Patient-specific iPSC-derived photoreceptor precursor cells as a means to investigate retinitis pigmentosa.

Next-generation and Sanger sequencing were combined to identify disease-causing USH2A mutations in an adult patient with autosomal recessive RP. Induced pluripotent stem cells (iPSCs), generated from the patient's keratinocytes, were differentiated into multi-layer eyecup-like structures with features of human retinal precursor cells. The inner layer of the eyecups contained photoreceptor precursor cells that expressed photoreceptor markers and exhibited axonemes and basal bodies characteristic of outer segments. Analysis of the USH2A transcripts of these cells revealed that one of the patient

Immunocytochemical localisation of hydroxyindole-O-methyltransferase in pineal photoreceptor cells of several fish species.

Melatonin is an internal "Zeitgeber," involved in the timing and control of a number of rhythmic functions and behaviours. Its synthesising cells remain to be identified in the fish pineal. The last step in the melatonin biosynthetic pathway is catalysed by the enzyme hydroxyindole-O-methyltransferase. An affinity-purified antibody, directed against chicken pineal hydroxyindole-O-methyltransferase, was used in the present study to identify the melatonin synthesising cells in four fish species: a primitive chondrostean (sturgeon), a saltwater teleost (dorado), and two freshwater teleosts (pike,

Synaptic contacts between bipolar and photoreceptor cells in the retina of Callionymus lyra L.

A combined light and electron microscopic study of Golgi-impregnated retinas of the marine teleost Callionymus lyra L. revealed mixed bipolar cells (M types) contacting rods and cones and pure cone bipolar cells (C types). Five types of mixed bipolar cells can be differentiated on the basis of their synaptic contacts. Two out of the five mixed bipolar cell types contact double cones, single cones, and rods (mixed, dark, pale, single [Mdps and midget-Mdps]). Their endbuds make narrow cleft junctions, with each type of photoreceptor, and in addition, two endbuds end centrally in the synaptic rib

Optimized Retinal Cell Dissociation for Downstream Analysis of Cone Photoreceptors.

The isolation of viable and homogenous retinal cells is essential for downstream molecular analysis, such as single-cell sequencing and transcriptomics. However, the dissociation of retinal cells can be challenging due to the intricate nature of retinal tissue and the fragility of its cells. Furthermore, cone photoreceptors are highly susceptible to degeneration. We developed an optimized protocol for retinal cell dissociation that improves the quality of cones for downstream molecular analysis. Our protocol employs a papain enzymatic digestion, mechanical dissociation, and filtration to obtai

TRPV4 affects visual signals in photoreceptors and rod bipolar cells.

INTRODUCTION: Mechanical sensitive channels expressed in mammalian retinas are effectors of elevated pressure stresses, but it is unclear how their activation affects visual function in pressure-related retinal disorders. METHODS: This study investigated the role of the transient potential channel vanilloid TRPV4 in photoreceptors and rod bipolar cells (RBCs) with immunohistochemistry, confocal microscopy, electroretinography (ERG), and patch-clamp techniques. RESULTS: TRPV4 immunoreactivity (IR) was found in the outer segments of photoreceptors, dendrites and somas of PKCα-positive RBCs and

Stimulation of AMPK Prevents Diabetes-Induced Photoreceptor Cell Degeneration.

Diabetic retinopathy (DR) is a kind of severe retinal neurodegeneration. The advanced glycation end products (AGEs) affect autophagy, and mitochondrial function is involved in DR. Adenosine-activated protein kinase (AMPK) is an important metabolic sensor that can regulate energy homeostasis in cells. However, the effect of AMPK in DR is still not fully understood. In this study, we investigated the effect of AMPK on diabetes-induced photoreceptor cell degeneration. In vivo, a diabetic mouse model was established by streptozotocin (STZ) injection. Haematoxylin-eosin (HE) staining was used to ob

RPE Cells Engulf Microvesicles Secreted by Degenerating Rod Photoreceptors.

