Experimental transmission of Karshi and Langat (tick-borne encephalitis virus complex) viruses by Ornithodoros ticks (Acari: Argasidae).

Selected species of mosquitoes and Ornithodoros ticks were evaluated for their potential to transmit Karshi and Langat (tick-borne encephalitis virus complex) viruses in the laboratory. Although there was no evidence of replication of Karshi virus in either of the two mosquito species tested [Ochlerotatus taeniorhynchus (Wiedemann) or Culex pipiens (L.)], Karshi virus replicated in and was transmitted by all three species of Ornithodoros ticks tested (Ornithodoros parkeri Cooley, Ornithodoros sonrai Sautet & Witkowski, and Ornithodoros tartakovskyi Olenev). When inoculated with Karshi virus, 9

Exploring the transcriptome of immature stages of Ornithodoros hermsi, the soft-tick vector of tick-borne relapsing fever.

Blood-feeding behavior has independently evolved in arthropods multiple times. Unlike hard ticks, soft ticks employ a rapid-feeding strategy for hematophagy, and there are comparatively limited studies on the transcriptomes of these organisms. This study investigates the soft tick Ornithodoros hermsi, conducting histopathological examinations at bitten skin sites and tick whole-body transcriptomic analyses across various developmental and feeding stages, including larvae, 1st-nymphal, and 2nd-nymphal stages. The results revealed the ability of O. hermsi to induce skin hemorrhage at the bite si

Immune responses of the argasid tick Ornithodoros moubata moubata induced by infection with the filarial worm Acanthocheilonema viteae.

Investigations were undertaken to determine whether the tick Ornithodoros moubata moubata mounted a detectable immune response to primary and secondary infections with Acanthocheilonema viteae. Uninfected control tick survival rate was 70%, but only 45% in the primary infection group. Post-secondary infection survival rate (82%) was comparable to controls, indicating that these selected ticks had some protective advantage. Mean A. viteae infective larvae recovery from ticks with secondary infections was 31.4% lower than expected, suggesting the development of immunity. SDS-PAGE of haemolymph f

[A case of experimental transmission of the causative agent of plague from Ornithodoros tartakovskyi ticks to fleas of the great gerbil].

Biological interrelations between ectoparasites from the colonies of great gerbil were studied experimentally. Hungry fleas Coptopsylla lamellifer are able to feed on the ticks Ornithodoros tartakovskyi infected with plague and in so doing be infected with plague agent. Such feeding is identified as "heterovampirism".

Ecological niche modeling and distribution of Ornithodoros hermsi associated with tick-borne relapsing fever in western North America.

Tick-borne relapsing fever in western North America is a zoonosis caused by the spirochete bacterium, Borrelia hermsii, which is transmitted by the bite of infected Ornithodoros hermsi ticks. The pathogen is maintained in natural cycles involving small rodent hosts such as chipmunks and tree squirrels, as well as the tick vector. In order for these ticks to establish sustained and viable populations, a narrow set of environmental parameters must exist, primarily moderate temperatures and moderate to high amounts of precipitation. Maximum Entropy Species Distribution Modeling (Maxent) was used

Spatial multi-criteria decision analysis for modelling suitable habitats of Ornithodoros soft ticks in the Western Palearctic region.

Ticks are economically and medically important ectoparasites due to the injuries inflicted through their bite, and their ability to transmit pathogens to humans, livestock, and wildlife. Whereas hard ticks have been intensively studied, little is known about soft ticks, even though they can also transmit pathogens, including African Swine Fever Virus (ASFV) affecting domestic and wild suids or Borrelia bacteria causing tick-borne relapsing fever (TBRF) in humans. We thus developed a regional model to identify suitable spatial areas for a community of nine Ornithodoros tick species (O. erraticu

Detection and molecular characterization of African swine fever virus recovered from Ornithodoros ticks of the Serengeti Ecosystem, Tanzania.

