A new syndrome of congenital generalized osteosclerosis and bilateral polymicrogyria.

We report a 29-week male fetus with healthy consanguineous parents. He showed a severe sclerosing bone disorder affecting all skeletal elements, resulting in insufficient modeling, generalized densification, and fragility of the skeleton. This skeletal dysplasia was associated with an abnormal craniofacial development (hypertelorism, severe microretrognathia, cleft palate, absent epiglottis, reduced number, and mineralization of teeth buds) and abnormal terminal phalanges. Neuropathologic examination showed bilateral fronto-parietal cerebral polymicrogyria. This syndrome appears to represent a

Osteosclerosis and Meningioma: Implicating the Tumor Suppressor Gene AMER1/WTX.

BACKGROUND/AIM: Meningiomas are common intracranial tumors with variable biological behavior and incompletely understood molecular drivers, particularly in sporadic cases. CASE REPORT: We present a unique case of a 29-year-old female with osteosclerosis harboring a germline mutation in the AMER1 (also known as WTX) gene - a tumor suppressor known to regulate the Wnt/β-catenin signaling pathway - who subsequently developed a large convexity meningioma. The patient had no recognized risk factors for meningioma, such as cranial irradiation, familial syndromes, or hormone exposure. Given AMER1's

Can transient neonatal osteosclerosis be differentiated from malignant infantile osteopetrosis?

Osteosclerosis in infancy requires careful evaluation as it may indicate the presence of osteopetrosis. Osteopetrosis is a rare disorder of high bone density due to impaired osteoclast resorption. Infantile forms of osteopetrosis have neurological and hematopoietic defects. However, some infants may present with a transient neonatal osteosclerosis (TNO) appearance without skeletal consequences. The aim of this case series is to compare TNO with infantile osteopetrosis. Radiographic reports of infants less than 12 months old were searched for the terms osteosclerosis and osteopetrosis from 2006

Automated Osteosclerosis Grading of Clinical Biopsies Using Infrared Spectroscopic Imaging.

Osteosclerosis and myefibrosis are complications of myeloproliferative neoplasms. These disorders result in excess growth of trabecular bone and collagen fibers that replace hematopoietic cells, resulting in abnormal bone marrow function. Treatments using imatinib and JAK2 pathway inhibitors can be effective on osteosclerosis and fibrosis; therefore, accurate grading is critical for tracking treatment effectiveness. Current grading standards use a four-class system based on analysis of biopsies stained with three histological stains: hematoxylin and eosin (H&E), Masson's trichrome, and reticul

Osteosclerosis in a thirty-four-year-old woman with primary hyperparathyroidism.

Hyperparathyroidism is an endocrine abnormality that frequently causes diffuse osteopenia in the bones. Osteosclerosis is a rare phenomenon in adults with primary hyperparathyroidism since the usual skeletal manifestation is generalized osteopenia. We describe a patient with generalized osteosclerosis in the jaws and skull in association with primary hyperparathyroidism, while the other skeletal bones had normal or decreased density.

Radiologic assessment and frequency of idiopathic osteosclerosis of jawbones: an interpopulation comparison.

BACKGROUND: Idiopathic osteosclerosis is a localized growth of compact bone with an unknown cause. This asymptomatic lesion is an insignificant finding and as such requires no treatment. It should be distinguished from other types of osteosclerosis created by inflammatory processes and systemic diseases. PURPOSE: To determine the prevalence and distribution of idiopathic osteosclerosis in the jawbones in Iran and to compare this prevalence and distribution with other populations. MATERIAL AND METHODS: This cross-sectional descriptive study was performed in the Dental School of Shahid Behesht

Changes in osteosclerosis of the osteoarthritic knee after high tibial osteotomy.

Radiographically demonstrated osteosclerosis was quantitatively evaluated in knees with medial compartmental osteoarthritis by dual xray absorptiometry. Bone density was measured in various areas of 144 knees with medial compartmental osteoarthritis, 23 knees of which were treated with high tibial osteotomy. The bone mineral density of the medial femoral and tibial condyles was greater than that of the lateral femoral and tibial condyles in all knees with medial compartmental osteoarthritis. The ratios of bone mineral density of the medial condyles to that of the lateral condyles were found to

Intramedullary osteosclerosis of the tibia with inflammatory features: Multimodality imaging findings.

