Immunosuppressive ergosterol-emodin hybrids from the endophytic Penicillium sp. XB152.

Ergostemodins A-D (1-4), four unprecedented heterodimers of ergosterol and emodin units featuring a unique 6/6/6/5/6/6/5/6 octacyclic skeleton, along with an unreported ergosterol, penicillisterene A (5), and a known ergosterol, acrocalysterol C (6), were isolated from the endophytic fungus Penicillium sp. XB152. Their structures were established by extensive spectroscopic analysis and single-crystal X-ray diffraction. A plausible biosynthetic pathway involving an intermolecular Diels-Alder reaction was proposed for 1-4. Compounds 1-6 were immunosuppressive against concanavalin A (ConA)-induce

Anti-Inflammatory Secondary Metabolites from Penicillium sp. NX-S-6.

Five new natural products, including two sorbicillinoids (1-2), one indolinone alkaloid (10), one tetracyclic steroid (11), and one α-pyrone derivative (14), were identified from the endophytic Penicillium sp. NX-S-6, together with thirteen known natural products. The structures of new compounds were unambiguously elucidated by comprehensive spectroscopic analyses (NMR, MS), as well as electronic circular dichroism (ECD) calculation. Notably, quinosorbicillinol (1) was identified as a rare hybrid sorbicillinoid incorporating a quinolone moiety, representing a unique structural scaffold in this

Acute abdomen: an unusual presentation of disseminated Penicillium marneffei infection.

Varied clinical presentations of Penicillium marneffei, an opportunistic pathogen in HIV disease has been rarely described in literature. We report a patient with advanced AIDS who presented to us with prolonged fever and had features of an acute abdomen. On radiologic imaging he had features of intestinal obstruction and mesenteric lymphadenitis. A diagnosis was made possible by endoscopic biopsies of the small bowel and bone marrow culture which grew P. Marneffei. He was treated with intravenous amphotericin for 2 weeks followed by oral itraconazole. This case is reported for its rarity and

Analysis of polymorphic microsatellite markers for typing Penicillium marneffei isolates.

Penicillium marneffei is an emerging opportunistic dimorphic fungal pathogen that is endemic in Southeast Asia. A typing method based on the analysis of size polymorphisms in microsatellite loci was investigated. Three loci available from the GenBank database were identified to harbor microsatellites. PCR primers flanking the microsatellite repeats were designed with one primer in the set fluorescently labeled. PCR products were then sized by automated capillary electrophoresis. As expected for a haploid fungus, a single band was observed for each microsatellite locus for all isolates. Polymor

Invasive infection due to penicillium species other than P. marneffei.

Infection caused by Penicillium spp. due to species other than P. marneffei is rare. We present three such cases of invasive disease. The first had chronic granulomatous disorder (CGD) with pulmonary infection caused by Penicillium spp. and he responded to amphotericin B therapy. Cases two and three were not known to be immunocompromised and both failed to respond to therapy. Case two had cerebral disease from an unknown source caused by P. chrysogenum. Case three probably acquired infection caused by P. decumbens peri-operatively and presented with paravertebral infection. The pertinent liter

Penicillium marneffei infection in patients infected with human immunodeficiency virus.

From June 1990 to August 1991, 21 patients infected with the human immunodeficiency virus (HIV) presented with systemic mycosis caused by Penicillium marneffei. Between August 1987 and August 1991, only five patients were observed who had P. marneffei infection but not HIV infection. The clinical presentation included fever, cough, and generalized papular skin lesions. For 11 of these 21 patients, the presumptive diagnosis of P. marneffei infection could be made by microscopic examination of Wright's-stained bone marrow aspirate and/or touch smears of skin specimens obtained by biopsy several

Penicamins A-L, Polyoxygenated Diterpenes from Penicillium camemberti JSB-7212.

Penicamins A-L (1-12), 12 highly oxygenated novel diterpenes, were obtained from the fungus Penicillium camemberti JSB-7212. Compounds 1-12 share the same 7/6/5 tricyclic skeleton as valparane-type diterpenes but differ in the absolute configurations at C-7, C-11, and C-14, as well as in the oxidation levels at C-6 and C-8, which were determined through extensive spectroscopic data interpretation. Stereochemical assignments of compounds 1, 2, and 4-12 were established by single-crystal X-ray diffraction, and the absolute configuration of 3 was determined by analysis of the NOESY data and bioge

Genomic insights into Penicillium chrysogenum adaptation to subseafloor sedimentary environments.

