inherited retinal degenerations such as retinitis pigmentosa (RP) affect around one in 4000 people and are the leading cause of blindness in working age adults in several countries. In these typically monogenic conditions, there is progressive degeneration of photoreceptors; however, inner retinal neurons such as bipolar cells and ganglion cells remain largely structurally intact, even in end-stage disease. Therapeutic approaches aiming to stimulate these residual cells, independent of the underlying genetic cause, could potentially restore visual function in patients with advanced vision loss

Bioengineered optogenetic model of human neuromuscular junction.

Functional human tissues engineered from patient-specific induced pluripotent stem cells (hiPSCs) hold great promise for investigating the progression, mechanisms, and treatment of musculoskeletal diseases in a controlled and systematic manner. For example, bioengineered models of innervated human skeletal muscle could be used to identify novel therapeutic targets and treatments for patients with complex central and peripheral nervous system disorders. There is a need to develop standardized and objective quantitative methods for engineering and using these complex tissues, in order increase t

Optogenetic Control of Airway Cholinergic Neurons In Vivo.

Dysregulation of airway nerves leads to airway hyperreactivity, a hallmark of asthma. Although changes to nerve density and phenotype have been described in asthma, the relevance of these changes to nerve function has not been investigated due to anatomical limitations where afferent and efferent nerves run in the same nerve trunk, making it difficult to assess their independent contributions. We developed a unique and accessible system to activate specific airway nerves to investigate their function in mouse models of airway disease. We describe a method to specifically activate cholinergic n

Optogenetic Hyperpolarization of Cardiomyocytes Terminates Ventricular Arrhythmia.

Cardiac defibrillation to terminate lethal ventricular arrhythmia (VA) is currently performed by applying high energy electrical shocks. In cardiac tissue, electrical shocks induce simultaneously de- and hyperpolarized areas and only depolarized areas are considered to be responsible for VA termination. Because electrical shocks do not allow proper control over spatial extent and level of membrane potential changes, the effects of hyperpolarization have not been explored in the intact heart. In contrast, optogenetic methods allow cell type-selective induction of de- and hyperpolarization with

Optogenetic Examination of Prefrontal-Amygdala Synaptic Development.

A brain network comprising the medial prefrontal cortex (mPFC) and amygdala plays important roles in developmentally regulated cognitive and emotional processes. However, very little is known about the maturation of mPFC-amygdala circuitry. We conducted anatomical tracing of mPFC projections and optogenetic interrogation of their synaptic connections with neurons in the basolateral amygdala (BLA) at neonatal to adult developmental stages in mice. Results indicate that mPFC-BLA projections exhibit delayed emergence relative to other mPFC pathways and establish synaptic transmission with BLA exc

Optogenetic control of contractile function in skeletal muscle.

Optogenetic stimulation allows activation of cells with high spatial and temporal precision. Here we show direct optogenetic stimulation of skeletal muscle from transgenic mice expressing the light-sensitive channel Channelrhodopsin-2 (ChR2). Largest tetanic contractions are observed with 5-ms light pulses at 30 Hz, resulting in 84% of the maximal force induced by electrical stimulation. We demonstrate the utility of this approach by selectively stimulating with a light guide individual intralaryngeal muscles in explanted larynges from ChR2-transgenic mice, which enables selective opening and

Roadmap for direct and indirect translation of optogenetics into discoveries and therapies for humans.

Optogenetics has transformed basic research on neural circuitry, led to diverse experimental insights into human brain function and dysfunction, and opened pathways for clinical translation based on new understanding of how specific cell types contribute to cognition and behavior. Many of these translational pathways do not rely on the direct application of optogenetics in humans, but rather develop and advance other treatment modalities by leveraging causal knowledge derived from optogenetic circuit neuroscience. However, a recent proof-of-principle study-showing that optogenetics applied dir

Integrating artificial intelligence and optogenetics for Parkinson's disease diagnosis and therapeutics in male mice.

Parkinson's disease (PD), a progressive neurodegenerative disorder, presents complex motor symptoms and lacks effective disease-modifying treatments. Here we show that integrating artificial intelligence (AI) with optogenetic intervention, termed optoRET, modulating c-RET (REarranged during Transfection) signalling, enables task-independent behavioural assessments and therapeutic benefits in freely moving male AAV-hA53T mice. Utilising a 3D pose estimation technique, we developed tree-based AI models that detect PD severity cohorts earlier and with higher accuracy than conventional methods. Em

Using optogenetics to dissect rod inputs to OFF ganglion cells in the mouse retina.

INTRODUCTION: Light responses of rod photoreceptor cells traverse the retina through three pathways. The primary pathway involves synapses from rods to ON-type rod bipolar cells with OFF signals reaching retinal ganglion cells (RGCs) via sign-inverting glycinergic synapses. Secondly, rod signals can enter cones through gap junctions. Finally, rods can synapse directly onto cone OFF bipolar cells. METHODS: To analyze these pathways, we obtained whole cell recordings from OFF-type α RGCs in mouse retinas while expressing channelrhodopsin-2 in rods and/or cones. RESULTS: Optogenetic stimulation

Optogenetics-enabled assessment of viral gene and cell therapy for restoration of cardiac excitability.

Multiple cardiac pathologies are accompanied by loss of tissue excitability, which leads to a range of heart rhythm disorders (arrhythmias). In addition to electronic device therapy (i.e. implantable pacemakers and cardioverter/defibrillators), biological approaches have recently been explored to restore pacemaking ability and to correct conduction slowing in the heart by delivering excitatory ion channels or ion channel agonists. Using optogenetics as a tool to selectively interrogate only cells transduced to produce an exogenous excitatory ion current, we experimentally and computationally q

From pineal photoreception to optogenetics: functional colour opponency based on a pineal bistable opsin.

