Turnover rates of photosynthesis determined by fluorescence kinetic analysis.

Maximum turnover rates of photosynthesis impose an upper limit on global primary production, yet traditional estimation methods remain tedious and prone to significant uncertainty. This research presents a new method and operational instrumentation to deduce the maximum turnover rates of photosynthesis from instantaneous kinetic analysis of chlorophyll fluorescence under saturating background light. Measurements of maximum turnover rates in model species of green algae and diatoms over a range of environmental conditions revealed that the rates vary from 150 to 600 s- 1 per PSII. These rates c

Red light promotes sea slug growth while blue light increases longevity of stolen chloroplasts.

In Metazoa, long-term sequestration of photosynthetically active algal chloroplasts - kleptoplasts - is a distinctive physiological trait of sacoglossan sea slugs. In this study, we examined the effects of the light spectrum on the body weight and photosynthetic activity of the sea slug Elysia crispata. Laboratory-reared specimens were exposed to equivalent irradiances of white, red, or blue light during feeding and starvation experiments. Sea slugs fed under red light increased their weight at a faster pace than specimens fed under white or blue light. At the end of the feeding trial, chlorop

The regulation of light quality on the substance production and photosynthetic activity of Dunaliella bardawil.

To study the influence and regulation of light quality on the microalgal photosynthetic activity and production of biomass and substances, green alga Dunaliella bardawil was cultured in this study under the monochromatic red light (7R0B), blue light (0R7B), and their combinations with different ratios (xRyB, x + y = 7), as well as a control of white light (W). The results demonstrated that the only advantage for control W was its chlorophyll-a (Chl-a) and Chl-b contents. All substance production at 7R0B were much lower than at control W, except of glycerol. Compared to control W, protein produ

Physiological and Metabolite Reconfiguration of Olea europaea to Cope and Recover from a Heat or High UV-B Shock.

To understand how olives reconfigure their metabolism to face stress shock episodes, plants from the economically relevant olive (Olea europaea cv. Cobrançosa) were exposed to high UV-B radiation (UV-B, 12 kJ m-2 d-1) or heat shock (HS, 40 °C) for two consecutive days. The physiological responses and some important lipophilic compounds were evaluated immediately (day 0) and 30 days after UV-B or HS episodes. Both treatments induced a reduction of the olive physiological performance, particularly increasing cell membrane damages and proline pool and at the same time reducing chlorophyll levels,

Effect of oxygen on the non-photochemical quenching of vascular plants and potential oxygen deficiency in the stroma of PsbS-knock-out rice.

The excessive and harmful light energy absorbed by the photosystem (PS) II of higher plants is dissipated as heat through a protective mechanism termed non-photochemical quenching (NPQ) of chlorophyll fluorescence. PsbS-knock-out (KO) mutants lack the trans-thylakoid proton gradient (ΔpH)-dependent part of NPQ. To elucidate the molecular mechanism of NPQ, we investigated its dependency on oxygen. The development of NPQ in wild-type (WT) rice under low-oxygen (LO) conditions was reduced to more than 50% of its original value. However, under high-oxygen (HO) conditions, the NPQ of both WT and Ps

Photosynthetic responses of Arabidopsis to SO2 were related to photosynthetic pigments, photosynthesis gene expression and redox regulation.

Sulfur dioxide (SO2) is one of the most common and harmful air pollutants. High concentrations of SO2 can induce a series of defensive responses in Arabidopsis plants. However, the role of photosynthesis in the plant response to SO2 stress is not clear. Here, we report the photosynthetic responses of Arabidopsis plants to SO2 stress. Exposure to 30 mg/m3 SO2 decreased stomatal conductance (Gs) and transpiration rate (Tr) but increased photosynthetic pigments and net photosynthetic rate (Pn). The contents of carbohydrates and sucrose were not altered. The transcript levels of most genes related

Porous translucent electrodes enhance current generation from photosynthetic biofilms.