African swine fever (ASF) is a highly lethal hemorrhagic disease of domestic pigs and Eurasian wild boars, caused by ASF virus (ASFV). In Africa, ASFV is maintained in a sylvatic cycle involving wild suids, primarily warthogs (Phacocherus africanus) and Ornithodoros ticks. Despite ASF outbreaks in Tanzania, the role of the sylvatic cycle in areas with high wildlife-livestock interactions remains understudied. This study aimed to detect and characterize ASFV in Ornithodoros ticks from warthog burrows in the Serengeti ecosystem. Soft ticks were collected from warthog burrows and screened for ASF

Isolation and Molecular Characterization of Tick-Borne Relapsing Fever Borrelia Infecting Ornithodoros (Pavlovskyella) verrucosus Ticks Collected in Ukraine.

BACKGROUND: Tick-borne relapsing fever (TBRF) is a neglected zoonotic bacterial disease known to occur on 5 continents. We report a laboratory-acquired case of TBRF caused by Borrelia caucasica, which is endemic in Ukraine and transmitted by Ornithodoros verrucosus ticks. METHODS: We isolated spirochetes and characterized them by partially sequencing the 16s ribosomal ribonucleic acid (rrs), flagellin (flaB), and deoxyribonucleic acid gyrase (gyrB) genes and conducting a phylogenetic analysis. RESULTS: These analyses revealed a close relationship of Ukrainian spirochetes with the Asian TBRF

Experimental transmission of Langat (tick-borne encephalitis virus complex) virus by the soft tick Ornithodoros sonrai (Acari: Argasidae).

Laboratory studies determined that the soft tick Ornithodoros sonrai Sautet & Witkowski is a competent vector of Langat (tick-borne encephalitis virus complex) virus. When ticks fed on suckling mice having a mean viremia of 10(7.2) plaque-forming units per ml, 52% (n = 208) became infected, and 84% (n = 87) of the infected ticks transmitted virus by bite when fed individually on suckling mice > or = 27 d after the infectious blood meal. Overall, 79 of 184 (43%) of ticks exposed to the original viremic mice individually transmitted virus by bite when tested up to 351 d after the infectious bloo

Range Modeling and Surveillance of Ornithodoros turicata Ticks: Implications for Detecting African Swine Fever Virus in the United States.

African Swine Fever virus (ASFv) is a re-emerging global swine disease that, if introduced to the United States, would cause severe economic consequences. The widespread presence of feral hogs in addition to the presence of competent tick vectors, specifically Ornithodoros turicata, fosters a greater risk of ASFv establishment. The specific aims of this study were to assess the geographic distribution of O. turicata, the spatial distribution of host species richness, and to identify localities that could be monitored for ASFv as part of a comprehensive surveillance system in the United States

Evaluation of the protective efficacy of Ornithodoros moubata midgut membrane antigens selected using omics and in silico prediction algorithms.

The African argasid tick Ornithodoros moubata transmits two important pathogens, the African swine fever virus and the spirochete Borrelia duttoni, the cause of human relapsing fever. To date, only conventional control measures such as widespread application of acaricides, strict control measures, and animal movement restrictions have been implemented to confine these diseases. Vaccines against tick infestations have the potential to be among the most efficacious interventions for the management of these diseases. Plasma membrane-associated proteins upregulated in tick midgut cells in response

Absence of transovarial transmission of Borrelia duttonii, a tick-borne relapsing fever agent, by the vector tick Ornithodoros moubata.

We examined the vector competence of the tick, Ornithodoros moubata, using laboratory-reared gerbils as hosts. Transmission of the relapsing fever agent Borrelia duttonii occurred efficiently from infected ticks to uninfected gerbils and from infected gerbils to uninfected ticks. Spirochetes were maintained stably in the ticks for at least 3 months, but they disappeared from the bloodstream of infected gerbils after three episodes of spirochetemia. We also examined transovarial transmission of B. duttonii during the gonotrophic cycle and filial generation. No spirochetes could be detected from

Relationships between the defensive systems of Iberian-breed swine and the European vector of African swine fever, Ornithodoros erraticus.

