In vitro and in vivo analyses of co-infections with peste des petits ruminants and capripox vaccine strains.

BACKGROUND: Peste des petits ruminants (PPR) and goat pox (GTP) are two devastating animal epidemic diseases that affect small ruminants. Vaccination is one of the most important measures to prevent and control these two severe infectious diseases. METHODS: In this study, we vaccinated sheep with PPR and POX vaccines to compare the changes in the antibody levels between animals vaccinated with PPRV and POX vaccines alone and those co-infected with both vaccines simultaneously. The cell infection model was used to explore the interference mechanism between the vaccines in vitro. The antibody l

Autophagy induction by the pathogen receptor NECTIN4 and sustained autophagy contribute to peste des petits ruminants virus infectivity.

Macroautophagy/autophagy is an essential cellular response in the fight against intracellular pathogens. Although some viruses can escape from or utilize autophagy to ensure their own replication, the responses of autophagy pathways to viral invasion remain poorly documented. Here, we show that peste des petits ruminants virus (PPRV) infection induces successive autophagic signalling in host cells via distinct and uncoupled molecular pathways. Immediately upon invasion, PPRV induced a first transient wave of autophagy via a mechanism involving the cellular pathogen receptor NECTIN4 and an AKT-

A thermostable presentation of the live, attenuated peste des petits ruminants vaccine in use in Africa and Asia.

The research objective was to develop a thermostable vaccine against peste des petits ruminants (PPR), a morbilliviral disease of small ruminants targeted for eradication that is a major constraint on the livelihoods of the rural poor throughout much of Africa and Asia. Although existing PPR vaccines provide life-long immunity, they require continuous refrigeration. This limits their utility in developing countries. Methods for the lyophilization of a related morbillivirus, rinderpest (RP), resulted in vaccine that could be used in the field for up to 30days without refrigeration which was a m

Concurrent vaccination of goats with foot and mouth disease (FMD) and peste des petits ruminants (PPR) booster vaccines.

Foot and mouth disease (FMD) remains subclinical and self-limiting in small ruminants, but risk of spread of infection to susceptible cohorts is of great epidemiological significance; therefore, small ruminants must be included in vaccination campaigns in FMD endemic regions. Three groups of goats already immunized against peste des petits ruminants (PPR) were vaccinated with FMD and PPR vaccines alone or concurrently. The specific antibody response against three FMD virus strains and PPR virus were evaluated by competitive enzyme-linked immunosorbent assay (cELISA). Goats concurrently vaccina

Mapping immune targets in peste des petits ruminants virus hemagglutinin: An integrated computational framework for vaccine candidate prioritization.

BACKGROUND: Peste des petits ruminants virus (PPRV) is a major transboundary viral pathogen of small ruminants and causes substantial economic losses in endemic regions. The hemagglutinin (H) protein mediates receptor recognition and host-cell attachment and is an important target for vaccine development. This study applied an integrated computational framework to identify conserved immunogenic regions within the PPRV H protein. METHODS: A total of 64 unique PPRV H protein sequences were analyzed using multiple sequence alignment, entropy-based conservation profiling, conservation-aware epito

Serosurveillance and participatory disease surveillance of Peste des Petits Ruminants (PPR) in Georgia and Armenia in 2019-2020.

Peste des petits ruminants (PPR) is a severe viral disease of small ruminants, endemic in Asia, the Middle East, and Africa, and a WOAH and FAO target for eradication. In the last decade, PPR emerged in countries in the Caucasus and Europe. Georgia's first outbreak in 2016 led to multi-year vaccination campaigns in Georgia and neighbouring, PPR-free, Armenia. This was followed by serological surveillance campaigns in 2019-2020 in both countries. In addition, in 2019, participatory diseases surveillance including interviews and focus-group discussions were conducted in four provinces of Georgia

Effect of injectable antioxidants on redox profile after Peste des petits ruminants (PPR) vaccination in Naïve goat kids.

