The cytotoxic effects of the organophosphates chlorpyrifos and diazinon differ from their immunomodulating effects.

Some organophosphate insecticides have immunomodulating capacities, but it is unknown whether different compounds within this class affect the immune system to the same extent. In this in vitro study, human immortalized T-lymphocytes or bronchial epithelial cells were treated with diazinon or chlorpyrifos in the absence or presence of cellular stress factors, thereby mimicking a stimulated immune system. Cytotoxicity was determined and cytokine release or cytokine-promoter studies were performed to study immunomodulatory effects of these chemicals, whereby the same concentrations of chlorpyrif

Staphylococcus aureus Preferentially Liberates Inorganic Phosphate from Organophosphates in Environments where This Nutrient Is Limiting.

Phosphate is an essential nutrient that Staphylococcus aureus and other pathogens must acquire from the host during infection. While inorganic monophosphate (Pi) is the preferred source of this nutrient, bacteria can also obtain it from phosphate-containing organic molecules. The Pi-responsive regulator PhoPR is necessary for S. aureus to cause infection, suggesting that Pi is not freely available during infection and that this nutrient must be obtained from other sources. However, the organophosphates from which S. aureus can obtain phosphate are unknown. We evaluated the ability of 58 phosph

The effect of trichlorfon and other organophosphates on prenatal brain development in the guinea pig.

The organophosphates trichlorfon, dichlorvos, dimethoate, soman, triortho-cresyl phosphate (TOCP), and the diethoxy-analogue of trichlorfon (O,O-diethyl 2,2,2-trichloro-1-hydroxyethylphosphonate, ethyl-trichlorfon), were administered to guinea pigs between day 42 and 46 of gestation. When the offsprings were examined at birth, there was a severe reduction in brain weight in the case of trichlorfon and dichlorvos, but not after treatment with the other organophosphates. The reduction in weight was most pronounced for cerebellum, medulla oblongata, thalamus/hypothalamus and quadrigemina. The eff

An assessment of organophosphate ester mixtures and the placental transcriptome.

Prenatal exposure to organophosphate ester (OPE) chemicals, commonly used as flame retardants and plasticizers, has been associated with adverse birth outcomes. The placenta is a critical fetal organ and therefore may be involved in pathogenesis of birth outcomes. The goal of this study was to evaluate associations of 10 maternal urinary OPE metabolites, individually and as a mixture, with the placental transcriptome at birth in the Conditions Affecting Neurocognitive Development and Learning in Early Childhood (CANDLE) study. Individual OPE metabolites were evaluated for associations with ind

Histopathological features of low-dose organophosphate exposure.

Organophosphate (OP) use remains largely available worldwide despite more strict regulatory measures, in agriculture, parks or households, leading to a daily low-dose exposure. The systemic dysfunction appears partly due to acetylcholinesterase inhibition, exhibiting a primary toxic effect on the endocrine system but also on the liver and kidneys, which are responsible for products metabolization and elimination. Prolonged OP exposure can be responsible for histopathological (HP) changes that can either evolve or worsen pre-existing conditions. We conducted an experimental study including six

Mechanism-based novel antidotes for organophosphate neurotoxicity.

This article describes current pursuits for developing novel antidotes for organophosphate (OP) intoxication. Recent mechanistic studies of benzodiazepine-resistant seizures have key consequences for victims of OP pesticide and nerve agent attacks. We uncovered why current therapies are not able to stop the OP-induced seizures and brain cell death and what type of drug might be better. OP exposure down regulates critical inhibitory GABA-A receptors, kills neurons, and causes massive neuroinflammation that will cause more neuronal death, which causes the problem of too few benzodiazepine recept

New insights into the organophosphate-induced intermediate syndrome.

Acute organophosphate (OP) poisoning can be deadlier than any other type of chemical poisoning. So far, only a few cases have been described that include extensive neurological complications. We present an outstanding case of severe oral OP poisoning with intermediate syndrome developed on the fourth day after hospital admission. The clinical picture involved weakness of the proximal upper and lower limb muscles and several muscles supplied by motor cranial nerves, but, what is peculiar, the distal upper and lower limb muscles were also affected (forearms, hands, legs, and feet). To our knowle

Pancreatic pseudocyst after acute organophosphate poisoning.

Acute organophosphate poisoning (OP) shows several severe clinical symptoms due to its strong blocking effect on cholinesterase. Acute pancreatitis is one of the complications associated with acute OP, but this association still may not be widely recognized. We report here the case of a 73-year-old man who had repeated abdominal pain during and after the treatment of acute OP. Hyperamylasemia and a 7-cm pseudocyst in the pancreatic tail were noted on investigations. We diagnosed pancreatic pseudocyst that likely was secondary to an episode of acute pancreatitis following acute OP. He was initi

Methyl parathion: an organophosphate insecticide not quite forgotten.

Methyl parathion (MP), a toxic organophosphate insecticide approved for outdoor use only, is classified by the World Health Organization (WHO) as a Category Ia (extremely toxic) and by the United States Environmental Protection Agency (U.S. EPA) as a Toxicity Category I (most toxic) insecticide. In several U.S. states in the late 1980s and early 1990s, toxic exposures were created by the illegal use of MP indoors by uncertified pest control operators. As the health effects of MP exposure became evident with increasing public awareness, intervention by the U.S. government, in collaboration with

Organophosphate-induced convulsions and prevention of neuropathological damages.