To discover whether the immune system of Iberian-breed pigs exerts any adverse action on Ornithodoros erraticus, 3 pairs of pigs were subjected to a weekly infestation over 12 wk with 1,000 larvae, 500 nymphs-1, or 200 adults. Each pair was bitten by only 1 developmental stage. Batches of parasites identical to the foregoing ones were fed weekly on control swine. In none of the 10 parameters studied for each of the batches fed weekly was any significant difference found that could be attributed to the state of sensitization of the animals in which, in a previous study, the presence of high tit

Transcriptomic Analysis of Salivary Glands of Ornithodoros brasiliensis Aragão, 1923, the Agent of a Neotropical Tick-Toxicosis Syndrome in Humans.

Tick salivary glands produce and secrete a variety of compounds that modulate host responses and ensure a successful blood meal. Despite great progress made in the identification of ticks salivary compounds in recent years, there is still a paucity of information concerning salivary molecules of Neotropical argasid ticks. Among this group of ticks, considering the number of human cases of parasitism, including severe syndromes and hospitalization, Ornithodoros brasiliensis can be considered one of the major Neotropical argasid species with impact in public health. Here, we describe the transcr

Soldado virus (Hughes group) from Ornithodoros (Alectorobius) capensis (Ixodoidea: Argasidae) infesting Sooty Tern colonies in the Seychelles, Indian Ocean.

In the summer of 1973, about 5,000 pairs of adult Sooty Terns abandoned a specific part of their breeding grounds on Bird Island in the Seychelles. Incubated eggs and newly hatched chicks were left unattended and the area was not reoccupied in 1974. Numerous Ornithodoros (Alectorobius) capensis occurred in the deserted part in both years, but few or none in the area where bird breeding was normal. Persons visiting the deserted area were bitten by many ticks and experienced severe pruritus persisting for several days. In 1973 and 1974, Soldado (SOL) virus (Hughes serogroup) was isolated from th

Subolesin/akirin orthologs from Ornithodoros spp. soft ticks: cloning, RNAi gene silencing and protective effect of the recombinant proteins.

Subolesin/akirin is a well characterized protective antigen highly conserved across vector species and thus potentially useful for the development of a broad-spectrum vaccine for the control of arthropod infestations including hard ticks, mosquitoes, sand flies and the poultry red mite Dermanyssus gallinae. Soft ticks could be also targeted by this vaccine if proved that the soft tick subolesin orthologs are conserved and induce protective immune responses too. However, to date no soft tick subolesin orthologs have been fully characterized nor tested as recombinant antigens in vaccination tria

A comparison of manual and carbon dioxide trap sampling of Ornithodoros soft ticks from warthog resting sites in South Africa.

BACKGROUND: In East and Southern Africa, the African swine fever (ASF) virus is maintained in an ancient sylvatic cycle involving warthogs (Phacochoerus spp.) and Ornithodoros soft ticks inhabiting warthog burrows. Although carbon dioxide (CO2) traps have previously been used to collect ticks from pigsties in Portugal, this method has never been tested in the context of the ASF sylvatic cycle in Africa. As warthogs adapt their resting site preferences in response to different levels of habitat transformation, our study aimed to evaluate the effectiveness of CO2 traps versus traditional manual

RNA-seq analysis and gene expression dynamics in the salivary glands of the argasid tick Ornithodoros erraticus along the trophogonic cycle.

BACKGROUND: The argasid tick Ornithodoros erraticus is the main vector of tick-borne human relapsing fever (TBRF) and African swine fever (ASF) in the Mediterranean Basin. Tick salivary proteins secreted to the host at the feeding interface play critical roles for tick feeding and may contribute to host infection by tick-borne pathogens; accordingly, these proteins represent interesting antigen targets for the development of vaccines aimed at the control and prevention of tick infestations and tick-borne diseases. METHODS: To identify these proteins, the transcriptome of the salivary glands o
