AChE and CoFe NPs-initiated cascade amplification for sensitive detection of organophosphorus pesticides.

Traditional methods for pesticide residue detection often rely on expensive instruments, are time-consuming, and lack cost efficiency. This investigation developed a fluorescence-based assay that utilizes cascade amplification through cholinesterase hydrolysis and thiamine (TH) oxidation for the detection of organophosphorus pesticides (OPs). A Prussian blue analog (CoFe NPs) with high catalytic activity for TH oxidation was synthesized. Cholinesterase hydrolyzes acetylthiocholine to produce thiocholine, which subsequently inhibits the CoFe NPs-catalyzed oxidation of TH. In the presence of par

Nanostructured photoelectrochemical biosensor for highly sensitive detection of organophosphorous pesticides.

A sensitive photoelectrochemical (PEC) biosensor for detection of organophosphorus pesticides (OPs) using the nanocomposite of CdSe@ZnS quantum dots (QDs) and graphene deposited on the ITO coated glass electrode as a photoactive electrode is presented. The integration of CdSe@ZnS/graphene nanocomposite with biomolecules acetylcholinesterase (AChE) as a biorecognition element yields a novel biosensing platform. Under visible light irradiation, the AChE-CdSe@ZnS/graphene nanocomposite can generate a stable photocurrent and the photocurrent is found to be inversely dependent on the concentration

Interaction between human serum esterases and environmental metal compounds.

Paraoxonase-1 (PON1) and cholinesterase (BChE) are two of the major human serum esterases. Although most of variation in PON1 activity results from genetic factors, there is growing evidence that environmental chemicals also modulate its activity. The aim of this study was to investigate whether environmental exposure to metal compounds has any influence on those esterases. A cross-sectional study was conducted in a representative sample of the general population of Andalusia, South of Spain. PON1 activity against different substrates (paraoxon, phenylacetate, diazoxon and dihydrocoumarin) and

Oral treatment of organophosphate poisoning in mice.

OBJECTIVE: Organophosphates are used as pesticides, herbicides, and chemical warfare agents. Treatment of organophosphate poisoning is with intravenous atropine and pralidoxime in addition to supportive care. This study determined the efficacy of oral agents in preventing death from organophosphate poisoning. METHODS: The organophosphate paraoxon (8 mg/kg) was used in a murine model with lethality at four and 24 hours as an end point. For oral treatment, 15 male Balbc mice were given either atropine sulfate (4 mg/kg), or a combination of atropine sulfate (4 mg/kg) with pralidoxime (100 mg/kg)

The effect of gallamine, gallopamil and nifedipine on responses to acetylcholine and carbachol in the taenia of the guinea-pig caecum.

The effects of gallamine, gallopamil and nifedipine on isotonic contractions of the isolated taenia of the guinea-pig caecum produced by acetylcholine (ACh) or carbachol (CCh) were investigated. Gallamine (0.1 to 0.3 mM) inhibited contractions produced by CCh more than those produced by ACh. The difference was still present after pretreatment of the tissue with paraoxon (10 microM for 20 min) to inhibit cholinesterases or in experiments carried out in the presence of tetrodotoxin (0.3 microM) to exclude possible ganglionic stimulation by the agonists. Gallopamil or nifedipine selectively inhib

Potential Common Mechanisms of Cytotoxicity Induced by Organophosphorus Pesticides via NLRP3 Inflammasome Activation.

The Multi-Threat Medical Countermeasure (MTMC) technique is crucial for developing common biochemical signaling pathways, molecular mediators, and cellular processes. This study revealed that the Nod-like receptor 3 (NLRP3) inflammasome pathway may be a significant contributor to the cytotoxicity induced by various organophosphorus pesticides (OPPs). The study demonstrated that exposure to six different types of OPPs (paraoxon, dichlorvos, fenthion, dipterex, dibrom, and dimethoate) led to significant cytotoxicity in BV2 cells, which was accompanied by increased expression of NLRP3 inflammasom

Highly-sensitive organophosphorus pesticide biosensors based on CdTe quantum dots and bi-enzyme immobilized eggshell membranes.

An optical biosensing method using CdTe quantum dots (QDs) and bi-enzyme-immobilized eggshell membranes for the determination of organophosphorus pesticides (OPs) has been developed. Increasing amounts of OPs led to a decrease of the enzymatic activity and thus a decrease in the production of hydrogen peroxide (H2O2), which can quench the fluorescence of the CdTe QDs. Under the optimum conditions, there was a good linear relationship between the enzyme inhibition percentage and the logarithm of paraoxon or parathion concentration in the range of 1.0 × 10(-11)-1.0 × 10(-6) mol L(-1). Organophosphorus compound detection on a cell chip with yeast coexpressing hydrolase and eGFP.

Organophosphorus compounds (OPs) such as pesticides, fungicides, and herbicides are highly toxic but are nevertheless extensively used worldwide. To detect OPs, we constructed a yeast strain that co-displays organophosphorus hydrolase (OPH) and enhanced green fluorescent protein (EGFP) on the cell surface using a Flo1p anchor system. OP degradation releases protons and causes a change in pH. This pH change results in structural deformation of EGFP, which triggers quenching of its fluorescence, thereby making this cell useful for visual detection of OPs. Fluorescence microscopy confirmed the hi

Lack of inhibition of glycolytic enzymes by the neurotoxic organophosphorus compounds mipafox and methamidofos.

Preincubation with Mipafox and Methamidofos as well as Paraoxon (used as control) did not cause inhibition of hexokinase, phosphofructokinase, glyceraldehyde-3-phosphate dehydrogenase and lactate dehydrogenase. This is in contrast with the inhibition of glycolysis by other neurotoxic compounds (hexacarbons, acrylamide, carbon disulfide).

