Other agents. Phencyclidine, marijuana, hallucinogens, inhalants, and anticholinergics.

Acute phencyclidine intoxication causes psychosis and a myriad of other symptoms and signs, some life-threatening. Anticholinergic poisoning is also a medical emergency, often requiring an intensive care unit. Marijuana and hallucinogens have rarely, if ever, resulted in direct overdose death, but intoxication can result in accidents or self-injury. Inhalants cause death from cardiac arrhythmia, suffocation, or accident. Each of these agents is associated with a variety of medical and neurologic complications, some of which are discussed at greater length elsewhere in this issue.

Acute phencyclidine intoxication: clinical patterns, complications, and treatment.

We describe four major and five minor clinical patterns of acute phencyclidine (PCP) intoxication and give the incidence of findings in each pattern. Major patterns were acute brain syndrome (248 cases; 24.8%), toxic psychosis (166 cases; 16.6%), catatonic syndrome (117 cases; 11.7%), and coma (106 cases; 10.6%). Minor patterns included lethargy or stupor (38 cases; 3.8%), and combinations of bizarre behavior, violence, agitation, and euphoria in patients who were alert and oriented (325 cases; 32.5%). Patients with major patterns of PCP toxicity usually required hospitalization and accounted

Effects of clozapine plus lamotrigine on phencyclidine-induced hyperactivity.

There is growing evidence from both uncontrolled and controlled clinical studies that lamotrigine (LTG) significantly augments clozapine (CLZ) in the treatment of refractory schizophrenia (RS) [Dursun, S.M., McIntosh, D., Milliken, H., 1999. Functional changes in neuronal systems induced by phencyclidine administration.

To analyze the mechanisms of PCP abuse, we investigated the changes in PCP-induced motivational properties and neuronal functions in animals. First, we determined that PCP-induced withdrawal syndrome may, in part, be produced by 5-HTergic neuronal systems. Second, using rats treated with subacute PCP, we established that subacute PCP may produce behavioral changes (stereotyped behaviors and hyperlocomotion), mediated by both dopaminergic, 5-HTergic neuronal, and NO systems. Third, using the place conditioning paradigm, we confirmed that (1) both dopamine-D1 and 5-HT2A receptors, but not sigma

Phencyclidine blocks voltage-dependent potassium currents in murine thymocytes.

We investigated the effect of phencyclidine (PCP) on the voltage-dependent K+ current in whole cell recordings from murine thymocytes. PCP caused a dramatic, reversible and dose-dependent decrease of the current. An analysis of the dose-response relationship suggests a single site of action for PCP with an IC50 of 4.7 microM. Dextrorphan and naloxone also inhibited the current, although their effects were of a lower magnitude than those of PCP. Neither 10 microM dextrorphan nor 100 microM naloxone antagonized the inhibitory action of 10 microM PCP. The analysis of the dose-response curve for P

Alterations in local cerebral glucose utilization induced by phencyclidine.

The effects of phencyclidine (PCP; 0.5, 1,5, 10 mg/kg, i.v.) on local cerebral glucose utilization (LCGU) in the rat were studied with the 2-deoxy-D-[1-14C]glucose method. Significant findings were obtained in 41 of 87 brain regions of PCP-treated rats (25-270% of control). Rates of LCGU increased throughout the limbic system, except the habenula. Although LCGU increased in most sensory structures, it decreased in specific layers of the somatosensory and auditory cortices and the inferior colliculus. Evidence was seen for dissociation between LCGU responses of specific thalamic relay areas and

The depressant effects of phencyclidine [1-(1-phenylcyclohexyl) piperidine, PCP] and three of its analogs (m-amino-PCP, m-nitro-PCP, and PCP-methyliodide) on the spontaneous action potential discharge of cerebellar Purkinje neurons in urethane-anesthetized rats were examined in this study. Both intraperitoneal injection and micro-pressure ejection were employed as routes of drug administration. The relative potency after parenteral administration corresponded closely with previous findings in behavioral test paradigms. Phencyclidine: behavioral and biochemical evidence against the anticholinergic hypothesis.

Phencyclidine (PCP) is known to have anticholinergic effects in various in vitro test systems and to inhibit the binding of muscarinic antagonists to rat brain membranes. In order to verify the anticholinergic properties of PCP, its interaction with oxotremorine (OXO), a muscarinic agonist, was studied in mice. OXO (1 mg/kg) in combination with PCP (10 mg/kg) was lethal in 100% of the mice studied. The lethality of this combination was completely reversed by 3.3 mg/kg methyl atropine bromide (MA), a quarternary muscarinic antagonist. Therefore, PCP appears to act as a muscarinic agonist in som

Is phencyclidine (PCP) abuse associated with organic mental impairment?

The performance of sober (average length of abstinence = 27 months) phencyclidine (PCP) abusers on neuropsychological measures of organicity was compared to that of polydrug users who were not experienced with PCP, and to controls who were not alcohol or drug abusers. Six of 12 PCP users, five of 12 polydrug users, and none of the controls showed neuropsychological impairments. The deficits in PCP users occurred despite negative medical-neurological history, and even though the PCP group abused other drugs previously associated with neuropsychological impairment less than the polydrug group. Sigma and phencyclidine receptors in the brain-endocrine-immune axis.

We have demonstrated the absence of PCP receptors and the presence of sigma receptors in PBLs and rat pituitary, adrenal, testis, and ovary that have kinetic and pharmacologic characteristics comparable with the sigma receptor found in the CNS. The physiologic significance of the sigma receptors in the various lymphoid and endocrine tissues remains to be determined, but their presence in such high densities suggests that endogenous sigma ligands may play an important role in regulating and integrating endocrine and immune responses. Thus, in addition to their central actions, sigma agonists in

Impact of intrauterine exposure to phencyclidine (PCP) and cocaine on neonates.

A total of 505 newborns who were exposed to illicit drugs during intrauterine life were studied to investigate the prevalence, growth parameters, newborn manifestations, and other effects of intrauterine exposure to cocaine and phencyclidine (PCP). The results support the hypotheses that both drugs have serious effects on growth parameters, but this effect was much more pronounced in the cocaine group than in the PCP group. Furthermore, the incidence of borderline microcephalic infants (head circumferences less than the 10th percentile) was much higher in the cocaine group. It also appears tha

Effects of phencyclidine-like drugs on punished behavior in rats.

The effects of phencyclidine (PCP) and a number of PCP-like drugs on punished responding in rats were compared to those of chlordiazepoxide and morphine. Twenty rats, trained to lever-press for food reinforcement, were tested under a modified Geller-Seifter procedure using a multiple schedule with responding in one component reinforced under a fixed-interval 60-sec schedule while each response in the other component resulted in both food and brief electric shock presentation. Chlordiazepoxide, PCP, etoxadrol, (+)-N-allylnormetazocine and (-)-beta-cyclazocine all resulted in selective increases

Electrophysiological effects of phencyclidine in the medial prefrontal cortex of the rat.

The effects of local applications of phencyclidine (PCP) and dopamine (DA) on neurons of the medial prefrontal cortex were investigated using single unit recording techniques. The activity of the majority of cells in the deeper layers of the medial prefrontal cortex was depressed by both phencyclidine and DA, whereas increases, as well as decreases, in the firing rates were observed in cells located in the superficial cortical layers. The stereospecificity of the responses of deeper cells to phencyclidine was demonstrated using the enantiomers of 1-(-1-phenylcyclohexyl)-3-methylpiperidine (PCM
