T598 and T601 phosphorylation sites of canine parvovirus NS1 are crucial for viral replication and pathogenicity.

Canine parvovirus-2 (CPV-2) is an important pathogen causing severe diseases in dogs and other wild carnivores. Phosphorylation of NS1 may be related to CPV-2 pathogenicity, but the exact mechanism is unclear. Here, we conducted parvovirus disease surveillance in Shaanxi Province of China and 51 fecal swabs were detected to be infected with CPV-2. The 7 CPV-2 strains were identified, all of which belonged to CPV-2c. The complete genome sequence of one of the strains (CPV-2c XY) was cloned into pKQLL plasmid to construct a full-length infectious clone plasmid pX-CPV-2c, which carried a genetic

Development of canine parvovirus-neutralizing monoclonal antibodies from natural host and their germline gene usage.

Canine parvovirus 2 (CPV-2) remains a leading cause of acute infectious gastroenteritis with high global morbidity in dogs. While murine neutralizing monoclonal antibodies (mAbs) are widely used for antiviral therapy, their efficacy is limited by immune rejection in canine recipients. Here, we developed an efficient single B-cell cloning platform to generate canine-derived neutralizing mAbs against CPV-2 and characterized their germline gene usage patterns. Specifically, using biotinylated CPV-2 virions as bait, CPV-2-binding B cells were singly isolated via fluorescence-activated cell sorting

Molecular epidemiology of canine parvovirus type 2 in Vietnam from November 2016 to February 2018.

BACKGROUND: Canine parvovirus type 2 (CPV-2) was first identified in the late 1970s; it causes intestinal hemorrhage with severe bloody diarrhea in kennels and dog shelters worldwide. Since its emergence, CPV-2 has been replaced with new genetic variants (CPV-2a, CPV-2b, and CPV-2c). Currently, information about the genotype prevalence of CPV-2 in Vietnam is limited. In the present study, we investigated the genotype prevalence and distribution of CPV-2 in the three regions of Vietnam. METHODS: Rectal swabs were collected from 260 dogs with suspected CPV-2 infection from northern, central, an

Multiplex Amplification Refractory Mutation System PCR (ARMS-PCR) provides sequencing independent typing of canine parvovirus.

Canine parvovirus-2 antigenic variants (CPV-2a, CPV-2b and CPV-2c) ubiquitously distributed worldwide in canine population causes severe fatal gastroenteritis. Antigenic typing of CPV-2 remains a prime focus of research groups worldwide in understanding the disease epidemiology and virus evolution. The present study was thus envisioned to provide a simple sequencing independent, rapid, robust, specific, user-friendly technique for detecting and typing of presently circulating CPV-2 antigenic variants. ARMS-PCR strategy was employed using specific primers for CPV-2a, CPV-2b and CPV-2c to differ

Three-year rabies duration of immunity in dogs following vaccination with a core combination vaccine against canine distemper virus, canine adenovirus type-1, canine parvovirus, and rabies virus.

Thirty-two seronegative pups were vaccinated at 8 weeks of age with modified-live canine distemper virus (CDV), canine adenovirus type-2 (CAV-2), and canine parvovirus (CPV) vaccine and at 12 weeks with a modified-live CDV, CAV-2, CPV, and killed rabies virus vaccine. An additional 31 seronegative pups served as age-matched, nonvaccinated controls. All test dogs were strictly isolated for 3 years after receiving the second vaccination and then were challenged with virulent rabies virus. Clinical signs of rabies were prevented in 28 (88%) of the 32 vaccinated dogs. In contrast, 97% (30 of 31) o

Influence of age and vaccination interval on canine parvovirus, distemper virus, and adenovirus serum antibody titers.

Canine core vaccine titer screenings are becoming increasingly popular in veterinary practice as a tool to guide vaccination decisions, despite a lack of supportive, peer-reviewed evidence-based literature. Additionally, it has been suggested that the canine core vaccine duration of host protective immunity can persist past the currently recommended vaccination interval. Thus, this study evaluated serum antibody titers against three core antigens in dogs with known vaccination histories and lifestyles, analyzing the effect of life stage, exposure risk, and time since last vaccination (TSLV). New canine parvovirus 2a infection in an imported Asian small-clawed otter (Aonyx cinereus) in Japan.

Post-import from the Republic of Indonesia to Japan in 2017, two juvenile, captive bred Asian small-clawed otters (Aonyx cinereus) exhibited gastrointestinal symptoms, including vomiting, diarrhea, and hematemesis, and died. One of them was examined postmortem. Microscopically, the small intestinal mucosa was necrotic with crypts lined by regenerating large epithelial cells. A gastric cardiac mucosal ulcerative lesion containing fungal yeasts and pseudohyphae morphologically indicated Candida spp. The lymph nodes exhibited marked lymphoid depletion. Canine parvovirus 2 (CPV-2) was isolated fro

Subacute massive necrotizing myocarditis by canine parvovirus type 2 infection with diffuse leukoencephalomalacia in a puppy.

