Occurrence of Cruzia tentaculata Larvae in the Land Snail Megalobulimus abbreviatus-Influence of Natural Parasite Load on Snail Physiology.

The nematode Cruzia tentaculata is reported here for the first time in the land snail Megalobulimus abbreviatus in southern Brazil. The snails were infected with a high prevalence of larvae up to the L3 stage (68.6%). Cysts were located mainly in the mantle (pulmonary cavity) and the nerve ring. No other helminths were identified in the collected snails. Some physiological aspects were compared between snails infected with L3 larvae and non-infected snails and among infected animals with different parasite loads. No differences were found in hemolymph protein, glucose, or urea content between

Assessing L. donovani Skin Parasite Load: A Proof of Concept Study of a Microbiopsy Device in an Indian Setting.

BACKGROUND: In the endgame of the elimination initiative of visceral leishmaniasis (VL) on the Indian subcontinent, one of the main questions remaining is whether asymptomatically infected individuals also contribute to transmission. We piloted a minimally invasive microbiopsy device that could help answer this question. While the potential of this device has been previously illustrated in Ethiopia, no such information is available for the setting of the Indian subcontinent. In this proof of concept study we aimed to assess 1) to what extent skin parasite load obtained with the new microbiopsy

Systemic inflammatory response syndrome in dogs naturally infected with Babesia canis: Association with the parasite load and host factors.

The common signs of canine babesiosis caused by an infection with Babesia canis are fever, anorexia, lethargy, pulse alterations, anemia, and occasionally mild icterus. Dogs with these clinical signs can be divided into two groups: those with acute-phase reaction and those with systemic inflammatory response syndrome (SIRS). Factors associated with the occurrence of SIRS in canine babesiosis have not been thoroughly researched. This article outlines a cross-sectional study of 54 client-owned dogs with an acute B. canis infection, and evaluates the differences in age, gender, laboratory finding

Seroprevalence and parasite load of Toxoplasma gondii in Mexican hairless pig (Sus scrofa) tissues from the Southeast of Mexico.

This study aimed to estimate the seroprevalence and determine the parasite load of Toxoplasma gondii (T. gondii) in the heart and tongue tissues and the semimembranosus/gracilis muscles in a population of Mexican hairless pig (MHP). A cross-sectional study was conducted in 81 MHP, 9-12 weeks of age, from 10 municipalities in Yucatan, Mexico. The prevalence was estimated by the detection of T. gondii IgG antibodies via an indirect ELISA assay. The parasite burden was also determined by testing genomic DNA from sampled tissues using quantitative real-time PCR (qPCR), which amplified a 62bp produ

Relationship between parasite load and immune responses in early stages of Leishmania donovani infection in inbred BALB/c mice.

The aim of this study was to determine if a correlation exists between Leishmania donovani parasite load and the corresponding humoral and cellular mediated immune responses in the early stages of Leishmania donovani infection in inbred BALB/c mice. Five groups of ten BALB/c mice each were inoculated intraperitoneally with stationary phase metacyclic promastigotes of Leishmania donovani at doses 1 x 10(2), 1 x 10(4), 10(6) and 1 x 10(8) respectively per mouse. Group five mice were not manipulated in any way and were left to serve as control. At weekly intervals, for five weeks, the mice were a

miR-148a regulation interferes in inflammatory cytokine and parasitic load in canine leishmaniasis.

Canine leishmaniasis (CanL) is a severe public health threat. Infected animals mediate transmission of the Leishmania protozoan to humans via the sandfly's bite during a blood meal. CanL progression depends on the degree of suppression of the immune response, possibly associated with microRNAs (miR), which can modulate mRNA translation into proteins and (consequently) regulate cell function. Increased miR-148a in splenic leukocytes (SL) of dogs with CanL was observed in previous studies, and in silico analysis, identified possible pathways involved in immune response regulation that are affect

Parasitic load and histological aspects in different regions of the spleen of dogs with visceral leishmaniasis.

Leishmania infantum causes from subclinical infection to severe disease in humans and dogs. The spleen is one of the organs most affected by the infection. Although evidence exists that the parasitic load distribution and histological alterations may not be homogeneous in the affected organs of naturally infected individuals, it has not been formally demonstrated using the current techniques used for studying the disease. In six dogs naturally infected with Leishmania, parasitic load and histological changes were compared in samples collected from the lower, middle and upper third of the splee

Body Condition Peaks at Intermediate Parasite Loads in the Common Bully Gobiomorphus cotidianus.

Most ecologists and conservationists perceive parasitic infections as deleterious for the hosts. Their effects, however, depend on many factors including host body condition, parasite load and the life cycle of the parasite. More research into how multiple parasite taxa affect host body condition is required and will help us to better understand host-parasite coevolution. We used body condition indices, based on mass-length relationships, to test the effects that abundances and biomasses of six parasite taxa (five trematodes, Apatemon sp., Tylodelphys sp., Stegodexamene anguillae, Telogaster o

High resolution melting real-time PCR for genotyping of Giardia lamblia assemblages A and B regardless of parasite load.

Giardia lamblia is a globally distributed fecal-oral transmitted enteroparasite. G. lamblia is phylogenetically divided into assemblages A to H. Assemblages A and B have high zoonotic potential, as reported in humans and other animals, including dogs and cats. Currently, G. lamblia genotyping is performed using gene sequencing, which is an expensive technique, especially in developing countries where higher frequencies of the protozoan have been reported. Real-time PCR with High Resolution Melting (qPCR-HRM) is a sensitive method for detecting polymorphisms and is used for differentiate specie

Immune-Pathological Correlates of Disease Severity in New-World Kala-Azar: The Role of Parasite Load and Cytokine Profiles.

