A 1,1,1-trichloroethane-degrading anaerobic mixed microbial culture enhances biotransformation of mixtures of chlorinated ethenes and ethanes.

1,1,1-trichloroethane (1,1,1-TCA) is a common groundwater pollutant as a result of improper disposal and accidental spills. It is often found as a cocontaminant with trichloroethene (TCE) and inhibits some TCE-degrading microorganisms. 1,1,1-TCA removal is therefore required for effective bioremediation of sites contaminated with mixed chlorinated organics. This study characterized MS, a 1,1,1-TCA-degrading, anaerobic, mixed microbial culture derived from a 1,1,1-TCA-contaminated site in the northeastern United States. MS reductively dechlorinated 1,1,1-TCA to 1,1-dichloroethane (1,1-DCA) and

Anaerobic osteomyelitis of long bones.

Nine cases of anaerobic osteomyelitis of long bones are reviewed, and data are tabulated for an additional 52 cases from the literature. Three major clinical presentations of this disease entity include an acute hematogenous form usually involving previously normal bones in younger patients; a chronic infection in which the anaerobic bacteria superinfect a fracture site already compromised by osteomyelitis due to aerobic organisms, usually Staphylococcus aureus; and an indolent infection at the site of indwelling prosthetic devices. This last form appears to be initiated at the time of surgery

The effect of maternal sodium ρ-perfluorous nonenoxybenzene sulfonate exposure on the gut microbiota in dams and offspring.

Sodium ρ-perfluorous nonenoxybenzene sulfonate (OBS) is increasingly used as an effective perfluoroalkyl/polyfluoroalkyl substances (PFASs) alternative across multiple industries. This study involves exposing pregnant C57BL/6 mice to OBS at concentrations of 0, 0.5, and 5.0 mg/L via drinking water during gestation and lactation. The investigation focused on analyzing gut microbiota in both dams and offspring after maternal OBS exposure. Results highlighted notable changes in the gut microbiota composition within the colonic content of both dams and offspring, the Bacteroidetes, Firmicutes, α-P

Mendelian randomization analyses of known and suspected risk factors and biomarkers for myasthenia gravis overall and by subtypes.

BACKGROUND: Myasthenia gravis (MG) is an autoimmune disease that affects neuromuscular junction. The literature suggests the involvement of circulating cytokines (CK), gut microbiota (GM), and serum metabolites (SM) with MG. However, this research is limited to observational trials, and comprehensive causal relationship studies have not been conducted. Based on published datasets, this investigation employed Mendelian Randomization (MR) to analyze the known and suspected risk factors and biomarkers causal association of MG and its subtypes. METHODS: This research used two-sample MR and linkag

Effects of Diets with Different Energy Levels at Cold Temperatures on Gut Microbiota and Metabolic State in Growing-Finishing Pigs.

In cold-temperature regions, particularly on family farms, threats to livestock health constrain the potential of livestock husbandry. This study aimed to explore the effects of different dietary energy levels, adjusted by oil addition, on gut microbiota and metabolic homeostasis at cold temperatures. Twenty-four healthy pigs were randomly divided into two groups and fed a basal diet (BD) or a basal diet supplemented with oil (OD, with net energy increased by 100 kcal/kg) for 103 days. The cold temperature and humidity were maintained at 14 ± 2 °C and 65 ± 10%, respectively. On day 103 of the

Engineered in situ biogeochemical transformation as a secondary treatment following ISCO - A field test.

ISCO using activated sodium persulphate is a widely used technology for treating chlorinated solvent source zones. In sensitive areas, however, high groundwater sulphate concentrations following treatment may be a drawback. In situ biogeochemical transformation, a technology that degrades contaminants via reduced iron minerals formed by microbial activity, offers a potential solution for such sites, the bioreduction of sulphate and production of iron sulphides that abiotically degrade chlorinated ethenes acting as a secondary technology following ISCO. This study assesses this approach in the

Design and Application of a Low pH Upflow Biofilm Sulfidogenic Bioreactor for Recovering Transition Metals From Synthetic Waste Water at a Brazilian Copper Mine.

