Phase separation is a common biological phenomenon in the liquid environment of organisms. Phase separation has been shown to be a key cause of many existing incurable diseases, such as the protein aggregates formed by phase separation of Alzheimer's Disease, Amyotrophic Lateral Sclerosis, Parkinson's disease, etc. Tracking the occurrence of phase separation in vivo is critical to many disease detection methods and solving many treatment problems. Its physicochemical properties and visual detection methods have flourished in the last few years in chemical biology, among which the fluorogenic t

The problems of extracting products efficiently from reaction workups are often overlooked. Issues such as emulsions and rag layer formation can cause long separation times and slow production, thus resulting in manufacturing inefficiencies. To better understand science within this area and to support process development, an image processing methodology has been developed that can automatically track the interface between liquid-liquid phases and provide a quantitative measure of the separation rate of two immiscible liquids. The algorithm is automated and has been successfully applied to 29 c

Plasma membrane heterogeneity has been tied to a litany of cellular functions and is often explained by analogy to membrane phase separation; however, models based on phase separation alone fall short of describing the rich organization available within cell membranes. Here we present comprehensive experimental evidence motivating an updated model of plasma membrane heterogeneity in which membrane domains assemble in response to protein scaffolds. Quantitative super-resolution nanoscopy measurements in live B lymphocytes detect membrane domains that emerge upon clustering B cell receptors (BCR

Thousands of genetic variants in protein-coding genes have been linked to disease. However, the functional impact of most variants is unknown as they occur within intrinsically disordered protein regions that have poorly defined functions1-3. Intrinsically disordered regions can mediate phase separation and the formation of biomolecular condensates, such as the nucleolus4,5. This suggests that mutations in disordered proteins may alter condensate properties and function6-8. Here we show that a subset of disease-associated variants in disordered regions alter phase separation, cause mispartitio

A ferritin particle consists of 24 ferritin proteins (FTH1 and FTL) and stores iron ions within it. During iron deficiency, ferritin particles are transported to lysosomes to release iron ions. Two transport pathways have been reported: macroautophagy and ESCRT-dependent endosomal microautophagy. Although the membrane dynamics of these pathways differ, both require NCOA4, which is thought to be an autophagy receptor for ferritin. However, it is unclear whether NCOA4 only acts as an autophagy receptor in ferritin degradation. Here, we found that ferritin particles form liquid-like condensates i

In Parkinson's disease with dementia, up to 50% of patients develop a high number of tau-containing neurofibrillary tangles. Tau-based pathologies may thus act synergistically with the α-synuclein pathology to confer a worse prognosis. A better understanding of the relationship between the two distinct pathologies is therefore required. Liquid-liquid phase separation (LLPS) of proteins has recently been shown to be important for protein aggregation involved in amyotrophic lateral sclerosis, whereas tau phase separation has been linked to Alzheimer's disease. We therefore investigated the inter

Polymeric membranes are used on very large scales for drinking water production and kidney dialysis, but they are nearly always prepared by using large quantities of unsustainable and toxic aprotic solvents. In this study, a water-based, sustainable, and simple way of making polymeric membranes is presented without the need for harmful solvents or extreme pH conditions. Membranes were prepared from water-insoluble polyelectrolyte complexes (PECs) via aqueous phase separation (APS). Strong polyelectrolytes (PEs), poly(sodium 4-styrenesulfonate) (PSS), and poly(diallyldimethylammonium chloride)

An important component of cellular biochemistry is the concentration of proteins and nucleic acids in non-membranous compartments1,2. These biomolecular condensates are formed from processes that include liquid-liquid phase separation. The multivalent interactions necessary for liquid-liquid phase separation have been extensively studied in vitro1,3. However, the regulation of this process in vivo is poorly understood. Here we identify an in vivo regulator of liquid-liquid phase separation through a genetic screen targeting factors required for Arabidopsis RNA-binding protein FCA function. FCA

We found that addition of N,N-dimethylformamide (DMF) induces phase separation of 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP)-water mixtures. The phase diagram of a DMF-HFIP-water ternary system at 298 K showed that phase separation occurs in a closed-loop area in the water-rich mole fraction range of x(H(2)O) > ∼0.4. To clarify the mechanism of DMF-induced phase separation of DMF-HFIP-water mixtures at the molecular level, small-angle neutron scattering (SANS) and (1)H and (13)C NMR measurements were conducted on the mixtures with varying DMF concentrations along a volume ratio of HFIP to water

