Ancient environmental microbiomes and the cryosphere.

In this review, we delineate the unique set of characteristics associated with cryosphere environments (namely, ice and permafrost) which present both challenges and opportunities for studying ancient environmental microbiomes (AEMs). In a field currently reliant on several assumptions, we discuss the theoretical and empirical feasibility of recovering microbial nucleic acids (NAs) from ice and permafrost with varying degrees of antiquity. We also summarize contamination control best practices and highlight considerations for the latest approaches, including shotgun metagenomics, and downstrea

A depositional taphonomy model for sedaDNA based on biotic profiles of contrasting sedimentation regimes.

Sedimentary ancient DNA (sedaDNA) has become an important tool in Quaternary science, but still little is understood of its taphonomy and whether sedaDNA represents the local vicinity or originates from distant sources. Here, we show key insights can be made about the physical origins of sedaDNA by integrating sedimentological and sedaDNA data into a sediment influx depositional model. We reconstruct contrasting taphonomic regimes of lacustrine, alluvial, terrestrial, and permafrost systems. Lacustrine systems show signals of regional catchment sources of sedaDNA, while alluvial, terrestrial,

The role of prokaryotic mercury methylators and demethylators in Canadian Arctic thermokarst lakes.

Permafrost soils are critical reservoirs for mercury (Hg), with the thawing process leading to the release of this element into the environment, posing significant environmental risks. Of particular concern is the methylated form of mercury, monomethylmercury (MMHg), known for its adverse effects on Human health. Microbial communities play a pivotal role in the formation of MMHg by facilitating Hg methylation and in the demethylation of MMHg, slowing the crossing of toxic threshold concentration in the environment. However, the specific microbes involved still need to be understood. This study

Solid phase thermal conductivity model of alpine meadow soils based on fractal theory.

The determination of thermophysical parameters is crucial in the hydro-thermal coupling calculations of permafrost. Alpine meadows can affect the heat exchange inside and outside the soil due to the well-developed root, which can have an impact on the stability of permafrost. At present, the research on the thermal conductivity of alpine meadows is less, and most of the research focuses on the thermal conductivity of rock and soil. In this paper, according to the distribution and development characteristics of roots in the soil, a theoretical model of thermal conductivity of root-soil in alpin

Responses of Soil Carbon Release to Freeze-Thaw Cycles Mediated by Carbon Availability at Regional and Global Scales.

Soil freeze-thaw cycles (FTCs) are widespread across mid- to high-latitude and high-altitude regions, with particularly high frequency in permafrost areas where they can enhance soil carbon release and potentially accelerate climate warming. However, the response of soil carbon release to FTCs and its determinants at the regional scale remain unclear due to the lack of direct experimental evidence. Here, based on topsoil samples along a 1000-km transect across the Tibetan permafrost region, we conducted a freeze-thaw microcosm experiment (5 cycles of 1-day freezing and 3-day thawing) to invest

Bioindicator "fingerprints" of methane-emitting thermokarst features in Alaskan soils.

Permafrost thaw increases the bioavailability of ancient organic matter, facilitating microbial metabolism of volatile organic compounds (VOCs), carbon dioxide, and methane (CH4). The formation of thermokarst (thaw) lakes in icy, organic-rich Yedoma permafrost leads to high CH4 emissions, and subsurface microbes that have the potential to be biogeochemical drivers of organic carbon turnover in these systems. However, to better characterize and quantify rates of permafrost changes, methods that further clarify the relationship between subsurface biogeochemical processes and microbial dynamics a

Ultraviolet radiation rather than inorganic nitrogen increases dissolved organic carbon biodegradability in a typical thermo-erosion gully on the Tibetan Plateau.

Permafrost thaw could lead to frozen carbon (C) being laterally transferred to aquatic systems as dissolved organic carbon (DOC). If this part of DOC has high biodegradability, it could be decomposed during the delivery process, release greenhouse gases to the atmosphere and trigger positive C-climate feedback. Thermokarst is an abrupt permafrost thaw process that can enhance DOC export and also impact DOC processing through increased inorganic nitrogen (N) and ultraviolet (UV) light exposure. Especially on the Tibetan Plateau, where thermokarst develops widely and suffers from serious UV radi

Inhibition of Methylmercury and Methane Formation by Nitrous Oxide in Arctic Tundra Soil Microcosms.

Climate warming causes permafrost thaw predicted to increase toxic methylmercury (MeHg) and greenhouse gas [i.e., methane (CH4), carbon dioxide (CO2), and nitrous oxide (N2O)] formation. A microcosm incubation study with Arctic tundra soil over 145 days demonstrates that N2O at 0.1 and 1 mM markedly inhibited microbial MeHg formation, methanogenesis, and sulfate reduction, while it slightly promoted CO2 production. Microbial community analyses indicate that N2O decreased the relative abundances of methanogenic archaea and microbial clades implicated in sulfate reduction and MeHg formation. Fol

Climatic Factors Influencing the Anthrax Outbreak of 2016 in Siberia, Russia.

In 2016, an outbreak of anthrax killing thousands of reindeer and affecting dozens of humans occurred on the Yamal peninsula, Northwest Siberia, after 70 years of epidemiological situation without outbreaks. The trigger of the outbreak has been ascribed to the activation of spores due to permafrost thaw that was accelerated during the summer heat wave. The focus of our study is on the dynamics of local environmental factors in connection with the observed anthrax revival. We show that permafrost was thawing rapidly for already 6 years before the outbreak. During 2011-2016, relatively warm year

Resolving circumarctic zero-curtain phenomena with AI-integrated earth observations.

