The potential of lumbrokinase and serratiopeptidase for the degradation of Aβ 1-42 peptide - an in vitro and in silico approach.

BACKGROUND: Alzheimer's disease (AD) is diagnosed with the deposition of insoluble β-amyloid (Aβ) peptides in the neuropil of the brain leading to dementia. The extracellular deposition of the fibrillar Aβ peptide on the neurons is known as senile plaques. Therefore, Aβ degradation and clearance from the human body is a promising therapeutic approach in the medication of AD. METHODS: In the current study, the enzyme lumbrokinase (LK) was extracted and purified from earthworm and its activity was utilized toward Aβ 1-42 amyloids degradation in vitro alongside with an additional enzyme serratio

Inhalable Artificial Polymeric Nucleases Degrading Neutrophil Extracellular Trap-DNAs and Alleviating Pulmonary Fibrosis.

Pulmonary fibrosis resulting from recurrent lung inflammation due to pathogen infection may lead to serious problems and death. Neutrophil extracellular traps (NETs), consisting of DNAs and proteins released by neutrophils in response to infection, are major pathogenesis factors for pathogen-associated pulmonary fibrosis. By mimicking nucleic acid hydrolase, polymeric artificial DNases bearing imidazole units (PEG-PIm) are developed to degrade the DNAs and thus deconstruct NETs, inhibiting pulmonary fibrosis. By tailoring the PIm segments with varied imidazole units, the polymer hydrolase with

Alpha-N-Benzoylarginine-2-naphthylamide hydrolase (cathepsin BI?) from rat skin. III. Substrate specificity, modifier characteristics, and transformation of the enzyme at acidic pH.

Some properties of rat skin benzoylarginine-2-naphthylamide hydrolase types I (preparations I and AI) and II (preparations II and NII) were studied. Both types were activated by dithiothreitol and EDTA, but responded differently to 1 mM KCN, when benzoylarginine-2-naphthylamide (BANA) was used as a substrate: type I was inhibited, while type II was activated. When leucine-2-naphthylamide was used as a substrate, both types were activated by KCN. Thiol proteinase inhibiting substances, like heavy metals, iodoacetic acid, 4-chloromercuribenzoic acid, and tosyllysine chloromethylketone, inhibited

Intestinal perfusion studies in tropical sprue. 1. Amino acid and dipeptide absorption.

Intestinal absorption of glycine 20 mmol/1, glycyl-glycine 10 mmol/1 plus L-leucine 10 mmol/1, and glycyl-L-leucine 10 mmol/1 has been studied by intestinal perfusion in 11 patients with tropical sprue and 10 control subjects. The patients with sprue had a significant reduction in the rate of absorption of glycine from a 20 mmol/1 solution, but there were no significant differences in the absorption of the other substances. The failure to demonstrate any difference in the absorption of these substances is probably related to their low concentration relative to the maximum absorptive capacity o

Peptidyl tRNA Hydrolase Is Required for Robust Prolyl-tRNA Turnover in Mycobacterium tuberculosis.

Enzymes involved in rescuing stalled ribosomes and recycling translation machinery are ubiquitous in bacteria and required for growth. Peptidyl tRNA drop-off is a type of abortive translation that results in the release of a truncated peptide that is still bound to tRNA (peptidyl tRNA) into the cytoplasm. Peptidyl tRNA hydrolase (Pth) recycles the released tRNA by cleaving off the unfinished peptide and is essential in most bacteria. We developed a sequencing-based strategy called copper sulfate-based tRNA sequencing (Cu-tRNAseq) to study the physiological role of Pth in Mycobacterium tubercul

Diphyllobothrium dendriticum (Cestoda: Diphyllobothriidae) in the Intestinal Tract of the Herring Gull Larus argentatus: Localization and Trophic Parameters.

We studied the activities both of digestive enzymes in the small intestine of the herring gull (Larus argentatus) and a tapeworm Diphyllobothrium dendriticum (Cestoda: Diphyllobothriidae) residing in the intestine. It was found that D. dendriticum infects the medial section of the small intestine of the herring gull. Such localization of D. dendriticum is caused by the maximal activity of proteases and glycosidases and by the high rate of membrane and cavitary digestion in this section. The activity of protease and glycosidase in gulls infected with D. dendriticum is decreased. The activity of

Intestinal dipeptidases and the dipeptide transport in the monkey and in man.

Among the powerful dipeptidases of the cytosol fraction the well-characterized 'master' dipeptidase of broad substrate specificity is an example of a 'true' dipeptidase. There are only a limited number of peptidases which together can hydrolyse the total range of the theoretically possible dipeptides. In the lumen, the dipeptides of dietary origin apparently enter the cell through a single transport system of broad specificity in the monkey and possibly also in man. Perfusion studies indicated impared absorption of glycine and Gly-Gly in Indian compared to English subjects, but the kinetic adv

Urinary peptidomics reveals proteases involved in idiopathic membranous nephropathy.

BACKGROUND: Idiopathic membranous nephropathy (IMN) is a cause of nephrotic syndrome that is increasing in incidence but has unclear pathogenesis. Urinary peptidomics is a promising technology for elucidating molecular mechanisms underlying diseases. Dysregulation of the proteolytic system is implicated in various diseases. Here, we aimed to conduct urinary peptidomics to identify IMN-related proteases. RESULTS: Peptide fingerprints indicated differences in naturally produced urinary peptide components among 20 healthy individuals, 22 patients with IMN, and 15 patients with other kidney disea

Proteolytic Activity and Substrate Specificity of Lake Geneva.

Lakewater microorganisms secrete proteases which contribute to the turnover of dissolved organic matter and the degradation of peptidic contaminants. However, little is known about the identities and substrate specificities of these proteases. Herein, we sought to characterize the global proteolytic fingerprint of the extracellular proteases present in Lake Geneva, the largest freshwater body in Central Europe. Using Multiplex Substrate Profiling by Mass Spectrometry (MSP-MS), we identified preferred enzymatic cleavage next to positively charged and certain nonpolar amino acids, while cleavage

Amelioration of Bleomycin and Methotrexate-Induced Pulmonary Toxicity by Serratiopeptidase and Fisetin.

Pulmonary toxicity by anticancer drugs often leads to discontinuation of therapy or switching the therapy to alternative drugs. In the present study, serratiopeptidase (SPTD) and fisetin (FST) were evaluated as chemoprotectant to counteract the pulmonary toxicity induced by BLM and MTX. Single dose of MTX (20 mg/kg) by intraperitoneal and BLM (5 mg/kg) by intra-tracheal route was administered on 7th day of study. SPTD (20 mg/kg), FST (25 mg/kg), and NAC (250 mg/kg) and combinations of SPTD + NAC, SPTD + FST, and FST + NAC were administered through oral gavage for 14 days. SPTD and FST showed s

Tracking the fate of pasta (T. Durum semolina) immunogenic proteins by in vitro simulated digestion.

The aim of the present study was to identify and characterize the celiacogenic/immunogenic proteins and peptides released during digestion of pasta (Triticum durum semolina). Cooked pasta was digested using a harmonized in vitro static model of oral-gastro-duodenal digestion. The course of pasta protein digestion was monitored by SDS-PAGE, and gluten proteins were specifically analyzed by Western blot using sera of celiac patients. Among the allergens, nonspecific lipid-transfer protein was highly resistant to gastro-duodenal hydrolysis, while other digestion-stable allergens such as α-amylase