Such organophosphorus (OP) compounds as diisopropylfluorophosphate (DFP), sarin and soman are potent inhibitors of acetylcholinesterases (AChEs) and butyrylcholinesterases (BChEs). The acute toxicity of OPs is the result of their irreversible binding with AChEs in the central nervous system (CNS), which elevates acetylcholine (ACh) levels. The protective action of subcutaneously (SC) administered antidotes or their combinations in DFP (2.0 mg/kg BW) intoxication was studied in 9-10-weeks-old Han-Wistar male rats. The rats received AChE reactivator pralidoxime-2-chloride (2PAM) (30.0 mg/kg BW),

Prevention of organophosphate intoxication with presynaptic cholinergic blockers.

In Vivo cardiovascular neuromuscular preparation of rats was used to study the prophylaxis and mechanism of diisopropyl fluorophosphate (DFP) intoxication by presynaptic cholinolytic agents (ganglionic blocking agents), including hexamethonium, trimethaphan and mecamylamine. The lethal action of DFP was partially or completely prevented by pretreatment of rats with hexamethonium (10 mg/kg), trimethaphan (80 mg/kg) and mecamylamine (30 mg/kg). Combined use of these drugs with 2-PAM (100 mg/kg) improved further the prophylaxis of DFP lethality. The mechanism of prophylaxis could be due to the re

Mechanisms of toxicological interactions involving organophosphate insecticides.

Administration of apparently nontoxic doses of organophosphorus compounds can greatly alter the toxicity of other compounds. These might include other organophosphates, as well as other drug or nondrug xenobiotics. Mechanisms of toxicologic interactions are discussed. Emphasis is placed upon organophosphate inhibition of noncritical tissue esterases and alterations in the metabolism and toxicity of selected ester and amide containing xenobiotics.

Acute pancreatitis induced by acute organophosphate poisoning.

A 32-year-old male with acute organophosphate poisoning developed hyperglycaemia, glycosuria and ketonuria soon after admission to hospital. Serum amylase estimations suggested a diagnosis of acute pancreatitis. He required insulin therapy to control his hyperglycaemia and the organophosphate poisoning was successfully managed by artificial ventilation, an infusion of pralidoxime and intermittent atropine. He was discharged on the 17th hospital day with no permanent physical sequelae.

Adult and embryo responses to organophosphate pesticides: azodrin.

Azodrin was applied to adult embryo chickens, Chukar Partridge, and Bobwhite Quail. Chronic exposure of adult birds to Azodrin mixed in their feed indicated that no a priori predictions could be made about one species based on the results of another; each had a different no effect (MACT) level. The chickens were between 25 and 100 ppm, the Chukar Partridge 5 and 25 ppm, and the Bobwhite quail less than 1.25 ppm. The chicken adults were most resistant, and the quail were least resistant to chronic exposure to Azodrin. Yolk-injected Azodrin caused the embryos of all three species to develop abno

Association of exposure to organophosphate pesticides with reproductive lifespan.

BACKGROUND: Organophosphate pesticides (OPs) have been found to have reproductive toxicity with overt endocrine disruption potential, yet the relationships between OPs with reproductive lifespan remain unexplored. OBJECTIVE: We aimed to investigate associations between urinary OP concentrations with reproductive lifespan. METHOD: This study included 1,356 postmenopausal women who participated in the National Health and Nutrition Examination Survey from 1999 to 2018 to evaluate the relationship between OP exposure and reproductive lifespan. Urine was analysed using gas chromatography coupled wi

Opsoclonus-myoclonus syndrome caused by organophosphate poisoning.

Opsoclonus-myoclonus syndrome is a combination of involuntary, arrhythmic, conjugate saccadic eye movements with myoclonus. The most common cause in adults is paraneoplastic encephalitis. Rarer causes include infections such as scrub typhus, and toxins such as organophosphates and cocaine. Organophosphates are one of the common poisonings in tropical countries such as India, causing both central and peripheral nervous system manifestations. We describe a middle-aged male farmer with unexplained altered consciousness and respiratory depression. After 2 days, he developed opsoclonus-myoclonus, a

[Effect of organophosphate pesticides poisoning on cognitive impairment].

It has been suggested that organophosphate pesticides can cause cognitive impairment, most of which are presented as mild cognitive impairment such as decreased attention or vigilance, reduced information processing speed and memory impairment. An early diagnosis and timely intervention in mild cognitive impairment, which may provide a better prognosis. Mild cognitive impairment in clinical work can be easily ignored for a long time. However, it have a particularly impact on patients' life and work, and even progress to irreversible neurodegenerative diseases such as Alzheimer's disease and Pa

Utilization of plasmapheresis for organophosphate intoxication: A case report.

Organophosphate (OP) compounds are extremely toxic chemicals that may be absorbed via skin, conjunctiva, gastrointestinal and respiratory systems. Treatment of OP poisoning is a critical and aggressive process which includes decontamination, antidote administration (atropin and oximes), mechanical ventilation support and extracorporeal elimination procedures if needed. Here we present a young female patient who was unintentionally poisoned by an OP (trichlorfon) after using it to moisture her skin. The importance of this patient is the dermal disease that makes her unprotective to dermal expos