Kala-azar is a protracted disease caused by the protozoan Leishmania infantum (zoonotic) or L. donovani (anthroponotic), transmitted by sandflies. Patients present with fever, anemia, and hepatosplenomegaly, potentially progressing to hemorrhaging, secondary infections, and death. Its pathogenesis is linked to an exaggerated cytokine response. We studied 72 hospitalized patients, analyzing clinical data and outcomes in relation to L. infantum DNA loads in blood and bone marrow, and plasma concentrations of IL-1β, IL-6, IL-8, IL-10, IL-12, TNF-α, and TGF-β. Cytokine levels were found to be elev

Molecular detection and parasite load of Trypanosoma cruzi in digestive tract tissue of Chagas disease patients affected by megacolon.

Chagas disease, caused by the Trypanosoma cruzi parasite in the Americas affects ∼ 7 million people, 30% with cardiac tissue damage and 10-15% with digestive disorders. In this study, we have developed a protocol to detect the presence of the parasite and estimate its load in resected dysfunctional tissue segments of chronically infected patients with digestive megacolon. We have included samples from 43 individuals, 38/5 with positive/negative serology for Chagas disease and digestive syndromes. Samples of 1.5 to 2.0 cm2 were taken from different points of the dysfunctional digestive tract in

Immunotherapy using anti-PD-1 and anti-PD-L1 in Leishmania amazonensis-infected BALB/c mice reduce parasite load.

Leishmaniasis is a neglected disease, for which current treatment presents numerous issues. Leishmania amazonensis is the etiological agent of cutaneous and diffuse cutaneous leishmaniasis. The roles of the programmed death-1 (PD-1) receptor on lymphocytes and its ligand (PD-L1) on antigen-presenting cells have been well studied in tumor and other infection models; but little is known about their roles in non-healing cutaneous leishmaniasis. In this study, we observed that L. amazonensis induced PD-1 expression on both CD4+ and CD8+ T cells and PD-L1 on dendritic cells on BALB/c mice. We teste

Increased level of soluble adenosine deaminase in bone marrow of visceral leishmaniasis patients: an inverse relation with parasite load.

Adenosine deaminase (ADA) which degrades adenosine to inosine, is known to be pro-inflammatory molecule in many diseases. Adenosine suppresses the functioning of the immune system and thus promotes dissemination of the parasite. In our previous finding, the level of soluble ADA in serum of visceral leishmaniasis (VL) was found to be increased as compared to healthy controls. However, it cannot be fairly interpreted unless their level is demonstrated at the disease site, where the parasite resides. We designed this study to correlate the level of soluble ADA (sADA) with parasitic load at the di

Correlation of parasite load determined by quantitative PCR to clinical outcome in a heart transplant patient with disseminated toxoplasmosis.

Disseminated toxoplasmosis is a life-threatening infection in transplant recipients, which results either from reactivation of latent infection or from organ-transmitted primary infection. Preventive measures and diagnostic screening methods differ between countries and are related to the seroprevalence of Toxoplasma spp. in the general population. Here we report a case of disseminated toxoplasmosis in a heart transplant recipient with previous immunity that occurred after cotrimoxazole prophylaxis for the prevention of Pneumocystis jirovecii pneumonia was stopped. Quantitative PCR proved usef

Relationship between fecal egg count, parasite load, and histopathological findings in the pancreas of cattle naturally infected with Eurytrema coelomaticum.

Eurytrematosis, a disease caused by Eurytrema spp., primarily affects the pancreas of mammals, including cattle. Although common in Africa, Asia and South America, Eurytrema coelomaticum has recently been detected in Argentina. The present study aimed to evaluate the relationship between parasite load, fecal egg counts (EPG) using two diagnostic techniques (BST and OST), and histopathological lesions in cattle naturally infected with Eurytrema. A total of 120 cattle from an endemic area were selected for the study, with both pancreatic and fecal samples collected for parasitological and histop

Recurrent visceral leishmaniasis relapses in HIV co-infected patients are characterized by less efficient immune responses and higher parasite load.

Visceral leishmaniasis (VL) and HIV co-infection (VL/HIV) has emerged as a significant public health problem in Ethiopia, with up to 30% of patients with VL co-infected with HIV. These patients suffer from recurrent VL relapses and increased mortality. Those with a previous history of VL relapses (recurrent VL/HIV) experience increased VL relapses as compared to patients with HIV presenting with their first episode of VL (primary VL/HIV). Our aim was to identify drivers that account for the higher rate of VL relapses in patients with recurrent VL/HIV (n = 28) as compared to primary VL/HIV (n =

Negative correlation between nuptial throat colour and blood parasite load in male European green lizards supports the Hamilton-Zuk hypothesis.

During female mate choice, conspicuous male sexual signals are used to infer male quality and choose the best sire for the offspring. The theory of parasite-mediated sexual selection (Hamilton-Zuk hypothesis) presumes that parasite infection can influence the elaboration of sexual signals: resistant individuals can invest more energy into signal expression and thus advertise their individual quality through signal intensity. By preferring these males, females can provide resistance genes for their offspring. Previous research showed that nuptial throat colour of male European green lizard, Lac