A simple, rapid and robust LC method was developed and validated for the enantiomeric separation of valsartan in bulk drug and formulation. The enantiomers of valsartan were resolved on a Chiralpak AD-H (amylose based stationary phase) column using a mobile phase consisting of n-hexane: 2-propanol: trifluoroacetic acid (85:15:0.2, v/v/v) at a flow rate of 1.0 mL/min. The resolution between the enantiomers was found to be not less than 3.2. The presence of trifluoroacetic acid in the mobile phase played an important role in enhancing chromatographic efficiency and resolution between the enantio

This report demonstrates a novel method to prepare fine polysaccharide glassy particles of uniform sizes under a condition without water/oil and water/air interfacial tension and cross-linking reagents. When a co-solution of dextran and polyethylene glycol (PEG) was frozen gradually, phase separation occurred during which dextran formed the dispersed phase and PEG remained in the continuous part. Fine dextran glassy particles were harvested after lyophilizing this frozen sample, followed by re-dissolving the continuous phase (PEG) in dichloromethane or acetonitrile. Desired mean particle diame

A thin-layer chromatographic system is described for the separation of diphenhydramine and bromodiphenhydramine which is based on a reverse phase paper chromatographic system developed by J. Vecerková (2,3) in 1962. Chromatographic systems for this separation described in the literature and several systems attempted in this laboratory using normal chromatography have not proven successful. The method using reverse phase thin-layer chromatography involves a stationary phase of mineral oil on silica gel G and a mobile phase of ethanol:water:ammonium hydroxide, 28% NH3, (55:43:2). A 10-cm length

We compare seven elastic models for polymer networks in the context of phase separation within a gel, including a new model that combines the finite extensible Arruda-Boyce model and the slip tube model for entangled chains. We study incompressible uniaxial stretch and compression, and three volume-changing constrained-dimension deformations, in which the material can only deform in the designated dimension(s), while the constrained direction(s) remain(s) the same. Each model responds differently to large deformations, and our proposed model successfully describes both strain softening and str

The distribution of the AB diblock copolymer in a phase-separating solution composed of immiscible A and B homopolymers in a common solvent has been investigated theoretically. We have utilized the mixing Gibbs energy density for the interfacial phase based on mean-field lattice theory to this four-component system. Distributions of the AB diblock copolymer in the A and B homopolymer-rich bulk phases and the interfacial region between the separating bulk phases are calculated as a function of the B-block content, degrees of polymerization of the copolymer and A and B homopolymers, as well as i

Protein condensates that form via phase separation typically become more viscous over time and can harden in a process referred to as "molecular aging." Several mechanisms have been identified for this phenomenon. Of these, the ones involving enhanced β-sheet or -strand interactions are of pathological relevance since they have been associated with neurodegeneration. Although there is much understanding of biopolymer phase behavior, an inclusive thermodynamic framework that unifies phase separation and β-sheet-based aging is lacking. We present a time-dependent, multi-component extension of as

Neurodegenerative diseases, such as Alzheimer's and Parkinson's, share a common pathological feature of amyloid structure accumulation. However, the structure-function relationship between these well-ordered, β-sheet-rich, filamentous protein deposits and disease etiology remains to be defined. Recently, an emerging hypothesis has linked phase separation, a process involved in the formation of protein condensates, to amyloid formation, suggesting that liquid protein droplets serve as loci for amyloid initiation. To elucidate how these processes contribute to disease progression, tools that can

Metal ions have emerged as critical factors influencing biomolecular phase separation and are increasingly implicated in the formation, maturation, and pathological transformation of condensates associated with amyloidogenic proteins. Recent studies have shown that metal ions can affect liquid-liquid phase separation and liquid-to-solid phase transition through multiple mechanisms, including electrostatic interactions, direct metal coordination, intermolecular bridging, and metal-induced conformational changes. Depending on the specific proteins and metal ions involved, these interactions can

Biomolecular condensates formed through phase separation are fundamental to the cellular organization. Although the physical principles underlying the phase separation of intrinsically disordered proteins are well understood, the molecular determinants of condensate formation in globular proteins remain elusive. Here, we employ holographic particle characterization, a label-free, high-throughput imaging technique, to investigate the self-assembly of bovine serum albumin (BSA), a model globular protein. We show that this technique reliably differentiates between liquid-like and gel-like condens

The effect of polypropylene glycol (PPG) on the phase structure formation and deformation properties of a photocurable system based on bisphenol-A 10-ethoxylate dimethacrylate (BPADMA) was studied. Optical interferometry established the complete compatibility of the components before polymerization, and the critical PPG concentration predicted as corresponding to the center of the phase inversion region (28 vol.%) was determined. SEM confirmed the formation of the predicted phase structure, characterized by interpenetrating continuous phases. FTIR spectroscopy showed that the introduction of P