Peste des petits ruminants (PPR) vaccination can induce oxidative and physiological stress in naïve goats, highlighting the need for supportive interventions to enhance post-vaccination resilience. The present study evaluated the effect of injectable immune modulators and antioxidants on the haematological, hormonal, and redox biomarkers in Rohilkhandi goats, a local registered goat breed of Uttar Pradesh, India, during Peste des petits ruminants (PPR) vaccination. Eighteen apparently healthy goats were divided into three groups (06 each): Control – vaccinated against PPR without supplementati

Genomic analysis of host - Peste des petits ruminants vaccine viral transcriptome uncovers transcription factors modulating immune regulatory pathways.

Peste des petits ruminants (PPR), is an acute transboundary viral disease of economic importance, affecting goats and sheep. Mass vaccination programs around the world resulted in the decline of PPR outbreaks. Sungri 96 is a live attenuated vaccine, widely used in Northern India against PPR. This vaccine virus, isolated from goat works efficiently both in sheep and goat. Global gene expression changes under PPR vaccine virus infection are not yet well defined. Therefore, in this study we investigated the host-vaccine virus interactions by infecting the peripheral blood mononuclear cells isolat

Experimental infection of sheep and goats with a recent isolate of peste des petits ruminants virus from Kurdistan.

Peste des petits ruminants (PPR) is a contagious viral disease of sheep and goats common in Africa and Asia. Its high morbidity and mortality has a devastating impact on agriculture in developing countries. As an example, an Asian lineage IV strain of PPRV was responsible for mass fatalities among wild goats in Kurdistan in 2010/2011. In separate experiments, three sheep and three goats of German domestic breeds were subcutaneously inoculated with the Kurdish virus isolate; three uninfected sheep and goats were housed together with the inoculated animals. All inoculated animals, all in-contact

MicroRNA Expression Profile in Peripheral Blood Lymphocytes of Sheep Vaccinated with Nigeria 75/1 Peste Des Petits Ruminants Virus.

Peste des petits ruminants (PPR) is one of the highly contagious transboundary viral diseases of small ruminants. Host microRNA (miRNA) expression patterns may change in response to virus infection, and it mainly works as a post-transcriptional moderator in gene expression and affects viral pathogenesis and replication. In this study, the change of miRNA expression profile in peripheral blood lymphocyte (PBMC) from sheep inoculated with PPR vaccine virus in vivo as well as primary sheep testicular (ST) cells inoculated with PPR vaccine virus in vitro were determined via deep sequencing technol

CAV1 up-regulated by peste des petits ruminants virus nucleocapsid protein impairs type I IFN response by cholesterol synthesis.

Peste des petits ruminants virus (PPRV) is a highly contagious pathogen that infects domestic and wild small ruminants, causing high mortality worldwide. PPRV has evolved multiple strategies to suppress the host antiviral immune response, thereby promoting viral replication. In this study, we demonstrate that caveolin-1 (CAV1) promotes PPRV replication. Mechanistically, CAV1 inhibits IFN-β production, ISG expression, and the IFN signaling pathway by promoting cholesterol synthesis. Importantly, we reveal that PPRV infection upregulates cellular CAV1 protein levels, and the viral nucleocapsid (

Molecular Detection of Peste des Petits Ruminants Virus (PPRV) in Goats and in Sheep in Ibadan, Oyo State, Nigeria.

Peste des petits ruminants (PPR) is a vaccine-preventable transboundary animal disease of goats and sheep majorly, and is regarded as a major constraint to small ruminant production especially in developing countries like Nigeria. Despite different strategies that have been employed to control PPR in Nigeria, cases of the disease are still reported in PPR-vaccinated and unvaccinated small ruminant farms. In this study, molecular detection of field PPR virus (PPRV) strains was carried out to determine the presence of PPRV. A total of 135 samples (45 oculo-nasal swabs and 90 tissue samples) were