Rhodopsin is mislocalized to the inner segment plasma membrane (IS PM) in various blinding disorders including autosomal-dominant retinitis pigmentosa caused by class I rhodopsin mutations. In these disorders, rhodopsin-laden microvesicles are secreted into the extracellular milieu by afflicted photoreceptor cells. Using a Xenopus laevis model expressing class I mutant rhodopsin or Na+/K+-ATPase (NKA) fused to Dendra2, we fluorescently labeled the microvesicles and found retinal pigment epithelial (RPE) cells are capable of engulfing microvesicles containing rhodopsin. A unique sorting mechani

Separation of photoreceptor cell compartments in mouse retina for protein analysis.

BACKGROUND: Light exposure triggers movement of certain signaling proteins within the cellular compartments of the highly polarized rod photoreceptor cell. This redistribution of proteins between the inner and outer segment compartments affects the performance and physiology of the rod cell. In addition, newly synthesized phototransduction proteins traverse from the site of their synthesis in the inner segment, through the thin connecting cilium, to reach their destination in the outer segment. Processes that impede normal trafficking of these abundant proteins lead to cell death. The study of

Review: pharmacological manipulation of docosahexaenoic-phospholipid biosynthesis in photoreceptor cells: implications in retinal degeneration.

Docosahexaenoic acid (22:6n-3, DHA) is derived in vertebrate animals from n-3 fatty acids present in the diet (i.e., alpha-linolenic acid, 18:3n-3 and/or other n-3-long chain polyunsaturated fatty acids) and is found in very high concentrations in phospholipids from membranes of the central nervous system. Disk membranes of photoreceptor outer segments and synaptic terminals display a preferential enrichment in DHA-phospholipids that appears to be necessary for normal excitable membrane functions. Because of the relevance of adequate DHA-phospholipid synthesis and sorting toward new assembled

Subfunctionalization of tbx2 paralogues during photoreceptor cell specification in zebrafish.

Zebrafish possess three distinct sources of retinal progenitors that produce identical photoreceptor subtypes throughout life. All photoreceptor progenitors simultaneously express multiple transcription factors specifying different identities, requiring mechanisms to repress alternative fates. Disruption of the tbx2 paralogues, tbx2a or tbx2b , resulted in a cell-fate switch of sws1 cones into rods. Here, we demonstrate that tbx2b was necessary for sws1 cone differentiation during embryogenesis and outgrowth at the retinal margin, but tbx2a was necessary during photoreceptor regeneration. Tran

[Advances in adaptive optical imaging of photoreceptor cell observation in the macular].

This article focuses on the application of adaptive optics (AO) technology in the observation of photoreceptor cells (PRs) in the macular area. Firstly, the principle and development process of AO technology are described. It can actively measure and correct optical aberrations to achieve high-resolution imaging. Since its invention, it has been increasingly used in ophthalmic research. Then, a detailed review is presented on the observation results of PRs by AO in healthy individuals and various ophthalmic diseases, including the performance characteristics of PRs in inherited retinal disease

Mouse Models of Inherited Retinal Degeneration with Photoreceptor Cell Loss.

Inherited retinal degeneration (RD) leads to the impairment or loss of vision in millions of individuals worldwide, most frequently due to the loss of photoreceptor (PR) cells. Animal models, particularly the laboratory mouse, have been used to understand the pathogenic mechanisms that underlie PR cell loss and to explore therapies that may prevent, delay, or reverse RD. Here, we reviewed entries in the Mouse Genome Informatics and PubMed databases to compile a comprehensive list of monogenic mouse models in which PR cell loss is demonstrated. The progression of PR cell loss with postnatal age

CD44 signaling in Müller cells impacts photoreceptor function and survival in healthy and diseased retinas.

