The rapid evolution of antibody‒drug conjugates (ADCs) and emerging X-drug conjugates (XDCs) has revolutionized targeted cancer therapy, yet their clinical translation is severely bottlenecked by the lack of human-relevant preclinical models. To bridge this translational gap, patient-derived organoids and organoids-on-chip (OoC) platforms recapitulate key features of human tumor biology, offering powerful tools for investigating cancer heterogeneity and improving the predictive evaluation of bioconjugated therapeutics. In this review, we systematically summarize the current landscape and chall

Despite their large societal burden, the development of therapeutic treatments for neurodegenerative diseases (NDDs) has been relatively unsuccessful. This is, in part, due to a lack of representative experimental models that reveal fundamental aspects of human brain pathology. Recently, assays for in vitro modeling of the human central nervous system (CNS) have significantly improved with the development of brain and spinal cord organoids. Coupled with induced-pluripotent stem cell and genome editing technologies, CNS organoids are a promising tool for studying neurodegeneration in a patient-

Drug sensitivity profiling on patient-derived organoids mirrors patient response for gastrointestinal cancers and is currently being evaluated in clinical trials to guide treatment decisions. However, long expansion times are typically required to achieve adequate cell numbers. Accordingly, miniaturization of organoid cultures can be leveraged to reduce assay time and increase the number of compounds tested. Microencapsulation by droplet microfluidics has been applied to produce miniaturized organoid cultures, but the predictive accuracy has yet to be fully established. Here, we show that drug

Retinal organoids (ROs) are three-dimensional in vitro models that replicate the specific cellular composition and inner structure of the retina. Currently, ROs derived from human pluripotent stem cells (hPSCs) have been shown to mimic both the structure and function of the human retina. Furthermore, ROs function as a powerful model system for researchers, facilitating the investigation of the pathogenesis and treatment strategies of retinal diseases. Despite their development for over a decade, ROs remain limited in terms of complexity and clinical application. This review summarizes recent a

Hard tissues are core components maintaining the mechanical integrity and physiological functions of organisms but have limited self-repair capacity. Traditional clinical strategies for hard tissue healing or regeneration face challenges, including donor scarcity. As engineered 3D cell culture systems that recapitulate tissue/organ-specific structures and functions, organoids outperform conventional 2D cell culture systems and animal models, thereby holding substantial potential for hard tissue healing or regeneration. Regrettably, constructing desired hard tissue organoids is highly demanding

Understanding the etiology of human neurological and psychiatric disorders remains challenging due to species-specific genomic architectures, prolonged developmental timelines, and the unique cytoarchitectural features of the human brain, which are inadequately replicated in conventional animal models. To address these limitations, human pluripotent stem cell (hPSC)-derived brain organoids have emerged as an ethically accessible experimental system that recapitulates critical aspects of early human neurodevelopment in vitro. These three-dimensional (3D) models mimic the generation of neural pr

Neural organoids (NOs), also known as brain organoids, are derived from human-induced pluripotent stem cells and are Microphysiological Systems (MPS) of the brain that can recapitulate key aspects of neurodevelopment. They enable in vitro studies of brain development and disease mechanisms, providing disease models for various neurodegenerative or neurodevelopmental/degenerative disorders like Alzheimer's disease, microcephaly, and autism. There are many protocols to generate NOs with different complexities and sizes, varying from 400 μm to several mm in diameter, with a starvation-induced nec

Here we describe organoid cultures derived from pig foliate taste papillae in which the cellular heterogeneity of the lingual epithelium is preserved. Pig taste organoids were maintained long term (18 passages) and continued to express taste stem cell markers (LGR4, LGR6, and SOX2) and taste receptor cell (TRC) markers (cytokeratin 20, ENTPD2, GNAT3, and OTOP1). We show insulin is necessary for optimum proliferation and differentiation of taste organoids. Some TRCs in the organoids contained insulin and the insulin-critical transcription factors MAFA and PAX4. However, we did not see any evide

The developing human cerebellum has a greater diversity of progenitor types than that of the mouse, necessitating a human-based model for studying cerebellar development and disease. Atamian et al.1 developed a 3D organoid model of cerebellar development, which recapitulates many cell types found in the developing human cerebellum, including Purkinje-neuron-like cells.