A sulfidogenic bioreactor, operated at low pH (4-5), was set up and used to remove transition metals (copper, nickel, cobalt, and zinc) from a synthetic mine water, based on the chemistry of a moderately acidic (pH 5) drainage stream at an operating copper mine in Brazil. The module was constructed as an upflow biofilm reactor, with microorganisms immobilized on porous glass beads, and was operated continuously for 462 days, during which time the 2 L bioreactor processed >2,000 L of synthetic mine water. The initial treatment involved removing copper (the most abundant metal present) off-line

Effects of fermented milk containing Lacticaseibacillus paracasei strain Shirota on constipation and gut microbiota: A randomised pilot study in Vietnam.

Fermented milk containing Lacticaseibacillus paracasei strain Shirota (LcS) has been shown to improve stool consistency in various countries; however, its effect on hard or lumpy stools (HLS) in the Vietnamese population remains unclear. We investigated the effects of LcS-fermented milk on constipated Vietnamese adults with a high prevalence of HLS. In a single-centre, open-label trial, 51 participants with HLS were randomised to receive one bottle per day of fermented milk containing ≥6.5 × 109 cfu of LcS for 4 weeks (probiotic group) or no intervention (control group), followed by a 2-week w

Identification of Dehalobacter reductive dehalogenases that catalyse dechlorination of chloroform, 1,1,1-trichloroethane and 1,1-dichloroethane.

Two novel reductive dehalogenases (RDases) that are highly similar to each other but catalyse distinct dechlorination reactions were identified from Dehalobacter-containing mixed cultures. These two RDases were partially purified from crude protein extracts of anaerobic dechlorinating enrichment cultures using blue native polyacrylamide gel electrophoresis. Gel slices were assayed for dechlorinating activity, and associated proteins were identified using liquid chromatography tandem mass spectrometry with the metagenome of the parent culture as the reference database. The two RDases identified

The Gut Microbiome in Autosomal Dominant Polycystic Kidney Disease: A Cross-Sectional Study.

KEY POINTS: Patients with autosomal polycystic kidney disease (ADPKD) display relevant alterations in gut microbiome signatures compared with a healthy control cohort. Gut microbiome alterations in patients with ADPKD were associated with specific markers of ADPKD disease progression. BACKGROUND: Changes in gut microbiota signatures have been associated with CKD and nephrolithiasis and may thus be a factor explaining variability of outcome in autosomal polycystic kidney disease (ADPKD). We aimed to characterize the intestinal microbiome in a cross-sectional study of patients with ADPKD and to

Refining the relationship between gut microbiota and common hematologic malignancies: insights from a bidirectional Mendelian randomization study.

BackgroundThe relationship between gut microbiota and hematologic malignancies has attracted considerable attention. As research progresses, it has become increasingly clear that the composition of gut microbiota may influence the onset and progression of hematologic malignancies. However, our understanding of this association remains limited.MethodsIn our study, we classified gut microbiota into five groups based on information at the phylum, class, order, family, and genus levels. Subsequently, we obtained data related to common hematologic malignancies from the IEU Open GWAS project. We the

Comprehensive assessment of HF-rTMS treatment mechanism for post-stroke dysphagia in rats by integration of fecal metabolomics and 16S rRNA sequencing.

BACKGROUND: The mechanism by which high-frequency repetitive transcranial magnetic stimulation (HF-rTMS) improves swallowing function by regulating intestinal flora remains unexplored. We aimed to evaluate this using fecal metabolomics and 16S rRNA sequencing. METHODS: A Post-stroke dysphagia (PSD) rat model was established by middle cerebral artery occlusion. The magnetic stimulation group received HF-rTMS from the 7th day post-operation up to 14th day post-surgery. Swallowing function was assessed using a videofluoroscopic swallowing study (VFSS).