Severe necrotizing myocarditis associated with canine parvovirus type 2 (CPV-2) infection and diffuse leukoencephalomalacia was recognized in a 4-week-old mongrel puppy. The cardiac lesions were characterized by severe diffuse myocardial degeneration and necrosis with occasional massive mineralization and distinct intranuclear inclusion bodies. Immunohistochemical examination revealed the presence of CPV-2 antigens in both the cytoplasm and nuclei of the myocytes. In the white matter of the cerebrum, moderate to severe diffuse necrosis with diffuse infiltration of gitter cells and occasional d

A modified live canine parvovirus strain with novel plaque characteristics. I. Viral attenuation and dog response.

A canine parvovirus (CPV) strain (C-780916) was found attenuated for pups at 80, but not after 51 serial passages in dog kidney cell (DKC) cultures. A variant viral population ('large plaque') emerged after prolonged cultivation in DKC cultures that may be associated with reduced native virulence. Dogs vaccinated with modified CPV developed high hemagglutination-inhibiting (HI) antibody titers within 4 days of incoluation and antibody persisted. Vaccinated animals shed small amounts of virus in the feces that spread to contact dogs. After five back-passages in dogs the modified strain was not

Increased survival in puppies affected by Canine Parvovirus type II using an immunomodulator as a therapeutic aid.

Canine parvovirus type II (CPV-2) infection induces canine parvoviral enteritis (CPE), which in turn promotes sepsis and systemic inflammatory response syndrome (SIRS). Mortality in this disease is usually registered within 48-72 h post-hospitalization, the critical period of the illness. It has been recently described that the use of an immunomodulator, whose major component is monomeric ubiquitin (mUb) without the last two glycine residues (Ub∆GG), in pediatric human patients with sepsis augments survival. It is known that CXCR4 is the cell receptor of extracellular ubiquitin in humans. This

Molecular Mechanisms Between Canine Parvovirus 2 and Host Interactions: From Viruses to Innate Immune Signaling Pathways.

Canine parvovirus 2 (CPV-2) is a major pathogen causing acute haemorrhagic enteritis and myocarditis in dogs. Since the discovery in the late 1970s, the original CPV-2 strain has evolved through VP2 mutations into new subtypes CPV-2a, 2b, and 2c. The viruses pose a persistent threat to canine health worldwide. This review summarizes the innate immunity evasion tactics employed by CPV-2, focusing on the interactions of various viral proteins and host signaling pathways. CPV-2 regulates the activation and inhibition of Toll-like receptors (TLRs), thereby influencing the downstream MyD88/TRIF sig

Engineering a recombinant VP2-Based neutralizing epitope vaccine candidate against canine parvovirus: a preliminary immunogenicity assessment.

BACKGROUND: Canine parvovirus (CPV) poses a severe threat to canine health, necessitating the development of safer and more effective vaccines. While traditional vaccines carry risks of virulence reversion and environmental contamination, subunit vaccines-especially neutralizing epitope vaccines-offer promising alternatives by eliciting targeted immune responses with enhanced safety. METHODS: We employed bacterial display technology to express 11 overlapping CPV VP2 gene fragments on the periplasmic membrane of E. coli. Key neutralizing antigenic epitopes of CPV VP2 were mapped using four neu

Canine Parvovirus is diagnosed and neutralized by chicken IgY-scFv generated against the virus capsid protein.

Canine parvovirus (CPV) can cause acute and highly contagious bloody enteritis in dog. To obtain antibodies against CPV, hens were immunized with virus-like particles (VLP) of CPV-VP2. The IgY single chain fragment variables (scFv) were generated by T7 phage display system and expressed in E. coli system. The titer of the primary scFv library reached to 1.5 × 106 pfu/mL, and 95% of the phages contained the target fragments. The CPV-VLP and CPV-VP2 protein showed similar reaction values to the purified scFv in the ELISA test, and the results of ELISA analysis using IgY-scFv toward CPV clinical

Administration with Vaccinia Virus Encoding Canine Parvovirus 2 vp2 Elicits Systemic Immune Responses in Mice and Dogs.

Canine parvovirus type 2 (CPV2) is a highly contagious cause of serious and often fatal disease in young dogs. Despite the widespread availability of attenuated vaccines, safer, more stable, and more effective CPV2 vaccine candidates are still under exploration. Vaccinia virus (VV) has already been proved to be a safe, stable, and effective vaccine vector. In this study, we generated a VV-based CPV2 vaccine candidate (VV-CPV-VP2) and then evaluated its immunogenicity in mice and dogs. The exogenous vp2 gene of CPV2, which replaced the major virulence gene hemagglutinin (ha) of VV, expressed ef