Research on human cerebellar development and disease has been hampered by the need for a human cell-based system that recapitulates the human cerebellum's cellular diversity and functional features. Here, we report a human organoid model (human cerebellar organoids [hCerOs]) capable of developing the complex cellular diversity of the fetal cerebellum, including a human-specific rhombic lip progenitor population that have never been generated in vitro prior to this study. 2-month-old hCerOs form distinct cytoarchitectural features, including laminar organized layering, and create functional con

Rapid advances in human brain organoid technologies have prompted the question of their consciousness. Although brain organoids resemble many facets of the brain, their shortcomings strongly suggest that they do not fit any of the operational definitions of consciousness. As organoids gain internal processing systems through statistical learning and closed loop algorithms, interact with the external world, and become embodied through fusion with other organ systems, questions of biosynthetic consciousness will arise.

INTRODUCTION: Organoids are three-dimensional cellular aggregates derived from stem cells or primary tissues that can self-organize into organotypic structures and showcase the physiological functions of that organ. Organoids typically comprise multiple organ-specific cell types that are responsible for organ function in vivo. They may also incorporate various cellular and molecular stromal components to recapitulate the in vivo microenvironment of the target organ.
METHODS: All articles related to thyroid-like organs were synthesized. Articles published between 1959 and 2023 were assessed, c

Alzheimer's disease (AD) is the primary cause of dementia, to date. The urgent need to understand the biological and biochemical processes related to this condition, as well as the demand for reliable in vitro models for drug screening, has led to the development of novel techniques, among which stem cell methods are of utmost relevance for AD research, particularly the development of human brain organoids. Brain organoids are three-dimensional cellular aggregates derived from induced pluripotent stem cells (iPSCs) that recreate different neural cell interactions and tissue characteristics in

COVID-19 is a systemic disease involving multiple organs, and clinically, patients have symptoms of neurological damage to varying degrees. However, we do not have a clear understanding of the relationship between neurological manifestations and viral infection due to the limitations of current in vitro study models. Brain organoids, formed by the differentiation of stem cells under 3D culture conditions, can mimic the structure of tiny cell clusters with neurodevelopmental features in different patients. The paper reviewed the history of brain organoids development, the study of the mechanism

Given the mismatch in size and morphology of intestinal organoids with native intestine, their application in intestinal segment regeneration has yet to be achieved. By engrafting ileal organoids in epithelium-removed colon, Sugimoto et al. recently constructed transplantable colonic segments with functional ileal epithelium and a subepithelial lymphovascular system, providing hope for the treatment of short bowel syndrome.

The obligate intracellular bacterium Chlamydia trachomatis is the leading cause of bacterial sexually transmitted infections. Once internalized in host cells, C. trachomatis undergoes a biphasic developmental cycle within a membrane-bound compartment, known as the inclusion. Successful establishment of the intracellular niche relies on bacterial Type III effector proteins, such as Inc proteins. In vitro and in vivo systems have contributed to elucidating the intracellular lifestyle of C. trachomatis, but additional models combining the archetypal environment of infection with the advantages of

The recent discovery of a new three-dimensional culture system for the derivation of cerebral organoids from human induced pluripotent stem cells provides developmental neurobiologists with the first example of a three-dimensional framework for the study of human brain development. This innovative approach permits the in vitro assembly of a human embryonic brain rudiment that recapitulates the developing human cerebrum. Organoids contain progenitor populations that develop to yield mature cortical neuron subtypes, potentially allowing investigators to study complex brain diseases that lack app

Human intestinal organoids (HIOs) recapitulate the architecture and cell diversity of intestinal epithelium, hence providing a system for modeling processes like regeneration and tumorigenesis. Here, we describe a detailed whole-mount immunostaining protocol for HIOs to visualize proliferative cell population. Briefly, this protocol includes preparation of HIOs, fixation and permeabilization of tissue, blocking of non-specific binding, antibody crosslinking, and visualization using a confocal microscope. Additionally, the protocol is broadly adaptable for investigating different antibodies, en

Proteomics, the large-scale study of proteins, enables the identification, quantification, and functional characterization of proteins, revealing post-translational modifications and protein interactions that are not apparent from transcriptomic data. Human organoids, which recapitulate the structural and functional complexity of native epithelial tissues, provide powerful tools to study disease mechanisms and personalize therapies. However, their culture poses challenges for efficient protein extraction and reproducible analysis. Here, we present a proteomics workflow optimized to maximize pr

Brain organoids represent three-dimensional structures that allow for human-specific studies in brain development, pathology and therapeutics. These self-organizing systems, formed through the differentiation of human pluripotent stem cells, can mimic important cellular and molecular events of brain development and therefore serve as a platform for the investigation of neurodevelopmental and neurodegenerative diseases, brain injuries, and tumorigenesis. Although brain organoids show promising perspectives in the study of human physiology, existing brain organoid platforms are hindered by issue