Intramedullary osteosclerosis (Ribbing disease) is a rare benign sclerosing bone disorder characterized by diaphyseal cortical thickening and endosteal sclerosis, most commonly involving the tibia. We report the case of a 30-year-old woman presenting with a one-year history of progressive bilateral tibial pain, initially affecting the right side. Radiography and computed tomography demonstrated asymmetric diaphyseal cortical thickening with medullary canal narrowing and metaphyseal sparing. Magnetic resonance imaging revealed marked cortical thickening associated with bone marrow edema and cir

Neurological manifestations in patients and disease carriers in an Italian family with osteosclerosis.

BACKGROUND: Hereditary cranial hyperostosis is a rare disease never described in Italy, so the neurological manifestations in patients and carriers of the disease have been little studied. METHODS: We describe the neurological and neuroimaging features of patients and carriers of the gene from a large Italian family with sclerosteosis. RESULTS: In this family, genetic testing detected the homozygous p.Gln24X (c.70C > T) mutation of the SOST gene in the proband and a heterozygous mutation in 9 siblings. In homozygous adults, severe craniofacial hyperostosis was manifested by cranial neuropath

A histomorphometric study of haematological disorders with respect to marrow fibrosis and osteosclerosis.

A retrospective investigation of 75 EDTA-decalcified Jamshidi biopsies collected over a 2-year period at Aarhus University Hospital was performed. The biopsies originated from 75 patients suffering from idiopathic myelofibrosis, other chronic myeloproliferative disorders, or other conditions with known associations with bone marrow fibrosis. The relative volumes of trabecular and woven bone, as well as haematopoietic and non-haematopoietic tissue, were estimated histomorphometrically. The degree of fibrosis was estimated semiquantitatively. Finally, the thickness of trabecular osteons was esti

[Vertebral osteosclerosis, sacro-iliitis and palmoplantar dermatitis. Apropos of a case].

The authors report a case of aseptic vertebral osteitis which they classify in the context of SAPHO in view of the association with unilateral sacro-ileitis and palmoplantar dermatitis. They stress the importance of recognizing this disease which is initially suggestive of either a bone disease (Paget's disease, bone metastasis) or infectious discospondylitis.

Pyruvate kinase deficiency anemia with terminal myelofibrosis and osteosclerosis in a beagle.

A 15-month-old male Beagle with chronic hemolytic anemia was found to have erythrocytic pyruvate kinase deficiency and, terminally, myelofibrosis and osteosclerosis. The dog's erythron was studied by procedures that enabled close comparison with congenital hemolytic anemia (pyruvate kinase deficiency) of Basenji dogs. The affected dog's sire, dam, and one littermate--each clinically and hematologically normal--were found to have 50% reduction in erythrocytic pyruvate kinase (PK) activity.

Sclerosing bone dysplasias: review and differentiation from other causes of osteosclerosis.

Sclerosing bone dysplasias are skeletal abnormalities of varying severity with a wide range of radiologic, clinical, and genetic features. Hereditary sclerosing bone dysplasias result from some disturbance in the pathways involved in osteoblast or osteoclast regulation, leading to abnormal accumulation of bone. Several genes have been discovered that, when disrupted, result in specific types of hereditary sclerosing bone dysplasia (osteopetrosis, pyknodysostosis, osteopoikilosis, osteopathia striata, progressive diaphyseal dysplasia, hereditary multiple diaphyseal sclerosis, hyperostosis corti

Difficulties in Differentiating Osteosclerosis in Patients With Multifocal Micronodular Pneumocyte Hyperplasia and Cancer.

A 52-year-old woman with multifocal micronodular pneumocyte hyperplasia in bilateral lungs and multiple sclerotic bone lesions (SBLs) visited our hospital. Tuberous sclerosis complex (TSC) was suspected but did not meet the diagnostic criteria. Ten years later, at age 62, the patient developed ureteral cancer. Cisplatin-containing chemotherapy ameliorated ureteral tumor, concomitant with exacerbation of SBLs. It was difficult to distinguish whether the exacerbation of SBLs was due to exacerbation of TSC or bone metastasis of cancer. The administration of cisplatin made the diagnosis even more

Diagnosis and treatment of intramedullary osteosclerosis: a report of three cases and literature review.

BACKGROUND: Intramedullary osteosclerosis (IMOS) is a rare condition without specific radiological findings except for the osteosclerotic lesion and is not associated with family history and infection, trauma, or systemic illness. Although the diagnosis of IMOS is confirmed after excluding other osteosclerotic lesions, IMOS is not well known because of its rarity and no specific feature. Therefore, these situations might result in delayed diagnosis. Hence, this case report aimed to investigate three cases of IMOS and discuss imaging findings and clinical outcomes. CASE PRESENTATION: All three