Across the circumarctic, permafrost landscapes store approximately 1700 billion metric tons of organic carbon-nearly twice atmospheric levels-yet reservoir stability depends on the zero-curtain, a subsurface thermal plateau that emerges when latent heat maintains soil temperatures near 0 °C during freeze-thaw phase transitions. The zero-curtain sustains liquid water through cryosuction within the active layer, enabling microbial activity to persist well into the cold season and regulating permafrost thermal stability. However, zero-curtain intensity, duration, and spatial extent remain inadequ

Drivers of Mercury Cycling in the Rapidly Changing Glacierized Watershed of the High Arctic's Largest Lake by Volume (Lake Hazen, Nunavut, Canada).

Across the Arctic, glaciers are melting and permafrost is thawing at unprecedented rates, releasing not only water to downstream aquatic systems, but also contaminants like mercury, archived in ice over centuries. Using concentrations from samples collected over 4 years and calibrated modeled hydrology, we calculated methylmercury (MeHg) and total mercury (THg) mass balances for Lake Hazen, the world's largest High Arctic lake by volume, for 2015 and 2016. Glacial rivers were the most important source of MeHg and THg to Lake Hazen, accounting for up to 53% and 94% of the inputs, respectively.

Unprecedented Increases in Total and Methyl Mercury Concentrations Downstream of Retrogressive Thaw Slumps in the Western Canadian Arctic.

Retrogressive thaw slumps (RTSs) are thermokarst features created by the rapid thaw of ice-rich permafrost, and can mobilize vast quantities of sediments and solutes downstream. However, the effect of slumping on downstream concentrations and yields of total mercury (THg) and methylmercury (MeHg) is unknown. Fluvial concentrations of THg and MeHg downstream of RTSs on the Peel Plateau (Northwest Territories, Canada) were up to 2 orders of magnitude higher than upstream, reaching concentrations of 1,270 ng L-1 and 7 ng L-1, respectively, the highest ever measured in uncontaminated sites in Cana

Effect of oil pollution on the ecological condition of soils and bottom sediments of the arctic region (Yakutia).

Oil and petroleum products are known to be among the most widespread soil pollutants. The risk of emergencies is sure to increase greatly in conditions of abnormally low temperatures. Oil and oil products are not only toxic to the environment, but can also have a negative impact on the state of the permafrost zone, accelerating the processes of permafrost degradation. The goal of the research was to study the soils and bottom sediments for oil pollution in the Arctic region of Yakutia. The research was carried out with using the complex of geochemical and microbiological methods of analysis. I

Metagenomic analysis reveals the diversity and distribution of antibiotic resistance genes in thermokarst lakes of the Yellow River Source Area.

Thermokarst lakes form as the results of ice-rich permafrost thawing and act as important water resources in cold regions. However, the distributions of antibiotic resistance genes (ARGs) in thermokarst lakes are far less studied. Using metagenomic sequencing approach, we provided the first study to document ARGs in thermokarst lakes of the Yellow River Source Area on the Qinghai-Tibet Plateau (QTP). The results revealed that both sediment and water of the thermokarst lakes harbor diverse ARGs. Multidrug resistance genes were the most diverse, while rifamycin resistance genes were the most abu

The dissemination of antibiotic resistance in various environmental objects (Russia).

Environmental objects (surface and groundwater, soil, bottom sediments, wastewater) are reservoirs in which large-scale multidirectional exchange of determinants of antibiotic resistance between clinical strains and natural bacteria takes place. The review discusses the results of studies on antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARG) isolated from environmental objects (water, soil, sewage, permafrost) of the Russian Federation. Despite the relevance of the topic, the number of available publications examining the resistomes of Russian water bodies and soils is s

Microbial Reduction of Methane Emissions from High-Altitude Thermokarst Lakes.

Thermokarst lakes, a typical landscape resulting from abrupt permafrost thaw, are expected to be a substantial CH4 source. Climate change perturbs CH4 dynamics in these systems, particularly through increasingly frequent wet-dry cycles in small thermokarst lakes. However, how wet-dry alternation alters microbial communities remains poorly understood, and quantifying the effects of microbial shifts on CH4 emissions from these lakes represents a key challenge. Here, by integrating field observations, laboratory incubation experiments, and amplicon sequencing, we show that seasonal thermokarst la

[Effects of nitrogen addition on the composition and biodegradability of dissolved organic matter in northern peatlands of the Greater Khingan Mountains, Northeast China.].

Dissolved organic matter (DOM) represents the most labile fractions of organic matter and drives ecosystem carbon turnover in northern peatlands. However, the effects of increased nitrogen (N) availability on DOM dynamics and the underlying mechanisms remain unclear in northern peatlands. We conducted a 14-year N addition experiment (0, 3, 6, and 12 g N·m-2·a-1) in a peatland located in the continuous permafrost region of the northern Greater Khingan Mountains. We examined the effects of N addition on DOM chemical composition and biodegradability in surface water and soil pore water (0-15 cm)

Factors Contributing to Anthrax Outbreaks in the Circumpolar North.

A 2016 outbreak of anthrax on the Yamal Peninsula in Siberia that led to the culling of more than two hundred thousand reindeer and killed one human, resulted in significant media interests and in the reporting was often linked to thawing permafrost and ultimately climate change. Here, we review the historic context of anthrax outbreaks in the circumpolar North and explore alternative explanations for the anthrax outbreak in Western Siberia. Further, we propose a convergence model where multiple factors likely contributed to the outbreak of anthrax, including an expanded population and discont
