Pathogenicity of avian reovirus variant in the immune organs of broiler chicks.

In recent years, the infection rate of avian reovirus (ARV) in chicks has been continuously increasing, seriously endangering the healthy development of China's chicken farming industry. However, the pathogenicity of this virus in immune organs has been less studied. In this study, an ARV strain (Chicken/XJ/China/2022/01/16) was isolated from arthritic broiler chicks in Xinjiang province and whole genome sequencing of the ARV strain was performed using next generation sequencing (NGS) technology, and sequence analysis showed that the strain was a type V variant of the ARV gene. In order to fur

Whole genome sequence analysis of turkey orthoreovirus isolates reveals a strong viral host-specificity and naturally occurring co-infections in commercial turkeys.

Avian orthoreoviruses (ARV) are an emerging threat for the poultry industry, both in the United States (US) and globally. ARV infections in turkeys have been associated with arthritis, lameness and neurological disorders, and cost the US economy approximately USD 33 million per year. There is not a commercial vaccine available and the shortage of turkey ARV (TRV) genomic data hinders the efforts to explore the molecular epidemiology of this virus, although several studies suggest a close relationship between European TRVs and TRVs present in the US. This study shows a snapshot of the genomic d

Avian Reoviruses from Clinical Cases of Tenosynovitis: An Overview of Diagnostic Approaches and 10-Year Review of Isolations and Genetic Characterization.

Reoviral-induced tenosynovitis/viral arthritis is an economically significant disease of poultry. Affected birds present with lameness, unilateral or bilateral swollen hock joints or shanks, and/or reluctance to move. In severe cases, rupture of the gastrocnemius or digital flexor tendons may occur, and significant culling may be necessary. Historically, vaccination with a combination of modified live and inactivated vaccines has successfully controlled disease. Proper vaccination reduced vertical transmission and provided maternal-derived antibodies to progeny to protect against disease, at a

Identification of Coronaviruses, Paramyxoviruses, Reoviruses, and Rotaviruses among Bats in Nigeria.

Bats are often consumed by some ethnic groups in Nigeria despite association of bats with many important emerging viruses. More than 300 bats representing eight species were captured during 2010-2011 in eight locations of northern Nigeria. Available fecal swabs (n = 95) were screened for the presence of arenaviruses, CoVs, paramyxoviruses (PMVs), reoviruses, rhabdoviruses, and influenza viruses using generic reverse transcription-polymerase chain reaction assays. Here, we document the detection of CoVs, PMVs, reoviruses, and rotaviruses (RVs) in Nigerian bats. The Nigerian bat CoVs are grouped

Mixed infection with reovirus and Chlamydophila in a flock of budgerigars (Melopsittacus undulatus).

Eleven budgerigars (Melopsittacus undulatus) from a zoological collection presented at necropsy with emaciation and splenomegaly or hepatomegaly or both. Polymerase chain reaction assays performed on liver and spleen samples were positive for Chlamydophila psittaci in 2 of 3 birds tested, and histologic findings in 2 additional birds were compatible with chlamydiosis. The aviary was subsequently closed to the public, and a 45-day treatment regimen with doxycycline in the seeds was initiated. No further deaths of birds with hepatomegaly or splenomegaly were observed after the first day of treat

Viral Heterogeneity in an Avian Reovirus Isolate Despite Plaque Purification and Differences in Segment-Wise Abundance Between Different Amplification and Purification Steps.

Avian reovirus (ARV) remains a significant concern in the poultry industry. Three rounds of plaque purification are routinely employed to obtain clonal populations of a viral isolate for experimental purposes. However, the effectiveness of this approach in achieving viral homogeneity has usually been taken for granted, especially in older publications, when there was no means of checking the purity. This study aimed to determine the purity of a plaque-purified ARV isolate (strain AL) using conventional PCR, Sanger sequencing, and whole-genome sequencing (WGS). WGS analysis of multiple samples

Pathogenicity and molecular evolutionary analysis of novel Avian orthreovirus genotypes IV and V in Upper Egypt.