Some photosynthetically active bacteria transfer electrons across their membranes, generating electrical photocurrents in biofilms. Devices harvesting solar energy by this mechanism are currently limited by the charge transfer to the electrode. Here, we report the enhancement of bioelectrochemical photocurrent harvesting using electrodes with porosities on the nanometre and micrometre length scale. For the cyanobacteria Nostoc punctiforme and Synechocystis sp. PCC6803 on structured indium-tin-oxide electrodes, an increase in current generation by two orders of magnitude is observed compared to

Disentangling the photochemical salinity tolerance in Aster tripolium L.: connecting biophysical traits with changes in fatty acid composition.

A profound analysis of A. tripolium photochemical traits under salinity exposure is lacking in the literature, with very few references focusing on its fatty acid profile role in photophysiology. To address this, the deep photochemical processes were evaluated by Pulse Amplitude Modulated (PAM) Fluorometry coupled with a discrimination of its leaf fatty acid profile. Plants exposed to 125-250 mm NaCl showed higher photochemical light harvesting efficiencies and lower energy dissipation rates. under higher NaCl exposure, there is evident damage of the oxygen evolving complexes (OECs). The thylakoid lumen Deg1 protease affects non-photochemical quenching via the levels of violaxanthin de-epoxidase and PsbS.

Non-photochemical quenching (NPQ), the dissipation of excess light energy as heat, has been long recognized as a major protective mechanism that minimizes the potential for oxidative damage to photosystem II (PSII) reaction centers. Two major positive contributors to NPQ are the carotenoid zeaxanthin, generated from violaxanthin by the enzyme violaxanthin de-epoxidase (VDE or NPQ1), and the thylakoid protein PsbS (NPQ4). The involvement of the lumenal Deg proteases in the repair of PSII from photoinhibition prompted us to further explore their possible role in other responses of Arabidopsis th

PsbE:Phe10 of Cytochrome b559 Modifies Inactivation and Recovery of Photosystem II in Response to High-Light Stress.

Cytochrome b559 (Cyt b559) is a Photosystem II (PS II) protein formed by PsbE (the α-subunit) and PsbF (the β-subunit) that is essential for the biogenesis of the photosystem and may contribute to photoprotection via cyclic electron flow; however, the precise roles of Cyt b559 and its molecular interactions with other proteins and cofactors remain unknown. We have introduced an Ala substitution in the α-subunit of Cyt b559 at PsbE:Phe10 in the cyanobacterium Synechocystis sp. PCC 6803. The conserved PsbE:Phe10 residue interacts with surrounding proteins and cofactors in the vicinity of the sec

Rational design and herbicidal activity of tenuazonic acid derivatives based on photosystem II D1 protein.

BACKGROUND: Natural products are an important source for the development of herbicides. Tenuazonic acid (TeA) is a natural phytotoxin produced by phytopathogenic fungi that strongly inhibits photosystem II (PS II) by preventing electron flow from the primary quinone acceptor (QA) of PS II to the secondary quinone acceptor (QB), thereby directly disrupting the key chain of plant photosynthesis. RESULTS: A series of TeA derivatives were rationally designed and synthesized based on the PS II D1 protein, and their herbicidal activities were evaluated. Compound C11 achieved > 90% control of Abutil

Paired metabolomics and volatilomics provides insight into transient high light stress response mechanisms of the coral Montipora mollis.

The coral holobiont is underpinned by complex metabolic exchanges between different symbiotic partners, which are impacted by environmental stressors. The chemical diversity of the compounds produced by the holobiont is high and includes primary and secondary metabolites, as well as volatiles. However, metabolites and volatiles have only been characterised in isolation so far. Here, we applied a paired metabolomic-volatilomic approach to characterise holistically the chemical response of the holobiont under stress. Montipora mollis fragments were subjected to high-light stress (8-fold higher t

Exposure to ultraviolet radiation delays photosynthetic recovery in Arctic kelp zoospores.

Seasonal reproduction in some Arctic Laminariales coincides with increased UV-B radiation due to stratospheric ozone depletion and relatively high water temperatures during polar spring. To find out the capacity to cope with different spectral irradiance, the kinetics of photosynthetic recovery was investigated in zoospores of four Arctic species of the order Laminariales, the kelps Saccorhiza dermatodea, Alaria esculenta, Laminaria digitata, and Laminaria saccharina. The physiology of light harvesting, changes in photosynthetic efficiency and kinetics of photosynthetic recovery were measured

Exploring thylakoid emergence: evolution of membrane biogenesis and photosystem II assembly in early-diverging cyanobacteria.

