Engineering CBM of β-Mannanase from Paenibacillus to Alter Product Chain Length for Preparing Mannan Oligosaccharide Prebiotics.

Biomass polysaccharides are explored as inexpensive raw materials for preparing value-added products, such as mannan oligosaccharides (MOS). However, the varied composition of MOS and the underlying mechanism have not been studied thoroughly yet. Here a β-mannanase from Paenibacillus sp. was engineered for hydrolyzing biomass polysaccharides and producing prebiotic MOS. Structural analysis and alanine scanning revealed two hot spots, Q66 and T88, in the carbohydrate binding module (CBM). The mutant T88Y demonstrated a 65% activity improvement and a 5-fold thermal stability enhancement at 70 °C

Molecular detection of Paenibacillus larvae, Melissococcus plutonius and Vairimorpha spp., in historical honey samples from South Brazil.

Honey bee colonies are threatened by several pathogens, including Paenibacillus larvae, Melissococcus plutonius, Vairimorpha apis, and Vairimorpha ceranae. Those infections affect bee health, survival, and productivity. The social behavior of the bees facilitates the spread of these pathogens, making disease management a significant challenge in apiculture. This study aimed to assess the historical presence of Paenibacillus larvae, Melissococcus plutonius, Vairimorpha apis, and Vairimorpha ceranae in honey samples from Rio Grande do Sul, Brazil. A total of 125 archival honey samples collected

Biogeography of Paenibacillus larvae, the causative agent of American foulbrood, using a new multilocus sequence typing scheme.

American foulbrood is the most destructive brood disease of honeybees (Apis mellifera) globally. The absence of a repeatable, universal typing scheme for the causative bacterium Paenibacillus larvae has restricted our understanding of disease epidemiology. We have created the first multilocus sequence typing scheme (MLST) for P. larvae, which largely confirms the previous enterobacterial repetitive intergenic consensus (ERIC)-polymerase chain reaction (PCR)-based typing scheme's divisions while providing added resolution and improved repeatability. We have used the new scheme to determine the

Neonatal Paenibacilliosis: Paenibacillus Infection as a Novel Cause of Sepsis in Term Neonates With High Risk of Sequelae in Uganda.

BACKGROUND: Paenibacillus thiaminolyticus may be an underdiagnosed cause of neonatal sepsis. METHODS: We prospectively enrolled a cohort of 800 full-term neonates presenting with a clinical diagnosis of sepsis at 2 Ugandan hospitals. Quantitative polymerase chain reaction specific to P. thiaminolyticus and to the Paenibacillus genus were performed on the blood and cerebrospinal fluid (CSF) of 631 neonates who had both specimen types available. Neonates with Paenibacillus genus or species detected in either specimen type were considered to potentially have paenibacilliosis, (37/631, 6%). We de

Potential synergistic antimicrobial efficiency of binary combinations of essential oils against Bacillus cereus and Paenibacillus amylolyticus-Part A.

The checkerboard method was used to study the potential interactions between eight essential oils (Basil, Cinnamon, Eucalyptus, Mandarin, Oregano, Peppermint, Tea tree, and Thyme) when used as antibacterial agents against Bacillus cereus LSPQ 2872 and Paenibacillus amylolyticus ATCC 9995. The minimum inhibitory concentration (MIC) of each essential oil (EO) and the fractional inhibitory concentration (FIC) index for the binary combinations of essential oils (EOs) were determined. According to FIC index values, some of the compound binary combinations showed an additive effect; however, Thyme/T

Isolation and identification of lipopeptide antibiotics from Paenibacillus elgii B69 with inhibitory activity against methicillin-resistant Staphylococcus aureus.

Two lipopeptide antibiotics, pelgipeptins C and D, were isolated from Paenibacillus elgii B69 strain. The molecular masses of the two compounds were both determined to be 1,086 Da. Mass-spectrometry, amino acid analysis and NMR spectroscopy indicated that pelgipeptin C was the same compound as BMY-28160, while pelgipeptin D was identified as a new antibiotic of the polypeptin family. These two peptides were active against all the tested microorganisms, including antibiotic-resistant pathogenic bacterial strains such as methicillin-resistant Staphylococcus aureus (MRSA). Paenibacillus polymyxa L62 synergistically regulates Pb2+ speciation and reconstructs soil microecological networks via surface complexation and PbS mineralization.