Retinitis pigmentosa (RP), an inherited retinal disease, affects 1,5 million people worldwide. The initial mutation-driven photoreceptor degeneration leads to chronic inflammation, characterized by Müller cell activation and upregulation of CD44. CD44 is a cell surface transmembrane glycoprotein and the primary receptor for hyaluronic acid. It is involved in many pathological processes, but little is known about CD44's retinal functions. CD44 expression is also increased in Müller cells from our Pde6bSTOP/STOP RP mouse model. To gain a more detailed understanding of CD44's role in healthy and

Retinal Injury Activates Complement Expression in Müller Cells Leading to Neuroinflammation and Photoreceptor Cell Death.

Retinal detachment (RD) is a neurodegenerative blinding disease caused by plethora of clinical conditions. RD is characterized by the physical separation of retina from the underlying retinal pigment epithelium (RPE), eventually leading to photoreceptor cell death, inflammation, and vision loss. Albeit the activation of complement plays a critical role in the pathogenesis of RD, the retinal cellular source for complement production remains elusive. Here, using C3 tdTomato reporter mice we show that retinal injury upregulates C3 expression, specifically in Müller cells. Activation of the comple

Induction of the unfolded protein response by constitutive G-protein signaling in rod photoreceptor cells.

Phototransduction is a G-protein signal transduction cascade that converts photon absorption to a change in current at the plasma membrane. Certain genetic mutations affecting the proteins in the phototransduction cascade cause blinding disorders in humans. Some of these mutations serve as a genetic source of "equivalent light" that activates the cascade, whereas other mutations lead to amplification of the light response. How constitutive phototransduction causes photoreceptor cell death is poorly understood. We showed that persistent G-protein signaling, which occurs in rod arrestin and rhod

Interaction and localization of the retinitis pigmentosa protein RP2 and NSF in retinal photoreceptor cells.

RP2 is a ubiquitously expressed protein encoded by a gene associated with X-linked retinitis pigmentosa (XLRP), a retinal degenerative disease that causes severe vision loss. Previous in vitro studies have shown that RP2 binds to ADP ribosylation factor-like 3 (Arl3) and activates its intrinsic GTPase activity, but the function of RP2 in the retina, and in particular photoreceptor cells, remains unclear. To begin to define the role of RP2 in the retina and XLRP, we have conducted biochemical studies to identify proteins in retinal cell extracts that interact with RP2. Here, we show that RP2 in

Syrian hamster prion protein (PrP(C)) is expressed in photoreceptor cells of the adult retina.

PrP(C), the cellular isoform of the prion protein (PrP) serves as a precursor to abnormal PrP isoforms which accumulate in diseases such as scrapie in sheep, and Creutzfeldt-Jakob disease in humans. Since prions can replicate in photoreceptors we surmised that PrP(C) must be expressed in these cells. Accordingly, monoclonal antisera directed against two epitopes of hamster PrP(C) produced retinal immunostaining in hamsters, and in mice bearing a hamster PrP transgene. Immunostaining was most prominent in the inner and outer segments of rod photoreceptors, coinciding with the earliest site of p

Intracellular staining of physiologically identified photoreceptor cells and hyperpolarizing interneurons in the teleost pineal organ.

The directly photosensory pineal organ of the rainbow trout functions primarily as a luminance detector. Its neutral output reflects the level of ambient illumination in an almost linear fashion over several orders of magnitude. It may thus transmit information about the daily light-dark cycle to central projection targets in the brain, and exert an important control over putative central oscillators. We have studied single neural elements in the explanted pineal organ of the rainbow trout by combining intracellular recording with intracellular injections of either the fluorescent dye Lucifer

Survival of some photoreceptor cells in albino rats following long-term exposure to continuous light.

Fischer albino rats, seven weeks of age, were exposed to continuous light at 65 foot-candle incident illuminance for up to 264 days. Other Fischer rats, seven months of age, were exposed to continuous light at 140 foot-candle incident illuminance for up to 147 days. In all cases, a small percentage of the photoreceptors survived. The identification of the surviving cells as photoreceptors was made by light microscopy on the basis of nuclear heterochromatin pattern and staining and by electron microscopy by the presence of ribbon synapses and ciliary basal bodies with ciliary filaments.