Avian orthoreovirus (ARV) imposes substantial economic losses on the global poultry industry, primarily through runting-stunting syndrome and viral arthritis. Despite its global prevalence, the molecular epidemiology and pathogenic landscape of ARV strains circulating in Upper Egypt remain largely unexplored. This study investigated the molecular characteristics, phylogenetic relationships, and pathogenic potential of ARV in broiler flocks across Assiut, Sohag, and New Valley governorates. Out of 140 clinical samples from chickens showing stunted growth and tendon swelling, 42 (30%) were posit

Optimized polypeptide for a subunit vaccine against avian reovirus.

Avian reovirus (ARV) is a disease-causing agent. The disease is prevented by vaccination with a genotype-specific vaccine while many variants of ARV exist in the field worldwide. Production of new attenuated vaccines is a long-term process and in the case of fast-mutating viruses, an impractical one. In the era of molecular biology, vaccines may be produced by using only the relevant protein for induction of neutralizing antibodies, enabling fast adjustment to the emergence of new genetic strains. Sigma C (SC) protein of ARV is a homotrimer that facilitates host-cell attachment and induce the

Novel goose parvovirus as a major risk factor for red skin and bristle feather syndrome in meat ducks: Epidemiological associations with duck circovirus and reovirus.

In recent years, a disease characterized by dorsal skin reddening and feather development disorder, termed "red skin and bristle feather syndrome" (RSBFS), has emerged in meat duck farms in China, resulting in substantial economic losses. To clarify the major etiological risk factor, their epidemiological patterns, and molecular characteristics, we collected 300 clinical samples from symptomatic and asymptomatic ducks across eight major duck-producing provinces in China between February and July 2025. Multipathogen screening was conducted using quantitative real-time PCR, followed by phylogene

Evidence of vertical transmission of novel duck orthoreovirus in ducks.

Since 2017, duck spleen necrosis caused by new variant duck orthoreovirus (N-DRV) infection had been observed in several provinces in China. This disease retards the growth and development of ducks, thereby reducing feed return rate. N-DRV infection causes damage to duck spleen and other immune organs, leading to immunosuppression and susceptibility to other pathogens. In this study, we successfully constructed a breeding duck artificial infection model and found that N-DRV infection can cause pathologic changes, such as ovarian hemorrhage, follicle atrophy, and fallopian tube bleeding in bree

Heterologous expression, immunogenic evaluation, and subunit vaccine potential of the σC protein from the Xinjiang avian reovirus (ARV) strain xj-1.1.

Avian reovirus (ARV) causes a range of diseases in poultry and results in significant economic losses for the poultry industry. To address the epidemic of ARV infection in yellow-feathered broilers in Xinjiang, this study conducted genetic and evolutionary analyses of a chicken-origin ARV field strain, designated ARV xj-1.1. The σC gene of this strain was expressed in vitro using both Escherichia coli and Pichia pastoris systems to compare expression characteristics and immunogenicity. Phylogenetic analysis revealed that ARV xj-1.1 belongs to genotype IV. The σC gene was successfully cloned an

Avian reoviruses: propagation, quantification, and storage.

Avian reoviruses (ARVs) are pathogens that cause significant morbidity among commercial poultry. ARVs are prototypic representatives of non-enveloped viruses that can cause cell-cell fusion. They belong to the Reoviridae family, which contains many highly pathogenic viruses. ARVs are ubiquitous in commercial poultry and are frequently isolated from the gastrointestinal and respiratory tracts of chickens with acute infections. The virus causes a range of disease states in chicken, including viral arthritis/tenosynovitis, gastroenteritis, hepatitis, myocarditis, "pale bird syndrome," runting-stu

Rapid and Visual Detection of Muscovy Duck Reovirus Using a Reverse Transcription Recombinase-Aided Amplification Assay in a Lyophilized Format for Point-of-Care Applications.

