Microtitre plate assay for paralytic shellfish toxins using saxiphilin: gauging the effects of shellfish extract matrices, salts and pH upon assay performance.

Saxiphilin is a hydrophilic protein with a high affinity and specificity for paralytic shellfish toxins (PSTs) found in the circulatory fluid of many invertebrates and ectothermic vertebrates. Saxiphilin has been found to be closely related to the iron binding transferrins, a group of proteins that range in molecular weight between 70 and 90 kDa. One saxiphilin isoform, that from the centipede Ethmostigmus rubripes, has been used to develop a microtitre plate assay for PSTs which relies upon detection of bound tritiated saxitoxin (STX). In this study, this assay was challenged with differing c

Environmental Drivers of Vibrio cholerae Abundances in Mobile Bay, Alabama.

Vibrio cholerae is the etiological agent of the illness cholera. However, there are non-O1/non-O139 V. cholerae (NOVC) strains that generally lack the toxin gene (ctx) and colonization factors that cause cholera. These NOVC strains are autochthonous members of estuarine environments and a significant cause of seafood-borne gastroenteritis in the United States. The objective of this study was to identify environmental parameters that correlate with NOVC prevalence in oysters, water, and sediment at three ecologically diverse locations in Mobile Bay, AL, including Dog River (DR), Fowl River (FR)

Field study on short-term changes in benthic environment and benthic microbial communities using pyrolyzed oyster shells.

To evaluate the effect of pyrolyzed crushed oyster shells (PCOS) on the remediation of sediments and microbial diversity, a field study was conducted in Buksin Bay, Tongyeong City, Republic of Korea. It was observed that after treatment with PCOS, the concentration of H2S in the sediment of the control site was 287 mg/L. Furthermore, it decreased up to 0 mg/L and remained so until the end of the field study, that is for a period of six months. Moreover, the concentrations of NO2-N + NO3-N, NH4-N, and PO4-P decreased sharply, and the oxidation-reduction potential (ORP) increased after PCOS trea

In situ dynamics of Vibrio parahaemolyticus and Vibrio vulnificus in water, sediment and triploid Crassostrea virginica oysters cultivated in floating gear.

AIMS: To understand spatial-temporal distribution of Vibrio parahaemolyticus and Vibrio vulnificus in triploid Crassostrea virginica in off-bottom aquaculture. METHODS AND RESULTS: Oysters, sediments and water were seasonally collected in Georgia, USA. V. parahaemolyticus and V. vulnificus were quantified with tlh/tdh/trh and vvhA genes, respectively. No tdh/trh genes were detected. Highest concentrations of tlh gene were observed in summer sediments > oysters > water (105 /g, 104 /g, 103 /ml). VvhA concentrations were similar in sediments and oysters but never exceeded ≥3 × 101 /ml in water.

Minimal effects of oyster aquaculture on local water quality: Examples from southern Chesapeake Bay.

As the oyster aquaculture industry grows and becomes incorporated into management practices, it is important to understand its effects on local environments. This study investigated how water quality and hydrodynamics varied among farms as well as inside versus outside the extent of caged grow-out areas located in southern Chesapeake Bay. Current speed and water quality variables (chlorophyll-a fluorescence, turbidity, and dissolved oxygen) were measured along multiple transects within and adjacent to four oyster farms during two seasons. At the scale of individual aquaculture sites, we were a

First detection and genetic characterization of Toxoplasma gondii in market-sold oysters in China.

Toxoplasma gondii oocysts in the water might be filtered through the gills of shellfish in the process of feeding, and can be concentrated in the digestive glands. Consumers might become infected through ingestion of the shellfish unless they are discharged or inactivated by the shellfish. Thus, the purpose of this study was to evaluate the presence of T. gondii in market-sold oysters in China under natural conditions using a molecular approach. A total of 998 oysters were collected from markets in four cities (Weihai, Qingdao, Yantai and Rizhao) of Shandong province, eastern China. Of these,

Molecular Characterization of Voltage-Gated Sodium Channels and Their Relations with Paralytic Shellfish Toxin Bioaccumulation in the Pacific Oyster Crassostrea gigas.

