Octopus 'hypnosis': inducing tonic immobility for studying local sensorimotor responses and arm-sucker coordination.

Effective methods of anesthesia for octopuses are essential to physiological studies as well as for their welfare in scientific research. However, commonly used forms of general anesthesia using ethanol, magnesium chloride, and similar agents have certain drawbacks. While these methods effectively induce still states in the octopus, they also affect the peripheral body and nervous system and are therefore less than optimal for studying local behavior in octopus arms and suckers. Further, stupefying effects outlast the anesthetized state. We explore an old, rarely used method of octopus 'hypnos

Systemic anti-cancer therapy associated with the occurrence of peripheral neurotoxicity and, specifically, peripheral neuropathy.

The investigation of peripheral neurotoxicity associated with systemic anticancer therapy (SACT) agents is often confined to a small range of chemotherapy agents. This study aimed to identify all SACT agents associated with peripheral neurotoxicity and, specifically, peripheral neuropathy, and to provide incidence estimates for the development of each type of neurotoxicity associated with each agent. Antineoplastic agents approved globally for clinical and/or research purposes were identified through triangulation of nine national and global drug product databases. The class of each agent was

MicroRNAs in chemotherapy-induced peripheral neuropathy: mechanisms and therapies.

Chemotherapy-induced peripheral neuropathy (CIPN) is a significant adverse effect of cancer therapies that profoundly disrupts the quality of life for patients. CIPN is characterized by sensory symptoms such as pain, tingling, and numbness, typically distributed in a "glove and stocking" pattern. Its underlying mechanisms remain incompletely understood, involving complex processes such as heightened neuronal excitability, alterations in ion channel function, neuroinflammation, and glial cell activation. MicroRNAs (miRNAs), small non-coding RNA molecules, play a pivotal role in regulating these

Recent Advances in Diagnosis, Management, Treatment, and Prevention of Neuropathies in Cancer Patients.

PURPOSE OF REVIEW: The purpose of this article is to provide an update on cancer-related neuropathies over the past five years, by reviewing the advances in pathophysiology and biology, diagnostic approaches, and management strategies. RECENT FINDINGS: New agents causing peripheral neuropathy include antibody-drug conjugates, combinations of immune-checkpoint inhibitor therapies, and targeted therapies. Development of axonal neuropathies has been found to be mediated through the protein sterile-α and Toll/interleukin 1 receptor motif containing protein 1 (SARM1). There have been emerging imag

Vasculitic neuropathy (VN) may affect the peripheral nervous system alone (non-systemic vasculitic neuropathy (NSVN)) or be part of a systemic vasculitis. Studies indicate that NSVN is ascommon as other inflammatory neuropathies but is underdiagnosed, probably becausethe clinical phenotype is very heterogenous and vary from sub-acute painful mononeuritis multiplex to progressive, symmetric polyneuropathy. Since the irreversible nerve damage can be reduced with immunosuppressants, early recognition of VN is important. More studies are needed to validate treatment and outcome measures.

Updates in the Treatment of Chemotherapy-Induced Peripheral Neuropathy.

Chemotherapy-induced peripheral neuropathy (CIPN) is a common toxicity associated with treatment with platinum-based agents, taxanes, vinca alkaloids, and other specific agents. The long-term consequences of this condition can result in decreased patient quality of life and can lead to reduced dose intensity, which can negatively impact disease outcomes. There are currently no evidence-based preventative strategies for CIPN and only limited options for treatment. However, there are several strategies that can be utilized to improve patient experience and outcomes as more data are gathered in t

Cryotherapy for the prevention of chemotherapy-induced peripheral neuropathy: A systematic review.

OBJECTIVE: Chemotherapy induced peripheral neuropathy (CIPN) is an adverse effect of certain chemotherapy agents that can result in dose reductions, permanent nerve damage, and chronic pain. Although pharmacological agents have been studied in this setting, there is no standard of care for the prevention of CIPN. Thus, the objective of this systematic review is to assess the efficacy and safety of cryotherapy for the prevention of CIPN. DATA SOURCES: PubMed (1946 to February 2020) and Embase (1947 to February 2020) were utilized to conduct a literature search using the following search terms:

Neurophysiological, nerve imaging and other techniques to assess chemotherapy-induced peripheral neurotoxicity in the clinical and research settings.

Chemotherapy-induced peripheral neurotoxicity (CIPN) is a common dose-limiting side effect of several anticancer medications. CIPN may involve multiple areas of the peripheral nervous system from the autonomic and dorsal root ganglia (DRG) to the axon and any peripheral nerve fibre type. Large diameter sensory myelinated (Aβ) fibres are more frequently involved, but motor, small myelinated (Aδ), unmyelinated (C) or autonomic fibres may also be affected. Here, we review the current evidence on techniques for the CIPN assessment in the clinical and experimental settings. Nerve conduction studies

Immunosuppressive treatment for non-systemic vasculitic neuropathy.

BACKGROUND: Non-systemic vasculitic neuropathy is a rare disabling disease that usually has a subacute onset of progressive or relapsing-remitting sensory or sensorimotor deficits. Asymmetry, pain and weakness are key features. The diagnosis can only be made by exclusion of other causes, the absence of systemic vasculitis or other rheumatic diseases, and the demonstration of vasculitis in a nerve or a combined nerve and muscle biopsy. There is a need for efficacious therapy to prevent disease progression and to improve prognosis. OBJECTIVES: To assess if immunosuppressive treatment in non-sys

Selected peripheral neuropathies associated with human immunodeficiency virus infection and antiretroviral therapy.