Muscovy duck reovirus (MDRV) causes substantial economic losses in the waterfowl industry, necessitating rapid and field-deployable diagnostic tools. In this study, we developed, for the first time, a lyophilized reagent-based reverse transcription recombinase-aided amplification (RT-RAA) assay targeting the σC gene of MDRV for point-of-care testing (POCT). The assay was optimized to operate at 39°C with a probe concentration of 100 nM. It achieved a detection limit of 1.03 × 101 copies/μL within 20 min, which is comparable to RT-quantitative polymerase chain reaction (qPCR) and superior to ex

A Novel Variant of Avian Reovirus Is Pathogenic to Vaccinated Chickens.

Avian reovirus (ARV) infections, characterized by severe arthritis, tenosynovitis, pericarditis, and poor weight gain, have become increasingly serious in recent years. The economic impact is significant as it causes growth inhibition and immunosuppression. Some commercial poultry in China have been widely vaccinated with available ARV vaccines; however, infections continue to occur even after vaccination. This study aimed to isolate a novel variant, ARV-SD19/11103, from the joint tissues of infected broiler chickens vaccinated with ARV vaccines in Shandong Province. Genetic evolution analysis

Development and evaluation of a live-attenuated novel duck reovirus vaccine strain E232-P100 conferring complete protection in ducklings.

Novel duck reovirus (NDRV) is a major pathogen that causes immunosuppression and secondary infections in ducklings, resulting in elevated mortality and severe economic losses to the global duck industry. However, there is currently no licensed vaccine available in China for the effective prevention and control of NDRV infections. To effectively control NDRV outbreaks, we developed a live-attenuated vaccine candidate against NDRV by serially passaging the NDRV E232 strain in specific-pathogen-free (SPF) chicken embryos and DF-1 cells. The resulting E232-P100 strain replicated efficiently in DF-

Evaluation of Pathogenicity and Antigenicity of Avian Reoviruses and Disease Control Through Vaccination.

Avian reoviruses are ubiquitous in poultry production worldwide and can be transmitted vertically or horizontally among chickens. The pathogenicity of reoviruses can range from very pathogenic viruses that affect multiple tissues and organs to apathogenic. Avian reoviruses have been associated with many disease presentations, and two of the most economically significant diseases are viral arthritis/tenosynovitis and viral enteritis. Viral arthritis/tenosynovitis has been recognized since the 1950s and essentially disappeared after development of attenuated live and inactivated vaccines in the

Molecular characterization of emerging avian reovirus variants isolated from viral arthritis cases in Western Canada 2012-2017 based on partial sigma (σ)C gene.

Viral Arthritis (VA), a disease caused by Avian Reovirus (ARV), has emerged as a significant cause of economic losses in broiler chicken flocks in Western Canada. These outbreaks were characterized by 4-13% morbidity, followed by a spike in mortality/culling that in extreme cases required total flock depopulation. From 2012-2017, 38 ARV isolates were recovered. Molecular characterization of a partial segment of the sigma (σ)C gene shows all six previously known ARV clusters in Western Canadian broiler chickens. The most numerous clusters were Cluster#4 and Cluster #5 while the most variable cl

Avian Reovirus σA Protein Inhibits Type I Interferon Production by Abrogating Interferon Regulatory Factor 7 Activation.

Type I interferon (IFN) response is the first line of host-based innate immune defense against viral infections. However, viruses have developed multiple strategies to counter host IFN responses, so they may continue infecting hosts via effective replication. Avian reovirus (ARV), an RNA virus, causes viral arthritis or tenosynovitis in chickens. Previous studies have shown that ARV is highly resistant to the antiviral effects of IFN. However, the underlying mechanisms that enable ARV to block the IFN pathway remain unclear. In this study, we found that ectopic expression of ARV protein, σA, s

Genomic sequences and pathogenic characteristics of two variant duck reoviruses associated with spleen necrosis.

Emerging variant novel duck reovirus (NDRV) strains that cause spleen swelling and necrosis have seriously threatened the waterfowl industry since 2017. However, there is no report about the complete genomic sequence of emerging variant strains isolated from Cherry valley ducks. In this study, we acquired the complete genome sequences of two variant NDRV strains, SD19/6201 and SD19/6202, and analyzed their genetic and evolutionary relationship with other orthoreoviruses. The phylogenetic analysis of σC showed that all the Chinese NDRVs were clustered into two distinct branches. The SD19/6201 s
