Do farnesol and tyrosol production in Candidozyma auris biofilms reflect virulence potential?

Candidozyma auris is associated with nosocomial outbreaks presenting high resistance rates to antifungal. Since its emergence in 2009, research has been carried out on the underlying quorum sensing molecules (QSMs), such as farnesol (Far) and tyrosol (Tyr). Our main goal was to quantify Far and Tyr in C. auris biofilms and assess their role in virulence using a Galleria mellonella larvae model. Among the tested strains, the most virulent CBS 12766 produced the highest QSMs levels, whereas the least virulent strain CBS 10913 produced the lowest. Notably, C. auris exhibited higher QSM production

ROS Stress and Cell Membrane Disruption are the Main Antifungal Mechanisms of 2-Phenylethanol against Botrytis cinerea.

2-Phenylethanol (2-PE), a common compound found in plants and microorganisms, exhibits broad-spectrum antifungal activity. Using Botrytis cinerea, we demonstrated that 2-PE suppressed mycelium growth in vitro and in strawberry fruit and reduced natural disease without adverse effects to fruit quality. 2-PE caused structural damage to mycelia, as shown by scanning and transmission electron microscopy. From RNA sequencing analysis we found significantly upregulated genes for enzymatic and nonenzymatic reactive oxygen species (ROS) scavenging systems including sulfur metabolism and glutathione me

[The resolution of racemic sec-phenethyl alcohol on cellulose tribenzoate-based CSP: influence of different alcohols in the mobile phase].

Several primary and secondary alcohols (ethanol, 1-propanol, 2-propanol, 1-butanol) were used as the mobile phase components separately, to investigate their effects on the capacity factor and stereoselectivity of sec-phenethyl alcohol enantiomers on cellulose tribenzoate-based CSP. The chiral recognition mechanism for the enantiomeric aromatic alcohols studied may involve: (1) the aromatic portion of the solute may insert into a chiral cavity of the CSP through a hydrogen bonding interaction between the solute's alcoholic hydrogen and the ester carbonyl group on the CSP; (2) the mobile phase

Effect of Lactiplantibacillus plantarum-mediated lactic fermentation on the characteristic flavor of sea buckthorn juice by volatolomics and metabolomics analysis.

Lactic fermentation enhances sensory quality and nutritional properties of fruit juice, though its comprehensive impact on sea buckthorn through Lactiplantibacillus plantarum fermentation (LPF) remains unreported. This study used volatolomics and metabolomics to explore mechanism of lactic fermentation improving flavor. LPF reduced total acidity (from 24.01 g/L to 15.79 g/L), increased total flavonoid by 20.35% and polysaccharide contents by 55.64%. It also increased the quantity of volatile compounds (from 36 to 41). β-Damascenone and ethyl isovalerate were key aroma compounds, while the incr

The concentration-dependent impact of quorum-sensing signal molecule 2-phenylethanol on the flavor metabolite profile of Lactiplantibacillus pentosus 1 in a simulated sour fish system.

2-Phenylethanol (2-PE) is a quorum-sensing molecule that influences microbial metabolism, but its specific role in lactic acid bacteria (LAB) fermentation remains underexplored. This study investigated the effects of 2-PE (0-750 μM) on Lactiplantibacillus pentosus 1 (Lp 1) fermentation in a simulated sour fish system. Bacterial growth, residual sugar content, organic acid profiles, and volatile flavor compounds, combined with metabolomic analysis were conducted. Results demonstrated that low to moderate concentrations of 2-PE inhibited Lp 1 growth and metabolic activity, while high concentrati

An engineered dual-functional L-DOPA decarboxylase enables a minimized hydroxytyrosol cascade.

Hydroxytyrosol has been proven beneficial to human health. However, the process involving the conversion of L-DOPA to 3,4-dihydroxyphenylacetaldehyde (3,4-DHPAA) in hydroxytyrosol biosynthesis typically required the simultaneous use of decarboxylase and oxidative deaminase. In addition, phenylacetaldehyde reductase from Solanum lycopersicum (SlPAR) in hydroxytyrosol biosynthesis exhibits poor thermal stability. In this study, we unexpectedly discovered that L-DOPA decarboxylase from Pseudomonas putida (PpDODC) exhibits weak dual-functional activity for both decarboxylation and oxidative deamin

Olive Oil Components as Novel Antioxidants in Neuroblastoma Treatment: Exploring the Therapeutic Potential of Oleuropein and Hydroxytyrosol.

In this review, we explored the therapeutic potential of oleuropein (OLE) and hydroxytyrosol (HT) in the treatment of neuroblastoma (NB). NB is an extracranial tumour that predominantly affects children aged between 17 and 18 months. Recurrence and drug resistance have emerged as the biggest challenges when treating NB, leading to a crucial need for new therapeutic approaches. Food of the Mediterranean Diet (MD) presents several health benefits, including that of cancer treatment. In this review, we emphasised olive oil since it is one of the main liquid ingredients of the MD. OLE is the princ

Chemicals Affecting Cyanobacterial Poly(3-hydroxybutyrate) Accumulation: 2-Phenylethanol Treatment Combined with Nitrogen Deprivation Synergistically Enhanced Poly(3-hydroxybutyrate) Storage in Synechocystis sp. PCC6803 and Anabaena sp. TISTR8076.

Various photoautotrophic cyanobacteria increase the accumulation of bioplastic poly(3-hydroxybutyrate) (PHB) under nitrogen deprivation (-N) for energy storage. Several metabolic engineering enhanced cyanobacterial PHB accumulation, but these strategies are not applicable in non-gene-transformable strains. Alternatively, stimulating PHB levels by chemical exposure is desirable because it might be applied to various cyanobacterial strains. However, the study of such chemicals is still limited. Here, 19 compounds previously reported to affect bacterial cellular processes were evaluated for their

Mechanism of synergistic DNA damage induced by caffeic acid phenethyl ester (CAPE) and Cu(II): Competitive binding between CAPE and DNA with Cu(II)/Cu(I).

Caffeic acid phenethyl ester (CAPE) is an active polyphenol of propolis from honeybee hives, and exhibits antioxidant and interesting pharmacological activities. However, in this study, we found that in the presence of Cu(II), CAPE exhibited pro-oxidative rather than antioxidant effect: synergistic DNA damage was induced by the combination of CAPE and Cu(II) together as measured by strand breakage in plasmid DNA and 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) formation, which is dependent on the molar ratio of CAPE:Cu(II). Production of Cu(I) and H2O2 from the redox reaction between CAPE and