A variety of peripheral neuropathies are associated with human immunodeficiency virus (HIV) infection. Although the incidence of certain forms of neuropathy is increased in HIV infection, in other cases, the association may be fortuitous. Different forms of peripheral neuropathy occur with increased frequency at particular stages of HIV disease. For example, inflammatory demyelinating neuropathy (IDP) is often the first manifestation of HIV disease, when CD4 lymphocyte counts are relatively high. As immunosuppression progresses and HIV viral load becomes uncontrolled, the incidence of distal s

[Polyradiculoneuritis and Campylobacter jejuni: clinical and physiopathological aspects].

Several explanations have been proposed to explain the relationship between axonal forms of acute auto-immune inflammatory polyradiculoneuritis and Campylobacter jejuni. The major hypothesis involving molecular imitation is based on the existence of common antigenic determinants (epitopes) in the lipopolysaccharides of the infectious agent and gangliosides, i.e. glycosphingolipides on the surface of the nervous system cells, especially peripheral nervous system cells. The purpose of this literature review is to improve understanding of the rather complex physiopathological mechanisms underlyin

Current understanding of the molecular mechanisms of chemotherapy-induced peripheral neuropathy.

Chemotherapy-induced peripheral neuropathy (CIPN) is the most common off-target adverse effects caused by various chemotherapeutic agents, such as cisplatin, oxaliplatin, paclitaxel, vincristine and bortezomib. CIPN is characterized by a substantial loss of primary afferent sensory axonal fibers leading to sensory disturbances in patients. An estimated of 19-85% of patients developed CIPN during the course of chemotherapy. The lack of preventive measures and limited treatment options often require a dose reduction or even early termination of life-saving chemotherapy, impacting treatment effic

Botulinum Toxin and Neuronal Regeneration after Traumatic Injury of Central and Peripheral Nervous System.

Botulinum neurotoxins (BoNTs) are toxins produced by the bacteria Clostridiumbotulinum, the causing agent for botulism, in different serotypes, seven of which (A-G) are well characterized, while others, such as H or FA, are still debated. BoNTs exert their action by blocking SNARE (soluble N-ethylmale-imide-sensitive factor-attachment protein receptors) complex formation and vesicle release from the neuronal terminal through the specific cleavage of SNARE proteins. The action of BoNTs at the neuromuscular junction has been extensively investigated and knowledge gained in this field has set the

Avapritinib for targeting KIT D816V-mutated acute myeloid leukemia relapsed after allogeneic hematopoietic stem cell transplant with extramedullary and central nervous system involvement.

Relapse of acute myeloid leukemia with CBFB::MYH11 fusion and KIT D816V mutation after allogeneic hematopoietic stem cell transplantation is uncommon and often associated with aggressive extramedullary spread and poor outcomes, particularly when the central nervous system is involved. We report a case of a 54‑year‑old man who developed extensive paravertebral, central nervous system, and multiorgan extramedullary relapse despite donor lymphocyte infusions and high‑dose chemotherapy. Avapritinib, a selective inhibitor of mutated KIT, was initiated as single‑agent therapy and resulted in rapid n

Disorders of spinal cord, nerve, and muscle.

Substance abuse may lead to acute or chronic injuries to the spinal cord or PNS in addition to injuries caused by trauma and infection. The most widespread of these conditions are the chronic myopathy and chronic peripheral neuropathy of alcoholism. Less common, but more striking, illnesses attributed to substance abuse include ATM, a subacute myeloradiculoneuropathy resembling combined systems disease, Guillain-Barré syndrome, plexitis, fulminant axonal neuropathy, and acute myoglobinuria with or without severe weakness. If the cause of such syndromes is not otherwise evident, careful probing

Clinical illness in a wild turkey with Laminosioptes cysticola infestation of the viscera and peripheral nerves.

Laminosioptes cysticola, the fowl cyst mite, was found in peripheral nerves and thoracic and abdominal viscera of an emaciated eastern wild turkey (Meleagris gallopavo) exhibiting severe torticollis, circling, loss of balance, and wing droop. Mites, sometimes accompanied by granulomatous inflammation, were abundant in brachial plexus and sciatic nerves. Mild lymphoplasmacytic perivascular cuffing was present in the cerebellum, but no direct evidence of mites or other infectious agents was found in the central nervous system. This is the first report of L. cysticola infestation in a wild turkey

Correlation between CNS Tuberculosis and the COVID-19 Pandemic: The Neurological and Therapeutic Insights.

The recent outbreak of Severe Acute Respiratory Syndrome-Coronavirus-2 (SARS-CoV-2) from Wuhan, China, was caused by a single-stranded RNA virus which has kept the entire world stranded. The outbreak was first diagnosed with respiratory illness, but recent findings of acute necrotizing hemorrhage of brain, brain encephalopathy, and the presence of the virus in the cerebrospinal fluid (CSF) have unveiled its neuroinvasivness. Various clinical features related to the central nervous system (CNS) and peripheral nervous system (PNS) due to COVID-19 infection are now identified. We demonstrate here

Toxic peripheral neuropathy associated with commonly used chemotherapeutic agents.

Peripheral neuropathy ranks among the most common non-haematological adverse effects of a number of effective chemotherapeutic agents, including platinum compounds, taxanes and vinca alkaloids. Newer agents, such as bortezomib, thalidomide and lenalidomide, frequently exert similar neurotoxic effects on peripheral nerves. Chemotherapy-induced peripheral neuropathy (CIPN) may result from a variety of mechanisms and may be related to causal factors, such as single dose per course, cumulative dose and risk factors including treatment schedule, prior or concomitant administration of other neurotox