The adsorption activity of Paenibacillus polymyxa L62 toward Pb2+ was evaluated in aqueous systems. Strain L62 achieved a Pb2+ removal efficiency of 95.97%. Its cell surface was enriched with functional groups such as -OH and -COO⁻, which promoted Pb2+ adsorption via complexation and ion exchange. In aqueous systems, XRD, FTIR, and EDS analyses suggested the possible formation of stable PbS crystalline precipitates on the cell surface. In soil incubation experiments, strain L62 was inoculated into Pb-contaminated soil at 1% (L1) and 3% (L3). Both treatments significantly promoted Pb transforma

Characterization of C3larvinA, a novel RhoA-targeting ADP-ribosyltransferase toxin produced by the honey bee pathogen, Paenibacillus larvae.

C3larvinA is a putative virulence factor produced by Paenibacillus larvae enterobacterial-repetitive-intergenic-consensus (ERIC) III/IV (strain 11-8051). Biochemical, functional and structural analyses of C3larvinA revealed that it belongs to the C3-like mono-ADP-ribosylating toxin subgroup. Mammalian RhoA was the target substrate for its transferase activity suggesting that it may be the biological target of C3larvinA. The kinetic parameters of the NAD+ substrate for the transferase (KM = 75 ± 10 µM) and glycohydrolase (GH) (KM = 107 ± 20 µM) reactions were typical for a C3-like bacterial tox

Tetracycline-resistance encoding plasmids from Paenibacillus larvae, the causal agent of American foulbrood disease, isolated from commercial honeys.

Paenibacillus larvae, the causal agent of American foulbrood disease in honeybees, acquires tetracycline-resistance via native plasmids carrying known tetracycline-resistance determinants. From three P. larvae tetracycline-resistant strains isolated from honeys, 5-kb-circular plasmids with almost identical sequences, designated pPL373 in strain PL373, pPL374 in strain PL374, and pPL395 in strain PL395, were isolated. These plasmids were highly similar (99%) to small tetracycline-encoding plasmids (pMA67, pBHS24, pBSDMV46A, pDMV2, pSU1, pAST4, and pLS55) that replicate by the rolling circle mec

Genomic diversity and tracing of Paenibacillus larvae in Australia: implications for American foulbrood outbreak surveillance in low-diversity populations.

Paenibacillus larvae is the causative agent of American foulbrood (AFB) in honeybees (Apis mellifera) and a devastating pathogen for honey and pollination industries worldwide. Despite this threat, a genomic survey of P. larvae has not been attempted within Australia. To examine the diversity of Australian populations, we sequenced 368 P. larvae genomes sourced primarily from south-eastern Australia. Multilocus sequencing typing analysis identified only 4 sequence types across all 368 samples, with 2 sequence types (ST18 and ST5) representing 96% of all isolates. In comparison to European-sour

Infected Nonunion of the Tibia Due to Paenibacillus turicensis in a Healthy Young Adult After an All-Terrain Vehicle Accident.

We present the case of a 19-year-old man with an open fracture of the tibia and fibula secondary to an accident with an all-terrain vehicle. He underwent operative excisional irrigation, debridement, and fixation on the day of injury. His course was complicated by nonunion of the tibia fracture. Infection is a common factor in fracture nonunion, even in patients who receive appropriate surgical and antimicrobial management. Paenibacillus turicensis, an organism adapted to survive in the environment via spore formation, was responsible for nonunion in our patient. A brief discussion of this unu

Functional and genomic analyses of plant growth promoting traits in Priestia aryabhattai and Paenibacillus sp. isolates from tomato rhizosphere.

This study investigated plant growth-promoting (PGP) mechanisms in Priestia aryabhattai VMYP6 and Paenibacillus sp. VMY10, isolated from tomato roots. Their genomes were initially assessed in silico through various approaches, and these observations were then compared with results obtained in vitro and in vivo. Both possess genes associated with the production of siderophores, indole acetic acid (IAA) and cytokinins (CKs), all of which have been shown to promote plant growth. The two strains were able to produce these compounds in vitro. Although both genomes harbor genes for phosphorus solubi