Effects of arecoline and cholinesterase inhibitors on cyclic guanosine 3',5'-monophosphate and adenosine 3'.5'-monophosphate in mouse brain.

In mice, Arecoline in vivo dose-dependently increased the cGMP concentrations of the cerebellum and the "cereberum" (= parts of cortex, hippocampus, hypothalamus, thalamus, striatum and midbrain) without influencing thecAMP levels. The cholinesterase inhibitors paraoxon and physostigmine caused an elevation only in "cerebrum", whereas the cGMP content of the cerebellum even decreased. Pretreatment with atropine prevented the rise in cGMP levels as well as the symptoms of cholinergic stimulation elicited by arecoline or paraoxon. Diazepam reduced cGMP levels below control values and blocked the

Thermostable esterase 2 from Alicyclobacillus acidocaldarius as biosensor for the detection of organophosphate pesticides.

Pesticides are the plague of modern times, although much needed in agriculture, causing damage to the entire ecosystem, including humans. The high operative costs and the requirement of specialized personnel for pesticide detection, incentive to develop alternative solutions such as the set up of cheap, rapid, and simple to use biosensors. In this work, we evaluate the possibility to use the esterase 2 from Alicyclobacillus acidocaldarius as a biosensor for the detection of specific organophosphate pesticides. With the recent demonstration of the very high affinity of esterase 2 toward paraoxo

Human liver paraoxonase (PON1): subcellular distribution and characterization.

The subcellular localization and different biochemical properties of a human hepatic microsomal enzyme that hydrolyses paraoxon (paraoxonase, PON1) were studied and compared to the paraoxon hydrolase activity found in human plasma as well as in rat liver and plasma. Having evaluated the influence of the postmortem interval by a parallel experiment performed in rats, we conclude that the paraoxonase activity was preferentially localized in the microsomal fraction. The enzyme reaction was optimized according to temperature, pH, buffer, ionic strength, substrate concentration, and enzyme protein

A heat stable paraoxonase (O,O-diethyl O-p-nitrophenyl phosphate O-p-nitrophenyl hydrolase) from Russell's viper venom.

Fractionation of Russell's viper venom revealed separate phosphohydrolase activities directed against p-nitrophenyl phosphate, bis(p-nitrophenyl) phosphate, p-nitrophenylthymidylic acid, and O,O-diethyl p-nitrophenyl phosphate (paraoxon). On gel fractionation, the first two activities are eluted ahead of the latter. They could be resolved further by phosphocellulose cation exchange chromatography. The hydrolytic activities directed against p-nitrophenylthymidylic acid hydrolyzing component is heat labile, while the paraoxon hydrolyzing component manifests an unusually high degree of heat stabi

Increased expression of a bacterial phosphotriesterase in Escherichia coli through directed evolution.

We devised a growth-based strategy for screening phosphotriesterase mutant libraries for variants with enhanced activity towards organophosphates that generate dimethyl phosphate when hydrolysed. Phosphotriesterase mutants were screened for activity by growing transformed Escherichia coli on agar plates containing methyl paraoxon as a sole phosphorus source. E. coli is capable of growth under these conditions when coexpressing the phosphotriesterase from Agrobacterium radiobacter P230 (OpdA) and the glycerophosphodiester phosphodiesterase from Enterobacter aerogenes (GpdQ). N-terminal PEGylation enhances organophosphorus hydrolase catalysis for a promising fast and long-acting prophylactic candidate.

Organophosphorus compounds (OPs) poisoning poses a significant health risk as an insecticide, and its potent toxicity is characterized by rapid onset and a very narrow window for intervention. Prophylactic medication is crucial for managing OPs poisoning, yet effective and safe drugs are still lacking. The enzyme Organophosphorus hydrolase (OPH) shows promise as a bioscavenger but faces challenges due to its short half-life and strong immunogenicity. Our research reveals that N-terminal PEGylation of OPH significantly extends its pharmacokinetic half-life, reduces immunogenicity, and, surprisi

Rational engineering of a native hyperthermostable lactonase into a broad spectrum phosphotriesterase.

The redesign of enzyme active sites to alter their function or specificity is a difficult yet appealing challenge. Here we used a structure-based design approach to engineer the lactonase SsoPox from Sulfolobus solfataricus into a phosphotriesterase. The five best variants were characterized and their structure was solved. The most active variant, αsD6 (V27A-Y97W-L228M-W263M) demonstrates a large increase in catalytic efficiencies over the wild-type enzyme, with increases of 2,210-fold, 163-fold, 58-fold, 16-fold against methyl-parathion, malathion, ethyl-paraoxon, and methyl-paraoxon, respect

Interaction of ethanol and the organophosphorus insecticide parathion.

Phosphorothioate insecticides such as parathion (O,O-diethyl-O-p-nitrophenyl phosphorothioate) undergo P450-dependent oxidative desulfuration, leading to both activation and detoxification of these compounds. Consequently, alterations in P450-dependent oxidative desulfuration may affect the acute toxicities of these insecticides. In the present study, pretreatment of mice with 15% ethanol in the drinking water for 6 days antagonized the acute toxicity of parathion, but not its toxic metabolite paraoxon (O,O-diethyl-O-p-nitrophenyl phosphate), suggesting that ethanol affected the oxidative desu

Phenylthioacetate: a useful substrate for the histochemical and colorimetric detection of cholinesterase.

Cholinesterase from frozen sections of honey bee (Apis mellifera, L.) brain hydrolyzes phenylthioacetate and acetylthiocholine to give virtually identical patterns of enzyme distribution with and without the use of paraoxon. In addition, phenylthioacetate is an economical substrate for in vitro and in vivo studies of cholinesterase. Phenylthioacetate offers the advantages of being easily synthesized and lipoid soluble, and appears to penetrate membranes easily.