Paralytic shellfish toxins (PST) bind to voltage-gated sodium channels (Nav) and block conduction of action potential in excitable cells. This study aimed to (i) characterize Nav sequences in Crassostrea gigas and (ii) investigate a putative relation between Nav and PST-bioaccumulation in oysters. The phylogenetic analysis highlighted two types of Nav in C. gigas: a Nav1 (CgNav1) and a Nav2 (CgNav2) with sequence properties of sodium-selective and sodium/calcium-selective channels, respectively. Three alternative splice transcripts of CgNav1 named A, B and C, were characterized. The expression

Evaluation of two virome probe hybridization capture panels for food safety surveillance.

In recent years, viromics has received growing attention for viral disease surveillance. This study set out to compare the VirCapSeq-VERT panel and the Comprehensive Viral Research Panel (CVR Panel) for probe hybridization capture of viral nucleic acids in oyster extracts, a main vehicle for the transmission of foodborne viruses. Using ten-fold serial dilutions of human norovirus (hNoV) GI.2 and GII.4 spike-in oyster extracts, both hybridization capture panels achieved detection levels down to 14 genome copies (gc) for hNoV GI.2 and 5 gc for hNoV GII.4. For hNoV GI.2, a genome coverage of ≥ 95

Amplicon-Based High-Throughput Sequencing Method for Genotypic Characterization of Norovirus in Oysters.

Norovirus is a highly diverse RNA virus often implicated in foodborne outbreaks, particularly those associated with shellfish. Shellfish are filter feeders, and when harvested in bays exposed to wastewater overflow or storm overflows, they can harbor various pathogens, including human-pathogenic viruses. The application of Sanger or amplicon-based high-throughput sequencing (HTS) technologies to identify human pathogens in shellfish faces two main challenges: (i) distinguishing multiple genotypes/variants in a single sample and (ii) low concentrations of norovirus RNA. Here, we assessed the pe

Metal stable isotopes in transplanted oysters as a new tool for monitoring anthropogenic metal bioaccumulation in marine environments: The case for copper.

Metal release into the environment from anthropogenic activities may endanger ecosystems and human health. However, identifying and quantifying anthropogenic metal bioaccumulation in organisms remain a challenging task. In this work, we assess Cu isotopes in Pacific oysters (C. gigas) as a new tool for monitoring anthropogenic Cu bioaccumulation into marine environments. Arcachon Bay was taken as a natural laboratory due to its increasing contamination by Cu, and its relevance as a prominent shellfish production area. Here, we transplanted 18-month old oysters reared in an oceanic neighbor are

Species-specific mechanisms of cytotoxicity toward immune cells determine the successful outcome of Vibrio infections.

Vibrio species cause infectious diseases in humans and animals, but they can also live as commensals within their host tissues. How Vibrio subverts the host defenses to mount a successful infection remains poorly understood, and this knowledge is critical for predicting and managing disease. Here, we have investigated the cellular and molecular mechanisms underpinning infection and colonization of 2 virulent Vibrio species in an ecologically relevant host model, oyster, to study interactions with marine Vibrio species. All Vibrio strains were recognized by the immune system, but only nonvirule

Identification of MMV Malaria Box inhibitors of Perkinsus marinus using an ATP-based bioluminescence assay.

"Dermo" disease caused by the protozoan parasite Perkinsus marinus (Perkinsozoa) is one of the main obstacles to the restoration of oyster populations in the USA. Perkinsus spp. are also a concern worldwide because there are limited approaches to intervention against the disease. Based on the phylogenetic affinity between the Perkinsozoa and Apicomplexa, we exposed Perkinsus trophozoites to the Medicines for Malaria Venture Malaria Box, an open access compound library comprised of 200 drug-like and 200 probe-like compounds that are highly active against the erythrocyte stage of Plasmodium falc

Distribution of butyltin and derivatives in oyster shells and trapped sediments of two estuaries in Cantabria (Northern Spain).