Most animals sense light through opsins, which are photosensitive G protein-coupled receptors that utilize light information to regulate various physiological functions. In vertebrates, a representative example of such functions is vision, which is based on photoreception by visual opsins expressed in rod and cone photoreceptor cells of the retina. Accumulating photobiological evidence has gradually revealed the significance of the molecular properties of these opsins, as well as those of signal transduction-related molecules such as transducin, rhodopsin kinases, and arrestins, and their coll

Optogenetics and chemogenetics: key tools for modulating neural circuits in rodent models of depression.

Optogenetics and chemogenetics are emerging neuromodulation techniques that have attracted significant attention in recent years. These techniques enable the precise control of specific neuronal types and neural circuits, allowing researchers to investigate the cellular mechanisms underlying depression. The advancement in these techniques has significantly contributed to the understanding of the neural circuits involved in depression; when combined with other emerging technologies, they provide novel therapeutic targets and diagnostic tools for the clinical treatment of depression. Acousto-optogenetics bandpass stabilizer: A programmable platform for mapping single-cell phenotypic life trajectories.

Photobleaching-induced signal decay undermines the accuracy of fluorescence-based drug screening across multiple disease models (e.g., HER2/EGFR-targeted therapies). Traditional phototoxicity mitigation strategies, primarily through reduced illumination, inevitably compromise imaging resolution and further providing false positive/false negative. Here, we introduce an innovative active photobleaching intervention strategy to solve these problems with the potential for broad applicability across a wide range of cell species. We employ the ultra-low-frequency (500-2000 Hz) acoustic modulation to

Non-Invasive Electroretinogram Recording with Simultaneous Optogenetics to Dissect Retinal Ganglion Cells Electrophysiological Dynamics.

Electroretinography (ERG) is a non-invasive electrophysiological recording technique that detects the electrical signaling of neuronal cells in the visual system. In conventional ERG recordings, the signals are considered a collective electrical response from various neuronal cell populations, including rods, cones, bipolar cells, and retinal ganglion cells (RGCs). However, due to the limited ability to control electrophysiological responses from different types of cells, the detailed information underlying ERG signals has not been analyzed and interpreted. Linking the features of ERG signals

Optogenetics-based localization of talin to the plasma membrane promotes activation of β3 integrins.

Interaction of talin with the cytoplasmic tails of integrin β triggers integrin activation, leading to an increase of integrin affinity/avidity for extracellular ligands. In talin KO mice, loss of talin interaction with platelet integrin αIIbβ3 causes a severe hemostatic defect, and loss of talin interaction with endothelial cell integrin αVβ3 affects angiogenesis. In normal cells, talin is autoinhibited and localized in the cytoplasm. Here, we used an optogenetic platform to assess whether recruitment of full-length talin to the plasma membrane was sufficient to induce integrin activation. A

Perturbation of nuclear proteins by optogenetic trapping in Drosophila ovary.

The eukaryotic nucleus hosts transcription factors and other numerous nuclear proteins essential for genome organization and function. Optogenetic tools have been applied to match nuclear protein dynamics but remain largely confined to transcription factors, leaving most nuclear proteins inaccessible for precise manipulation. Here, we engineered an optogenetic tool, nuclear localization sequence (NLS)-light-activated reversible inhibition by assembled trap (LARIAT), by fusing a NLS to LARIAT, enabling light-inducible clustering of diverse GFP-tagged nuclear proteins in Drosophila ovarian cells

An optogenetic actuator for reversible alkalization of acidic presynaptic compartments.

Precise control of synaptic vesicle luminal pH offers a way to analyze proton-driven mechanisms involved in neurotransmitter loading, vesicle maturation, and synaptic transmission. Current genetically encoded tools either monitor vesicular pH or optogenetically acidify vesicles; here, we address this gap with a light-driven alkalizing actuator. Here, we developed syp-NsXeR, a genetically encoded optogenetic actuator based on the inward proton pump xenorhodopsin NsXeR, targeted to acidic presynaptic compartments through synaptophysin. Confocal imaging demonstrated enrichment of the construct at

Optogenetic stimulation of the dorsal striatum bidirectionally controls seizures.

Despite a century of development of antiseizure medications, up to a third of people with epilepsy do not achieve seizure freedom with drug therapy. Deep brain stimulation is of growing use, but just as with pharmacotherapy, is not universally effective. Identifying new targets for deep brain stimulation-and in particular sites that are effective against a range of seizure types-may close this gap. Engagement of the basal ganglia experimental seizures was first observed almost 75 y ago. However, the role of the basal ganglia's input nucleus, the striatum, in seizure control is relatively under

Optogenetic stimulation of dorsal striatum bidirectionally controls seizures.

Engagement of the striatum (caudate/putamen) and other basal ganglia nuclei during seizures was first observed over 75 years ago. Basal ganglia output nuclei, and the substantia nigra pars reticulata, in particular, have well-established anti-seizure effects across a large array of experimental models. However, striatal control of seizures is understudied. To address this gap, we used optogenetic approaches to activate and inactivate neurons in the dorsal striatum of Sprague-Dawley rats submitted to the gamma-butyrolactone (GBL) model of absence epilepsy, amygdala kindling model of temporal lo