Thylakoid membranes (TM) in cyanobacteria and chloroplasts host the light-dependent reactions of oxygenic photosynthesis. Gloeobacterales, the earliest-diverging cyanobacterial lineage, lack TM and perform photosynthesis in the cytoplasmic membrane (CM), representing an ancestral state relative to other cyanobacteria (Phycobacteria). This study investigates the evolutionary origin of TM. Phylogenomic analyses were performed across a phylogenetically diverse set of cyanobacteria, including extensive representation of basal lineages (Gloeobacterales, Thermostichales, Gloeomargaritales, and Pseud

Introduction of a phenylalanine sink in fast growing cyanobacterium Synechococcus elongatus PCC 11801 leads to improved PSII efficiency, linear electron transport, and carbon fixation.

Cyanobacteria are investigated for fundamental photosynthesis research and sustainable production of valuable biochemicals. However, low product titer and biomass productivities are major bottlenecks to the economical scale-up. Recent studies have shown that the introduction of a metabolic sink, such as sucrose, 2,3-butanediol, and 2-phenyl ethanol, in cyanobacteria improves carbon fixation by relieving the "sink" limitation of photosynthesis. However, the impact of light intensity on the behavior of this sink-derived enhancement in carbon fixation is not well understood and is necessary for t

Functional aspects of the photosynthetic light reactions in heat stressed Arabidopsis deficient in digalactosyl-diacylglycerol.

Plants are often submitted, in their natural environment, to various abiotic stresses such as heat stress. However, elevated temperature has a detrimental impact on overall plant growth and development. We have examined the physiological response of the dgd1-2 and dgd1-3 Arabidopsis mutants lacking 30-40% of digalactosyl-diacylglycerol (DGDG) exposed to heat constraint. These mutants, which grow similarly to wild type under normal conditions, were previously reported to be defective in basal thermotolerance as measured by cotyledon development. However their functional properties were not desc

Effects of Ca2+ and EGTA on P680*+ reduction kinetics and O2 evolution of Photosystem II.

We report for the first time significant changes in the P680*+ reduction kinetics of Photosystem II (PS II) in which the 17 and 23 kDa extrinsic polypeptides are intact, in the presence of Ca(2+) or ethylene glycol bis (beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) which were added to vary the Ca(2+) concentration from 5 microM to 30 mM. The decrease in the extent of normal P680*+ reduction decay with lifetimes of 40-370 ns and a corresponding increase in the extent of kinetics with lifetimes of 20-220 micros was interpreted as being due to electron transfer from Y(Z) to P680*+ bein

Export of vacuolar manganese by AtNRAMP3 and AtNRAMP4 is required for optimal photosynthesis and growth under manganese deficiency.

Manganese (Mn) is an essential element, acting as cofactor in numerous enzymes. In particular, a Mn cluster is indispensable for the function of the oxygen-evolving complex of photosystem II. Metal transporters of the Natural Resistance-Associated Macrophage Protein (NRAMP) family have the ability to transport both iron and Mn. AtNRAMP3 and AtNRAMP4 are required for iron mobilization in germinating seeds. The results reported here show that, in adult Arabidopsis (Arabidopsis thaliana) plants, AtNRAMP3 and AtNRAMP4 have an important role in Mn homeostasis. Vacuolar Mn accumulation in mesophyll

Influence of the diadinoxanthin pool size on photoprotection in the marine planktonic diatom Phaeodactylum tricornutum.

The pool size of the xanthophyll cycle pigment diadinoxanthin (DD) in the diatom Phaeodactylum tricornutum depends on illumination conditions during culture. Intermittent light caused a doubling of the DD pool without significant change in other pigment contents and photosynthetic parameters, including the photosystem II (PSII) antenna size. On exposure to high-light intensity, extensive de-epoxidation of DD to diatoxanthin (DT) rapidly caused a very strong quenching of the maximum chlorophyll fluorescence yield (F(m), PSII reaction centers closed), which was fully reversed in the dark.