BACKGROUND: Penicillium chrysogenum is a filamentous fungal species with diverse habitats, yet little is known about its genetics in adapting to extreme subseafloor sedimental environments. RESULTS: Here, we report the discovery of P. chrysogenum strain 28R-6-F01, isolated from deep coal-bearing sediments 2306 m beneath the seafloor. This strain possesses exceptional characteristics, including the ability to thrive in extreme conditions such as high temperature (45 °C), high pressure (35 Mpa), and anaerobic environments, and exhibits broad-spectrum antimicrobial activity, producing the antibi

Proteomics and Penicillium chrysogenum: Unveiling the secrets behind penicillin production.

Discovery, industrial production and clinical applications of penicillin, together with scientific findings on penicillin biosynthesis and its complex regulation, are model milestones of the historical evolution of the most recognized 'magic bullet' against microbial infections available in the worldwide market. Thousands of tons of penicillin produced nowadays are the result of a huge number of technical, industrial and scientific tackled and solved challenges. This combination of, sometimes unsuspected, findings has given Proteomics the chance to support the understanding of the physiology o

Structural variation among wild and industrial strains of Penicillium chrysogenum.

Strain selection and strain improvement are the first, and arguably most important, steps in the industrial production of biological compounds by microorganisms. While traditional methods of mutagenesis and selection have been effective in improving production of compounds at a commercial scale, the genetic changes underpinning the altered phenotypes have remained largely unclear. We utilized high-throughput Illumina short read sequencing of a wild Penicillium chrysogenum strain in order to make whole genome comparisons to a sequenced improved strain (WIS 54-1255). We developed an assembly-fre

Penicillium morneffei, an emerging AIDS-related pathogen--a RIMS study.

Twenty-seven HIV-infected patients with penicilliosis were included to study the clinical, laboratory and therapeutic features of HIV-infected patients with disseminated Penicillium marneffei infection. Diagnosis was based on microscopy and culture. CD4 counts were assessed by flow cytometry. Males (23 out of 27) predominated while the ages ranged from 20 to 54 years (mean 31.5 years). The most common manifestations were fever, weight loss, anaemia and skin lesions. CD4 counts were low in all the patients. The response to treatment with intravenous amphotericin B followed by itraconazole was e

Penicillium marneffei infection in patients infected with human immunodeficiency virus.

Opportunistic infections are major causes of morbidity in patients with human immunodeficiency virus (HIV) infection. Penicillium marneffei infection is very common in HIV-infected individuals in northern Thailand. The patients usually present with fever, anemia, weight loss, skin lesions, generalized lymphadenopathy, and hepatomegaly. The average number of CD4+ T-lymphocytes at presentation was 63.8 cells/mm3. Diagnosis depends on familiarity with the clinical syndromes and support from the mycology laboratory. The response to antifungal treatment of the patients is generally good, but depend

The influence of inoculation with Penicillium and yeast on taste enhancement in Jinhua ham.

This study investigated the effect of Penicillium (Penicillium aethiopicum & Penicillium chrysogenum, PP), yeast (Candida parapsilosis & Rhodotorula mucilaginosa, CM) and mixture (MI) inoculation on the taste development of Jinhua ham. The results showed that Penicillium and yeast significantly increased acidic protease activity and reduced TVB-N values compared to the control. Three kinds of inoculation promoted the degradation of both myofibrillar and sarcoplasmic proteins, leading to a accumulation of peptides and free amino acids. It was strongly detected by the electronic tongue sensors P

New colours for old in the blue-cheese fungus Penicillium roqueforti.

Penicillium roqueforti is used worldwide in the production of blue-veined cheese. The blue-green colour derives from pigmented spores formed by fungal growth. Using a combination of bioinformatics, targeted gene deletions, and heterologous gene expression we discovered that pigment formation was due to a DHN-melanin biosynthesis pathway. Systematic deletion of pathway genes altered the arising spore colour, yielding white to yellow-green to red-pink-brown phenotypes, demonstrating the potential to generate new coloured strains. There was no consistent impact on mycophenolic acid production as