Distribution of butyltin compounds (BTs) and derivatives (monobutyltin, MBT; dibutyltin, DBT and tributyltin, TBT) was analysed in Crassostrea gigas oyster shells and the sediments trapped in the shell chambers, from two different estuaries in Cantabria (Northern Spain), with very different environmental conditions, where previous data have not been reported. Inorganic tin analysis in oyster shells was performed in order to study the degradation of BTs. Shell thickening and losses in biological growth are related with the presence of TBT, and were determined using three morphological indexes.

Increase of bone mineral density by calcium supplement with oyster shell electrolysate.

The effect of calcium supplementation, in patients with osteoporosis is still a matter of controversy. Oyster shell electrolysate (OSE) was reported to raise serum calcium and increase urinary calcium excretion in vitamin D-deficient states more readily than calcium carbonate. Since the effect of calcium salts on osteoporosis depends heavily on its bioavailability, the effect of 900 mg/day calcium as OSE was tested in 12 elderly osteoporotic females, using radial bone mineral density measured by single photon absorptiometry and spinal trabecular bone density measured by quantitative computed t

Glycosylation of oyster peptides generates supramolecular assembly of glycopeptides to relieve DSS-induced colitis.

The incidence of colitis is increasing annually, however, effective treatment options remain limited. Oyster peptides are a food source that has shown promising nutritional strategy for colitis alleviation. However, their clinical translation is hindered by poor stability and rapid in vivo metabolism. To address these limitations, we employed glycosylation modification to enhance the stability and functional properties of oyster peptides. Using a Maillard reaction-driven supramolecular assembly approach, we synthesized glycopeptides with a glycosylation degree of 42.5%. In a murine model of de

Study on defluorination performance and mechanism of Fe-Al modified oyster shell substrate coupled with microbially induced calcium precipitation for enhanced ecological floating beds.

Fluorine is the most reactive nonmetallic element. Low fluoride concentrations in drinking water prevent dental caries, while excessive fluoride leads to fluorosis. To overcome the low fluoride (F⁻) removal efficiency and high cost of conventional ecological floating beds (EFBs), this study developed two slow-release materials (carbon-releasing and calcium-releasing) and fabricated an Fe-Al modified substrate primarily using oyster shells as the raw material, while integrating microbial-induced calcium precipitation technology to enhance the treatment efficiency of the "substrate-microbe" syst

Characterization of Oyster Protein Hydrolysate-Iron Complexes and their In Vivo Protective Effects against Iron Deficiency-Induced Symptoms in Mice.

Iron is one of the trace mineral elements, and iron deficiency is a common phenomenon that negatively influences human health. Food-derived iron supplements were considered excellent candidates for improving this syndrome. In this work, oyster-protein hydrolysates (OPH) and ferrous chloride successfully formed the OPH-Fe complex (6 mg/mL, 40 °C, 30 min), where the main binding sites involved were the carboxyl and amino groups. The OPH-Fe complex showed no obvious changes in the secondary structure, while the iron changed the morphological appearance and also showed fluorescence quenching, an u

[Effects of Oyster Shell Powder and Lime on Availability and Forms of Phosphorus and Enzyme Activity in Acidic Paddy Soil].

Soil acidification improvement in the main grain production regions of southern China is an important issue to enhance the quality of cultivated land and promote grain yield. In order to explore the effects of oyster shell powder and lime on acidity and availability and inorganic forms of phosphorus in acidic paddy soil, a pot experiment was performed using oyster shell powder and lime amendments with dosages of 0.05%, 0.10%, and 0.15%. The results showed that both oyster shell powder and lime significantly (P<0.05) increased the pH and decreased exchangeable acid content of paddy soil. The im
